  • Lignocellulose and lignolic compounds were absolutely given much weight In the biosphere, and their degradation was essential for continuous biological carbon circulation. Whereas aerobic cellulolytic microorganism dissolved the cellulose into their elements in the first stage, strict anaerobic cellulolytic microorganism's role was taken I increasing interest through the recent research. It was reviewed that anaerobic microbial degradation process of lignocellulose and its derivatives (cellulose, lignin, oligolignol and monoaromatic compound), and function of anaerobic microorganism on the. environmental ecology.

고농도 암모니아성 질소 폐수의 효과적인 혐기성 처리를 위한 영향 인자 평가 (Estimation of influening factors for efficient anaerobic digestion of high strength ammonia-nitrogen wastewater)

  • In this study, the influencing factors for efficient anaerobic digestion of high strength ammonia-nitrogen wastewater removal were investigated by testing biochemical methane potential test. In the influencing factors, the trace metals which could increase activity of anaerobic microorganisms, microbial concentration and types were evaluated. In the results, trace metals supplementation showed gas production amount higher than those without addition of trace metals. Among the tested trace metals, B, Ni, and Se were preferable to gas production. In the result of gas production according to the microbial concentration, the amount of gas production was proportional to the microbial concentration. In addition, the shortest lag time and the fastest gas production rate were achieved when the highest microbial concentration was tested. granule-type microorganism produced more gas than suspended-type microorganism. In conclusion, the efficient anaerobic digestion for high strength ammonia-nitrogen wastewater removal could be achieved by applying necessary trace metals injection and high concentration granule type microorganism.

혐기성 조건에서 담체에 부착된 미생물의 관찰 (Microscope Examination of Attached Biofilm under Anaerobic Conditions)

  • Microstructural examinations were performed on the anaerobic biofilm from reactor filled with PE support media. Optical microscope, SEM and fluorescent microscope were used for qualitative and morphological studies on the attached microorganism under anaerobic condition. Microorganisms were attached in crevices where protection from shear forces of surfaces where easy to contact with support media surface. A hypothesis for biofilm accumulation occurs on a surface such as polymer support media is presented schematically : 1st step ; cell-support media attachment, 2nd step ; cell-support media attachment and cell-cell attachment, 3rd step ; attached biofilm from neighboring crevices joins together and growing, 4th step ; mature and irregualar biofilm was formed. In SEM photographs, shape and structures of biofilm were observed, but microorganism species and methanogens were not identified. A large number of methanogenic bacteria were identified on the surface of PE substratum by fluorescence under 480nm of radiation and it was estimated that methanogenic bacteria was related to initial attachment of bacteria under anaerobic condition.

Tolerance of Anaerobic Granular Sludge to Oxygen

  • To study the tolerant capacity of anaerobic granular sludge (AGS) to oxygen using semi -dynamic batch experiment, the aerating time, pH of the basal media, reductive inorganic materials, microorganism, and microorganism metabolite were investigated. When the aerating time was higher or lower than 0.5 h, the producing gas activity of sludge was lower than that of the control. The oxygen tolerance of the experimental sludge was the highest at the initial pH 7.2. The producing gas activity of sludge I was higher than that of sludge II. And storage at $4^{\circ}C$ can low the lose of the oxygen tolerance capacity of granular sludge. The producing gas activity of sludge was the highest when KI was added. The growth of aerobic microorganisms and some metabolite could increase the producing gas activity of granular sludge.

생물학적 처리에 의한 돈사폐수의 인제거 (Biological Removal Phosphorus Containing Swine Wastewater)

  • The studies of swine wastewater treatment aim to development of process using soil microorganism. Removal rate of swine wastewater containing organic matter was 99 percent in case of high loading rate. Microorganism was devoted to improve the treatment efficiency of the process. According to the result obtained from biological treatment of high loading rate swine wastewater. Hydraulic retention time was 2.3 days in unit process of biological phosphorus removal. BO $D_{rm}$ / $P_{rm}$ ratio was 1122 in room temperature anaerobic process and 355.6 in mesophilic anaerobic process. And then phosphorus removal rate mesophilic anaerobic process was 3 time as much as than room temperature acaerobic process.

Effect of Aeration on Denitrification by Ochrobactrum authropi SY509

  • Aeration was found to affect the biological denitrification by Ochrobactrum authropi SY509. Although cell growth was vigorous under 1 vvm of aeration and an agitation speed of 400 rpm in a 3-L jar fermentor, almost no nitrate was removed. Yet under low agitation speeds (100, 200, and 300 rpm), denitrification occurred when the dissolved oxygen was exhausted shortly af-ter the inoculation of the microorganism. Ochrobactrum authropi SY509 was found to express highly active denitrifying enzymes under anaerobic conditions. The microorganism also synthesized denitrifying enzymes under aerobic conditions (1 vvm and 400 rpm), yet their activity was only 60% of the maximum level under anaerobic conditions and the nitrate removal efficiency was merely 15%. However, although the activities of the denitrifying enzymes were inhibited in the presence of oxygen, they were fully recovered when the conditions were switched to anaerobic conditions.

