Anti-allergic Action of Some Medicinal Plants (수종 생약의 항알레르기 작용)

  • Anti-allergic action of each water extracts of some crude drugs was investigated in mice and rats. The activity of hyaluronidase which was used in the screening test of anti-allergic action was inhibited significantly by Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Cinnamomi Ramulus, Glycyrrhizae Radix and Scutellariae Radix. The 48-hour homologous passive cutaneous anaphylaxis(48-hr PCA) in mouse ear was inhibited significantly by intraperitoneal(i.p.) injection of Amomi Semen, Cimicifugae Rhizoma, and ketotifen, a comparative drug of an anti-allergic action. The increase of vascular permeability induced by histamine or serotonin was inhibited significantly by i.p. injection of Amomi Semen, Cimicifugae Rhizoma, Cinnamomi Ramulus and ketotifen. In rat dorsal skin, the increase of vascular permeability which was induced by histamine, serotonin or compound 48/80 was inhibited significantly by i.p. injection of Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Scutellariae Radix and ketotifen. Armeniacae Semen and Liriopis Tuber which had not inhibited hyaluronidase activity did not inhibit 48-hr PCA and the increase of histamine, serotonin or compound 48/80-induced vascular permeability in mice and rats. These results suggest that each water extract of Amomi Semen and Cimicifugae Rhizoma has anti-allergic action.

Effects of Amomi Semen Butanol Fraction on Gastritis and Helicobacter pylori (위염 및 Helicobacter pylori에 미치는 Amomi Semen Butanol 추출물의 영향)

  • In a preliminary screening of plant extracts for the antigastritic and anti- Helicobacter pylori (H. pylori) actions in rats, the ethanol extract of Amomi Semen (AS) showed positive activity in HCl Ethanol-induced gastric lesions and H. pylori. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent butanol fraction significantly reduced HCl, Ethanol-induced gastric lesions at the oral dose of 350 mg/kg. Also butanol fraction has an inhibitory effect on the growth of H. pylori $(MIC=1.43\;{\mu}g/mL)$. In pylorus ligated rats, butanol fraction showed decrease in the volume of gastric secretion and acid output, of which effects were stronger in other fractions. We isolated 6 subfractions by column chromatography. The protective effects of 6 subfractions of Amomi Semen were also significant in the HCl, Ethanol induced gastric lesion model. These results might suggest that they had inhibitory action in gastric lesion through inhibition of gastric acid secretion. Butanol fraction of AS can be applied as treatment of H. pylori. Butano fractions and ethanol extract of AS was carried out or the development of a new gastroprotective supplementary product.

Activity of Medicinal Plants on Proliferation and Differentiation of Osteoblasts (생약의 조골 세포 증식과 분화 검색)

  • Osteoblasts play an important role in bone metabolism by bone formation. Natural medicines having a stimulatory activity on osteoblast proliferation and differentiation can improve bone diseases such as osteoporosis. The methanol extracts of 159 herbal medicines were screened for the stimulatory activity on osteoblast proliferation by MTT assay and differentiation in the presence of ascorbic acid and $\beta$-glycerophosphate. Among the tested extracts, Alpiniae Semen, Amomi Semen, Anemarrhenae Rhizoma, Bambusae Folium, Cannabis Semen, Dalbergiae odoriferae Lignum, and Luffae Fructus Retinervus showed relatively strong stimulatory activity on osteoblast proliferation, whereas Amomi Semen showed strong stimulatory activity on osteoblast differentiation.

  • In order to search for antioxidants from the plants, eighty-two kinds of herbal medicines were investigated. The MeOH extracts of Euryales Semen, Alpiniae Officinari Rhizoma. Drynariae Rhizoma, Sophorae Flos, Trachelospermi Caulis, Crassirhizomae Rhizoma, Euphorbiae lathyridis Semen, Lini Semen, Myristicae Semen, Epimedii Herba, Santali Lignum rubrum, Perillae Herba, Amomi Tsao-Ko Fructus and Garanii Herba showed potent antioxidative activities using the 1,1-diphenyl-2-picrylhydrazyl free radical generating system. Effects of Amomi, Semen Extract on Synthesis of Insulin-like Growth Factor-1 and Anti-wrinkle in Skin (사인추출물의 인슐린 유사 성장인자-3의 합성과 피부 노화 개선에 대한 효과)

  • We screened several materials to stimulate IGF-1 promoter activity using luciferase reporter assay and found that Amomi Semen extract (ASE) among them is the most powerful stimulator We also studied about the anti-wrinkle effect of ethanolic extract of Amoni Semen in vitro and in vivo. Semi-quantitative RT-PCR showed that the extract elevated the presence level of IGF-1 mRNA. And $[^3H]$ proline incorporation and semi-quantitative RT-PCR showed that the extract increased the expression of type-I collagen compared with vehicle in vitro and in vivo, respectively. Significant inhibition of MMP-1 expression was determined by ELISA and Western blot. Finally, topical treatment of the extract on hairless mouse's dorsal skin expanded the volume of collagen and dermal thickness. These results suggest that Amomi Semen may be a good candidate for improving extracellular matrix through the increase of collagen expression and inhibition of MMP-1 expression. Moreover, this study enables us to guess that IGF-1 stimulated by the extract may be involved in the mechanism of anti-wrinkle effect of it.

