Left Ventricle Segmentation Algorithm through Radial Threshold Determination on Cardiac MRI (심장 자기공명영상에서 방사형 임계치 결정법을 통한 좌심실 분할 알고리즘)

  • The advance in medical technology has decreased death rates from diseases such as tubercle, pneumonia, malnutrition, and hepatitis. However, death rates from cardiac diseases are still increasing. To prevent cardiac diseases and quantify cardiac function, magnetic resonance imaging not harmful to the body is used for calculating blood volumes and ejection fraction(EF) on routine clinics. In this paper, automatic left ventricle(LV) segmentation is presented to segment LV and calculate blood volume and EF, which can replace labor intensive and time consuming manual contouring. Radial threshold determination is designed to segment LV and blood volume and EF are calculated. Especially, basal slices which were difficult to segment in previous researches are segmented automatically almost without user intervention. On short axis cardiac MRI of 36 subjects, the presented algorithm is compared with manual contouring and General Electronic MASS software. The results show that the presented algorithm performs in similar to the manual contouring and outperforms the MASS software in accuracy.

  • The elution of di(2-ethylhexyl) phthalate (DEHP) from flexible bags into human blood or transfusion was studied. The conditions of determination of DEHP using high performance liquid chromatography (HPLC) was established as follows: Condition I-column, ${\mu}-Bondapak^{TM}\;C_{18}$; mobile phase, methanol: water=91 : 9 ; flow rate, 1.2ml/min; wavelength, 254nm; injection volume, $10{\mu}l$. Condition II-column, Lichorsorb RP-18$(10{\mu}m)$; mobile phase, methanol: water=94 : 6 ; flow rate, 1.1ml/min; wavelength, 254nm; injection volume, $10{\mu}l$. DEHP was found to be migrating from PVC blood and total parentral nutrient bags into methanol, but not into anti, coagulant drug solution.

A simple and novel equation to estimate the degree of bleeding in haemorrhagic shock: mathematical derivation and preliminary in vivo validation

  • Determining blood loss [100% - RBV (%)] is challenging in the management of haemorrhagic shock. We derived an equation estimating RBV (%) via serial haematocrits (Hct1, Hct2) by fixing infused crystalloid fluid volume (N) as [0.015 × body weight (g)]. Then, we validated it in vivo. Mathematically, the following estimation equation was derived: RBV (%) = 24k / [(Hct1 / Hct2) -1]. For validation, non-ongoing haemorrhagic shock was induced in Sprague-Dawley rats by withdrawing 20.0%-60.0% of their total blood volume (TBV) in 5.0% intervals (n = 9). Hct1 was checked after 10 min and normal saline N cc was infused over 10 min. Hct2 was checked five minutes later. We applied a linear equation to explain RBV (%) with 1 / [(Hct1 / Hct2) -1]. Seven rats losing 30.0%-60.0% of their TBV suffered shock persistently. For them, RBV (%) was updated as 5.67 / [(Hct1 / Hct2) -1] + 32.8 (95% confidence interval [CI] of the slope: 3.14-8.21, p = 0.002, R2 = 0.87). On a Bland-Altman plot, the difference between the estimated and actual RBV was 0.00 ± 4.03%; the 95% CIs of the limits of agreements were included within the pre-determined criterion of validation (< 20%). For rats suffering from persistent, non-ongoing haemorrhagic shock, we derived and validated a simple equation estimating RBV (%). This enables the calculation of blood loss via information on serial haematocrits under a fixed N. Clinical validation is required before utilisation for emergency care of haemorrhagic shock.

Rapid Determination of Volatile Organic Compounds in Human Whole Blood Using Static Headspace Sampling with Gas Chromatography and Mass Spectrometry

  • Headspace (HS) and headspace solid-phase microextraction (HS-SPME) were studied for extracting volatile organic compounds (VOCs) from whole blood, with chemical and instrumental variables being optimized for maximum sensitivity: incubation at $60^{\circ}C$, equilibration for 30 min, pH 11, and 2 mL injection volume. Both techniques provided accurate analyses, with detection limits of 0.05-0.1 ng $mL^{-1}$ and 0.05-0.5 ng $mL^{-1}$. HS showed better sensitivity, reproducibility, and analysis times than HS-SPME. Overall levels of chloroform in whole blood were found to be 0.05-5.84 ng $mL^{-1}$; detected levels of benzene were 0.05-2.20 ng $mL^{-1}$.

