Improving Texture and Storage Stability of Chinese-Style Pork Jerky by the Addition of Humectants

  • Chinese-style pork jerky with different levels (3, 6 and 9%) of glycerol or sorbitol were prepared. Moisture content, water activity ($a_w$) and shear value decreased with the addition of glycerol or sorbitol. During storage at $28^{\circ}C$ thiobarbituric acid (TBA) value of all samples declined with the addition of glycerol or sorbitol during storage time. After storage at room temperature ($28^{\circ}C$) for 180 days, the volatile basic nitrogen (VBN) of all treated products were less than the control group. In addition, the mold and yeast growth were inhibited slightly with the addition of glycerol or sorbitol. The absorption isotherm of pork jerky with different levels of glycerol at $25^{\circ}C$ had an aw above 0.75, and moisture contents at 6 and 9% which were higher than the control group. The moisture content was less than the control group with the addition of sorbitol. The chewiness desirability score of pork jerky with 6% glycerol was higher than the other treatments. However, no differences in hardness desirability score due to treatments were detected by sensory panelists.

  • The glycerol carbonate was synthesized by glycerol and urea using metal oxide catalysts. The physical properties of the prepared metal oxide catalysts were investigated by X-ray diffraction (XRD), specific surface area analysis (BET), field emission scanning electron microscopy (FE-SEM) and temperature programmed desorption (TPD). In addition, we confirmed the conversion of the glycerol and the yield of the glycerol carbonate according to characteristics of metal oxide catalysts. From XRD and FE-SEM analysis, the crystallite size and crystallinity of metal oxide catalysts decrease with addition of Al. In addition, the Zn-Al mixed metal oxide had higher catalytic activity than the pure ZnO due to decreased side reaction in the synthesis of glycerol carbonate.

  • Prepared polyurethane resin for buffalo leather coating on surface was synthesized with addition reaction glycerol which had different mole ratio. Mechanical properties of the synthesized polyurethane resin were measured by the SEM, FT-IR, UTM. Growing concerns in the evnironment-friendly polymer resin, we have synthesized low late obtained solvent water dispersion resin to be coating on buffalo leather. The increase of aliphatic trihydric alcohol glycerol mole %, abrasion resistance and tensile strength had demonstrated reduce properties. On the contrary, elongation and flexibility properties had highly increased. In the result of toluene solvent resistance, there was no effect of increased or decreased by the ratio of glycerol mole %.

  • Effects of the glycerol addition and removal medium containg 10% sucrose on the mouse embryo survival after freezing in a liquid introgen container were determined using the FDA test. The summarized results are the following. 1. The FDA score was higher (P<0.05) when embryos were frozen in the glycerol addition medium with sucrose that the one without it(3.4 vs 3.0). 2. No difference in the score was found between the glycerol removal medium containing 10% sucrose(3.2) and PBS+10% sucrose(3.3). 3. The score was higher(P<0.01) at morular stage than blastocyst stage of embryos.

  • Glycerol-spore formation of wild myxobacteria isolated in Korea has been tested. One third of the wild myxobacterial isolates belonged to the genus Myxococcus and Corallococcus produced a large number of glycerol-spores in the presence of glycerol or dimethyl sulfoxide. However, two third of the other wild myxobacterial isolates belonged to the genus Myxococcus and Corallococcus produced a negligible number of spores under the same condition. In addition, the myxobacteral isolates belonged to the other genus, such as Stigmatella, Archangium, and Cystobacter, failed to produce a noticeable number of glycerol-spores.

  • Multiple ejaculates were collected from four male mongrel dogs. The second fraction and the small volume of third fraction from the ejaculates were divided and treated as follows : control; addition of the egg-yolk Tris extender to the semen at $37^{\circ}C$. group I; Removal of seminal plasma, group II; addition of the glycerolated extender at $4^{\circ}C$, group III Removal of seminal plasma and addition of glycerolated extender at $4^{\circ}C$. The semen cooled to $4^{\circ}C$ was equlibrated for 2hrs and preserved in refrigerator at $4^{\circ}C$. The preserved semen was evaluated for kinetics, morphology, motility and thermoresistance daily for 3 days. 1. The kinectics after preserved days 2 and 3 of group I was significantly higher than that of control(p<0.05). 2. There were no significant difference in abnormal morphology of each group between the periods of storage. 3. The motility after preserved day 1 and days 3 of group I was significantly higher than that of others(p<0.05), and the molity after preserved days 2 of group I and III was signficantly higher than that of others(p<0.05). 4. When the molity of preserved semen was measured during incubation at $37^{\circ}C$, the motility of four groups was declined at similar rates. There was no effect of removal of seminal plasma and glycerol addition on thermoresistance.