Antibiotic effects of Medicinals resolving dampness with aroma on vaginal microorganisms (방향화습약(芳香化濕藥)이 질내(膣內) 미생물(微生物)에 미치는 영향)

  • Purpose : This study was conducted to investigate effects of 7 herbs among medicinals resolving dampness with aroma on vaginal microorganisms. Methods : Staphylococcus aureus, Methicillin- resistant Staphylococcus aureus, Candida albicans and Gardnerella vaginalis were used for vaginal pathogenic microorganisms. Lactobacillus gasseri, Streptococcus spp. and Escherichia coli HB101 were used for vaginal flora. Medicinals resolving dampness with aroma, Pogostemonis Herba, Amomi Cardamomi Fructus, Amomi Semen, Atractylodis Rhizoma, Ammomi Tsao-ko Fructus, Alpiniae Katsumadaii Semen and Magnoliae Cortex were used in this study. In vitro antimicrobial activities were observed by optical density and colony test. Results : The optical density showed that Alpiniae Katsumadaii Semen and Magnoliae Cortex among medicinals resolving dampness with aroma had antimicrobial effects on pathogenic vaginal microorganisms (Methicillin-resistant Staphylococcus aureus). Pogostemonis Herba, Ammomi Tsao-ko Fructus had antimicrobial effects on Gardnerella vaginalis. The colony test showed that Alpiniae Katsumadaii Semen, Magnoliae Cortex among medicinals resolving dampness with aroma had antimicrobial effects on Methicillin-resistant Staphylococcus aureus. Pogostemonis Herbs, Alpiniae Katsumadaii Semen had antimicrobial effects aganist Gardnerella vaginalis. The optical density showed that Magnoliae Cortex among medicinals resolving dampness with aroma had antimicrobial effect on normal vaginal microorganisms (Streptococcus spp.). The colony test showed that medicinals resolving dampness with aroma had no antimicrobial effect on normal vaginal microorganisms. Conclusion : From this study, we could suggest that Pogostemonis Herba, Ammomi Tsao-ko Fructus, Alpiniae Katsumadaii Semen, Magnoliae Cortex of medicinals resolving dampness with aroma are available to antimicrobial agent of pathogenic vaginal microorganisms in vitro.

  • In vitro sensitivity testing was performed for 21 kinds putative anticancer drugs selected from references and information. Cellular damage of P815 mastocytoma cells following exposure to water extracts of drugs was evaluated by colony formation assay. Highly effective drugs with more than 50% inhibition of colony formation were seven (Houttuyniae Herba, Sanguisorbae Radix, Nepetae Herba, Manitis Squama, Lonicerae Flos, Amomi Semen, Polyporus), though not more effective than BCNU. According to the results of $^3H-thymidine$ incorporation assay for determination of selective cytotoxicity, 3 of these drugs (Houttuyniae Herba, Polyporus, Manitis Squama) were found to be low cytotoxic to normal mouse lymphoid cells. These findings suggest that the above 3 drugs may be used for effective anticancer drugs in vivo.

  • Dalwoneum is the one of Chinese traditional prescription used for the treatment of liver disease. This prescription consists of Arecae Semen (12 g), Magnoliae Cortex (9 g), Amomi tsao-ko Fructus (3 g), Anemarrhenae Rhizoma (12 g), Paeoniae Radix (9 g), Scutellariae Radix (12 g), and Glycyrrhizae Radix (3 g). Water extract of Dalwoneum showed the significant hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. (omitted)

${\beta}-Glucuronidase-inhibitory\;Activity$ and Hepatoprotective Effect of Herbal Medicines (생약의 ${\beta}-Glucuronidase$ 저해와 간장보호효과)

  • Inhibitory effect of water and ethyl acetate extract of 60 kinds of herbal medicines was investigated on ${\beta}-glucuronidase$. Among water extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Termaliae Fructus, Amomi Tsa-ko Fructus and Arecae Semen were also potent inhibitors. Among ethyl acetate extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Nelumbinis Semen. Ephedrae Radix and Termaliae Fructus were also potent inhibitors. The extract of Galla Rhois had potent hepatoprotective effect on $CCl_4-induced$ hepatotoxicity of rats. These results suggest that the ${\beta}-glucuronidase$ seems to be closely related to the liver injury, which could be prevented by the inhibitor of ${\beta}-glucuronidase$.

Screening of Korea Traditional Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (한약재의 최종당화산물 생성저해활성 검색)

  • Advanced glycation end products (AGEs) are largely involved in the pathogenesis of diabetic complications. As part of our ongoing project directed toward the discovery of preventive and/or delay agents for diabetic complications from natural sources, 92 Korean traditional herbal medicines have been investigated with an in vitro evaluation system using AGEs inhibitory activity. Of these, 17 herbal medicines exhibited a significant inhibitory activity against AGEs formation. Particularly, 9 herbal medicines, i.e., Cinnamomi Cortex, Artemisiae Argyi Herba, Ammoni Tsao-ko Fructus, Menthae Herba, Amomi Semen, Polygoni Avicularis Herba, Lycopi Herba, Salviae Radix, and Nelumbinis Semen showed more potent inhibitory activity (2-4 fold) than the positive control aminoguanidine.
