Development of Analytical Method and Validation using HPLC/PDA for Discrimination between Artemisiae Argyi Folium and Artemisiae Iwayomogii Herba

  • In this study, we described the new developed method to simultaneously discriminate two herbal drugs of Artemisiae Argyi Folium and Artemisiae Iwayomogii Herba using eight marker compounds (1 - 8) on an HPLC-PDA system. The developed method was applied to quantify the major components of two herbal drugs. The pattern analysis successfully discriminated and evaluated different components between Artemisiae Argyi Folium and Artemisiae Iwayomogii Herba. Results were used for classification of different species from collected samples.

Antimicrobial Effect on the Periodontal Pathogens and Anti-inflammatory Effect of Artemisiae Iwayomogii Herba (한인진(韓茵蔯)의 치주염세균에 대한 항균효과 및 항염효가)

  • Objectives : The purpose of this study was to evaluate on the antimicrobial effect on the periodontal pathogens and anti-inflammatory effect of Artemisiae Iwayomogii Herba. Artemisiae Iwayomogii Herba has been used for treating as Artemisiae Capilaris Herba in Korea. Methods : Artemisiae Iwayomogii Herba was prepared by extracting medicinal herb with water. We investigated antimicrobial activity by the minimun inhibitory concentration (MIC) test. We also investigated inhibition of IL-$1{\beta}$-induced collagenase-l(MMP-l), stromelysin-1(MMP-3), interleukin-6 gene expression in human gingival fibroblasts. Results : The antimicrobial effect of Artemisiae Iwayomogii Herba was evaluated with MIC against periodontopathogens; Porphyromonas gingivalis 2561, W50, A7A1-28, 9-14K-1, Prevotella intermedia28, and Actinobacillus actinomycetemcomitans Y4, MICs of Artemisiae Iwayomogii Herba were 0.156 mg/ml, 0.625 mg/ml, 0.313 mg/ml, 1.25 mg/ml, 10 mg/ml and 10 mg/ml. The anti-inflammatory effect of Artemisiae Iwayomogii Herba was evaluated with Influence of herbs on the IL-$1{\beta}$-induced expression of MMP-1, MMP-3, interleukin-6, IL-$1{\beta}$ increased MMP-1, MMP-3, interleukin-6 mRNA levels. Artemisiae Iwayomogii Herba significantly inhibited IL-$1{\beta}$-induced MMP-1, MMP-3, interleukin-6 gene expressions in a dose-dependent manner. Conclusions : These results suggested that Artemisiae Iwayomogii Herba might reduce the excessive proteolytic capacity of the gingival fibroblast during inflammation and could be developed a new drug in periodontitis.

The Effects of Artemisiae Iwayomogii Herba, Curcumae Radix, and Aurantii Fructus Immaturus Complex Extract (ACA) on Dyslipidemia-related Factor Expression and Anti-oxidation in HepG2 Cells (인진(茵蔯), 울김(鬱金), 지실(枳實) 추출물(ACA)이 HepG2 세포에서 나타나는 이상지질혈증 관련 인자 발현 및 항산화에 미치는 영향)

  • Objective: To investigate the effect of Artemisiae Iwayomogii Herba, Curcumae Radix, and Aurantii Fructus Immaturus complex extract (ACA) on dyslipidemia-related factor expression and anti-oxidation in HepG2 cells. Method: After treatment with ACA in the HepG2 cells, DPPH, ABTS radical scavenging activity, ROS production, and glutathione (GSH) production were measured. The free fatty acid, lipid peroxidation (MDA), ACAT1, and HMG-CoA reductase mRNA expression were measured in the HepG2 cells after treatment with ACA. Results: 1. DPPH, ABTS radical scavenging activity increased in an ACA concentration-dependent manner. 2. ACA significantly decreased ROS production in comparison to the control group. 3. ACA significantly increased glutathione production. 4. ACA significantly decreased free fatty acid and lipid peroxidation (MDA) in the HepG2 cells. 5. ACA decreased the mRNA expression of ACAT1 and HMG-CoA reductase. Conclusion: These results suggest that Artemisiae Iwayomogii Herba, Curcumae Radix, and Aurantii Fructus Immaturus complex extract (ACA) inhibits dyslipidemia-related factor expression and that it is effective in anti-oxidation. A future in vivo experiment with ACA is needed to investigate the effect on anti-dyslipidemia. It is expected that ACA is effective in anti-dyslipidemia and applied to cardiovascular disease, ischemic heart disease, stroke, etc.

