Cytotoxic Sesquiterpenoid from the Seeds of Amomum xanthioides

  • As parts of our continuing search for biologically active compounds from medicinal plants, we investigated the constituents of the seeds of Amomum xanthioides and isolated a sesquiterpenoid, a nerolidol derivative from its MeOH extract. The chemical structure was determined by spectroscopic methods, including 1D and 2D NMR to be ($2S^*$,$2'R^*$,$5'S^*$)-2-(5'-ethenyltetrahydro-5'-methylfuran-2'-yl)-6-methylhept-5en-2-ol (1). Compound 1 was isolated for the first time from nature source. Compound 1 exhibited a good cytotoxicity against SK-OV-3 and SK-MEL-2 cells ($IC_{50}$: 16.7 and $8.6\;{\mu}M$, respectively) using a SRB bioassay. In this study, we also determined the absolute configuration of 2 reported in previous paper.

사인의 항당뇨 작용에 관한 연구 (Study no the Antidiabetic Effect of Amomum xanthioides Extract)

  • The antidiabetic effect of Amomum xanthioides(A. xanthioides) extract was investigated. Alloxan caused the hyperglycemia and hypoinsulinemia by a selective destruction of pancreatic ${\beta}$-cell. Pretreatment of mouse with A. xanthiodies extract for 2 days prior to alloxan administration completely protected hyperglycemia induced by alloxan. In addition, administration of A. xanthioides extract to alloxan-induced diabetic mouse significantly abolished hyperglycemia, hypoinsulinemia, and, the reduction of size and number of insulin-secreting cells induced by alloxan. Administration of A. xanthioides extract to alloxan-induced diabetic mouse rapidly increased pancreatic Reg gene expression to 7 days, and then decreased. In alloxan-diabetic mouse. Reg gene expression was increased at 3 days after alloxan injection, and sustained until 24 days. The present results indicate that A. xanthioides extract contains potentially effective components exhibiting both protection and treatment of alloxan-induced diabetes. These results suggested that the antidiabetic effect of A. xanthoides extract may be mediated through the regeneration of pancreatic ${\beta}$-cells.

  • Seven monoterpenes, three sesquiterpenes, three phenolics and one flavonoid were isolated from the MeOH extract of Amomum xanthioides. Their structures were determined by spectroscopic methods to be caryophyllene oxide (1), bornyl acetate (2), nerolidol (3), spathulenol (4), (-)-borneol (5), (+)-5-endohydroxycamphor (6), vanillic acid (7), protocatechuic acid methyl ester (8), betulabuside A (9), (1R,4S,6R)-6-hydroxyfenchan-2-one-6-O-$\beta$-D-glucopyranoside (10), (1S,4R,6S)-6-hydroxybornan-2-one-6-O-$\beta$-D-glucopyranoside (11), (1R,2S,4S,5R)-angelicoidenol 2-O-$\beta$-D-glucopyranoside (12), 1-O-vanilloyl-$\beta$-D-glucopyranoside (13), and quercetin-3-rhamnopyranoside (14). Compounds 6-14 were isolated for the first time from this plant source. Compounds 3 and 4 exhibited moderate cytotoxicity against four human cancer cell lines in vitro using a SRB bioassay.

  • Non-alcoholic fatty liver disease (NAFLD), especially including non-alcoholic steatohepatitis (NASH) is one of the common diseases with 25% of prevalence globally, but there is no thera-peutic access available. Amomum villosum var. xanthioides (Wall. ex Baker) T.L.Wu & S.J.Chen (AX), which is a medicinal herb and traditionally used for treating digestive tract disorders in Asia countries. We aimed to examine pharmacological effects of ethyl acetate fraction of AX (AXEF) against ER stress-induced NASH mice model using C57/BL6J male mice by tunicamycin (TM, 2 mg/kg) injection focusing on the oxidative stress. Mice were orally administrated AXEF (12.5, 25, or 50 mg/kg), silymarin (50 mg/kg) or distilled water daily for 5 days, and outcomes for fatty liver, inflammation, and oxidative stress were measured in serum or liver tissue levels. AXEF drastically attenuated hepatic ER stress-induced NASH which were evidenced by decreases of li-pid droplet accumulations, serum liver enzymes, hepatic inflammations, and cell death signals in the hepatic tissue or serum levels. Interestingly, AXEF showed potent antioxidant effects by quenching of reactive oxidative stress and its final product of lipid peroxide in the hepatic tissue, specifically increase of metallothionein (MT). To confirm underlying actions of AXEF, we ob-served that AXEF increase MT1gene promoter activities in the physiological levels. Collectively, AXEF showed antioxidant properties on TM-induced ER stress of NASH by enhancement of MTs.

