Retinal prostheses are being developed to restore vision for the blind with retinal diseases such as retinitis pigmentosa (RP) or age-related macular degeneration (AMD). Among the many issues for prosthesis development, stimulation encoding strategy is one of the most essential electrophysiological issues. The more we understand the retinal circuitry how it encodes and processes visual information, the greater it could help decide stimulation encoding strategy for retinal prosthesis. Therefore, we examined how retinal ganglion cells (RGCs) in in-vitro retinal preparation act together to encode a visual scene with multielectrode array (MEA). Simultaneous recording of many RGCs with MEA showed that nearby neurons often fired synchronously, with spike delays mostly within 1 ms range. This synchronized firing - narrow correlation - was blocked by gap junction blocker, heptanol, but not by glutamatergic synapse blocker, kynurenic acid. By tracking down all the RGC pairs which showed narrow correlation, we could harvest 40 functional connectivity maps of RGCs which showed the cell cluster firing together. We suggest that finding functional connectivity map would be useful in stimulation encoding strategy for the retinal prosthesis since stimulating the cluster of RGCs would be more efficient than separately stimulating each individual RGC.

Objective : The subgenual cingulate cortex, a part of default-mode network, has been known to playa key role in the pathophysiology of depression. The previous studies have reported abnormal functional connectivity between the subgenual cingulate cortex and other brain regions in the patients with depression. The goal of this shldy was to explore the resting-state functional connectivity of the subgenual cingulate cortex between the patients with depression and healthy subjects. Methods : Twenty patients with major depression and age- and sex-matched 20 healthy subjects underwent 5-minute resting state fMRI scans. The functional connectivity map in each subject was acquired using seed-based correlation analysis with the seed located in the subgenual cingulate cortex (Talairach coordinates; x=-10, y=5, z=-10). The functional connectivity maps were calculated using AFNI and compared between the patient and healthy subject group via two-sample T-test using 3dttest++ in AFNI package. Results : Functional connectivity was decreased between the subgenual cingulate cortex and both sides of fusiform gyrus in depressed subjects. Connectivity was also decreased between the subgenual cingulate cortex and the left cerebellum in the patient group. There was no correlation between the severity of depression and the degree of functional connectivity between the subgenual cingulate cortex and the regions showing decreased functional connectivity. Conclusion : Decreased resting-state functional connectivity between the subgenual cingulate cortex and both sides of fusiform gyrus, and decreased connectivity between the subgenual cingulate cortex and the left cerebellum found in the patients with major depression in comparison to the healthy subjects might be related to abnormal emotional and cognitive processing of depressed patients.
Objective: In this study, we used resting-state fMRI data to map differences in functional connectivity between a comprehensive set of 8 distinct cortical and subcortical brain regions in healthy controls and Internet addicts. We also investigated the relationship between resting state connectivity strength and the level of psychopathology (ex. score of internet addiction scale and score of Barratt impulsiveness scale). Background: There is a lot of evidence of relationship between Internet addiction and impaired inhibitory control. Clinical evidence suggests that Internet addicts have a high level of impulsivity as measured by behavioral task of response inhibition and a self report questionnaire. Method: 15 Internet addicts and 15 demographically similar non-addicts participated in the current resting-state fMRI experiment. For the connectivity analysis, regions of interests (ROIs) were defined based on the previous studies of addictions. Functional connectivity assessment for each subject was obtained by correlating time-series across the ROIs, resulting in $8{\times}8$ matrixs for each subject. Within-group, functional connectivity patterns were observed by entering the z maps of the ROIs of each subject into second-level one sample t test. Two sample t test was also performed to examine between group differences. Results: Between group, the analysis revealed that the connectivity in between the orbito frontal cortex and inferior parietal cortex, between orbito frontal cortex and putamen, between the orbito frontal cortex and anterior cingulate cortex, between the insula and anterior cingulate cortex, and between amydgala and insula was significantly stronger in control group than in the Internet addicts, while the connectivity in between the orbito frontal cortex and insula showed stronger negative correlation in the Internet addicts relative to control group (p < 0.001, uncorrected). No significant relationship between functional connectivity strength and current degree of Internet addiction and degree of impulsitivy was seen. Conclusion: This study found that Internet addicts had declined connectivity strength in the orbitofrontal cortex (OFC) and other regions (e.g., ACC, IPC, and insula) during resting-state. It may reflect deficits in the OFC function to process information from different area in the corticostriatal reward network. Application: The results might help to develop theoretical modeling of Internet addiction for Internet addiction discrimination.
Purpose: One of the suggested potential mechanisms of tinnitus is an alteration in perception in the neural auditory pathway. The aim of this study was to investigate the difference in laterality in functional connectivity between tinnitus patients and healthy controls using resting state functional MRI (rs-fMRI). Materials and Methods: Thirty-eight chronic tinnitus subjects and 45 age-matched healthy controls were enrolled in this study. Connectivity was investigated using independent component analysis, and the laterality index map was calculated based on auditory (AN) and dorsal attention (DAN), default mode (DMN), sensorimotor, salience (SalN), and visual networks (VNs). The laterality index (LI) of tinnitus subjects was compared with that of normal controls using region-of-interest (ROI) and voxel-based methods and a two-sample unpaired t-test. Pearson correlation was conducted to assess the associations between the LI in each network and clinical variables. Results: The AN and VN showed significant differences in LI between the two groups in ROI analysis (P < 0.05), and the tinnitus group had clusters with significantly decreased laterality of AN, SalN, and VN in voxel-based comparisons. The AN was positively correlated with tinnitus distress (tinnitus handicap inventory), and the SalN was negatively correlated with symptom duration (P < 0.05). Conclusion: The results of this study suggest that various functional networks related to psychological distress can be modified by tinnitus, and that this interrelation can present differently on the right and left sides, according to the dominance of the network.

