Cytochemical and Biochemical Characteristics of Cellular Adhesion in Amoeba proteus (Amoeba proteus의 표면흡착에 관한 세포화학 및 생화학적 특성)

  • The effects of proteases, neuraminidase and EDTA on adhesion of amoebae on the substratum, ultrastructure and biochemical composition of the cell surface were studied by concanavalin A (con A) cytochemistry and SDS PAGE. By con A cytochemistry the glycocalyx of the plasmalemma was easily subdivided into outer filamentous (F) layer and the inner amorphous (A) layer. On treatment with neuraminidase, amoebae attached to the substratum and spreaded better than untreated cells exposing the more con A binding sites in A- and F-layer. When the cells were treated with trypsin or proteinase K, cells stayed unattached for 12 and 48 hr, respectively. Con A binding sites of A layer and all of those glycoproteins were removed by proteinase K. On the other hand, trypsin damaged all of the con A binding sites in both A- and F-layer without significant change in PAS-stained profile of the plasmalemma. Some of the mucopolysaccharides of the cell surface were released by these enzymes and EDTA. When the cells were incubated with monovalent con A they did not attch on the substratum and cytolysed. From these results adhesion of amoebae on the substratum appears to be mediated by the interaction of the glycoproteins and mucopolysaccharides of the A layer.

Concanavalin A Mediated Calcium Changes on Expansion and Hatching of the Mouse Blastocyst (Concanavalin A를 매개로한 세포내 Calcium의 변화와 생쥐 포배의 팽창과 부화)

  • Objective: The oligosaccharide moieties of glycoproteins and proteoglycans have a vital function in blastocyst differentiation. Concanavalin (ConA), a lectin, is known to bind on the preimplantation embryos, especially on blastocyst. In this study, we investigated whether ConA can modulate the trophoblast development and about the regulating mediator. Also, we investigated whether expansion is enough for hatching procession of the mouse blastocyst. Method: Embryos were collected at 72 h post hCG injection and chemicals were treated after 24 h (96 hr post hCG injection). ConA or calcium ionophore A23187 were exposed to blastocyst and than analysis the developmental process for 48 hr. Intracellular free-$Ca^{2+}$ concentration in trophectoderm was measured with confocal laser microscope after exposing to ConA or calcium ionophore A23187. ConA-pretreated blastocyst exposed to the calcium ionophore A23187 and then analyzed the developmental process. Otherwise ouabain was treated to the blastocyst to block the $Na^+/K^+$-ATPase activity. Results: In contrast to the control blastocyst, the ConA-exposed blastocysts developed beyond the expansion stage with significantly high rate (90.4%) at 12 h post administration. ConA induced an increase the intracellular $Ca^{2+}$ concentration in trophectoderm. Calcium ionophore A23187 also stimulated expansion of blastocyst. Most of the control blastocysts developed to the hatching stage at 144 h post hCG injection. However, strongly 65% of the ConA-exposed embryos were arrested at expanded stage at same time point. The developmental progression rates to hatching stage of both ConA- and calcium ionophore A23187-expose blastocysts were significantly lower than that of the control. However ConA-pretreated embryos developed to the hatching stage like control embryos. Ouabain showed a tendency to delayed the progress to expansion stage but did not inhibit the development to the hatching stage. Conclusion: ConA-mediated expansion is the result of the increase of intracellular free-calcium in blastocyst stage embryo. It is suspected that expansion of the blasocyst is a essential indirect factor in hatching and the calcium may triggering the cellular mechanisms for the both expansion and hatching progression.

Effects of Low Crude Protein Diets Supplemented with Synthetic Amino Acids on Performance, Nutrient Utilization and Carcass Characteristics in Finishing Pigs Reared Using a Phase Feeding Regimen

