Antidepressant-like effect of chlorogenic acid isolated from Artemisia capillaris Thunb.

  • Artemisia capillaris Thunb. is widely used in the herbal medicine field. This study describes the antidepressant effect of a flavonoid (chlorogenic acid) isolated from the Artemisia capillaris Thunb. The expression of the pituitary gland and hypothalamic POMC mRNA or plasma ${\beta}$-endorphin levels were increased by extract of Artemisia capillaris Thunb. or its flavoniod administered orally. In addition, antidepressant activity was studied using the tail suspension test (TST), the forced swimming test (FST) and the rotarod test in a chronically restrained immobilization stress group in mice. After restraint stress (2 h/day for 14 days), animals were kept in a cage for 14 days without any further stress, but with drugs. Mice were fed with a diet supplemented for 14 days and during the behavioral test period with chlorogenic acid (30 mg/kg/day). POMC mRNA or the plasma ${\beta}$-endorphin level was increased by the extract of Artemisia capillaris Thunb. and its flavoniod. In addition, the immobility time in TST and FST was significantly reduced by chlorogenic acid. In the rotarod test, the riding time remained similar to that of the control group at 15 rpm. Our results suggest that the flavonoid (chlorogenic acid) isolated from Artemisia capillaris Thunb. shows a potent antidepressant effect.

사철쑥과 비쑥의 분류학적 실체 (Taxonomic identities of Artemisia capillaris Thunb. and A. scoparia Waldst. & Kit.)

  • 사철쑥(Artemisia capillaris Thunb.)과 비쑥(A. scoparia Waldst. & Kit.)의 분류형질인 뿌리 형태, 잎 크기, 두상화서 크기는 사철쑥의 연속변이에 포함되어 두 분류군을 구분하는 유용한 형질이 아니며, 다른 분류학적 중요형질인 생육형에 있어서 관찰된 국내의 모든 표본과 생체들은 반관목 또는 다년초로서, 비쑥의 형질인 1, 2년초는 없는 것으로 확인되었다. 따라서 국내에 분포하는 개체 중 기존에 비쑥으로 알려진 다년초의 개체들은 사철쑥의 변이이며, 1, 2년초인 비쑥은 국내에 분포하지 않는 것으로 판단된다.

Artemisia capillaris Thunb. inhibits melanin synthesis activity via ERK-dependent MITF pathway in B16/F10 melanoma cells

  • Genus Artemisia occurs as a hardy plant and has a wide range of culinary and medicinal features. In this study, we aimed to describe the melanin inhibitory activity of one Artemisia species, i.e., Artemisia capillaris Thunb. Ethanol extracts of fermented Artemisia capillaris (Art.EtOH.FT) and non-fermented Artemisia capillaris (Art.EtOH.CT) were tested for their ability to inhibit tyrosinase activity and melanin pigmentation. Both extracts showed dose-dependent inhibition against ${\alpha}$-melanocyte stimulating hormone-stimulated melanin formation and tyrosinase activity, without cytotoxicity. At $100{\mu}g/mL$, both extracts showed greater inhibition than kojic acid, the positive control. Protein expressions of microphthalmia-associated transcription factor (MITF), tyrosinase (TYR), tyrosinase-related protein 1 (TRP-1), and tyrosinase-related protein 2 (TRP-2) at the transcriptional level were determined by using real-time and semi-quantitative polymerase chain reaction. To complete the mechanistic study, presences of upstream elements of MITF, the phosphorylated-extracellular signal-regulated kinase (p-ERK), and phosphorylated-mitogen-activated protein kinase kinase (p-MEK) were confirmed by using western blot analysis. Expressions of p-TYR, p-TRP-1 and p-TRP-2, downstream factors for p-ERK and p-MITF, were translationally inhibited by both extracts. Art.EtOH.FT induced more potent effects than Art.EtOH.CT, especially signal transduction effects. In summary, Artemisia capillaris extracts appear to act as potent hypopigmentation agents.

Simultaneous Determination of Four Compounds from Artemisia capillaris using High Performance Liquid Chromatography-Ultraviolet Detector (HPLC-UVD) and Their Quantitative Study in Artemisia Genus

  • Artemisia capillaris Thunb. (Compositae) is a native herb of East Asian countries and has used for the treatment of jaundice, high liver fever, and digestive diseases for a long time, as well as being developed as the source of herbal preparations until now. The major components from A. capillaris were chlorogenic acid (1) and its derivatives substituted with caffeoyl moieties, such as 3,5-dicaffeoylquinic acid (2) and 4,5-dicaffeoylquinic acid (3), and coumarins, such as scoparone. In the study, four compounds, chlorogenic acid, 3,5-dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid and scoparone (4) in the 70% ethanolic extract of A. capillaris were simultaneously determined by using HPLC-UVD system. This method was validated with the terms of linearity, precious and accuracy according to ICH guidelines. The developed method was successfully applied for the quantitative analysis of Artemisia genus, A. capillaris, A. iwayomogi, A. princeps, and A. argyi, distributed in Korea.