  • The purpose of this study is to collect biosignal data in a non-invasive and non-restrictive manner using a BCG (Ballistocardiogram) sensor, and utilize artificial intelligence machine learning algorithms in ICT and high-performance computing environments. And it is to present and study a method for developing and validating a data-based blood glucose prediction model. In the blood glucose level prediction model, the input nodes in the MLP architecture are data of heart rate, respiration rate, stroke volume, heart rate variability, SDNN, RMSSD, PNN50, age, and gender, and the hidden layer 7 were used. As a result of the experiment, the average MSE, MAE, and RMSE values of the learning data tested 5 times were 0.5226, 0.6328, and 0.7692, respectively, and the average values of the validation data were 0.5408, 0.6776, and 0.7968, respectively, and the coefficient of determination (R2) was 0.9997. If research to standardize a model for predicting blood sugar levels based on data and to verify data set collection and prediction accuracy continues, it is expected that it can be used for non-invasive blood sugar level management.

Bleeding Efficiency and Meat Oxidative Stability and Microbiological Quality of New Zealand White Rabbits Subjected to Halal Slaughter without Stunning and Gas Stun-killing

  • A study was conducted to compare the effect of halal slaughter without stunning and gas stun killing followed by bleeding on residual blood content and storage stability of rabbit meat. Eighty male New Zealand white rabbits were divided into two groups of 40 animals each and subjected to either halal slaughter without stunning (HS) or gas stun-kill (GK). The volume of blood lost during exsanguination was measured. Residual blood was further quantified by determination of haemoglobin content in Longissimus lumborum (LL) muscle. Storage stability of the meat was evaluated by microbiological analysis and measuring lipid oxidation in terms of thiobarbituric acid reactive substances (TBARS). HS resulted in significantly higher blood loss than GK. HS had significantly lower residual haemoglobin in LL muscle compared to GK. Slaughter method had no effect on rabbit meat lipid oxidation at 0, 1, and 3 d postmortem. However, at 5 and 8 days of storage at $4^{\circ}C$, significant differences (p<0.05) were found, with meat from the GK group exhibiting significantly higher levels of MDA than that from HS. At day 3, greater growth of Pseudomonas aeroginosa and E. coli were observed in the GK group (p<0.05) with B. thermosphacta and total aerobic counts remained unaffected by slaughter method. At days 5 and 7 postmortem, bacterial counts for all tested microbes were affected by slaughter method, with GK exhibiting significantly higher growth than HS. It can be concluded that slaughter method can affect keeping quality of rabbit meat, and HS may be a favourable option compared to GK due to high bleed out.

Effects of External Whole-Body gamma-Irradiation on Blood Picture and Growth in Young Rabbits (Cobalt-60 gamma 선(線) 외부전신조사(外部全身照射)가 어린 토끼의 혈액상(血液像)과 성장률(成長率)에 미치는 영향(影響))

  • In the event a population of animals is exposed to ionizing radiation, proper disposition of animals will minimize loss to the farmer and protect the public from unwholesome meat and dairy products. Clinical response is an important factor in considering salvage of animals for food following exposure to ionizing radiation whether the dose is lethal or not. It is the purpose of this report to present the discussions of blood picture and growth of y ung growing rabbits exposed to cobalt-60 gamma-irradiation. The experimental animals were Hyrnalayan rabbits of 65 days old. The body weight for all animals of the time of irradiation was approximately 450g. The 96 rabbits used in the experiment were allotted t groups of eight. Rabbits in one group served as controls and the others were exposed to single doses of 100, 200, 300, 400, 500, 600 and 700 Roentgens (R). The exposure does rate averaged 15 R per minute. Central blood was obtained by heart puncture. Erythrocyte and leukocyte enumerations and, determinations of hemoglobin in blood and hematocrit value were made in the usual manner. Erythrocyte counts, concentrations of hemoglobin in blood, and hematocrit readings dropped to the lowest value between 1 and 4 weeks postirradiation, returning towards the control values. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. There was a progressive decrease with increasing radiation does in the above values. Mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration were not affected by the irradiation. After irradiation of each does the numbers of total leukocyte and the absolute numbers of various types of white cell began to decrease rapidly, and reached a minimum after 2 weeks, returning towards normal levels. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. The reductions in total leukocyte and differential counts as the radiation does increases were apparent. Relative neutrophilia was observed in each irradiated group because of more expense of lymphocytes after irradiation. Growth rate of the rabbits was not affected by 100 R of whloe-body irradiation, while rabbits irradiated with 200, 300, 400 and 500 R showed marked depression of growth rate. As the radiation does increases, a depression of growth rate was apparent. The rabbits exposed to 600 and 700 R ceased from growing and recovery was not recognized during the experimental period.