  • This experiment was conducted to investigate the effect of feeding a low CP diet supplemented with synthetic amino acids on performance, nutrient utilization and carcass characteristics of finishing pigs fed under a three-phase feeding regimen. Ninety-six finishing pigs (Landrace$\times$Large White$\times$Duroc), $55.75kg{\pm}0.65$ of initial body weight, were blocked by weight and sex and allotted to four dietary treatments in a randomized block design. There were six pens per treatment and four pigs per pen. Pigs were fed a 16%-14%-12% CP (for phase I-II-III, respectively), sequence of diets. Dietary treatments were 1) Control, 2) Con+L (a sequence of diets reduced in CP by l percentage unit with lysine (L) supplementation, 3) Con+LMT (a sequence of diets reduced in CP by 2 percentage unit with LYS, methionine (MET) and threonine (THE) supplementation) and 4) Con+LMTT (a sequence of diets reduced in CP by 3 percentage unit with LYS, MET, THR and tryptophan (TRP) supplementation). The finishing period (55 to 105 kg) was divided into three phases (55 to 72 kg, 72 to 90 kg and 90 to 105 kg). Pigs fed either the control or Con+L diet grew faster (p<0.05) than pigs fed the Con-LMT or Con+LMTT diet. There was no difference in ADFI among dietary treatments. Phosphorus (P) digestibility was lowest in the control group and highest in the Con+LMTT group (p<0.05). Within each phase, no significant differences in dry matter (DM) and CP digestibilities were found. Although some amino acid digestibilities were affected by dietary treatments, digestibilities of essential amino acids (EAA), non-essential amino acids (NEAA) and total amino acid were not significantly influenced by dietary treatments. For the entire experiment periods, Con+L, Con+LMT and Con+LMTT treatments resulted in 13.4, 18.8 and 21.6% lower total N excretion compared with the control. Con+LMT and Con+LMTT treatments showed significantly lower BUN concentration compared with the control and Con+L treatment (p<0.05), but there was no significant difference in BUN concentration between pigs fed the control and Con+L treatment or between pigs fed Con+LMT and Con+LMTT treatments (p>0.05). Carcass length, backfat thickness and carcass grade were not significantly affected by dietary treatments (p>0.05). In conclusion, reducing dietary CP level by 1 percentage unit and supplementing only LYS at each phase could be a very beneficial feeding strategy for finishing pigs fed under a three phase feeding regimen in terms of both environmental and economical aspects.

The Effect of the Curcumae Longae Rhizoma (CLR) Extract on the Hepatocellular Carcinogenesis and Acute Liver Damage Induced by Diethylnitrosamine (DENA) and CCl4 in Rats (강황추출물이 Diethylnitrosamine과 CCl4로 유발된 흰쥐의 간암과 간 손상에 미치는 영향)

  • Objective : In order to investigate the effect of Curcumae Longae Rhizoma(CLR) extract on the hepatocellular carcinogenesis and acute liver damage induced by diethylnitrosamine(DENA) and $CCl_4$ in rats. Methods : Experimental groups were subdivided into four; normal group (Nor), acute liver damage and hepatocellular cancer inducing control group (Con), and CLR extract 200mg/kg/day (CAA) or 400mg/kg/day (CAB) administered groups to Con. Thereafter the changes of the body weight, the liver weight and the weight of liver/100g body weight, total cholesterol, HDL cholesterol, triglyceride, the activities of AST, ALT, ALP, LDH, AFP, SOD, catalase were measured. And we observed by optical and electron microscopy. Result : 1. The body weight was decreased in Con compared with Nor for 5 weeks, but increased in Con compared with Nor from 6 week to 9 week. During experimental period of total 9 weeks, CAA and CAB were increased compared with Con. 2. The liver weight was increased significantly (p<0.05) in Con compared with Nor. The weight of liver/100g body weight was increased significantly (p<0.05) in Con compared with Nor and decreased significantly (p<0.05) in CAB compared with Con. 3. The level of total cholesterol was increased in Con and CAA compared with Nor, but there was not statistically significant. The level of triglyceride was decreased in Con compared with Nor. But increased in CAA and CAB compared with Con. The level of HDL-cholesterol was significantly increased (p<0.05) in CAA and CAB compared with Con. 4. The activities of AST, ALT were increased in Con compared with Nor, but decreased in CAA compared with Con, significantly decreased (p<0.05) in CAB compared with Con. 5. The activities of ALP, LDH were increased in Con compared with Nor, but decreased in CAA and CAB compared with Con. 6. The activities of AFP was increased significantly (p<0.05) in Con compared with Nor, but decreased significantly (p<0.05) in CAA and CAB compared with Con. 7. The activities of SOD were increased in Con, CAA and CAB compared with Nor, but decreased in CAA and CAB compared with Con. The activities of Catalase was more increased in CAA and CAB compared than Con. 8. The results of light microscopical observation, a number of hepatocytes were damaged in Con compared with Nor and CAB. 9. According to the electron microscopical observation, irregular nuclear membrane, condensed nucleoplasm was observed in Con, the experimental group was observed in the nucleus of the well-preserved and evenly developed nucleoplasm. Conclusions : These results suggest that administration of CLR extract suppress or retard on the hepatocellular carcinogenesis and acute liver damage induced by DENA and $CCl_4$ in rats.

