Bibliographic study about the origin of Allii Macrostemi Bulbus (해백(薤白)의 기원에 관한 문헌적 고찰)

  • Objective: The aim of this study was to analyze the origin and characteristics of Allii Macrostemi Bulbus. Methods: Bibliographies on Allii Macrostemi Bulbus that focused on the origin of the material were examined. Based on this analysis, an inventory was prepared and the characteristics of the original material are presented. Results: Korean and foreign literature were reviewed to prepare an inventory, and Based on these results, the state of plants either currently cultivated or distributed as medicines, , and are summarized. 1. The origin of Allii Macrostemi Bulbusis the scaly stem of Allii macrozoon Bunge or Allii bakeri Regel. 2. The current white water mark in circulation is the scaly stem of the sandalae Allii macromoleon Bunge. 3. Scilla sinensis(Lour.) Merr. is. 4. The properties of Scilla sinensis(Lour.) Merr. are cool and bitter, healing the forests, mowing the ointment, stopping the oxen, and curing the absence of distractions and stuffy symptoms and removal of the placenta after childbirth. Conclusions: The origin of Allii Macrostemi Bulbusis the scaly stem of Allii macrozoon Bunge or Allii bakeri Regel.

Effects of Bulbus Allii Macrostemi Extract on PKC activity in Pulmonary Vascular Endothelial Cells Damaged by XO/HX (산소자유기에 의해 손상된 혈관내피세포의 PKC 활성도에 미치는 해백 추출물의 효과(I))

  • To investigate the protective effect of Bulbus Allii Macrostemi (BAM) on the damage by pulmonary vascular endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen free radical, Neutral Red (NR) and protein kinase c (PKC) activity assay were used. The results were obtained as follows ; The viability of vascular endothelial cells treated with XO/HX was decreased. And activation of PKC represented a maximal increase in group treated with XO/HX for 15 mins in vasvular pulmonary endothelial cells. But pretreated groups with BAM extracts were not inhibited the increase of PKC activation by XO/HX in a dose-dependent fashion. These results show that XO/HX elicits toxic effects in cultured pulmonary vascular endothelial cells, and suggest that BAM extract is very effective in the prevention of XO/HX-induced PKC activation.

Effects of Guaruhaebaekbanha-tang Extract on Beating Rate and LDH Activity in Cultured Rat Myocardial Cells (과루해백반하탕 추출물이 배양 심근세포의 박동수와 LDH 활성도에 미치는 영향)

  • To certify the protective effect of herbal medicine against oxygen free radical-induced myocardiotoxicity, cytotoxicity was measured using MTT, LDH activity and Beating rate assay in the presence of Guaruhaebaekbanha-tang(GHBT) extracts or single constituents of this prescription. Myocardial toxicity was evaluated in neonatal rat myocardiocytes in cultures. In the present study, xanthine oxidase/hypoxanthine(XO/HX) resulted in a decrease in cell viability, increases in LDH activity in culture medium and decreases in beating rate in cultured myocardial cells. In the effect of GHBT extract, it showed the prevention from the XO/HX-induced cardiotoxicity by the increases of cell viability and beating rate as well as the decrease of LDH activity. In the protective effect of Fructus Trichosanthis(FT), Bulbus Allii Macrostemi(BAM) and Rhizoma Pinelliae(RP), all the extracts were significantly effective in the protection of XO/HX-induced cardiotoxocity in cultured myocardial cells by the increase of beating rate as well as th decrease of LDH activity. From these results, they show that XO/HX is cardiotoxic in cultured myocardial cells derived from neonatal rat, and it suggests that GHBT, FT, SAM, RP extracts are positively effective in the blocking in XO/HX-induced cardiotoxicity.

In the effect of JHGT extract and its single constituents, which are Fructus Ponciri Seu Aurantii Immaturus (FPSAI), Cortex Magnoliae Officinalis (CMO), Bulbus Allii Macrostemi (BAM), Ramulus Cinnamomi (RC) and Fructus Trichosanthis (FT), they showed the prevention from the XO/HX-induced cardiotoxicity by the decrease of LDH activity and lipid peroxidation. From these results, they show that XO/HX is cardiotoxic in cultured myocardial cells derived from neonatal rat, and it suggests that JHGT, FPSAI, PT, CMO, BAM, RC and FT extracts are positively effective in the blocking in XO/HX-induced cardiotoxicity.