사인의 열수 추출물이 B16F10 흑색종세포의 멜라닌형성에 미치는 영향 (Inhibition of Aqueous Extract from Amomum xanthioides on ${\alpha}$-melnocyte Stimulating Hormone Induced Melanogenesis in B16F10 Cell)

  • To develope skin-whitening or therapeutic agents against hyperpigmentation, aqueous extract from Amomum xanthioides (AEAX) was evaluated for melanogenesis inhibitory activity in B16F10 melanoma cell. The treatment with AEAX at the 0.5 and 1.0 mg/ml level significantly inhibits the biosynthesis of melanin compared with untreated control. The AEAX-treated cells at the 1.0 mg/ml level were more efficient than commercial arbutin at 0.1 mg/ml. The tyrosinase activity also significantly decreased in AEAX-treated cells at the 0.5 and 1.0 mg/ml level. The Western analyses confirmed the significantly decreased expression of tyrosinase and tyrosinase-related protein-1 by AEAX treatment. These results indicate that AEAX may contribute to the inhibition of melanin biosynthesis through regulating tyrosinase activity and expression and serve as a new candidate in the design of new skin-whitening or therapeutic agents.

생약재의 항충치 및 항산화효과 탐색 (Anticariogenic and Antioxidant Activities from Medicinal Herbs)

  • 생약재를 이용하여 천연 자원에서 항충치와 항산화 물질을 탐색하고 부가가치를 높이고자 하였다. 41종의 생약재 20 g을 ethanol 300 mL에 추출하고 농축하여 전량을 20 mL로 하여 시료로 사용하였다. 생약재의 phenolic compound의 함량을 측정하였으며, phenolic compound의 함량은 tannic acid로 환산하여 나타냈다. 가장 많은 phenolic compound 함량은 측백으로 $115.24{\mu}g/mL$이었으며, 목과, 토복령, 목단, 소엽 , 팔각향이 각각 $97.49{\mu}g/mL,\;97{\mu}g/mL,\;96.27{\mu}g/mL,\;96.56{\mu}g/mL,\;94.40{\mu}g/mL$이었다. Streptococcus mutans와 타액을 배양한 균주 5종에 대하여 생약재 ethanol 추출물로 항균성효과 실험을 하였다. 그 결과 Streptococcus mutans에서는 팔각향과 공사인이 98%로 높았으며, S-1과 S-2에서는 팔각향이 98%, 계피가 94%로 높은 항균효과가 있었다. S-3는 목과 95, S-4는 희첨 97%이었고, S-5에서는 연자육이 94%로 항균효과가 높았다. Streptococcus mutans와 타액을 배양한 균주 5종에서 glucosyltransferase(GTase) 효소를 분리하여 생약재에 의한 활성저해효과를 측정하였다. Streptococcus mutans의 GTase에서는 팔각향과 공사인이 94%로 활성저해효과가 가장 높았다. S-1에서 분리한 GTase에 대해서는 공사인이 95%로 효소활성 저해 효과가 가장 높았으며, S-2는 팔각향이 59%로 가장 높았다. S-4의 GTase는 목과가 96%이고, 작약과 팔각향이 95%였고, S-5의 GTase에서는 측백이 90%로 효소활성 저해효과가 높았다. 생약재들의 항산화성을 알아보기 위하여 DPPH radical scavenging를 알아보았으며, 시료는 $100{\mu}g/mL,\;200{\mu}g/mL,\;300{\mu}g/mL$를 첨가하여 측정하였다. 시료 $100{\mu}g/mL$에서 측백이 81.08%로 항산화활성이 가장 높았으며, 토복령, 계피가 각각 80.45%, 76.63%로 항산화활성이 높았다. 시료 $300{\mu}g/mL$에서는 연자육이 82.79%로 가장 높았으며, 감초, 계피, 공사인, 목과, 목단, 소엽, 애엽, 인진, 정향, 지각, 창이자, 천초, 측백, 토복령, 헛개나무, 홍화자 등 또한 항산화활성이 높았다. 생약재 ethanol추출물 중 항충치, 항산화효과가 높은 생약재들은 대부분 많은 phonolic compound함량을 나타내고 있어 phenolic compound는 항충치 및 항산화효과에 관련이 있는 것으로 생각된다.

