Screening for Biological Activity of Crude Extract and Bioactive Fractions from Brachyglottis monroi

  • The effects of crude extract and bioactive fractions from Brachyglottis monroi on biological activity were investigated. The crude ethanol extract inhibited the growth of the Gram positive bacterium Bacillus subtilis (ATCC Strain number 19659, 1 mm zone at 150 μg/disk) and the dermatophyte Trichophyton mentagrophytes (ATCC 28185, 2 mm zone at 150 μg/disk), and toxic to P388 tumor cells (IC50 23.96 μg/ml at 75 μg/disk). Cytotoxic activity was strongly showed by Fr. 6 (P388 IC50 19.67 μg/ml at 75 μg/disk).

  • The effects of bioactive fractions from Brachyglottis monroi on biological activity were investigated. this bioactive subfraction 6-5 is the most cytotoxic to P388 murine leukaemia cell lines. A comparison of IC/sub 50/ values of these subfraction in cancer cell lines showed that their susceptibility to these subfractions decreased in the following order; Fr. 6-5 > Fr. 6-3 > Fr. 6-6 > Fr. 6-1 > Fr. 6-2 > Fr. 6-4 by the MTT method. Silica gel flash column chromatography concentrated the cytotoxic activity in subfraction 6-5 which eluted with 30% and 40% ethyl acetate : hexane gave a major bioactive (51 mg, P388 IC/sub 50/ 8,286 ng/mL at 7.5 ㎍/disc).

  • 뉴질랜드의 자생식물인 Brachyglottis monroi와 Trichocolea hatcheri의 에탄올추출물에 대한 생리활성을 조사한 결과, 그람음성균인 Bacillus subtilis (ATCC 19659, 1 mm inhibition zone at $150{\mu}g/disc$)와 피부곰팡이균인 Trichophyton mentagrophytes (ATCC 28185, 6 mm inhibition zone at $150{\mu}g/disc$)에 대한 억제효과가 관찰되었으며, 이들 추출물은 P388 murine leukaemia cells ($IC_{50}$ $23.96{\mu}g/mL$)에서도 세포독성이 나타났다. B. monroi의 에탄올추출물의 항바이러스효과는 T. hatcheri의 에탄올추출물보다 강한 활성이 관찰되었으며, T. hatcheri의 에탄올추출물의 항균효과는 B. monroi의 에탄올추출물보다 강한 활성을 나타났다. 그렇지만 P388 murine leukaemia cells ($IC_{50}>12,500{\mu}g/mL$)에 대한 세포독성은 아주 미미한 것으로 나타났다.

  • The inhibitory effect of 13(E)-Labd-13-ene-8$\alpha$ 15-diol(1), isolated from the ethanol extract of Brachyglottis monroi, on the proliferation of human breast cancer MDA-MB231 cells was examined. Compound (1) at concentration as high as 16$\mu$/$m\ell$ has inhibited the proliferation of MDA-MB231 and this cytotoxic effect was increased in a time and dose-dependent manners. (omitted)

  • The cytotoxic activity of 13(E)-Labd-13-ene-8$\alpha$, 15-diol(1), isolated from the ethanol extract of Brachyglottis monroi was evaluated against tumor cell lines such as P388, SNU-C4 MDA-MB231, B 16 melanoma and A549 in vitro. By mean of spectral analysis particularly by the aid of various two dimensional NMR experiments, 1H-NMR and 13C-NMR signals of (1) was completely assigned, and thus the structure of (1) was established unambiguously.   • The effects of crude extracts from medicinal plants on biological activity were investigated. The crude ethanol extract of H. paucistipula inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (2 mm inhibition zone at 150 ㎍/disc) and the dermatophyte Trichophyton mentagrophytes ATCC 28185, (7 mm inhibition zone at 150 ㎍/disc), and toxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, (IC/sub 50/ 2.48 ㎍/㎖ at 75 ㎍/disc). This crude ethanol extract of H. paucistipula is the strongest antimicrobial and cytotoxic activities against P388 murine leukaemia cells ATCC CCL 46 P388D1.

Anticancer and Antimicrobial Activities of 13(E)-labd-13-ene-8α,15-diol from Brachyglottis monroi

  • In a previous study, we reported that 13(E)-labd-13-ene-$8{\alpha}$,15-diol (13E) possesses antiviral and anticancer activities. In this study, the anticancer and antimicrobial activities of 13(E) were evaluated against 4 cancer cell lines and 6 bacteria. 13(E) showed inhibitory effect on a variety of cancer cell lines. The $IC_{50}$ values was 8.3-21.3 ${\mu}g/mL$. 13(E) was the most effective growth inhibitor of murine leukaemia cell lines P388, producing approximately 8.3 ${\mu}g/mL$ of $IC_{50}$ in the cytopathic effect (CPE) method. 13(E) also inhibited the growth of the gram-positive bacteria (Staphylococcus aureus, Bacillus cereus and Listeria monocytogenes) and gram-negative bacteria (Vibrio parahaemolyticus, Escherichia coli and Salmonella enteritidis) with a range of minimum inhibitory concentration (MIC) values from 0.092 to 0.598 mg/mL and gram-negative bacteria were more sensitive to the compound (MIC, 0.092 mg/mL).
