Production and Evaluation of Anti-Gastrin Serum for Radioimmunoassay (방사면역측정을 위한 항 Gastrin 혈청의 생산 및 평가)

  • In order to produce antibody for use in radioimmunoassay of gastrin in physiological concentration, four rabbits of New Zealand white were immunized with synthetic human gastrin-17-I conjugated to hemocyanin with EDC. Among them, only one rabbit produced antibody that could bind 50% of $^{125}I-gastrin$ at a final dilution of 1:25,000. $^{125}I-gastrin$ was prepared with synthetic human gastrin-17-I and $NaI^{125}$ by lactoperoxidase technique. The product was then purified on a column of Sephadex Gl5/G5O (7:3, w/w) followed by a column of DEAE sephadex A-25. The specific radioactivity of the purified $^{125}I-gastrin$ was in the range of 347-1429 ${\mu}Ci/nmole$ when determined by the self-displacement method. The effective affinity constant $(K_{eff})$, total binding sites (N), heterogeneity index $({\alpha})$ and average affinity constant $(K_{0})$ of the anti-gastrin serum calculated from Scatchard plot as well as Sips plot were $1.77{\times}10^{11}/M$, 255 nM, 0.84 and $0.79{\times}10^{11}/M$, respectively. When radioimmunoassay was performed with the anti-gastrin serum, it was confirmed that the mean concentration of gastrin immunoreactivity in plasma was increased by feeding in humans and rats, and also increased by bombesin administration in rats. The results indicate that the anti-gastrin serum produced in the present investigation is suitable for radioimmunological determination of gastrin in physiological concentration.

Early Gastrin Response to Single Glucose and Casein Feeding (포도당 및 Casein 액 단일투여에 의한 조기 Gastrin 반응)

  • The early plasma gastrin responses to single oral glucose or casein solution were studied in the same normal subjects on different days. After an overnight fast, glucose or casein solution was ingested within few minutes at the breakfast time. The plasma gastrin responses to these solutions were compared and contrasted with the concentration when the subjects received glucose solution intravenously. Results were as follows: 1) Rapid intravenous glucose infusion did not produce any changes in the plasma gastrin concentration. 2) Plasma gastrin concentration rose and peaked within 10 minutes after an oral liquid ingestion and then decreased substantially by 15 minutes, but remained slightly above fasting levels at 60 minutes. 3) There was no significant difference between the mean plasma gastrin concentrations after glucose of casein ingestion, but each fluid produced a significant increase in serum gastrin above fasting levels. 4) The subjects who produced high plasma gastrin response to glucose solution did so to casein solution. Conversely a low response to one solution reflected a low response to the other solution. 5) From the above results, authors discussed that individual responsibility rather than the sorts of meals is the factor in the determination of the magnitude of the early gastrin response.

Plasma Gastrin Concentration after a Carbohydrate Meal and a Protein Meal in Normal Human Subjects (식후 정상 한국인의 혈장 gastrin 농도)

  • This study was conducted to investigate the effect of the ingestion of rice and hamburger meals upon the plasma gastrin concentration in normal human subjects in Korea. Eight normal human subjects including male and female with the mean age of 28 years (range: 20-40 years) were studied. After an overnight(about 15 hrs) fast each subject ingested a rice meal and a hamburger meal on different days. The rice meal consisted of 250 g boiled rice, 50 g vegetables and 200 ml barley tea, corresponding to 6.8 g protein, 0.5 g fat and 81g carbohydrate and the hamburger meal consisted of 200 g hamburger, 50 g vegetables ana 200 ml milk, corresponding to 43 g protein, 43 g fat and 9 g carbohydrate. The venous blood samples were drawn before and after the ingestion of the test meal at the following times: -30, 0, 15, 30, 45, 60, 90, and 120 min, for measurement of gastrin by radioimmunoassay. The following results were obtained : 1) Plasma gastrin concentration in response to the ingestion of the rice or the hamburger meal increased significantly compared with the concentration in fasting state. 2) The increase of the plasma gastrin concentration after the hamburger meal was significantly higher than that after the rice meal. 3) There was a significant linear correlation between the postprandial peak plasma gastrin concentration after the rice meal and the concentration after the hamburger meal in each subject. It is inferred from the above results that a carbohydrate meal as well as a protein meal has a stimulatory effect on gastrin release in normal human subjects.

