Anti-stress and Anti-nociceptive Effects of the Aqueous Extracts of Chungsimyeonja-eum against Immobilization Stress and Incision Pain in Rats (청심포자음(淸心蒲子飮)의 구속 스트레스 및 절개 통증에 대한 항스트레스 작용과 진통효과)

  • The aqueous extract of Chungsimyeonja-eum has been used to treat palpitation, anemia, chronic fatigue, hypertension, and stroke. It also possesses various pharmacological effects including hypotensive, blood circulating, sedative, anti-inflammatory, diuretic, and anti-stress activities. In the present results, the latency time of plantar test was decreased in the immobilization stress and incusion pain-induction group. However the paw withdrawal latency values were increased in the immobilization stress and incision pain-induction groups after treatment with the aqueous extract of Chungsimyeonja-eum. The c-Fos expression in the paraventricular nucleus (PVN), and 5-hydroxytryptamine (5-HT) and tryptophan hydroxylase (TPH) in the dorsal raphe were enhanced in the immobilization stress and incision pain-induction groups. However, the treatment with the aqueous extract of Chungsimyeonja-eum suppressed the immobilization stress and incision pain-induced increase of c-Fos, 5-HT, and TPH expressions. Here in this study, we have demonstrated the protective effects of Chungsimyeonja-eum on immobilization and incision pain-induced stress. The present study revealed that Chungsimyeonja-eum treatment diminishes immobilization and pain stress.

Protective Effects of Chungsimyeonja-eum on Glutamate-induced Apoptosis in C6 Glial Cells (Glutamate로 유도된 C6 glial 세포 자멸사에 대한 청심연자음(淸心蓮子飮)의 보호효과)

  • Objective : The water extract of Chungsimyeonja-eum (CSYJE) has traditionally been used in treatments of heart diseases and brain diseases in Oriental medicine. However, little is known about the mechanism by which CSYJE protects neuronal cells from injury damages. Therefore, in this study we attempted to elucidate the mechanism of the cytoprotective effect of the CSYJE extract on glutamate-induced C6 glial cell death. Methods : Cultured cells were pretreated with CSYJE and exposed to glutamate, cell damage was assessed by using MTT assay and propidium iodide (PI), probe 2',7'-dichlorofluorescein diacetate (DCF-DA) staining. Western blotting was performed using anti-procaspase-3 and anti-PARP, respectively. Result : We determined the elevated cell viability by CSYJE extract on glutamate-induced C6 glial cell death. Glutamate induced DNA fragmentation on C6 glial cells but pre-treatment with CSYJE inhibited DNA fragmentation. One of the main mediators of glutamate-induced cytotoxicity was known to generation of reactive oxygen species (ROS). Pre-treatment with CSYJE inhibited this ROS generation from glutamate-stimulated C6 glial cells. Also, we identified that the ROS-induced DCF-DA green fluorescence was reduced by CSYJE pre-treatment. The critical markers of apoptotic cell death are the cleavages of procaspase-3 protease and PARP proteins, so we checked the expression level and cleavages of procaspase-3 protease and PARP proteins. Glutamate-treated C6 glial cells showed the cleavages of procaspase-3 protease and PARP proteins and followed the reduction of expression of these proteins. Conclusion : These findings indicate that CSYJE may prevent cell death from glutamate-induced C6 glial cell death by inhibiting the ROS generation and procaspase-3 and PARP expression.

A Study on Change of Marker Compounds and Biological Activity in Chungsimyeonja-eum Decoction Depending on A Storage Temperature and Periods (보관 온도 및 기간에 따른 청심연자음 전탕액의 지표성분과 약리 활성 변화에 관한 연구)

  • Objectives : Chungsimyeonja-eum (CSYJE; Qingxinlianzi-yin in Chinese; Seishinrenshi-in in Japanese), a traditional herbal medicine, has been used for treating mouth dryness, hyperuresis. This study was designed to determine preservation period of CSYJE. We investigated the stability and biological activity of CSYJE depending on the preservation temperature and periods. Methods : CSYJE decoction was preserved for 0-6 or 12 months at room temperature (RT, $23{\pm}1^{\circ}C$) or refrigeration ($4^{\circ}C$). To evaluate the stability of CSYJE decoction, pH and dissolved solids content were measured. High-performance liquid chromatography (HPLC) analysis was performed to determine marker compounds-liquiritin apioside, liquiritin, baicalin, wogonoside, and glycyrrhizin-in CSYJE. To determine anti-inflammatory effect of CSYJE, prostaglandin $E_2$ ($PGE_2$) was measured in LPS-stimulated RAW 264.7 macrophages. Results : Both at RT and $4^{\circ}C$, pH was decreased depending on the preservation periods. There was no changes in dissolved solids content depending on the preservation temperature and periods. Both at RT and $4^{\circ}C$, the contents of liquiritin apioside and liquiritin were slightly increased at 1 month of storage. The level of baicalin was decreased time-dependently and the disappearance rate at RT is larger than at $4^{\circ}C$. CSYJE inhibited $PGE_2$ production in LPS-stimulated RAW 264.7 cells and maintained inhibitory effect by 12 months both at RT and $4^{\circ}C$. Conclusions : Based on the disappearance rate of the baicalin in CSYJE, the preservation period is recommended within 8 months for RT and 12 months for $4^{\circ}C$.

A Case of Conversion Disorder with Blackout (일시적 혼수를 나타낸 전환장애 1례)

  • Objectives : This case report presents a 68-year-old female patient. Her chief complaint was fatigue and depressed mood. During the therapeutic period, she experienced a sudden onset of unconsciousness. After two days, she became alert. We diagnosed her with conversion disorder. Methods : We treated her with Korean traditional herbal medicine (Chungsimyeonja-eum, Soum-In Bojungyikgi-tang, Hyeongbangjiwhang-tang) and oriental psychotherapy for 21 days. The effects of treatment were measured by Beck Depression Inventory (BDI), Zung Depression Rating Scale (SDS), and State-Trait Anxiety Inventory (STAI-X-1). Results : After treatment, the chief complaint and accompanying symptoms have been subsided and improved. Further, BDI, SDS, STAI-X-1 score were decreased. Conclusions : In this case, we recognized that Korean traditional herbal medicine and oriental psychotherapy could be effective for clinical symptoms of conversion disorder.