생약재 추출물의 hyaluronidase 저해 및 라디칼 소거 활성 검색 (Screening of Hyaluronidase Inhibitory and Free Radical Scavenging Activity in vitro of Traditional Herbal Medicine Extracts)

  • 본 연구에서는 항염증 및 항산화 활성을 갖는 유용한 물질 을 검색하기 위해 40여 가지 생약재를 에탄올 추출하여 H HAase 억제 및 라디칼 소거 활성을 측정하였다 .. HAase 억제 활성을 측정한 결과, I mg/mL의 농도에서 황기, 두충, 오미 자, 황금, 오가피, 목과, 목단피, 산사가 50% 이상의 저해활 성을 나타내었고, 이들에 대하여 n-hexane, chloroform, ethyl a acetate, water로 순차적으로 용매분획하여 활성을 측정한 결 과, 모든 시료의 ethyl acetate와 water 분획에서 높은 활성을 보였다 항산화 활성은 DPPH 라디칼에 대한 전자공여능과 F Fenton 반응에 의해 발생한 hydroxyl radical에 대한 소거능으 로 평가하였다 .. DPPH 라디칼에 대한 전자공여능은 복분자, 목단피, 백작약, 차전자, 마가목 에탄올 추출물이 I mg{mL 놓도에서 90% 이상으로 대조군으로 이용한 BHA와 Q - -tocopherol과 유사한 활성을 나타내었다 .. 2-deoxyribose 산화 법에 따른 hydroxyl 라디칼 소거활성에서 대부분의 에탄올 추출물 시료에서 모두 높은 활성을 나타내었다. 활성올 갖는 시료에 대하여 지질과산화 억제 활성을 측정한 결과 linoleic a acid와 phosphatidylcholine liposome 기질 모두에 대하여 오미 자 추출물은 가장 높은 억제 활성을 나타내었다.

제호탕(醍醐湯)의 현대적 재현 (Modern Reproducing of Jehotang Method)

  • The "tang[tɑ:ŋ]" in Korean pronounciation means the beverage made of boiled medicinal herbs. The"Jeho-tang", the name of Drink in this abstract, is described in a variety of medical books including the "Dongeui Bogam" as being effective for illness from the summer heat in promoting digestion, curing the heatstroke and bringing it to a halt. The Drink was used as the Royal gifts granted to retainers and royal families on the Day of Dano-festival on the fifth of the fifth month of the year according to the lunar calendar, the items of encouragement for those who worked hard in sacrifices, and the awards for students of Confucianism who were proficient in their learning. The Jeho-tang used in this study was scientifically cooked again after a long time in history through looking at the methods written in the documents concerned with the Drink such as the "Dongeui Bogam" and the "Taste of Korea". In preparation of the medicinal herbs for the Drink, the powder of thinner than 30 mesh of the "Prunus mume", which is a species of Asian plum in the family of Rosaceae, and those of 50 mesh of the "Santalum album", which is the fragrant wood of trees in the genus Santalum and the "Amomum Xanthioides", which is produced in Vietnam and is the name of a kind of herb medicines, being very effective in the desease caused from heatstrare, were used. The sugar concentration of the honey boiled down long time at low heat was $82.43^{\circ}Bx$. When cooking in a double boiler, the inner part of the liquid for the Drink was kept at $80^{\circ}C$ for 12 hours to make it finished in a state of ointment. In the general composition of the finished Jeho-tang, the moisture content was 24.4%, 1.3% of crude fat, 1.4% crude protein and 0.7% ash, along with pH3.2. The acceptance on the whole was come out to be the highest in the sample diluted with the drinking water of 7-fold of the Jeho-tang, indicating that the 7-fold's addition of water was optimum level for drinking. In the Drink cooked by a vaccum pressure extractor for herb medicine, which was developed to improve the art of cooking, the longer the time of pressure was, the less the heterogeneous feeling at tongue was and the more the glossiness of the Drink was. The Jeho-tang cooked under pressure for 7 hours received an excellent evaluation in its acceptability in every way.
