  • Angiotensins and bradykinin play important roles in blood pressure regulation and in other physiological and pathological conditions. The present work is aimed to investigate potential antagonists against either angiotensin or bradykinin from plant sources. Each methanol extract of selected 25 plants was solvent fractionated to three fractions. The effects of total of 75 plant samples against the contractions induced by angiotensin I or bradykinin in rat ileum were measured. The results are summarized.

Inhibition of angiotensin converting enzyme increases PKCβI isoform expression via activation of substance P and bradykinin receptors in cultured astrocytes of mice

  • Background: Angiotensin-converting enzyme inhibitor (ACEi) inhibits the catalysis of angiotensin I to angiotensin II and the degradation of substance P (SP) and bradykinin (BK). While the possible relationship between ACEi and SP in nociceptive mice was recently suggested, the effect of ACEi on signal transduction in astrocytes remains unclear. Objectives: This study examined whether ACE inhibition with captopril or enalapril modulates the levels of SP and BK in primary cultured astrocytes and whether this change modulates PKC isoforms (PKCα, PKCβI, and PKCε) expression in cultured astrocytes. Methods: Immunocytochemistry and Western blot analysis were performed to examine the changes in the levels of SP and BK and the expression of the PKC isoforms in primary cultured astrocytes, respectively. Results: The treatment of captopril or enalapril increased the immunoreactivity of SP and BK significantly in glial fibrillary acidic protein-positive cultured astrocytes. These increases were suppressed by a pretreatment with an angiotensin-converting enzyme. In addition, treatment with captopril increased the expression of the PKCβI isoform in cultured astrocytes, while there were no changes in the expression of the PKCα and PKCε isoforms after the captopril treatment. The captopril-induced increased expression of the PKCβI isoform was inhibited by a pretreatment with the neurokinin-1 receptor antagonist, L-733,060, the BK B1 receptor antagonist, R 715, or the BK B2 receptor antagonist, HOE 140. Conclusions: These results suggest that ACE inhibition with captopril or enalapril increases the levels of SP and BK in cultured astrocytes and that the activation of SP and BK receptors mediates the captopril-induced increase in the expression of the PKCβI isoform.

Wogonin attenuates vascular remodeling by inhibiting smooth muscle cell proliferation and migration in hypertensive rat

  • Wogonin, extracted from the roots of Scutellaria baicalensis Georgi, has been shown to suppress collagen deposition in spontaneously hypertensive rats (SHRs). This study was performed to investigate the role and mechanism of wogonin underlying vascular remodeling in SHRs. After injection of SHRs with 40 mg/kg of wogonin, blood pressure in rats was measured once a week. Masson's trichrome staining was conducted to observe the changes in aortas and mesenteric arteries. Vascular smooth muscle cells (VSMCs) isolated from rat thoracic aortas were treated with Angiotensin II (Ang II; 100 nM) in the presence or absence of varying concentrations of wogonin. The viability and proliferation of VSMCs were examined using Cell Counting Kit-8 assay and 5-ethynyl-2'-deoxyuridine assay, respectively. The migration of VSMCs was examined using wound healing assay and transwell assay. We found that wogonin administration alleviated hypertension, increased lumen diameter, and reduced the thickness of the arterial media in SHRs. Ang II treatment enhanced the viability of VSMCs, which was inhibited by wogonin in a concentration-dependent manner. Wogonin reversed Ang II-induced increases in the viability, proliferation, and migration of VSMCs. Moreover, wogonin inhibited Ang II-induced activation of mitogen-activated protein kinase (MAPK) signaling in VSMCs. Overall, wogonin repressed the proliferative and migratory capacity of VSMCs by regulating the MAPK signaling pathway, thereby attenuating vascular remodeling in hypertensive rats, indicating that wogonin might be a therapeutic agent for the treatment of vascular diseases.

만성폐쇄성폐질환에서 ACE 유전자 다형성과 기관지 과민성의 연관성 (Polymorphisms of Angiotensin-converting Enzyme Gene Associated in Patients with COPD with or without Bronchial Hyperresponsiveness)

  • 목 적 : ACE 유전자 다형성에 따라 폐를 포함한 여러 조직에서 ACE 활성 및 농도 등에 차이를 나타내고 이러한 차이가 COPD 등의 만성 호흡기 질환에서 질환의 발생 및 임상 표현형의 차이를 유발할 것으로 추정되어 지고 있다. 따라서 이 연구는 ACE 유전자 다형성이 COPD 환자에게서 동반될 수 있는 기도 과민반응 등의 기관지 천식 요소의 발현 유무와 연관성이 있는지 알아보고자 하였다. 방 법 : 100명의 COPD 환자들을 대상으로 기도 과민성의 동반 유무에 따라 두 군으로 분류하였고, PCR 방법을 통하여 ACE 유전자형을 검사하여 두 군 간의 차이를 알아보았다. 결 과 : COPD 환자에서 기도 과민반응 유무에 따른 ACE 유전자형의 분포에 차이는 없었고, COPD의 임상적 단계에 따른 각 군 간의 의미 있는 차이도 보이지 않았다. 결 론 : 이러한 연구 결과는 ACE 유전자 다형성에 따른 차이가 COPD 환자에게서 나타나는 기도 과민성 등의 천식 요소의 발현과 연관이 없음을 시사하는 소견이라 할 수 있다.