The Determination of Blood-Brain Barrier Permeability and Pharmacokinetics of a Rat Transferrin Receptor Monoclonal Antibody by Brain Perfusion Method and Intravenous Injection Technique in Mice (마우스에서 뇌관류법과 정맥투여법에 의하여 흰쥐 트란스페린 단일항체의 체내동태 및 혈액-뇌 관문 투과성의 검토)

  • Brain drug targeting through the blood-brain barrier (BBB) in vivo is possible with peptidornirnetic monoclonal antibodies that undergo receptor-mediated transcytosis through the BBB. Monoclonal antibody to the rat transferrin receptor, such as the OX26 was studied in rats as a transport vector through BBB on the transferrin receptor. But, OX26 is not an effective brain delivery vector in mouse. In the present studies, rat monoclonal antibody, 8D3 to the mouse transferrin receptor were evaluated for brain drug targeting vector intransgenic mouse model. Pharrnacokinetic parameters in plasma and organ uptakes were determined at varioustimes after i.v. bolus injection of [$^{}125}I$] 8D3 in Balb/c mice. Brain uptake of [$^{}125}I$] 8D3 was also studied with an internal carotid artery perfusioncapillary depletion method. After i.v. injection of [$^{}125}I$] 8D3, plasma concentrations declined biexponentially with elimination half lift of approximately 2.2 hours. Brain uptake of [$^{}125}I$] 8D3 was $0.50{\pm}0.09$ persent of injected dose per g brain after 2 hours i.v. injection. After perfusion 5 min the apparent volume of distibution of [$^{}125}I$] 8D3 in brain was $22.3 {\mu}l/g,$ which was 4.8 fold higher than the intravascular volume. These studies indicate rat monoclonal antibody to the mouse transferrin receptor, 8D3 may be used for brain drug targeting vector in mice.

A Characteristics of Phagocytic Plaque on Staphylococcus aureus Layer Formed by Leukocytes of the Alcoholics (Staphylococcus aureus를 이용한 주정중독환자 백혈구의 Phagocytic Plaque 형성력)

  • This study was conducted to develop a method for direct determination of phagocytic activities in human circulatic systems and to measure the phagocytic activities in human leukocytes from the alcoholics, since phagocytic activity was considered to be very important in human immune mechanism at early stage for the health care of the alcoholics. The subjects for this study were 130 among which 95 males and 3 females were diagnosed as alcoholism and 32 was healthy blood donors. A thin layer of heat-killed Staphylococcus aureus Cowan I was placed on a plastic dish and reacted with whole blood to measure the phagocytic plaque formation by human leukocytes. In order to determine the health conditions of the subjects, some clinical laboratory tests, such as white blood cell counts, hemoglobin contents (Hgb), mean corpuscular volume of red blood cells(MCV), serum electrophoresis, B and T-lymphocytes, T-lymphocyte subtypes and phytohemagglutination test were also implemented. Compared to the non-alcoholism, new and old alcoholic inpatients showed statistically significant differences on levels of Hgb and MCV (p<0.05), but showed that T and B-lymphocyte numbers decreased and Helper T cell/Suppressor T cell ratio ($1.6{\pm}0.8$%) increased. Compared to non-alcoholism, phagocytic plaque activities of leukocytes from alcoholic patients decreased significantly and an unusual pattern in phagocytic plaque was observed, showing a strange body and chain shaped phagocytosis. Based upon these results, it is concluded that a phagocytic-plaques of Staphylococcus aureus Cowan I by leukocytes was very simple and useful method for the early immunological determination of phagocytic activities in alcoholic patients without requiring any special equipments.