  • 80% extract of Gamigwaruhaebaekbaekju-Tang (GGHBT), Gagamgwaruhaebaekbaekju-Tang (GGGHBT) and Gamigwaruhaebaekwhanggum-Tang (GGHWT) remarkably showed inhibitory effects on HMG-CoA reductase, lipid peroxidation of rat liver and LDL oxidation, and DPPH free radical scavenging effect in a dose-dependent manner. Especially, GGHWT which is formulated with Trichosanthis Fructus, Pinelliae Tuber, Aurantii Immatures Fructus, Magnoliae Cortex, Allii Macrostemi Bulbus, Cinnamomi Ramulus and Scutellariae Radix on the basis of Gwaruhaebaekbaekiu-Tang listed on the traditional medicinal references showed more effective hypocholesterolemic activities in vitro bioassay than the other prescriptions.

Effect of Simple Formulas of Muscle Section in Donguibogam on Myogenic Regulatory Factors and IGF-1 Expression in C2C12 Cells

  • Simple formulas (單方) of muscle section in Donguibogam (東醫寶鑑) have long been prescribed for strengthening muscle and/or prevention of age-related muscle loss. However, biological activity and mechanisms by which they influence myoblast differentiation have not been studied. Therefore, in this study, we evaluated the effects of 14 simple formulas on myoblast differentiation in C2C12 myoblast cells under non-cytotoxic ($0.5mg/m{\ell}$) conditions. C2C12 cells were treated with water extracts of simple formulas for 72 h, and RT-PCR was performed to determine the gene expression levels of myogenic regulatory factors (MRFs), including myoD, myogenin, MRF4, myf5, and insulin like growth factor-1 (IGF-1). Treatment with Colocasiae Rhizoma (CR), Pini Semen (PS), and Sesami Semen (SS) resulted in a significant increase in expression of myogenin in C2C12 cells. Treatment with Allii Macrostemi Bulbus (AM), Colocasiae Rhizoma (CR), and Pini Semen (PS) also resulted in increased expression of MRF4 in C2C12 cells. In addition, enhanced expression of IGF-1 was observed in treatment with Eucommiae cortex (EC), Dioscoreae Rhizoma (DR), Colocasiae Rhizoma (CR), Pini Semen (PS), and Sesami Semen (SS) in C2C12 cells. These results indicate that simple formulas of muscle section in Donguibogam could potentially enhance myoblast differentiation at least in part via increasing expression of myogenin, and/or MRF4 and/or IGF-1.

A Review Study of Researches on Three Decoctions Including Fructus trichocanthis and Bulbus allii macrostemi in Geum-Gwe-Yo-Ryak (<금궤요략(金匱要略)> 내(內) 과루(瓜蔞)와 해백(薤白)이 포함된 3방제(方劑)에 대한 연구동향(硏究動向) 고찰(考察))

  • Objectives : The purpose of this study is to review the effect of Guaruhaebaekbaekju-tang (GHBJ), Guaruhaebaekbanha-tang (GHBH) and Jisilhaebaekgyeji-tang (JHGJ) by summarizing Korean and English articles. Methods : The study was based on 23 papers published since 1995. Papers were searched on seven domestic electronic databases including Koreantk, NDSL, RISS, KISS, OASIS, Koreamed and Library of Korea University and three international electronic databases including PUBMED, Embase and Science Direct. Results : 1. 23 research papers were collected, including 13 in vitro studies, 6 in vivo studies, 2 case reports and 2 literature studies. 2. GHBJ, GHBH and JHGJ showed significant myocardial protective and anti-hyperlipidemic and anti-inflammatory effects. In addition, researches on anti-oxidant, thrombosis, and atherosclerosis have been conducted in various ways. 3. It can be confirmed that these studies are related to Chest and back pain (胸背痛), Insomnia by chest paralysis (胸痺不得臥), Stabbing pain that radiates from chest to back (心痛徹背), Chest numbness (心中痞), Obstruction of qi in the chest (留氣結在胸), Fullness sensation in chest (胸滿) among the terms. Conclusion : Through these results, close association was found between classic description and modern research about GHBJ, GHBH and JHGJ, and the possibility of providing objective evidence was confirmed. To improve the quality of the study, large scale studies will be required to evaluate the authority of GHBJ, GHBH and JHGJ.
