Improvement Effect of the Eucommia ulmoides Extracts on CIA-induced Rheumatoid Arthritis Animal Models (두충 추출물의 류마티스관절염 동물모델에 대한 개선 효과)

  • The present study aimed to evaluate the effect of Eucommia ulmoides extracts on rheumatoid arthritis biomarker in a CIA-induced DBA/1 mice. For evaluation, Eucommia ulmoides extracts was administered orally at dose of 100 mg/kg/day for 4 weeks after production of an animal model of rheumatoid arthritis and we confirmed the treatments' effects based on serum biomarker, radiological, structural parameter analysis. Compared to the negative control group, the Eucommia ulmoides extracts treatments significantly reduced the serum level of inflammation and immunoglobulin markers (i.e., TNF-α, IgG, and hs-CRP), and significantly decreased the monocyte count of white blood cells. Furthermore, the Eucommia ulmoides extracts treatments effectively preserved the joint destruction, and little the joint deformation. Moreover, compared to the negative control group, the Eucommia ulmoides extracts treatments increased the bone volume, and significantly decreased bone inflammation. The results indicate that the Eucommia ulmoides extracts improved rhrumatoid arthritis symptoms. Thus, the Eucommia ulmoides extracts may be a novel therapeutic option for the management of rheumatoid arthritis.

  • Purpose : In this study we tested the hypothesis that eucommia ulmoides (EU) extract would improve disuse-induced muscle atrophy following hindlimb immobilization. Method : The hindlimb immobilization was performed with casting tape to keep the left ankle joint in a fully extended position. The rats in EU treated group were orally administrated with eucommia ulmoides water extract. After 2 weeks of immobilization, all animals were sacrificed, and the whole gastrocnemius muscles were dissected from both legs. The morphology of right and left gastrocnemius muscles in both EU and control groups were assessed by hematoxylin and eosin staining. Results : Eucommia ulmoides extract represented the significant protective effects against the reductions of the gastrocnemius muscles weight and average cross section area to compared with Control group. Conclusion : Eucommia ulmoides has protective effects against immobilization induced muscle atrophy.

Effects of the Dietary Mixture of Eucommia ulmoides Oliver on Osteoporosis (두충[Eucommia ulmoides Oliver] 조성물이 골다공증에 미치는 효과)

  • Inhibitory effects of Eucommia ulmoides Oliver against osteoporosis were studied with ovaryectomized rats. The diet mixture A of calcium, vitamin D$_3$, and vitamin K$_1$and the additional mixture B with extract of Eucommia ulmoides Oliver were fed for 7 months to ovaryectomized rats. The urine content of pyridinoline and deoxypyridinoline was used as bone biochemical markers. Concentrations of deoxypyridinoline of ovaryectomized rats fed with the mixture A and B decreased 9.0% and 26.9%, respectively, when they were compared to the ovaryectomized control. The trabecular bone areas of proximal tibias of ovaryectomized rats were measured and compared to the ovaryectomized control. The areas fed with the mixture A and B increased 9.9% and 42.0%, respectively, that proved the inhibitory effect of Eucommia ulmoides Oliver against osteoporosis.

  • In this study, Eucommia ulmoides Oliver extracts was studied in order to see any effects on the ${\beta}$-hexosaminidase release suppression of RBL-2H3 cells and on the expression of filaggrin, transglutaminase-1 (TGase-1) and cornified cell envelope (CE) related to the recovery of HaCaT keratinocyte skin barrier. Results showed that Eucommia ulmoides Oliver extracts reduced ${\beta}$-hexosaminidase release in RBL-2H3 cells and increased the effects of Eucommia ulmoides Oliver extract on the expression of filaggrin, transglutaminase-1 (TGase-1) and cornified cell envelope (CE) in HaCaT keratinocytes. Taken together, these results suggested that Eucommia ulmoides Oliver extract may be applicable for keratinocyte differentiation.

