Effect of Proliferation Inhibition on B16/F10 Melanoma Cell by Chloroform Extract from Cornis fructus (산수유 클로로포름 추출물에 의한 B16/F10 melanoma세포의 증식억제효과)

  • Cornis fructus have various biological effects and major chemical components have been tannins, saponins, ursolic acids, gallic acids, linoleic acids, morronisides, cornins and loganins. The main aim of the present study is measurment the effect of chloroform extract from Cornis fructus on proliferation inhibition and Cell death. Cells were cultured in the presence of chloroform extracts from Cornis fructus for 48 h. after 48h treatment of B16/F10 melanoma cells with chloroform extracts, the cells were observed a dose-dependent inhibitions of cell viability with cell death in their proliferation. the cells were estimated cell viability, cell number, total DNA fragmentation and chromatin condensation in a dose-dependent manner. It also caused cell death as measured by cell morphology, DNA fragmentation and nucleus chromatin condensation. therefore, these results suggest that chloroform extracts from C. fructus is inhibitory proliferation and is related to cell death in this cells.

Effects of Methanol Extract from Cornis fructus on Melanogenesis (산수유 메탄올 추출물이 B16/F10 Melanoma 세포주의 멜라닌 생성에 미치는 영향)

  • Cornis fructus has been traditionally used for treating allergy, asthma, hepatitis, and chronic nephritis. It major chemical constituents have been saponins, phenolic acids and loganin. The main aim of the present study was to examine the effect of methanol extract from Cornis fructus on melanogenesis. Cell were cultured in the presence of methanol extracts from Cornis fructus for 48 h, and there were estimated total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. Methanol extract from Cornis fructus increased the melanin content and tyrosinase activity in a dose-dependent manner. Particularly, it was observed that only methanol extract $200\;{\mu}g/ml$ stimulated the melanin secretion in B16/F10 melanoma cells by 152% at 48 h treatment and the activity of tyrosinase was increased by 261% in the presence of same concentration.

  • Cornis fructus were extracted by successive extractions and then fractionated with chloroform extract to get active fractions. This study was performed to determine the cytotoxic effect of chloroform extract from Cornis fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. All extracts did not exhibit cytotoxicity in HIH 3T3 fibroblasts. Chloroform extract exhibited antitumor activity in A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. Futher fractionation with chloroform extract was performed to obtain effective fractions. (omitted)

All extracts did not exhibit cytotoxicity in NIH 3T3 fibroblasts. Chloroform extract exhibited antitumor activity in A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. Futher fractionation with chloroform extract was performed to obtain effective fractions. 3 fraction showed the strongest cytotoxic effect against A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. These results suggest that 3 fraction of the chloroform extract from Cornis fructus possessed bioactive material of antitumorous agents.

  • Fruits of Cornus Officinalis have been used as an astrinent, tonic and haemostatic in chinese medicine, contain a large amount of hydrolyzable tannins. The main aim of the present study was to examine the effect of Corni Fructus on melanogenesis. Cells were cultured in the presence of water extracts from Corni Fructus for 48 h, and there were estimated total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. Water extract from Corni Fructus increased the melanin content and tyrosinase activity in a dose-dependent mammer. Especially, It was observed that 100 μg/ml only water extract stimulated melanin secretion in B16/F10 melanoma cells by 130% at 48 h treatment and activity of tyrosinase increased by 160% in presence of same concentration.

Antioxidative Effects of Cornis fructus Extract on Chromium Trioxide Toxicity (Chromium Trioxide의 독성에 대한 산수유 추출물의 항산화 효과)

  • This study examined the antioxidative effects of Cornis fructus (CF) extract on cultured C6 glioma cells. For this purpose, cytotoxicity analysis of chromium trioxide ($CrO_3$) was performed and the protective effects of the CF extract on $CrO_3$-induced cytotoxicity was examined after the C6 glioma cells were cultured for 48 hours. The antioxidative effects, such as electron donating activity (EDA) and lactate dehydrogenase (LDH) activity were also analyzed. In this study, $CrO_3$ decreased the cell viability in a dose dependent manner. The $XTT_{50}$ value was determined to be $33{\mu}M$ after the cells were treated for 48 hours at a concentrations of $20{\sim}40{\mu}M$ $CrO_3$. The catalase (CAT) antioxidant increased significantly the cell viability that had been decreased by $CrO_3$-induced cytotoxicity. Regarding the protective effect of the CF extract, the cell viability of the CF extract was increased significantly compared to that of $CrO_3$ only. In addition, the CF extract showed antioxidative effects, such as EDA and an inhibitory effect on the LDH activity. These findings suggest that the cytotoxicity of $CrO_3$ is correlated with oxidative stress, and the CF extract effectively prevented $CrO_3$-induced cytotoxicity through the antioxidative effects. In conclusion, natural products, such as the CF extract may be a useful therapeutic agent for the prevention or treatment of toxicity induced by heavy metals via oxidative stress.
