Antimicrobial Effect of Citrus unshiu Markovich Extracts on Food-Borne Pathogens (청피 추출물이 식중독 유발 미생물의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effects of the Citrus unshiu Markovich extracts against food-borne pathogens. First, the Citrus unshiu Markovich was extracted with methanol at room temperature, and fractionation of the methanol extracts from Citrus unshiu Markovich was carried out by using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Citrus unshiu Markovich extracts was determined using the paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Citrus unshiu Markovich showed the highest antimicrobial activity against Bacillus cereus and Shigella sonnei. A synergistic effect was found in combined extracts of Citrus unshiu Markovich and Hedyotis diffusa Willd as compared to each extract alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Citrus unshiu Markovich against Bacillus cereus and Shigella sonnei. The ethyl acetate extract of Citrus unshiu Markovich showed strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. The 5,000 ppm of ethyl acetate extract from Citrus unshiu Markovich retarded the growth of Bacillus cereus more than 24 hours and Shigella sonnei up to 24 hours. The ethyl acetate extracts of Citrus unshiu Markovich have shown an antimicrobial effect against Bacillus cereus and Shigella sonnei.

Evaluation on antioxidative and antiaging effect of immature fruits from Citrus unshiu Markovich (미숙감귤의 항산화 및 항노화 활성에 대한 평가)

  • Objective : This study was designed to investigate effect of immature fruits from Citrus unshiu Markovich to mature them on antioxidative and antiaging-related enzyme activities in vitro. Methods : Fruits from Open field-cultivated C. unshiu Markovich (ripe and unripe fruits of Citri) made a purchase in Jeju island. We measured total polyphenol and flavonoid contents in mature C. unshiu Markovich (MC) and immature C. unshiu Markovich (IMC) 70% ethanol extract. DPPH free radical, ABTS radical, and hydroxyl radical scavenging activities in both extracts were determined. Also, reducing power, SOD-like activities, elastase inhibition, and collagenase inhibition activities of these extract were assayed at 5, 1, 0.5, and 0.1 mg/㎖. Results : The total polyphenol contents in the ethanol extract of IMC were higher than those of MC (26.99 ± 0.89 mg/TAEg). And total flavonoid contents in them were very similar to both extract (MC; 7.25 ± 0.4 mg/RUEg and IMC; 7.44 ± 0.18 mg/RUEg). DPPH free radical scavenging activities, ABTS radical scavenging capacity, reducing power, and SOD-like activities of IMC-treated group showed significant increase compared to those of MC group in all treated concentrations. Particularly, DPPH free radical scavenging and elastase inhibition activities in IMC-treated group at 5 mg/㎖ showed similar with positive control group. Besides, IMC extract at 5 mg/㎖ concentration surpassed positive control (BHA or Vit. C) in SOD-like activities and ABTS radical scavenging capacity. Conclusion : These results suggest that IMC ethanol extract may be used as a natural antioxidant and a antiaging material for development of functional foods and cosmeceuticals.

Changes in the Constituents and Antioxidant Activity in Accordance with the Processing Conditions of Citrus unshiu Markovich (진피의 수치 조건에 따른 성분 및 항산화 활성 변화)

  • Processing of medicinal plants is one of the processing methods for reducing of toxicity or improving of effect on medicinal plants. In this study, we studied the changes in the constituents and antioxidant activity in accordance with the processing conditions by the salt water of Citrus unshiu Markovich. Changes in antioxidant activity was measured by the DPPH method and changes in the components were analyzed by high performance liquid chromatography. As a result, the content of the main constituents (narirutin, hesperidin) and total polyphenol were increased by increasing the concentration of salt water. Moreover, antioxidant activity was increased gradually in proportion to the increase of the total polyphenol content.

  • [ ${\beta}$-Cryptoxanthin ] contents were determined from Citrus unshiu Markovich fruits grown in a greenhouse and open field of Jeju Island, off the southern coast of Korea. In a greenhouse and open field, the ${\beta}$-cryptoxanthin content in the peel was greatly increased by harvesting citrus fruits in the late season from August through November. However, ${\beta}$-cryptoxanthin content in the flesh was gradually increased and was superior to that of the citrus fruits grown in a greenhouse. ${\beta}$-Cryptoxanthin was efficiently purified from the flesh of citrus fruits harvested in the late harvesting season in November. The ${\beta}$-cryptoxanthin contents in the peel and flesh of citrus fruits harvested from a greenhouse in November were 0.89 mg% and 0.35 mg%, respectively, and in that obtained from an open field were 1.12 mg% and 0.35 mg%, respectively.