In this study, an evaluation method was proposed to determine the restoration section in the riverine ecobelt project. The target river for this study is the Hongcheon river in Kangwon-do. The Hongcheon river of 96 km was divided longitudinally into subsections of 2 km. The analysis through map and aerial photograph as well as field surveys were performed in 48-sub-sections. The fragmentation items were classified into connectivity and ecological functionality. The connectivity fragmentation was also divided into two items: the area discontinuity of the land use and the line discontinuity of river bank and road. Also, the ecological functional fragmentation was evaluated by using the items of river channel, river bed, vegetation, and the obstruction of river flow. These items was modified from those in the previous literature. From map analyses and field surveys, the fragmentation score was kept with each items in 48 sub-sections of Hongcheon river. The fragmentation rate was made from the total score in each section. The results showed that sections from F1 to G2 was evaluated to have high rates of all connectivity and functionality fragmentation of 1st or 2nd rate. Other sections have high connectivity fragmentation of 2nd rate, but low functional fragmentation. Thus, these sections are evaluated to be excludible in restoration site. This study seems to make a contribution to evaluate the fragmented sections for the riverine ecobelt project.
Neuronal connectivity determines brain function. Therefore, understanding the full map of brain connectivity with functional annotations is one of the most desirable but challenging tasks in science. Current methods to achieve this goal are limited by the resolution of imaging tools and the field of view. Macroscale imaging tools (e.g., magnetic resonance imaging, diffusion tensor images, and positron emission tomography) are suitable for large-volume analysis, and the resolution of these methodologies is being improved by developing hardware and software systems. Microscale tools (e.g., serial electron microscopy and array tomography), on the other hand, are evolving to efficiently stack small volumes to expand the dimension of analysis. The advent of mesoscale tools (e.g., tissue clearing and single plane ilumination microscopy super-resolution imaging) has greatly contributed to filling in the gaps between macroscale and microscale data. To achieve anatomical maps with gene expression and neural connection tags as multimodal information hubs, much work on information analysis and processing is yet required. Once images are obtained, digitized, and cumulated, these large amounts of information should be analyzed with information processing tools. With this in mind, post-imaging processing with the aid of many advanced information processing tools (e.g., artificial intelligence-based image processing) is set to explode in the near future, and with that, anatomic problems will be transformed into informatics problems.
Gene set enrichment analysis (GSEA) is a popular tool to identify underlying biological processes in clinical samples using their gene expression phenotypes. GSEA measures the enrichment of annotated gene sets that represent biological processes for differentially expressed genes (DEGs) in clinical samples. GSEA may be suboptimal for functional gene sets; however, because DEGs from the expression dataset may not be functional genes per se but dysregulated genes perturbed by bona fide functional genes. To overcome this shortcoming, we developed network-based GSEA (NGSEA), which measures the enrichment score of functional gene sets using the expression difference of not only individual genes but also their neighbors in the functional network. We found that NGSEA outperformed GSEA in identifying pathway gene sets for matched gene expression phenotypes. We also observed that NGSEA substantially improved the ability to retrieve known anti-cancer drugs from patient-derived gene expression data using drug-target gene sets compared with another method, Connectivity Map. We also repurposed FDA-approved drugs using NGSEA and experimentally validated budesonide as a chemical with anti-cancer effects for colorectal cancer. We, therefore, expect that NGSEA will facilitate both pathway interpretation of gene expression phenotypes and anti-cancer drug repositioning. NGSEA is freely available at www.inetbio.org/ngsea.