  • Objectives : Propolis and Hovenia dulcis Thunb. has been used as treatment of diseases in the Korean medicine. In this study, we investigated that the hepatoprotective effects of Propolis oral administration according to mixture with Hovenia dulcis Thunb. on ${\gamma}-GTP$, GOT, GPT, Total bilirubin, LDH, ALP, Total cholesterol, Triglyceride, SOD, activity of catalase and Glutathione Peroxidase in galactosamine (GalN)-induced liver in rats. Methods : The animals were divided into 5 groups. Control, the liver injury-induced and not treated group. Pro1, liver injury and administrated propolis. Pro2, liver injury and administrated propolis capsule. Pro3, liver injury and administrated mixture of propolis capsule with Hovenia dulcis Thunb. Pro4, liver injury and administrated mixture of Propolis capsule with Hovenia dulcis Thunb. and Artemisia capillaris Thunb.. Animals were treated by Oral administration of Propolis, Hovenia dulcis Thunb., and Artemisia capillaris Thunb. mixture ltime 2 days for 14 days. Results : The Pro1 group was significantly increased on ${\gamma}-GTP$ and activity of Glutathione peroxidase but decreased on GOT in serum as compared with the control group. The Pro2 group was significantly increased on WBC, RBC, Hct, HGB in serum and activity of CuZnSOD as compared with the control group. The Pro 3 group was decreased on Total bilirubin, increased on LDH, WBC, RBC, Hct and HGB in serum as compared with the control group. The Pro 4 group was decreased on GOT in serum as compared with the control group. Conclusion : By evaluating the liver function and lipid metabolism, Pro3 had a hepatoprotective effect on the prevention of hepatotoxity.

Genetic variation and relationship of Artemisia capillaris Thunb.(Compositae) by RAPD analysis

  • Randomly Amplified Polymorphic DNA (RAPD) was performed to define the genetic variation and relationships of Artemisia capillaris. Fifteen populations by the distributions and habitat were collected to conduct RAPD analysis. RAPD markers were observed mainly between 300bp and 1600bp. Total 72 scorable markers from 7 primers were applied to generate the genetic matrix, and 69 bands were polymorphic and only 3 bands were monomorphic. The genetic dissimilarity matrix by Nei's genetic distance (1972) and UPGMA phenogram were produced from the data matrix. Populations of Artemisia capillaris were clustered with high genetic affinities and cluster patterns were correlated with distributional patterns. Two big groups were clustered as southern area group and middle area group. The closest OTUs were GW2 and GG1 in middle area group, and GB1 from southern area group was clustered with OTUs in middle area group. RAPD data was useful to define the genetic variations and relationships of A. capillaris.

Chemical Constituents from the Aerial Parts of Artemisia capillaris Thunb. and Their Anti-allergic and Anti-inflammatory Effects

  • Artemisia capillaris Thunb. (Compositae) is a traditional medicinal plant with various pharmacological activities. To elucidate new anti-allergic and anti-inflammatory constituents, the aerial parts of A. capillaries were investigated to afford a new compound, (6E,8E)-6-methylundeca-6,8-diene-2,5,10-trione (17) together with 19 known compounds (1 - 16, 18 - 20). The structures of these compounds were determined by extensive spectroscopic analyses including 1D, 2D NMR, HREIMS, and optical rotation [α]D. The absolute configuration of compound 2 was determined to be S form for the first time. All isolates (1 - 20) were tested their inhibitory effects on interleukin 2 (IL-2) expression in T cells and NO production in lipopolysaccharide (LPS)-stimulated RAW246.7. Among them, compounds 10, 11, 19, and 20 reduced IL-2 expression in a dose-dependent manner. In addition, compound 10 also inhibited NO production with an IC50 value of 37.3 ± 0.4 μM.