  • This study classified 40 of 10 weeks-old male Sprague Dawley rats which have osteoporosis caused by glucocorticoid(GC) induction into four groups of 10 rats and made the subjects medicated them Eucommia ulmoides Oliver and participate in treadmill exercise. Group I was non-treatment after GC induction; Group II was treadmill exercise after GC induction; Group III was Eucommia ulmoides Oliver feeding after GC induction; Group IV was treadmill exercise and Eucommia ulmoides Oliver feeding after GC induction. In the bone mineral density and bone strength test, there was a statistically significant difference between the group I, II, III compared to group IV(p<0.05). In the osteocalcin level as one of bone formation indexes, there was a statistically significant difference between the group I, II, III compared to group IV(p<0.05). In respect to the calcium there was a statistically significant difference between group I, II, III compared to group IV(p<0.05). In respect to histological findings, the group IV showed mild femur disruption and lytic change on femur section. The above results suggests that Eucommia ulmoides Oliver medicated and treadmill exercise is effective to prevention and treatment of osteoporosis.

Antioxidant, Antidiabetic and Cytotoxic Effects of Eucommia ulmoides Oliv. Bark in vitro

  • The aim of this study is to investigate in vitro biological properties of Eucommia ulmoides Oliv. bark. Ethyl acetate(EtOAc) fraction from aqueous extract of Eucommia bark showed strong antioxidant activity of $IC_{50}$ 19.2 ${\mu}g/ml$ by 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical-scavenging assay. The Eucommia bark extract showed $\alpha$-glucosidase inhibitory activity and inhibited growth of human liver cancer cell, suggesting its potential biological value of anticancer.

  • Chemical composition of the Eucommia ulmoides bark, including extractives, proximate, mineral, fatty acid and monosaccharide compositions, was studied The most abundant mineral was calcium (533.17 mg/l00 g). $\alpha-linolenic$ acid (24.7%) and linoleic acid (24.3%), showed higher contents among the fatty acids. Major monosaccharides of E. ulmoides balk were arabinose (13.94 mg/g), xylose (18.91 mg/g) and glucose (119.7 mg/g). From the bark of E ulmoides, four compounds were isolated and their structures were elucidated as caffeic acid (I), kaempferol (II), quercetin (III) and isoquercitrin (IV) by spectroscopic analysis such as NMR and MS, including cellulose TLC and other chemical evidence such as hydrolyzation and acetylation. The antioxidant activities of four isolated compounds were evaluated by DPPH free radical scavenging, hydroxyl scavenging and reducing power assays. The results indicated that all the isolated compounds showed higher DPPH radical scavenging activity than $\alpha-tocopherol$ and BHT that were used as positive controls and these four compounds exhibited considerable reducing power and hydroxyl radical (OH) scavenging activity. Considering from the results above, it suggests that the E. ulmoides bark is a potential natural source of antioxidant material.

Isolation of Grb2-Shc Interaction Inhibitory Compounds from the Leaves of Eucommia ulmoides O. (두충(Eucommia ulmoides O.)잎으로부터 Grb2-Shc 결합저해 활성물질의 분리)

  • The MeOH extracts obtained from the leaves of Eucommia ulmoides were solvent-fractionated with EtOAc, n-BuOH, and $H_20$, successively. From the EtOAc extract showing Grab2-Shc inhibitory activity, a flavonol and a lignan compounds were isolated through repeated silica gel column chromatographies. By interpretation of several spectral data and adaptation of acetylation method, the chemical structures of the compounds were determined as 5,7,3',4'-tetrahydroxy-flavonol (quercetin), and 4,4',8',9-tetrahydroxy-3,3'-dimethoxy-7,9'-cyclolignan {(-)-olivil)}, respectively. The compounds exhibited $IC_{50}$ values in Grb2-Shc inhibitory activity to be 93 and $210\;{\mu}mole/l$, respectively.

  • Geniposide, an iridoid glucoside, was isolated from the cortex of Eucommia ulmoides by repealed silica gel chromography and identified by its spectral data. The quantitative determination of geniposide with HPLC has been developed and this method provided a tool for standardization of the commercial Eucommiae Cortex.