양돈장 분뇨의 혐기성 부숙 과정에서 첨가제에 따른 오염물질 및 악취 저감 효과 (The Effect of Reduction of Contaminants and Odor according to the Additives in the Anaerobic Maturation Process of Piggery Slurry)

  • The changes of contaminants and odor corresponding to anaerobic maturation process of piggery slurry were investigated by applying the additives, such as different kinds of complex microorganism products and deodorants containing microorganism activating agents. The pHs during 20-day anaerobic maturation were operated stably without great change regardless of the additives, although they were rather lower in the case that the additives were contained than the case that they were not contained. The effects of removing CODcr, $NH_3-N$, T-N, and T-S in case that the additives were not contained, were not so great during the 20-day operation and so it would be difficult to remove the organic materials and nitrogen ingredients simply with anaerobic maturation process. However, in case of anaerobic maturation process that the additives were contained, their average removal rates were improved with the values of $49\%,\;63.5\%,\;48.5\%,\;and\;30.7\%$ for above each of items, even if the 20-day of short-term maturation period was applied. Especial1y, odor intensity with the additives was lowered continuously during the operation period and it had more than two times of lowering effect compared to that without those.

부착성 미생물을 이용한 질산화 및 탈질특성에 관한 연구 (Study on the Characteristics of Nitrification and Denitrification using Attached Microorganism)

  • In this research, characteristics of nitrification and denitrification using the microorganism attached on sponge and plates were examined. The denitrification and nitrification performance were investigated under the anaerobic and aerobic condition for about 2 months. Because the basins of denitrification and nitrification were connected in series, wastewater was flowed from denitrification basin to nitrification one. The 90% of influent flowrate was returned from nitrification basin to denitrification one. Most of organic material was removed in nitrification basin, wherease the only exact amount of organics required in denitrification process was removed in denitrification one. This experiment resulted in that heterotrophic bacteria existing in aerobic basin governed the removal efficiency of organic compounds. In case the influent BOD concentration into nitrification basin was 80mg/l, it did not affect to accumulation of nitrifying bacteria, the balance of heterotrophic bacteria was proved to be an important factor in nitrification/denitrification method such as anaerobic and aerobic cycling type.

  • 토양 미생물을 이용한 고농도 축산폐수의 과부하 처리를 위한 공정개발 연구결과 유기물 제거율은 99%이었다. 상온 혐기조에서 $0.7kg\;BOD/m^3/d$ 이하에서 처리수의 BOD 농도는 150mg/L 이하를 이었다. 상온 혐기조를 운전한 실험에서 후속 폭기조의 물질 대사계수(km)는 $0.73hr^{-1}$, 세포증식계수(Y)는 0.433 g/VSS/g BODrm/d 였으며 중온으로 운전한 경우 후속 폭기조에서 각각 $1.45hr^{-1}$과 0.776 g VSS/g BODrm/d 로 반응계수 값이 두 배에 가깝게 개선되었다.

Evaluation of the Degradation of Carbohydrate-based Material During Anaerobic Digestion for High-efficiency Biogas Production

  • Purpose: In this study, the potential for biogas production, degradation rates, and lag-phase of diauxic growth of carbohydrate-based material, which is one of the proximate compositions, were investigated. Methods: This study was conducted using starch as a carbohydrate-based material. In experimental condition 1, the biogas potential of carbohydrate-based material was measured. In experimental condition 2, the effect of feed to microorganism ratio (F/M ratio) on lag-phase of diauxic growth from carbohydrate-based material was tested. Biochemical methane potential tests were performed at five different feed to microorganism ratios (0.2, 0.4, 0.6, 0.8, and 1.0) under mesophilic conditions. The biogas production patterns, lag-phase, total volatile fatty acids to total alkalinity ratio (TVFA/TA ratio), and time required for 90 percent biogas production were used to evaluate biogas production based on the biochemical methane potential tests. Results: In experimental condition 1, unlike previous studies, biogas was produced in the TVFA/TA ratio ranging from 1.131 to 2.029 (approximately 13-19 days). The methane content in the biogas produced from the digesters was 7% on day 9 and increased rapidly until approximately day 27 (approximately 72%). In experimental condition 2, biogas yield was improved when the feed to microorganism ratio exceeded 0.6, with an initial lag-phase. Conclusions: Even if the TVFA/TA ratio was greater than 1.0, the biogas production was processed continuously, and the $CO_2$ content of the biogas production was as high as 60%. The biogas yield was improved when the F/M ratio was increased more than 0.6, but the lag-phase of carbohydrate-based material digestion became longer starting with high organic loading rate. To clarify the problem of the initial lag-phase, our future study will examine the microbial mechanisms during anaerobic digestion.