Antimicrobial Effects Of Herbs For Removing Dampness And Promoting Urination Against Vaginal Microbe (이수삼습약(利水參濕藥)의 질내(膣內) 미생물(微生物)에 대한 항균효과(抗菌效果))

  • Purpose : This study was conducted to investigate the antimicrobial effects of herb for removing dampness and promoting urination against vaginal microbes. Methods : Staphylococcus aureus. Methicillin-resistant Staphylococcus aureus(MRSA), Candida albicans and Gardnerella vaginalis were used for vaginitis-induced microbes. Lactobacillus gasseri, Streptococcus spp. and Escherichia coil HB101 were used for normal vaginal florae. And herbs for removing dampness and promoting urination(Dianthi herbs. Tokoro Rhizoma, Saururi Herbs, Pyrrosiae Folium, Artemisiae Iwayomogii Herba, Plantaginis Semen, Tetrapanacis Medulla, Polygoni Avicularis Herba, Malvae Semen, Akebiae Caulis, Kochiae Fructus, Lygodii Spora) were used. Antimicrobial activities were estimated by the change of optical densities and colony test in vitro. Results : Plantaginis Semen, Artemisiae Iwayomogii Herba and Lygodii Spora had the antimicrobial susceptibility and selective toxicity against MRSA and Gardnerella vaginalis. Polygoni Avicularis Herba had the antimicrobial susceptibility and selective toxicity against Staphylococcus aureus. MRSA and Gardnerella vaginalis. Malvae Semen and Kochiae Fructus had the antimicrobial susceptibility and selective toxicity against MRSA. Dianthi Herba had the antimicrobial susceptibility and selective toxicity against Gardnerella vaginalis. Conclusion : According to these results, we can suggest that Plantaginis Semen, Artemisiae Iwayomogii Herba, Lygodii Spora, Polygoni Avicularis Herba, Malvae Semen, Kochiae Fructus and Dianthi Herba would be available to the antimicrobial agent for vaginitis-induced microbe in vitro.

  • Objectives: To investigate the effect of fermented extract of Artemisiae Iwayomogii Herba, Curcumae Longae, Crataegi Fructus and Salviae Miltiorrhizae Radix (FMH) on anti-inflammation associated with dyslipidemia and anti-oxidation in RAW264.7 and HUVEC cells. Methods: The total polyphenols, total flavonoids, DPPH radical scavenging activity, ABTS radical scavenging activity, and cytotoxicity of FMH were measured. RAW264.7 cells treated with FMH were tested for production of NO, and for cytokine and LTB4 levels and HUVEC cells treated with FMH were examined for production of cDNA of genes related to inflammation. Results: 1. FMH contained polyphenols and flavonoids. The DPPH and ABTS radical scavenging activity of FMH increased in a concentration-dependent manner. 2. FMH treatment inhibited the production of nitric oxide (NO), cytokines, and LTB4 in RAW264.7 cell when compared to the untreated control group. 3. FMH decreased the transcription of pro-inflammatory genes, whereas it increased transcription of anti-inflammatory genes, in HUVEC cells. Conclusion: FMH is effective as an antioxidant and for treatment and prevention of dyslipidemia, atherosclerosis, ischemic heart disease, stroke, and other cardiocerebrovascular diseases.

Antioxidant and Anti-dyslipidemic Effect of Artemisiae iwayomogii Herba, Curcumae longae Radix, and Plantaginis Semen Complex Extract(ACP) on HepG2 Cells (HepG2 cell에서 한인진, 울금, 차전자 추출물(ACP)의 항산화 및 항이상지질혈증 효과)

  • This study was performed to investigate the antioxidant and antidyslipidemic effects of Artemisiae iwayomogii Herba, Curcumae longae Radix and Plantaginis Semen complex extract(ACP) on HepG2 cells. We measured total polyphenols, total flavonoids, radical scavenging activity, and ABTS radical scavenging activity of ACP to evaluate its antioxidant activity. HepG2 cells were treated with ACP. Then, we evaluated ROS production; intracellular GSH content; GPx, GR, SOD, and catalase activities; free fatty acids and MDA levels; and mRNA expression levels of ACAT1 and HMG-CoA reductase. Results: ACP contains polyphenols and flavonoids and increased the DPPH and ABTS radical scavenging activities in HepG2 cells in a dose dependent manner. Also, ACP significantly reduced ROS production in HepG2 cells compared to the control group and significantly increased the GSH content, and elevated the enzyme activities of GPx, GR, and catalase in HepG2 cells compared to the control group. In addition, ACP reduced the mRNA expression of ACAT1 and HMG-CoA reductase in HepG2 cells compared to that in the control group. Conclusion: These results suggest that ACP has an antioxidant effect and may suppress the expression of dyslipidemia - associated genes and thus may be useful for the improvement of dyslipidemia.

