Boron Analysis in High Carbon Graphitized Steel using Neutron Autoradiography (Neutron Radiography를 이용한 고탄소흑연강에서 붕소 분석)

  • To study the distribution of boron and the boron effect for nucleation of graphite in high carbon steel, neutron induced autoradiography method is used. High carbon steel is easy to make the graphitization by addition of boron. It is easy to analysis of boron distribution using neutron radiography with neutron fluence of $1.9$\times${\times}10^{13}/cm^2$in the boron added high carbon steel. By the neutron induced autoradiography technique, it was found that the distribution of boron depended on boron content, graphitiging temperature and time. And by the analysis of secondary ion mass spectroscopy (SIMS) and electron probe micro analysis (EPMA), boron or boride were acted at nucleation site of graphite in high carbon steel.

  • The behavior of the non-equilibrium grain boundary segregation of boron in low carbon steel was studied through a particle tracking autoradiography. The behavior of the non-equilibrium grain boundary segregation of boron during continuous cooling was compared with the isothermal kinetics of the non-equilibrium grain boundary segregation of boron at the holding temperature using an effective time method. On the basis of the experiments, the cooling rate dependence of the non-equilibrium segregation of boron was explained using the time dependence of the non-equilibrium segregation of boron in low carbon steel. The experimental observations for the cooling rate dependence of the grain boundary segregation of boron are in good agreement with the time dependence of the grain boundary segregation of boron. The mechanisms of the non-equilibrium segregation of boron during cooling in low carbon steel are also discussed.

Distribution of the Muscarinic Cholinergic Receptors and Characterization in the Brain of Wistar Rats and Spontaneously Hypertensive Rats (SHR Strain) by Digital Autoradiography (Digital Autoradiographic System을 이용한 선천성고혈압에서의 Muscarinic Cholinergic Receptor 분포 및 특성)

  • Using in vitro autoradiography with a digital autoradiography system and radioreceptor assay, the distribution and the binding characteristics of the muscarinic cholinergic receptors (mAChR) were studied in regions of rat brain. Radioreceptor assay revealed that mAChR could be measured with saturation binding assay in the brain and heart homogenates: No difference in Kd or Bmax of the brain or heart was found between the normal Wistar rats and SHR rats. Specific binding of $^3H$ quinuclidinyl benzilate (QNB) increased and saturation was reached by 2 hours after incubation with slide-mounted brain tissue. The distribution of mAChR was heterogeneous along the fields of brain. Affinity (Kd) of mAChR was not different significantly among cortex, hippocampus and caudate-putamen. No difference was found between normal rats and SHR strain. More receptors (Bmax) were found in the cortex and hippocampus than in the caudate-putamen in normal rats. More receptors were found in the cortex and caudate-putamen in SHR rats than in normal rats. Radioreceptor assay and digital autoradiographic analysis of affinity and number of mAChR gave the same results. With the above findings, we concluded that we could use digital autoradiographic system with $^3H$-QNB in the characterization of mAChR of rats and that the cortex and caudate-putamen of SHR strain rats have more receptors than those of normal rats.

Neural Adaptation of Beta Adrenergic Receptor Subtypes after Chronic Imipramine Treatment: A Quantitative Autoradiographic Study

  • This study compares the subtypes of central beta adrenergic receptors (ARs) of brains of untreated rats with those of imipramine-treated rats. Beta adrenergic receptors were measured by quantitative autoradiography of the binding of $^3H$-dihydroalprenolol ($^3H$-DHA) in coronal sections of rat brain. Repeated treatment of rats with imipramine significantly reduced the binding of $^3H$-DHA to beta-1 AR in many brain areas, especially throughout the cerebral cortex, hippocampus, thalamus, and amygdala. Significant reductions of the binding of $^3H$-DHA to beta-2 AR were not found in any area of the brain. These data suggests that a selective down-regulation of beta-1 AR may be involved in the adaptive changes occurring after prolonged imipramine treatment.

  • By using autoradiography and phosphate medium, high phosphate accumulating bacteria were isolated from the soil of protected cultivation area and activated sludge. Selected strain, PO8, was gram-negative, rod/spherial(0.5~0.6$\times$1.0~1.2${\mu}{\textrm}{m}$ in size) and non-motile. PI4, another selected strain, was gram-negative, rod(0.4~0.5$\times$1.5~1.6${\mu}{\textrm}{m}$ in size) and motile. It also had flagella. According to their morphological, physiological and biochemical properties, the stains were identified as Acinetobacter lwoffi PO8 and Chromobacterium lividum PI4, respectively. A. lowoffi PO8 and C. lividum PI4 cultured in the P-1 medium containing 150ppm phosphate were able to uptake high phosphate up to 92% and 85%, respectively after 24 hours at 3$0^{\circ}C$ during liquid culture.

  • The purpose of this study is to investigate the effect of Shudihuang water extracts on the cGMP production of medulla and cortical membranes, receptors for atrial natriuretic peptide in the kidney by in vitro autoradiography in rats for 8, 16 weeks. The cGMP production of medullary and cortical membranes of the kidney decreased after the administration of Shudihuang water extracts. The density of receptors for atrial natriuretic peptide in the kidney decreased after the administration of Shudihuang water extracts only for 16 weeks. These results suggest that the long term administration of Shudihuang water extracts has decreased plasma renin activity and plasma levels of aldosterone modulated cGMP production of medullary and cortical membranes, density of receptors for atrial natriuretic peptide in the kidney.
