  • Two flavone glycosides have been obtained from the aerial part of Cirsium japonicum var. ussuriense Kitamura(Co,positae) in Korea and identified by means of spectral analysis as linarin and cirsimarin. When 10-2mg/ml of cirsimas was added, lipid peroxide formation in the rat liver decreased by 12% compare to control.

  • Cirsium japonicum var. ussuriense (CJ) is used as a folk medicine for diuretic, arthritis, dyspepsia, and bleeding in Korea. Until now, more than fifteen kinds of CJ were identified and among them, Cirsium japonicum var. nakaianum Nakai was selected as experimental material. Both that from The Medical Herb Garden of Seoul National University located at Gayang-dong, Koyang city and native that from a hillock near Dangjin, Chungnam were collected and analyzed to determine whether it is varied in efficacy on their habitats. Extracts and fractions of CJ had a strong effect of antibacterial activity on H. pylori and antioxidant effects. And the CJ from Garden and Dangjin were showed to be more efficient in acute gastritis through the HCl${\cdot}$ethanol-induced gastric lesion test and chronic gastritis through the indomethacin-induced gastric lesion test, respectively. This study proved that the effects of CJ are varied by their habitats. Also, these results suggest that the CJ has potentials for use as functional food and medicine.

Anti-Obesity Effects of Water Extracts from Different Organs of Cirsium japonicum var. ussuriense (엉겅퀴 부위별 열수 추출물의 항비만 효과)

  • Cirsium japonicum var. ussuriense are often used in treatment of human diseases such as hemorrhage, blood congestion and inflammation in Korea. However, there was not report on the anti-obesity efficacy of water extracts from different organs of C. japonicum var. ussuriense. Here, we investigated the antioxidant effects of water extracts of flowers, leaves and roots from C. japonicum var. ussuriense. Total polyphenol amounts of the flower extract showed higher than those of leaf and root extract. Flower extract also showed the high antioxidant activities such as DPPH radical scavenging activity and reducing power. We also investigated the anti-obesity effects of water extracts of flowers, leaves and roots from C. japonicum var. ussuriense in 3T3-L1 cells and high fat diet-induced obesity mice. The mice were divided into four groups [high fat diet (HFD) control, HFD + leaf extract, HFD + flower extract and root extract] and administered with each extract (200 mg/kg) for 12 weeks. The flower and leaf extract significantly suppressed the levels of oil red O and triglyceride content. The flower and leaf extract also significantly reduced the triglyceride, total cholesterol and lower density lipoprotein levels of plasma as well as body and abdominal fat weight. Furthermore, oral glucose tolerance in the flower and leaf extract groups were significantly ameliorated in comparison to the high fat diet group. Therefore, these results indicate that the flower and leaf extract could ameliorate obesity and attenuate blood glucose level in high fat diet-induced obesity mice. We conclude that this study may provide positive insights into water extracts of flowers and leaves from C. japonicum var. ussuriense as a functional food ingredient for treatment of obesity.

  • Cirsium japonicum var. ussuriense is often used in treatment of human disease such as hemorrhage, blood congestion and inflammation. This study was accomplished to evaluate the antioxidant properties of the leaf (CLE) and flower (CFE) extracts of C. japonicum var. ussuriense to protect normal human red blood cells (RBC) and plasma samples against oxidative damage in vitro. CLE and CFE were prepared by extracting with hot water. In red blood cells and plasma, oxidative hemolysis and lipid peroxidation induced by the aqueous peroxyl radical generator [2,2'-azobis (2-amidinopropane) dihydrochloride, AAPH] were significantly suppressed by CLE or CFE in a dose-dependent manner at the same time. CLE and CFE also prevented the depletion of cytosolic antioxidant glutathione (GSH) in RBC. These results suggest that the leaves and flowers of C. japonicum var. ussuriense may have the antioxidant properties.

  • Cirsium japonicum var. ussuriense is known to have a variety of biological activities, including anti-inflammatory, analgesic activity and antipyretic activity. In this study we investigated the role of polyacetylene compound, 1-Heptadecene-11, 13-diyne-8, 9, 10-triol (PA) from the root of Cirsium japonicum var. ussuriense as an immune-modulator. PA was evaluated as inhibitors of some macrophage functions involved in the inflammatory process. We tested the effect of PA on the production of pro-inflammatory cytokines, interleukin-1beta (IL-$1{\beta}$) and tumor necrosis factor-alpha (TNF-$\alpha$), and nitric oxide (NO) in murine macrophage cell line, RAW264.7. There was no effect on cytokine production of macrophages by PA itself. However, PA inhibited lipopolysaccharide (LPS)-induced IL-$1{\beta}$ and TNF-$\alpha$ production by macrophages at a dose dependent manner. PA also suppressed the NO production of macrophages by LPS. LPS-induced NF-${\kappa}B$ activity was decreased by treatment of PA. Therefore, these results suggest that PA has anti-inflammatory effect by inhibiting the NF-${\kappa}B$ activation.