  • Lipomyces starkeyi KSM 22 produces dextranase and amylase (DXAMase). The addition of 0.02% (w/v) 2-deoxy-D-glucose or 0.5% (w/v) glycerol into a 1% (w/v) starch medium increased the final activity of DXAMase produced 2.5 fold or 2.4 fold, respectively, compared to that of a 1% (w/v) starch medium. This activity was similar to that produced with 1% (w/v) dextran. The stability of purified OXAMase at 40$\^{C}$ for 3 weeks was tested using the various enzyme stabilizers. With the addition of 25% (v/v) glycerol, 90.9% of initial activity was left after 3 weeks. For practical use, the addition of 1% (v/v) glycerol with 50 mM of CaCl$_2$ or KH$_2$PO$_4$was adequate and maintained 73.4% of the initial activity under the test conditions used.

Biological production of 1,3-propanediol using crude glycerol derived from biodiesel process (바이오디젤 부산물인 폐글리세롤을 이용한 생물학적 1,3-propanediol 생산)

  • The production of 1,3.propanediol (1,3-PD) was investigated with Klebsiella pneumoniae DSM2026 and K. pneumoniae DSM4799 using crude glycerol obtained from biodiesel industry. Crude glycerol was used without prior purification to investigate effects of impurities in crude glycerol on 1,3-PD production. In the batch cultures, 1,3-PD production with crude glycerol was $1.1{\sim}2.5$ times higher than that with pure glycerol, indicating that crude glycerol is even a better substrate than pure glycerol for 1,3-PD fermentation. When glucose was added, 1,3-PD production and yield decreased in spite of enhanced cell growth. Furthermore, the addition of glucose was found to increase 2,3-butanediol, a by-product, significantly because of the change in metabolism in the presence of glucose. In semi-batch cultures without glucose addition, 26 g/L 1,3-PD was produced with crude glycerol, which was $2{\sim}3$ times higher than that with pure glycerol. Based on our results, it was clearly shown that crude glycerol is an effective substrate for biological 1,3-PD production, making it more feasible to produce 1,3-PD at a lower price.

The Effect of Paljeong-San Pharmacopuncture Treatment on Glycerol-Induced Acute Renal Failure in Rats (팔정산(八正散) 약침(藥鍼)이 글리세롤로 유발된 흰쥐의 급성 신부전에 미치는 영향)

  • Objective: The primary objective this study was to evaluate the effect of Paljeong-san (PJS) pharmacopuncture treatment on against the glycerol-induced acute renal failure in rats. Methods: Glycerol injection decreased GFR (glomerular filtration rate) and increased urine volume, serum creatinine, BUN level, urine albumine secretion and fractional excretion of Na+, K+. PJS was selected in the basis of invigorating kidney which can eliminate pathogens. Rats were treated with PJS pharmacopuncture on Shin-shu (BL23) and Chon-chu (ST25) point for 3 days, followed by 50% concentration of glycerol injection ($5m{\ell}/kg$ body weight). Results: After the 3 days treatment period, Paljeong-san (PJS) pharmacopuncture treatment improved renal function. In addition, Glycerol injection increased lipid peroxidation, and decreased Na-K-ATPase in renal cortex and which were prevented by PJS treatment. Conclusion: This study suggests that Paljeong-san (PJS) pharmacopuncture treatment show favorable effect on glycerol-induced acute renal failure in rats.

Effects of Various Cryoprotectants on the Survival of Frozen Mouse Embryo (항동해제의 종류가 동결 생쥐배의 생존성에 미치는 영향)

  • These experiments were carried out to clarify the effects various kinds of cryoprotectants which were frequently used in freezing embryos of domestic animals on the survival of frozen-thawed mouse embryos. As cryoprotectant, glycerol, DMSO and methanol were used and the procedures of adding them in medium were practiced by one-step or six-step adding method. Morphologically normal mouse embryos developed to blastocyst by in vitro culture after freezing and thawing were transferred to pseudopregnant recipients by surgical procedures. The results obtained in these experiments were summarized as follows: 1. The survival rates of the frozen-thawed 8-cell embryos, morulas and blastocysts following one-step addition of glycerol were 83.6, 80.3 adn 70.3%, respectively, while following six-step addition of glycerol, 69.2, 56.3 and 66.7% respectively. 2. When glycerol, DMSO and methanol were used as cryoprotectant under the same condition of freezing and thawing, the survival rates of frozen-thawed embryos were 74.0, 76.1 and 37.6%, respectively. 3. The implantation rate of embryos transferred to pseudopregnant recipients after freezing and thawing was 49.2%.