인진분획물이 인체간세포의 $TGF{\beta}1-induced$ Apoptosis에 미치는 영향 (Effects of Five Fractions of Artemisia capillaris THUNB on $TGF{\beta}1-induced$ Apoptosis in HepG2 Cells)

  • Objectives: This study was camed out to examine the effect of five fractions of aqueous extract from Artemisia capillaris THUNB(ACT), on TGF, 1-induced apoptosis, cell viability, cell cycle progression and mRNA expression of apoptosis-related genes in human hepatocyte cell line HepG2. Methods: This study employed Tryphan blue exclusion assay, DNA fragmentation assay, Cpp32 protease activity assay and Quantitative RT-PCR analysis. Results: In the Tryphan blue exclusion assay, the butanol fraction of ACT with $TGF{\beta}$, l showed magnificent (Nice word, ut is it appropriate in a medical abstract\ulcorner) viability and the H2O fraction of ACT with $TGF{\beta}$, l also showed higher viability than only $TGF{\beta}$, l-treated group. DNA fragmentation assay showed that the butanol fraction and the H2O fraction carried inhibitory effects on apoptosis induction, with the butanol fraction displaying greater effects. The Cpp32 protease activity assay showed that the butanol fraction decreased Cpp32 protease activity. The H2O fraction of ACT had no significant effect on the Cpp32 protease activity. Quantitative RT-PCR showed that the butanol fraction suppressed Bax, p 15/INK4B, p21/Waf1, PAI-1 and increased Bcl-2 gene. Conclusions: The data shows that butanol fraction of ACT increases the hepatocyte viability and has the hepatocellular protective effect by the suppression of $TGF{\beta}$, l induced-apoptosis through gene regulation.

네트워크 약리학을 활용한 심상성 건선에 대한 인진호(茵蔯蒿)의 잠재적 작용 기전 탐색 연구 (A Network Pharmacology-based Study to Explore the Potential Mechanism of Artemisia capillaris Thunb. for Psoriasis Vulgaris)

  • Objectives : The purpose of this study is to investigate the potential mechanism of Artemisia capillaris Thunb. for psoriasis vulgaris. Methods : We conducted the network pharmacological analysis. It contains the process that search the compounds of the herb, the target proteins of the compounds, related genes of psoariasis vulgaris and the pathway/terms of the common protein lists between psoriasis vulgaris and Artemisia capillaris Thunb.. Results : 13 compounds and 30 protein targets of Artemisia Capillaris Herba were searched. And 997 psoriasis-related genes were searched. The common proteins were 11, and the core genes were 3; AKT1, CASP3, MAPK8. The related pathway/terms of 11 proteins were analyzed. ω-hydroxylase P450 pathway(60%), nitric oxide(NO) biosynthetic process(20%) were resulted. Also, 19 proteins of Artemisia Capillaris Herba were analyzed, and sterol homeostasis(78.95%), sterol biosynthetic process(15.79%), Type 2 diabetes mellitus(5.26%) were resulted. Conclusion : The Artemisia Capillaris Herba can potentially act through the ω-hydroxylase P450 pathway and nitric oxide(NO) biosynthetic process for psoriasis. Also, the metabolism of sterol biosynthesis and homeostasis can be involved in a roundabout way for psoriasis.

사철쑥 추출물의 면역세포의 생육증진 및 세포독성 (Enhanced Immune Activity and Cytotoxicity of Artemisia capillaris Thunb. Extracts against Human Cell Lines)

  • 사철쑥 추출물은 정상폐세포인 HEL299 세포주에 $0.5\;g/{\ell}$ 이하의 농도로 투여시는 정상세포 생존율을 80% 이상으로 유지시켜 정상세포에 대한 안정성이 유지되었으며, 암세포 생육억제활성은 증류수 및 에탄올 추출물 모두 $0.5\;mg/m{\ell}$ 이상의 농도에서 유방암세포주인 MCF7에 대하여 88%이상, 폐암세포주인 A549에 대하여서는 모든 추출물에서 $0.5\;mg/m{\ell}$의 농도 이상에서 65%이상의 저해를 나타내었다. 또한 간암세포주인 Hep3B에 대하여는, 에탄올 추출물이 87% 이상, 위암세포주인 AGS에 대하여 증류수 추출물이 약 85% 이상의 높은 암세포 생육억제활성을 나타내었다. 또한 정상세포독성에 대한 암세포의 억제율을 나타내는 selectivity에 있어서 각 추출물이 $0.1\;mg/m{\ell}$에서 $1.0\;mg/m{\ell}$의 농도에서 1.5 이상의 수치를 나타내었다. B와 T 세포주의 생육촉진 실험 결과 배양 기간이 증가함에 따라 세포의 생육이 촉진되었으며, 배양 5일째 $0.5\;mg/m{\ell}$의 농도에서 B세포의 생장을 최고 1.2배 이상 증가, T-cell을 $1.2{\sim}1.5$배 이상 생장을 증가시켰으며, 배양 6일째 에탄올 추출물이 IL-6를 $67\;pg/m{\ell},\;TNF-{\alpha}$를 물 추출물이 $68\;pg/m{\ell}$의 분비 하였다.
