  • Kidney allograft transplantation is the most effective method of renal replacement for end stage renal disease patients. Still, it is another kind of 'disease', requiring immunosuppression to keep the allograft from rejection(allograft immune reaction). Immune system of the allograft recipient recognizes the graft as a 'pathogen (foreign or danger)', and the allograft-recognizing commanderin-chief of adaptive immune system, T cell, recruits all the components of immune system for attacking the graft. Proper activation and proliferation of T cell require signals from recognizing proper epitope(processed antigen by antigen presenting cell) via T cell receptor, costimulatory stimuli, and cytokines(IL-2). Thus, most of the immunosuppressive agents suppress the process of T cell activation and proliferation.

The Histologic Findings and the Expression of Laminin in the Mucosa of the Rat Trachea During (백서 기관 점막의$SO_2$ 노출 후 회복과정의 조직학적 관찰 및 laminin의 발현에 관한 연구)

  • Background and Objectives : Sulfur dioxide gas is one of the major airborne Pollutants noxious to human in industrialized countries. The most vulnerable areas in the human respiratory system were the trachea and main bronchi and a gradient of decreasing damage was observed in the peripheral tracheobronchial tree. Induced functional alteration was increased mucosal permeability, and morphological changes were epithelial sloughing, intracellular edema, mitochondrial swelling, widened intercellular spaces, and ciliary cytoplamic extrusions. The laminins are a family of extracellular matrix glycoproteins localized in the basement membrane. Their primary role is cell-matrix attachment, but many additional biologic activities, including Promoting cell growth and migration, tumor growth and metastasis, wound repair, and graft survival, have been demonstrated. Materials and Methods : Histologic changes and expression of laminin in tracheal mucosa sacrificed at 1 day, 2 day, 3 day, 1 week, 2 weeks, and 3 weeks after continued SO2 exposure of 250 ppm for 30 minutes a day(to 7week) were studied in rats. In this study, mild immune reaction for laminin was noted at the apical cytoplasm of epithelial cells and basement membrane one day after a 7 week $SO_2$ exposure. The cilia and nucleoi of epithelial cells were normal and no immune reaction was noted in Goblet cells. The lamina propria of the tracheal tissue was infiltrated by monocytes and lymphocytes. Results : At 24 hours after exposure, all tracheal cells except Goblet cells revealed a mild immune reaction for laminin. No immune reactions were noted in the basement membrane. At 72 hours after exposure, mild or moderate immune reactions for laminin was seen in the tracheal cell cytoplasm. Irregular faint immune reaction for laminin was noted in the basement membrane. At 1 week after exposure, strong immune reaction for laminin was detected over all tracheal cells, and the basement membrane was seen clearly. At 2~3 weeks after exposure, strong immune reaction for laminin was seen in all tracheal epithelial cells except Goblet cells and a mild immune reaction was partly revealed in the basement membrane. Conclusion : Our study suggests that 502 produces histologic damage on the tracheal mucosa. Longer duration after exposure of $SO_2$ makes more progressive healing on the tracheal mucosa and increased immunoreactivity for laminin.

  • Erythrosine used as a colouring agent in drugs, cosmetics and foods in Korea, was examined for its effects on murine immune system and methemoglobin formation. As immunotoxicologic assay parameters, we adopted circulating leukocytes and immunoorgan weights for pathotoxicology, IgM plaque forming cells and arthus reaction for humoral immunity, delayed hypersensitivity reaction of cell mediated immunity and carbon clearacnce for macrophage function. Erythrosine's effects were observed as follows; 1. Ery throsine showed no significant effects on circulating leulocyte counts and relative immunoorgan weight. 2. Erythrosine diminished IgM plaque forming cells. 3. Erythrosine decreased arthus reaction, in the dose dependent manner. 4. Erythrosine had no significant effect on delayed hypersensitivity. 5. Phagocytic and corrected phagocytic index were not affected. 6. Methemoglobin content was similar in the test and control groups.

  • Both immune reaction and hypersensitivity reaction are occurred by the same mechanism, the antigen and antibody reaction. The favorable result of this reaction towards a host is called clinically an immune reaction and the opposite results is called an hypersensitivity reaction. Type IV hypersensitivity reaction is a delayed type which is related to the cellular immune reaction and a contact hypersensitivity is included in this type. Various dental materials such as metal (mercury, nickel, chrome, cobalt), resin and eugenol are etiologic substances. Patch test kit is composed of test substance with a controlled concentration which respond only to a susceptible patient and an aluminum chamber, and etiologic substances for hypersensitivity can be easily and comfortably found just by applying the kit to the patient's skin. In this case report, the patch test was performed to a patients with oral lichen planus and the allergen, restorative material was found. After removal of the matching restoration from the patient's mouth, the symptom was improved.

  • The immune system is comprised of cells and molecules whose collective and coordinated response to the introduction of foreign substance is referred to as the immune response. Defense against microbes is mediated by the early reaction (innate immunity) and the late response (adaptive immunity). Innate immunity consists of the epithelial barrier, phagocytes, complement and natural killer cells. Adaptive immunity, a more complex defense reaction, consists of activation of later-developed lymphocytes that, when stimulated by exposure to infectious agents, increase in magnitude and defensive capabilities with each successive exposure. In this review we discuss recent advances in important primary immune deficiency disorders of innate immunity (chronic granulomatous disease, leukocyte adhesion deficiency) and adaptive immunity (severe combined immune deficiency, Wiskott- Aldrich syndrome).

