Suppression of Immediate Hypersensitivity by Methyleugenol (메틸유제놀에 의한 즉시형 과민 반응의 억제)

  • We studied the action of methyleugenol on immediate hypersensitivity. Methyleugenol completely inhibited systemic anaphylaxis induced by compound 48/80 in mice. Methyleugenol al so inhibited local anaphylaxis induced by anti-dinitrophenyl (DNP) IgE. Moreover, methyleugenol dose-dependently inhibited histamine release in peritoneal mast cells activated by compound 48/80 or anti-DNP IgE. These results suggest that the inhibitory effect of methyleugenol on anaphylaxis induced by compound 48/80 or anti-DNP IgE is due to, in part at least, the membrane stabilization of mast cells.

  • The effect of aqueous extract of Magnoliae Cortex (Magnoliaceae) (MCAE) on the immediate-type allergic reaction was investigated. MCAE inhibited compound 48/80 induced systemic anaphylactic reaction in rats. MCAE (0.1 and 1 g/kg) also significantly inhibited local immunoglobulin E (lgE)-mediated passive cutaneous anaphylactic (PCA) reaction. MCAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) 1gE. Moreover, MCAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-mediated tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ production. These results indicate that MCAE inhibits immediate-type allergic reaction in vivo and in vitro.

  • We studied the effects of the powders of Spirulina platensis (SPP) on anaphylactic reactions. SPP inhibited systemic anaphylaxis induced by compound 48/80 in rats. SPP also inhibited local anaphylaxis activated by anti-dinitrophenyl(DNP) IgE. Moreover, SPP dose-dependently inhibited histamine release in rat peritoneal mast cells activated by compound 48/80 or anti DNP-IgE. These results suggest that SPP may contain compounds with actions that inhibit mast cell degranulation in the rat.

  • We studied the effects of sulfasalazine(SSZ) on anaphylaxis. SSZ was found to exhibit a inhibitory activity on the compound 48/80-induced anaphylaxis. SSZ also inhibited local a naphylaxis activated by anti-dinitrophenyl(DNP) IgE. Moreover, SSZ dose-dependently inhibited histamine ralease in rat peritoneal mast cells activated by compound 48/80 or anti DNP IgE. We investigated the effects of SSZ on cAMP of rat peritoneal mast cell. The level of cAMP in rat peritoneal mast cells, when SSZ was added, transiently and significantly increased approximately 16-fold compared with that of basal cells. These results suggest that the antianaphylactic action of SSZ may be associated with an increase in the intracellular cAMP content of the mast cells as the result of an inhibition of the cAMP phosphodiesterase.

  • The purpose of this research was to investigate the effects of supercritical fluid extract of Kamichungbieum (SFE) on allergic reaction. SFE (500 mg/kg) inhibited the systemic anaphylaxis induced by compound 48/80 or platelet activating factor and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-lgE and DNP-human serum albumin (HSA) in vivo. Also, SFE inhibited the SRSC-induced delayed type hypersensitivity and inhibited the hind paw edema induced by histamine. In addition, SFE inhibited the permeability of evans blue induced by acetic acid and inhibited the writhing syndrome induced by acetic acid. These results indicate that SFE may be useful for the prevention and treatment of allergy related disease.

  • The effect of aqueous extract of Salvia plebeia R. Br. (Labiatae) (AESP) on immunoglobulin (lgE) antibody mediated allergic reactions in rats was investigated. AESP inhibited passive cutaneous anaphylaxis (PCA) when intravenously, intraperitoneally, and orally administered. AESP dose-dependently inhibited histamine release from rat peritoneal mast cells activated by anti-DNP IgE antibody. Moreover, AESP had an inhibitory effect on anti-DNP IgE antibody induced $TNF-{\alpha}$ production from RPMC. These results suggest that AESP inhibit the IgE-mediated allergic reaction in rats.

  • We studied the effect of aqueous extract of Magnolia officinalis bark (Magnoliaceae) (MOAE) on the immediate hypersensitivity reaction. MOAE (0.01 to 1g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in rats. MOAE (0.1 and 1g/kg) also significantly inhibited local immunoglobulin E (lgE)-mediated passive cutaneous anaphylactic reaction. When MOAE was pretreated at concentrations ranging from 0.01 to 1g/kg, the levels of plasma histamine were reduced in a dose-dependent manner. MOAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) Igl. The level of cyclic AMP (CAMP) in RPMC, when MOAE was added, significantly increased compared with that of the normal control. Moreover, MOAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP Igl-induced tumor necrosis factor-$\alpha$ production from RPMC. These results indicate that MOAE inhibits immediate hypersensitivity reaction in vivo and in vitro.

Effect of Samul-Tang on the Allergic Inflammatory Response (사물탕(四物湯)이 알러지 염증반응에 미치는 영향)

  • Samul-Tang (SMT) has been used for nourishing of the blood, hematopoiesis as a herbal medicine history. The purpose of this study is to find out anti-allergic inflammatory reaction of SMT. To clarify the mechanism, the effect of SMT on vascular permeability of rat cutaneous tissue and histamine and cytokines (IL-6, IL-8, TNF-${\alpha}$) release from mast cells were observed. The results are the pretreatment with SMT significantly decreased the compound 48/80-induced degranulation and histamine release from RPMC, SMT also inhibited the anti-DNP lgE-induced increment of vascular permeability of rat cutaneous tissue. SMT significantly reduced the PMA plus A23187-induced increment of expression of IL-6, IL-8, and TNF-${\alpha}$ in HMC-1 Cell. The Present study provide evidence that SMT inhibits mast cell-derived inflammatory allergic reactions by blocking histamine release and pro-inflammatory cytokine expression, and suggest the mechanisms of action. Furthermore, in vivo and in vitro anti-allergic effect of SMT suggests a possible therapeutic application of this agent in inflammatory allergic diseases.