Search for Plant-originated Antibacterial Compounds Against Pathogen (Acidovorax avenae subsp. citrulli) of Watermelon Bacterial Fruit Blotch (수박 과실썩음병 병원균(Acidovorax avenae subsp. citrulli)에 대한 식물유래 항균 활성물질 탐색)

  • 133 Species of medicinal plants were used for the development of natural agrichemicals with anti-microbial activity against Acidovorax avenae subsp. citrulli, a pathogen of bacterial fruit blotch in watermelon. The MeOH-extracts of these medicinal plants were examined for anti-microbial activity by bioassay. The MeOH-extract of Citrus unshiu Markovich had the strongest antibacterial activity against Acidovorax avenae subsp. citrulli. To identify anti-microbial compounds from Citrus unshiu Markovich, solvent-fractionation was used. The fraction of hexane, which showing the highest value of anti-microbial activity, was analyzed by GC-MS. Each mass spectra, corresponding to each peak of chromatogram, was compared to mass database of Wiley library. As a result, d-Limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, terpineol, palmitic acid, 9,12-octadecadienoic acid, Linolenic acid, and stigmasterol were identified. Among them, d-Limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, and terpineol confirmed to be shown the anti-microbial activity by bioassay. Especially, d-Limonene and ${\gamma}$-terpinene found to have strong activity. In conclusion, we thought d-limonene and ${\gamma}$-terpinene from Citrus unshiu Markovich. Latin, had anti-microbial activity against Acidovorax avenae subsp. citrulli and could be candidates for the control agents for the control of bacterial fruit blotch in watermelon.

Anti-Toxoplasmosis Effect of Citrus Unshiu Markovich against Toxoplasma Gondii (한약제 진피의 항 톡소포자충 효과 확인)

  • Toxoplasma gondiiis a widespread apicomplexan parasite which is able to infect virtually all warm-blooded vertebrates. Twenty-two percent of the U.S. population is infected, but severe disease in adults is mainly limited to immunosuppressed patients. In patients with acquired immunodeficiency syndrome(AIDS), T. gondii causes a life-threatening opportunistic infection, with Toxoplasma encephalitis as its most severe manifestations. T. gondii is also known to cause congenital infection and is among the pathogens with the highest incidence of complications in pregnancies. Despite its clinical importance, only very few therapeutic drugs against T. gondii are available, all of which target the rapidly dividing tachyzoites, leaving the dormant encysted bradyzoite stage unaffected. We searched 15 traditional medicines that have anti-inflammatory effect from dongyibogam and Traditional Chinese medicine. In vitro studies were performed with HeLa cell cultures, with quantification of Toxoplasma growth by a cell proliferation assay. The result of experiment shows the selectivity of Citrus unshiu Markovich is 6.0. This is higher than sulfadiazine (selectivity was 1.63). For in vivo studies, mice were acutely infected intraperitoneally with $10^5$ tachyzoites of the virulent RH strain and then treated per orally for 4 days from 6 hours postinfection. Efficacy was assessed by sequential determination of parasite burdens in peritoneal cavity. In vivo, Citrus unshiu Markoviche inhibited Toxoplasma growth at a concentration of 10㎎/㎏ of body weight per day, the inhibition ratio was estimated to be 64.01%.