  • The effect of bile deprivation on serum lipid and gastrin contents was investigat\ulcornered after choledocho-urinary cystostomy in Sprague-Dawley rats. Bile deprivated rats were compared with sham operated control group. Gastrin levels in serum and antral tissue were measured and serum lipid concentrations were also measured. Gastrin levels of serum and tissue after bile deprivation were increased significantly compared with those of the controL At the end of 1st and 2nd week after bile deprivation, serum cholesterol and triglyceride contents were significantly lower han those of the control. By 4th week, there was no significant difference between two groups. Increases in serum and antral gastrin levels temporarily coincided well with decreases in serum lipid contents after bile deprivation. These results suggest that there is increase in biosynthesis and release of gastrin and decrease in fat absorption at early stage of bile deprivation.

A Study on Fasting Serum Gastrin level in Normal Subjects and Various Gastric Diseases by Radioimmunoassay (각종(各種) 위질환(胃疾患)에 있어서의 혈청(血淸) gastrin 치(値)에 관(關)한 임상적(臨床的) 연구(硏究))

  • The fasting serum gastrin levels were measured by radioimmunoassay in 32 normal healthy subjects without recognized gastrointestinal symptoms and 90 patients with various gastric diseases, who were visited or admitted to National Medical Center from February to June, 1981. The following results were obtained; 1) The fasting mean serum gastrin level in normal subjects was $55.9{\pm}26.3pg/ml$, and there was no difference between male and female. 2) The gastrin levels in gastric and duodenal ulcer were $85.0{\pm}28.4$ and $76.0{\pm}43.0pg/ml$ respectively, and it's values were elevated as compared with normal subjects. In gastric ulcer, the gastrin level was elevated than that of duodenal ulcer, but no significant difference in each other. 3) The gastrin level in stomach cancer was $89.5{\pm}42.2pg/ml$, and it's values were markedly elevated as compared with values in normal subjects. 4) The gastrin level in gastritis was $73.4{\pm}37.4pg/ml$, and it's values were elevated as compared with values in normal subjects. 5) The gastrin level in post-gastrectomy state was $50.3{\pm}16.3pg/ml$, and it's values were slightly decreased as compared with values in normal subjects.

Effects of Long Term Stimulated Acupuncture at ST 36 on the Serum Gastrin Level in Rats (족삼리 장기 매침이 정상 백서의 혈중 Gastrin 농도에 미치는 영향)

  • To investigate the effect of acupuncture on Zusanli (ST 36) in this study, gastrin level of serum by radioimmunoassay was measured at 1, 3, 6 weeks in rats. In other groups, 1 mg/day omeprazole was administered for 1, 3, 6 weeks. Omeprazole increased significantly gastrin level of serum at 1, 3, 6 weeks in a time dependent manner. Acupuncture applied to the ST 36 acupoint decreased at 1, 3, 6 weeks. Furthermore, acupuncture applied to other acupoint, Yanglinquan (GB 34) did not produced significant effect. When the common peroneal nerve was dissected, acupuncture of ST 36 produced the change of gastrin level. These data suggest that acupuncture decreased gastrin level of serum in point specific way and that effect was not related with surrounding nerve.

CR2945, $\beta$-2-( [2-(8-azaspiro [4.5] dec-8-ylcarbony!)-4.6-dimethylphenyl]amino-2-oxoethyl] -(R)-1-naphthalenepropanoic acid. is known to have an inhibitory effect on a gastrin receptor of colorectal cancer cells. As the human pancreatic cancer cells (BxPC-3) express gastrin receptors. interruption of binding of gastrin with gastrin receptor of human pancreatic cancer cells by CR2945 inhibits the growth of human pancreatic cancer cells. Effect of Medial Amygdala on Gastric Acid Secretion and Plasma Gastrin Concentration in Conscious Rats (흰쥐에서 내측 편도체가 위산 분비와 혈장 Gastrin 농도에 미치는 영향)