Effect of Betaine on Immune Response in Laying Hens (비태인이 산란계의 면역 반응에 미치는 영향)

  • This experiment was conducted to determine the effect of betaine on immune response in laying hens. A total of 72 ISA-brown laying hens were divided into four groups of 18 hens each and fed corn-soybean meal based diets with addition of 0, 300, 600 and 1,200 ppm betaine for four weeks. The effect of betaine on splenocyte proliferations with mitogens, concanavalin A(Con A) and pokeweed mitogen(PWM), were assayed after incubation using [3H] thymidine uptake. Proliferations of splenocyte were significantly increased by activation of mitogen Con A or PWM. Mitogen effects of Con A were increased by Con A plus betaine injection(0.1 mM), whereas PWM effects did not affect in PWM plus betaine injection(0.1 mM) in vitro. Splenocyte of laying hens fed betaine tended to proliferate in the presence of PWM, but appeared to be slightly suppressed in the presence of Con A in vivo. Proliferation of splenocytes which were stimulated by Con A or Con A+betaine injection(0.1 mM) were increased in dietary 600 ppm betaine, but inhibited in dietary 1,200 ppm betaine supplementation. Spleen weights and sheep red blood cell(SRBC) titers of hens fed betaine tended to increase compared to those of control, but were not significantly different. These results suggested that betaine could increase splenocyte proliferation in vitro.

A Study on the Converter Drive by Means of Series and Parallel Connection (콘버어터 직병렬 접속 운전에 관한 연구)

  • This paper presents a method of improving the power factor and , the waveform of A.C line currents and the output waveforms of AC to DC fully bridge converter systems which is achieved by connecting CON I.II and CON III.IV in series and parallel. The basic principle of the CON I.II.III.IV's operation is simple and feasibility of thease converter is proved by experiments. CON I and CON III are constructed of natural commutation, CON II and CON IV are constructed of forced commutation. The experimental results show that the power factor and waveform of the source currents and the output waveforms are improved by the method of connecting converter in series and parallel and driving it in I, II, III, IV quadrant.

Effect of Agariemycetes Supplementation on Egg Quality and Fecal Noxious Gas Concentration in Laying Hens (사료내 Agariemycetes 첨가 급여가 산란계의 계란 품질 및 분내 유해 가스 농도에 미치는 영향)

  • This study was conducted to evaluate effects of dietary Agariemycetes on egg quality and fecal noxious gas concentration in laying hens. A total of two hundred forty laying hens were randomly allocated to four treatments with five replicates of twelve chicks per pen. The experiment lasted 42 days with a 7-day adjustment period. Dietary treatments were as follows: 1) CON (basal diet), 2) A1 (basal diet + Agariemycetes 0.1%), 3) A3 (basal diet + Agariemycetes 0.3%) and 4) A5 (basal diet + Agariemycetes 0.5%). There was no significant difference in egg production and egg weight between CON and Agariemycetes treatments. During the second week of the experiment, eggshell breaking strength was higher in A3 and A5 treatments than that in CON treatment (P<0.05). Besides, eggshell breaking strength in P5 treatment was higher when compared with that in CON treatment (P<0.05) in the sixth week. During the second week of the experiment, eggshell thickness was higher in Agariemycetes treatments than CON treatment (P<0.05). Also, A5 treatment was higher than other treatments in the fourth week of the experiment (P<0.05). At the end of the second week, yolk color unit in A5 treatment was lower compared with CON treatment (P<0.05). During the fourth week of the experiment, CON treatment was higher than A3 treatment (P<0.05). During the second week of the experiment, Haugh unit in A3 treatment was greater than that in CON treatment (P<0.05). At the end of six weeks, Haugh unit in CON treatment was lower than other treatments (P<0.05). Fecal ammonia and hydrogen sulfide were increased in Agariemycetes treatments when compared with CON treatment (P<0.05). Compared with CON and A5 treatments, fecal mercaptans was higher than in other treatments in first of the experiment (P<0.05). In conclusion, this study indicated that the addition of Agariemycetes in the diet improved the egg quality and decreased the feacal noxious gas concentration in laying hens.