Evaluation of Anti-inflammatory and Antioxidant Abilities of Complex Extracts Produced from Leonurus japonicus Houtt., Houttuynia cordata Thunberg, and Citrus unshiu Markovich (익모초, 어성초, 진피 복합 추출물의 항염증 및 항산화 평가)

  • This study aimed to confirm the potential as a material for food, medicine, and cosmetics by measuring the physiological activity of the complex extract(Leonurus japonicus Houtt., Houttuynia cordata Thunberg and Citrus unshiu Markovich). The complex extract was with hot water, The evaluation of cytotoxicity, antioxidant, anti-inflammatory, and MAPKs of the extract was performed by using ELISA and western blot. Total polyphenols and flavonoid contents of complex extracts were 126.16 mg/g and 105.84 mg/g, respectively. The scavenging activities of DPPH and ABTS radical significantly increased in a dose-dependent in complex extracts. Compared to the control group, the complex extracts treatments significantly reduced concentrations of nitric oxide and inflammatory cytokine (i.e., IL-1β, TNF-α, and IL-6). Furthermore, the complex extracts treatments significantly reduced protein expression levels of MAPKs (i.e., ERK, JNK, and p38). The results indicate that the complex extract can be used for oxidative damage and inflammation. Thus, the results of this study can be used as basic data for composite materials for food, medicine, and cosmetics.

New Record of Paratrichodorus minor (Colbran, 1956) Siddiqi, 1974 and Redescription of P. porosus (Allen, 1957) Siddiqi, 1974 (Triplonchida: Trichodoridae) from Citrus unshiu Markovich in Korea

  • During the survey of plant parasitic nematode of Citrus orchard in Korea, Paratrichodorus minor (Colbran, 1956) Siddiqi is founded from the soil of Citrus unshiu Markovich for the first time in Korea. The body length of females is $428.34{\sim}506.22{\mu}m$ long: a = $12.08{\sim}22.79$, b = $4.02{\sim}5.02$, V(%) = $38.46{\sim}60.68$. The excretory pore is located near the base of esophagus. Additionally, the morphological characteristics on P. porosus (Allen, 1957) Siddiqi, 1974 recorded in 1976 on Korea is redescribed in detail.

Potentials of Chenpi on Metabolic Syndrome: A Review

  • Objectives: Metabolic Syndrome (MetS) is strongly related with central obesity, hypertriglyceridemia, low high-density lipoprotein-cholesterol (HDL-C), hyperglycemia, and hypertension. This study reviewed the potential of Chenpi in treatment of MetS through amelioration of co-related diseases, such as diabetes mellitus, hyperlipidemia, obesity, hepatic steatosis, and inflammation. Methods: Six electronic databases (Oriental Medicine Advanced Searching Integrated System (OASIS), Korean Traditional Knowledge Portal, Korea Institute of Oriental Medicine (KIOM), Research Information Sharing Service (RISS), PubMed, and Embase) were used to search for in vitro, in vivo, and clinical research that discusses the potential effects of Chenpi (Citrus unshiu Markovich, Citrus reticulata Blanco) on diabetes mellitus, hyperlipidemia, obesity, hepatic steatosis, and inflammation. Results: This review suggests the potential of Chenpi as a candidate for the treatment of metabolic syndrome through improvement of co-related diabetes, hyperlipidemia, obesity, hepatic steatosis, and inflammation. However, comparison of the results of each study was limited by a lack of quantification of the experimental materials.

Anticancer activity of chloroform extract of Citrus unshiu Markovich peel against glioblastoma stem cells (교모세포종 암줄기세포에 대한 진피 소수성 추출물의 항암 활성)

  • Glioblastoma is the most common primary malignant brain tumor and has an extremely poor prognosis. Glioblastoma stem cells (GSCs) contribute to tumor initiation, recurrence, and resistance to therapy, and are thus a key therapeutic target. The peel of Citrus unshiu Markovich has been used in traditional medicine in East Asia to treat various diseases. In this study, we investigated the anticancer activity and molecular mechanism of the chloroform extract of this natural product (CECU) in U87MG GSCs. The results show that CECU inhibited the proliferation, tumorsphere formation, and migration of U87MG GSCs by causing cell cycle arrest at the G0/G1 phase and apoptosis. In addition, CECU downregulated key cancer stemness regulators, including CD133, Oct4, Nanog, integrin α6, ALDH1A1, and STAT3 signaling in U87MG GSCs. Furthermore, CECU significantly suppressed in vivo tumor growth of U87MG GSCs in a chorioallantoic membrane model. Therefore, CECU can be utilized as a natural medicine for the prevention and treatment of glioblastoma.