Protective Effects of a Composite Preparation (Samulchungkan-Tang) of Crude Drugs on Hepatic Injury Induced by Toxic Drugs in Rats (생약복합제제(生藥複合製劑)(사물청간장(四物淸肝腸))의 흰쥐 약물성(藥物性) 간(肝) 장해(障害)에 대한 보호작용(保護作用))

  • This study was attempted to investigate the effects of Composite Preparation (Samulchungkan-Tang) extract (SCTE), Scutellarias Radix extract (SRE), Artemisiae iwayomogii Herba extract (AIHE), Artemisia capillaris Flos extract (ACFE), Paeaniae Radix extract (PRE) and Gardeniae Fructus extract (GFE) on the activities of GOT, GPT, ALP and LDH, and Content of total cholesterol in serum of $CCl_4$ and ${_D}-galactosamime$ intoxicated rats, and bile flow in rats. 1) In $CCl_4-intoxicated$ rats-The activities of S-GOT and S-GPT which were elevated by $CCl_4$ were significantly decreased in dose of SCTE 450 mg/kg, ACFE 600 mg/kg and GFE 300 mg/kg, respectively as compared to $CCl_4$ intoxicated rats. ALP activity increased by $CC1_4-treatment$ was markedly decreased in dose of SCTE 450 mg and 600 mg/kg, SRE 400 mg/kg, AIHE 400 mg/kg, ACFE 600 mg/kg and PRE 300 mg/kg, and LDH activity in SCTE 450 mg and 600 mg/kg, ACFE 600 mg/kg and GFE 300 mg/kg, respectively compared to $CCl_4$ treated rates. ACFE 400 mg/kg and PRE 300 mg/kg decreased the content of total cholesterol increased by $CCl_4$, the liver weight in all sample administered groups was decreased significantly as compared to $CCl_4$ treated groups. 2) In ${_D}-galactosamine$ intoxicated rats-Sample of SCTE 450 mg and 600 mg/kg, SRE 400 mg/kg, AIHF 400 mg and 600 mg/kg, ACFE 600 mg/kg, PRE 300 mg/kg and GFE 300 mg and 500 mg/kg decreased the activities of S-GPT, ALP and LDH which was increased by ${_D}-galactosamine$ intoxication, compared to ${_D}-galactosamine$ intoxicated groups. In S-GOT activity elevated by ${_D}-galactosamine$ was significantly decreased by SCTE 450 mg/kg, ACFE 600mg/kg, AIHE 600 mg/kg, PRE 300 mg/kg, GFE 300 mg and 500 mg/kg. However, SCTE 600 mg/kg, SRE 400 mg/kg, and AIHE 400 mg/kg were not effected significantly. 3) In bile secretion-SCTE 450 mg and 600 mg/kg, ACFE 600 mg/kg and GFE 500mg/kg increased significantly the amount of bile secretion as compared to normal groups, but AIHE 400 mg/kg, SRE 400 mg/kg, and PRE 300 mg/kg did not effected significantiy.

Screening of Anti-angiogenic Activity from Plant Extracts (식물 추출물의 혈관신생 억제 효능 검색)

  • We examined anti-angiogenic effects of water extracts from 32 plant materials (20 Korean medicinal plants and 12 western herbs) using cell-based anti-angiogenic assay, HUVEC tube formation assay, and then we found that 7 plant extracts inhibited HUVEC tube formation strongly. The plant materials which showed anti-angiogenic effects are Cinnamomi Ramulus, Atractylodis Rhizoma alba, Polygalae Radix, Myristicae Semen, Artemisiae Iwayomogii Herba, leaves of Rosmarinus officinalis, and leaves of Melissa officinalis. We also investigated inhibitory effects of these anti-angiogenic herbal extracts on MMP (matrix metalloproteinase) activity which has important roles in angiogenesis. Among extracts tested in this study, water extract of Melissa officinalis showed the most potent anti-angiogenic and MMP inhibitory activity.

Kagam-bojungikgitang Inhibits LPS-induced Inflammatory Mediators in RAW 264.7 Macrophages (가감보중익기탕의 LPS 유도 염증성 매개물에 대한 억제 효과)

  • Kagam-bojungikgitang is the water extracts prepared from Ginseng Radix, Astragali Radix. Angelicae gigantis Radix, Astractylodis Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisiae iwayomogii Herba, and Scutellariae Radix. This is a modified prescription of Bojungikgitang, which has been used for the treatment of indigestion, and immunological disease in oriental countries. In this study, the effects of Kagam-bojungikgitang and Bojungikgitang on the production of prostaglandin E₂ (PGE₂) and the expression of cyclooxygenase-2 (COX-2) were examined using RAW 264.7 macrophages activated with lipopolysaccharide (LPS). Both prescriptions dose-dependently reduced the release of PGE2 and expression of COX-2 caused by stimulation of LPS without cytotoxic effect. Kagam-bojungikgitang's inhibitory effects were better than Bojungikgitang in PGE2 production and COX-2 expression. Moreover, Kagam-bojungikgitang also attenuated markedly the production of tumor necrosis factor (TNF)-α, and IL-6 than Bojungikgitang in LPS-stimulated RAW 264.7 macrophages. These results suggest that Kagam-bojungikgitang decreases PGE2 and pro-inflammatory cytokine production in macrophages and these properties may contribute to the anti-inflammatory activity of Kagam-bojungikgitang.