Enfrentamiento con el trauma a través de análisis de personajes femeninos en (영화, <루시아>의 여성 인물 분석을 통한 트라우마 직면의 문제)

  • El cine como medio de narrar el trauma histórico de los cubanos se formó especialmente en los años sesenta gracias al Instituto Cubano del Arte e Industria Cinematográficos y se sitúa en la cumbre. En el cine hecho por el director cubano Humberto Solás el discurso de historia, trauma y su memoria tiende a relatar básicamente sobre el mensaje de no olvidar el trauma histórico a través de buscar la manera eficaz e intrínseca de narrarlo. El hecho de no olvidar el trauma histórico y de poder enfrentarse con los elementos causantes del trauma significa el poder de tener la posibilidad probable de adquirir la mirada objetiva para observar la experiencia 'histórica' y la memoria 'cotidiana', y sus relaciones. Si consideramos el hecho de que la tendencia de la víctima suele querer olvidar la memoria del trauma por el miedo y dolor de enfrentarse con la representación del trauma, el enfrentarse con el trauma representado ya puede significar que la víctima esté en un trámite de conseguir la penetración con la cual supera lo negativo que le ofrece el trauma a la víctima. Este artículo se dedica especialmente para averiguar la estrategia del director cubano para realizar este fin de encontrar la manera adecuada en narrar el trauma cubano para que los cubanos puedan adquirir la penetración a través de la identificación y la simpatía entre los personajes del cine y los espectadores, enfocando con detalle en los carácteres de los protagonistas femeninas que son tres diferentes individuos en el mismo número de episodios. Cada protagonista, Lucía en cada episodio experimenta el trauma personal con diversos motivos, pero a pesar del dolor y del miedo del trauma histórico se despiertan los espectadores, quienes se identifican con la protagonista a través de la simpatía y la identificación, llegándose al momento de conseguir la penetración con la cual mira el futuro de la sociedad cubana con el objetivismo libre de los traumas históricos.

Inhibition of Con A-induced Iymphocyte proliferation by peritoneal exudate of Toxoplasma gondii-infected mice (톡소포자충 감염 마우스 복수액에 의한 Con A 유도 림프구 증식 억제효과)

  • The presence of biological response modifiers (BRMI-like effect was confirmed in peritoneal exudate (PE) of ToxopLnsmo gondii-infected ICR mice which inhibited Concanavalin A (Con A)-induced peritoneal Iymphocyte (PL) proliferation. During 5 days of PL incubation with $10{\;}\mu\textrm{g}/ml$ Con A with or without PE, 3H-thymidine uptake was measured for the last 24 hrs. Compared to uninduced control, PL proliferated by 7.3-fold with Con A induction_ When PE of infected mice was added, PL proliferation was inhibited by $74.0{\;}{\pm}{\;}11.9%$ whereas inhibition by PE of normal mice was $16.4{\;}{\pm}8.3%$. Inhibitory effect of PE increased exponentially from 3 days up to 4-5 days of survival after the infection. Inhibitory activity of PE was decreased concentration-dependently. Also the inhibition was diminished when the PE was treated with heat of $95^{\circ}C$ for 10 min orprecipitated with 10% trichloroacetic acid (TCA). In SDS-PAGE of PE, many minor bands appeared newly. Heat-labile protein molecule in PE exerted inhibitory activity to Con A- induced Iymphocyte proliferation.

Evaluation of Houttuynia cordata and Taraxacum officinale on Growth Performance, Nutrient Digestibility, Blood Characteristics, and Fecal Microbial Shedding in Diet for Weaning Pigs

  • A total of 144 pigs ((Landrace${\times}$Yorkshire)${\times}$Duroc)] with an average initial BW of $8.45{\pm}0.57$ kg were used in a 5-wk growth trial. Pigs were randomly allocated to 4 treatments with 9 replications per pen in a randomized complex block design. Dietary treatments included: i) CON (basal diet), ii) ANT (CON+tylosin 1 g/kg), iii) H1 (CON+H. cordata 1 g/kg) and iv) T1 (CON+T. officinale 1 g/kg). In this study, pigs fed the ANT and T1 treatment had a higher (p<0.05) average daily gain (ADG) and gain:feed (G:F) ratio than those fed CON and H1 treatment. Dietary ANT and T1 treatment led to a higher energy digestibility than the CON group. No difference (p>0.05) was observed on the growth performance and apparent total tract digestibility with H1 supplementation compared with the CON treatment. The inclusion of ANT treatment led to a higher (p<0.05) lymphocyte concentration compared with the CON treatment. Dietary supplementation of herbs did not affect (p>0.05) the blood characteristics (white blood cell (WBC), red blood cell (RBC), IgG, lymphocyte). No difference was observed on (p<0.05) fecal microbial shedding (E. coli and lactobacillus) between ANT and CON groups. Treatments H1 and T1 reduced the fecal E. coli concentration compared with the CON treatment, whereas the fecal lactobacillus concentration was not affected by the herb supplementation (p>0.05). In conclusion, the inclusion of T. officinale (1 g/kg) increased growth performance, feed efficiency, energy digestibility similarly to the antibiotic treatment. Dietary supplementation of T. officinale and H. cordata (1 g/kg) reduced the fecal E. coli concentration in weaning pigs.