Purpose: This study examines construction core plans for the users of vertical-typed general hospitals to effectivly use the flow line. Methods: The study sampled representative 9 hospitals, calculated the depth value through Convex Map of Space Syntax and Justified Graph according to the determination of form of construction cire, and analyzed its functional connectivity. Results: The analysis of the connectivity between operation core part and emergency part of core space with high importance in the hospitals showed that the types of hospital and hospital have the lowest depth value in the spatial phase diagram, where central treatment part and outpatient part are arranged well vertically. Elevators for patients at these hospitals are close to operation and emergency parts actually separated from the elevators for passengers. For shortening of flow line of patients and private movement environment, however, it is desirable to arrange the elevators for patients to be adjacent to the operation parts and to arrange the emergent patient entrances more effectively to separate them from the flow line of visitors and guardians. Implications: Consideration should be taken into account for the effective flow line design. This study hopefully may serve as a stepping stone for the standard design of horizontal/vertical flow line.

Purpose: To report the use of multiband accelerated echo-planar imaging (EPI) for resting-state functional MRI (rs-fMRI) to achieve rapid high temporal resolution at 3T compared to conventional EPI. Materials and Methods: rs-fMRI data were acquired from 20 healthy right-handed volunteers by using three methods: conventional single-band gradient-echo EPI acquisition (Data 1), multiband gradient-echo EPI acquisition with 240 volumes (Data 2) and 480 volumes (Data 3). Temporal signal-to-noise ratio (tSNR) maps were obtained by dividing the mean of the time course of each voxel by its temporal standard deviation. The resting-state sensorimotor network (SMN) and default mode network (DMN) were estimated using independent component analysis (ICA) and a seed-based method. One-way analysis of variance (ANOVA) was performed between the tSNR map, SMN, and DMN from the three data sets for between-group analysis. P < 0.05 with a family-wise error (FWE) correction for multiple comparisons was considered statistically significant. Results: One-way ANOVA and post-hoc two-sample t-tests showed that the tSNR was higher in Data 1 than Data 2 and 3 in white matter structures such as the striatum and medial and superior longitudinal fasciculus. One-way ANOVA revealed no differences in SMN or DMN across the three data sets. Conclusion: Within the adapted metrics estimated under specific imaging conditions employed in this study, multiband accelerated EPI, which substantially reduced scan times, provides the same quality image of functional connectivity as rs-fMRI by using conventional EPI at 3T. Under employed imaging conditions, this technique shows strong potential for clinical acceptance and translation of rs-fMRI protocols with potential advantages in spatial and/or temporal resolution. However, further study is warranted to evaluate whether the current findings can be generalized in diverse settings.

In this study, an ecological fragmentation improvement effect from different riverine ecobelt width was analyzed. The target river for this study is the Hongcheon river in Kangwondo. This section of 11 km among the Hongchenon river was reported to be seriously fragmented in the previous literatures. The Hongcheon river of 11 km was divided longitudinally into subsections of 1 km. The analysis through map and aerial photograph as well as field surveys were performed in 11 subsections(A-K). Using the fragmentation index, the connectivity fragmentation such as river bank, road, and land use and the ecological functional fragmentation were evaluated. The results showed that I-section has the highest fragmentation rate. The Hongcheon river in the present status without ecobelt was analyzed to has the fragmentation of 2nd rate from the mean fragmentation value of 11-subsections. Also, when the riverine ecobelt of 10 m-50 m was restored, the improvement effect of connectivity fragmentation was relatively small, while for the ecological fragmentation, the improvement effect of 12.4-25.1% was predicted. Also, the ecobelts of 10 m and 30 m were evaluated to have a relatively great improvement effect. Especially, for the ecobelt of 30m width, the fragmentation rate was mitigated form 2nd rate to 3rd rate.