  • Cirsium japonicum var. ussuriense is often used in treatment of human disease such as hemorrhage, blood congestion and inflammation. The present study has been undertaken to investigate the effect of the leaf extract from C. japonicum var. ussuriense (CLE) on the development of collagen-induced arthritis (CIA) in DBA1/J mice. CLE administration suppressed markedly the arthritis incidence and arthritis score in CIA mice. Also, CLE significantly suppressed the release of $PGE_2$, TNF-${\alpha}$, IL-4 and IL-6 in CIA mice. However, CLE significantly increased the production of IL-10, but not IL-4. These results suggest that CLE suppress inflammatory mediators and regulates Th1 and Th2 cytokines. These properties may contribute to the anti-arthritis action of CLE.

  • Objective: The roots, leaves, flowers, stems and seeds of Cirsium japonicum var. ussuriense are often used in treatment of human diseases such as hemorrhage, blood congestion and inflammation. Focusing our attention on natural and bioavailable sources of antioxidants and anti-inflammation, we undertook to investigate the antioxidant and anti-inflammatory properties of Cirsium japonicum var. ussuriense used as a folk medicine in Korea. Methods: The extracts of the leaves, stems, flowers, seeds and roots from C. japonicum var. ussuriense were prepared by extracting with water or 80% ethanol. Total flavonoids and polyphenols were measured by a colorimetric assay. The free radical scavenging activity of the extract was analyzed by the DPPH (1,1-diphenyl-2-picryl hydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and Griess reagent assay. An oxidative product of nitric oxide (NO), was measured in the culture medium by the Griess reaction. The level of prostaglandin $E_2$ ($PGE_2$) was measured by enzyme-linked immunosorbent assay. The expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were measured by Western blot analysis. Results: Total flavonoid and polyphenol amounts of the leaves (CLE) and flowers (CFE) showed higher than those of the seed extract (CSE), stem extract (CSTE) and roots (CRE). CLE and CFE also showed the high antioxidant activities such as DPPH, NO-like and ABTS radical scavenging activity. An antioxidant activities of these water extracts showed higher than those of 80% ethanol extracts. We investigated the anti-inflammatory effects of CLE on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. CLE significantly suppressed the levels of the inflammatory mediators such as NO and prostaglandin $E_2$ ($PGE_2$) in dose dependant. Furthermore, the levels of iNOS and COX-2 protein expressions were markedly suppressed by the treatment with CLE extract in a dose dependent manner. Conclusions: These results suggest that CLE water extract has a higher anoxidant and anti-inflammatory activity, these properties may contribute to the oxidative and inflammatory related disease care.

Effects of Cirsium japonicum var. ussuriense Extract on Tumor Immunity

  • Objective : The present study is aimed to elucidate the effects of Cirsium japonicum var. ussuriense on immunomodulation and the potential as an herbal remedy for cancer treatment. Method : It was performed through measurement of effects Cirsium japonicum var. ussuriense extract (CJE) on NO production, NK cell cytotoxicity and cytokine gene expressions related with macrophage and NK cell activity. Result : 1. CJE did not show any direct cytotoxic effects on 7250, HT1080, Hep G2 and CT-26 cells. 2. CJE activated macrophages partially to product NO and up-regulated gene expressions for iNOS in RAW 264.7 cells. 3. CJE promoted cytotoxicity of NK cells against YAC-1 cells at higher concentration than 200 ${\mu}g/ml$. 4. CJE up-regulated gene expressions for $IL-1{\beta}$, IL-2, iNOS, $IFN-{\gamma}$ and $TNF-{\alpha}$ in mice splenocytes. 5. CJE inhibited lung tumor metastasis induced by CT-26 cell transplantation compared with the control group. Conclusion : It could be concluded that CJE is an effective herbal drug for immune modulating and anti-cancer treatment by promoting activity of macrophages and NK cells.

  • The present study has been undertaken to investigate the effect of the extract of Cirsium japonicum var. ussuriense leaves (CLE) on blood circulation in a rat model of topical ferric chloride ($FeCl_3$)-induced carotid artery damage. $FeCl_3$ treatment seriously damaged the carotid artery such as the walls of the artery, blood flow and inflammation. However, CLE administration has ameliorated blood circulation and suppressed vessel inflammation. CLE administration also has ameliorated the $FeCl_3$-induced artery tissue damage. Furthermore, CLE significantly suppressed the expression of adhesion molecules. These results suggest that CLE ameliorate blood circulation through suppress inflammatory mediator and adhesion molecule production.

Antioxidant and Anticancer Activities of Methanol and Water Extracts from Leaves of Cirsium japonicum

  • Potential antioxidant activities of methanol and water extracts of Cirsium japonicum var. ussuriense (CJ) leaves were examined. The reducing power and hydroxyl raical-scavenging activity assays showed that the methanol extract had a significantly higher activity than the water extract. In addition, the methanol extract showed a concentration-dependent reducing power, ranging from 0.228 to 1.072($0.1{\sim}0.5\;mg/mL$), as well as a high DPPH free radical-scavenging activity($EC_{50}=40.25\;{\mu}/mL$). The total phenolic(as tannic acid) and flavonoid(as quercetin) contents of the extract were 62.41 mg/g and 13.48 mg/g, respectively. The cytotoxic activity indicated that the methanol extract has an inhibition activity in the stomach carcinoma cell (35.40%), suggesting that the methanol extract of CJ leaves could be used as a potential source of pharmaceutical material.