IMMUNE REACTION OF SPECIFIC SERUM ANTIBODIES TO PORPHYROMONAS ENDODONTALIS ANTIGEN IN PATIENTS WITH PERIAPICAL LESION (치근단 병소가 있는 환자에서 Porphyromonas endodontalis 항원에 대한 혈청 특이 항체의 면역 반응 연구)

  • Porphyromonas endodontalis is a black-pigmented anaerobic Gram-negative rod which is associated with endodontal infections and this microorganism possesses a potential for pathogenicity. The purpose of this study was to compare the membrane components of Porphyromonas endodontalis and Porphyromonas gingivalis and to study the immune reaction patterns of Porphyromonas endodontalis with patients with periapical lesion. Porphyromonas endodontalis (ATCC 35406), Porphyromonas gingivals serotypea (381), serotype b(W50), serotype c(A7A1-28) were cultured in anaerobic condition. Rabbit antisera were prepared by intravenous injection of formalized whole cells and human sera were obtained from patients and dental students. Indirect immunofluorescence method was used to study on the cross reaction between Porphyromonas endodontalis and Porphyromonas gingivalis serotype a, b, c antigen. Total membrane protein profiles of Porphyromonas endodontalis antigen were studied by sodium dodecyl sulfate polyacrylamide gel electrophoresis and the reactivity of antigenic components of Porphyromonas endodontalis against sera of patients and rabbit anti-Porphyromonas endodontalis antisera were assessed by Immunoblotting method. The following results were obtained : 1. Antigens of Porphyromonas endodontalis has multiple antigenic components, and both patients with periapical lesion and normal healthy individual showed immune response to this. 2. Patients group and healthy individual group showed a diversity of immune reaction pattern but they showed immune response against 43kd protein. 3. Patients with periapical lesion showed more diverse immune response than healthy individual and in some patients, much more bands appeared to lower molecular weight protein. 4. According to indirect immunofluorescence and Immunoblotting study, Porphyromonas endodontalis did not share common antigen with Porphyromonas gingivalis serotype a, b, c.

Effects of Squalene on the Immune Responses in Mice(I): Humoral Immune Responses of Squalene

  • Effects of squalene on humoral immune system in mice were investigated. Squalene exhibited significant increases in the circulating leukocyte counts and relative spleen and thymus weights of the mice. However, the relative liver weight was slightly decreased. Hemagglutination titers (HA) were signficantly enhanced by squalene while Arthus reaction was not affected. Splenic plaque forming cells (PFC) were also greatly increased by squalene, especially at doses of 50 and 100 mg/kg of it.

  • Some of organophosphate pesticides which are the most heavily used in Korea, were examined for their effects on the murine immune system. Immunotoxicological assay parameters adaopted in this study were Arthus reaction for humoral immunity, delayed type hypersensitivity reaction for cell mediate immunity, carbon clearance for macrophage function and susceptiility to tumor challenge. Subchronic exposure of rodents to the pesticides resulted in the marked suppression of immune functions and enhancement of susceptibility to tumor challenge. Among the pesticides tested (fenitrothion, fenthion, diazinon and EPN), fenitrothion was the most suppressive in Arthus and delayed type hypersensitivity reaction.

  • This study was performed to investigate the effect of olive oil diet on the immune response in ICR male mice. Experimental diets of 4 groups were fed ad libitum to the ICR male mice for 27 days. The results of this study were summarized as followings: 1. 10% Olive oil diet group as compared with the control diet group significantly decreased liver weight rate but significantly increased hemagglutination titer (HA), Arthus reaction, delayed type reaction (DTH), rosette forming cell (RFC), and phagocyte activity. 2. 20% Olive oil hypersensitivity diet group as compared with the control diet group significantly increased body weight gain, liver weight rate, and HA but significantly decreased Arthus reaction, DTH, RFC, phagocyte activity, and peripheral circulating white blood cell (WBC). 3. 30% Olive oil diet group as compared with the control diet group significantly increased liver weight rate but significantly decreased body weight gain, Arthus reaction, plaque forming cell (PFC), DTH, RFC, phagocyte activity, and WBC. The results showed that the increase of olive oil doses significantly decreased humoral and cellular immune responses, phagocyte activity, and WBC.

EFFECTS OF A CYTOTOXIC SUBSTANCE, PANAXYTRIOL FROM PANAX GINSENG C.A. MEYER ON THE IMMUNE RESPONSES IN NORMAL MICE

  • The effects of panaxytrion as known to be a cytotoxic substance isolated from Panax ginseng on the immune responses were examined. The i.p. administration of panaxytriol to normal mice for 6 consecutive days as doses of 5, 10 and 20 mg/kg suppressed the increase of body weight dose-dependently but did not affect the weight ratio of immunoorgans to body weight, No significant changes were observed in the humoral immune responses as measured by Arthus reaction and plaque forming cells and in the cellular immune response as measured by delayed hypersensitivity as well as phagocytic activity of reticuloendotherial system. These results suggested that panaxytriol, a cytotoxic substance to cancer cells, has no detrimental effects on the immune function in normal mice.