  • This study was undertaken to investigate the effect of medial amygdala on the gastric acid secretion and plasma gastrin concentration in the rats with chronic gastric fistula. After the medial nucleus of amygdala was damaged bilaterally by radiofrequency a. c. through stereotaxically inserted electrodes, the gastric juice was collected in the basal and histamine-stimulated states for 1 hour. The gastric juice was also collected while the medial nucleus of amygdala was stimulated with biphasic square wave in the both states. After the collection of the gastric juice, blood samples were drawn from the abdominal aorta for the radioimmunoassay of plasma gastrin. The results were as follows: 1) The damage of the medial amygdala significantly decreased the gastric juice volume and the acid output in the histamine-stimulated state. 2) The electrical stimulation of the medial amygdala significantly increased the gastric juice volume and the acid output in the histamine-stimulated state, and the acid output in the basal state. 3) The damage of the medial amygdala significantly decreased the plasma gastrin concentration but the electrical stimulation of the medial amygdala did not affect the plasma gastrin concentration. It is therefore suggested that the medial amygdala has a facilitatory influence on the histamine-stimulated gastric acid secretion in rats, and the influence may not be attributed to gastrin release.

Effects of ST36, BL21 on the Serum Gastrin Level, Endocrine Cells and Mucus of Gastric Mucosa In Rats (족삼리(足三里) 위유(胃兪) 혈(穴)이 흰쥐 혈중 Gastrin 농도, 위점막의 내분비세포 및 점액에 미치는 영향)

  • Objective : The purpose of this study is To investigate the effects of acupuncture and moxibustion at Zusanli(ST36) and Weishu(BL21) Methods : serum gastrin level by radioimmunoassay was measured at 5 days after acupuncture and moxibustion of those acupoints. Gastric endocrine cell(G cell and Histamine immunoreactive density) by avidin-biotinylated complex(ABC) technique, histological examinations(Alcian Blue-PAS Stain; Alcian blue-Periodic Acid Schiff reagent) of the gastric mucosa were also performed. Acupuncture applied to the ST36 acupoint increased gastrin level of serum, but moxibustion did not produced significant effect. All of acupuncture and moxibustion at BL21 acupoint increased gastrin level of serum significantly. In moxibustion at ST36 and BL21, the number of gastrin secreting cells in gastric mucosa, the density of immunoreactive histamin secreting cells and the density of body mucosa stained by PAS were decreased compare to acupuncture at ST36 and BL21. In acupuncture and moxibustion at BL21, the density of pylorus mucosa stained by PAS were increased compare to the groups applied to ST36. In the density of body mucosa stained by AB, moxibustion at BL21 and ST36 were increased compare to the other groups. Results : These data suggest that acupuncture and moxibustion at BL21 increased gastrin level of serum and those effects were more potent than acupuncture at ST36.

Effect of Treatment with Manual and Electro-Acupuncture at ST 36 and GB 34 on the Serum Gastrin Level in Rats (족삼리와 양릉천의 침, 전침과 경혈단위 전침기구 자극이 혈중 Gastrin 농도에 미치는 영향)

  • It is well known that acupuncture is effective for treatment of gastric disease in human. For years, we have been utilizing a electro-acupuncture(EA) in attempt to manage gastric symptoms. However, the data on clinical effectiveness is conflicting and the action of acupoints is unclear. The purpose of this study was to investigate the effects of manual and electro acupuncture(EA) at acupoints of ST 36 and GB 34 on the serum gastrin level by radioimmunoassay in rats. All of acupuncture treatment was applied to the acupoints for 30 min during 3 days. Manual acupuncture with both acupoints resulted in increased gastrin serum level when compare to control group treated with only anesthesia. Either high (100 Hz) and low (2 Hz) frequency EA at ST 36 decreased gastrin serum level compared with control group, but EA at GB 34 conversely increased. In EA with device was designed stimulating only acupoint(New EA), either low frequency on ST 36 and high frequency on GB 34 unusually increased gastrin serum level compared to control group. The results of this experiment suggest that many factors, such as the selection of acupoints, the manipulation and the specification of acupuncture device, are capable of affecting acupuncture effects.
