Survey research on effect of Chinensis galla, Lithospermi radix, Salviae miltiorrhizae radix - containing cosmetics for improving dark circles (오배자.자초.단삼 함유 화장품의 다크써클 개선 효능에 관한 설문조사)

  • Objective : This clinical research was conducted to test patients with dark circles by external application with Chinensis galla, Lithospermi radix and Salviae miltiorrhizae radix in cosmetics. Methods : We gave prototype cosmetics to 27 patients who volunteered for this clinical research in Kyungwon university from October 14th, 2009 to November 11th. Left inferior palpebra area of patients was treated with Chinensis galla, Lithospermi radix and Salviae miltiorrhizae radix in cosmetics(Experimental group) and right inferior palpebra area of patients was treated with placebo(Control group) for 4weeks. We observed change of darkness of inferior palpebra and subjective satisfaction. Also taking digital image and color meter were used to evaluate the effects of Chinensis galla, Lithospermi radix and Salviae miltiorrhizae radix in cosmetics. Results : Overall evaluation was slightly positive. It is more effective on Right inferior palpebra than on Left inferior palpebra to improve darkness of inferior palpabra in this clinical research. Conclusion : We can guess that cosmetics with Chinensis galla, Lithospermi radix and Salviae miltiorrhizae radix has some therapeutical effects in reducing the symptoms of dark circles.

Study on Antioxidant Effect of Lithospermi Radix on Liver Cells Isolated from Oxidatively Stressed Rat (산화동물모델 흰 쥐 간세포에 대한 자근(紫根)의 황산화 효과 연구)

  • Objective : This study purposed to investigate the anti-oxidative effect of Lithospermi Radix (root of Lithospermum erythrorhizon S.) on liver cells isolated from oxidatvely stressed rat by AAPH. Method : We investigate effects of Lithospermi Radix(LR) and its fractions on normal liver cells' proliferation. And the amounts of SOD, GSH, catalase, NO, MDA production by liver cells isolated from the oxidatively stressed rat by AAPH also were measured after incubation with various fractions of LR extraction. Results : LR and its fracitons showed no toxicity on the normal liver cells from rat. LR and its fracitons increased the activity of SOD and reduced the amounts of NO and MDA in the liver cells from the oxidatively stressed rat. Conclusion : Lithospermi Radix could be supposed to have antioxidant effect on liver cells with no toxicity.

  • The present study was undertaken to compare the pharmacological activities of crude Lithospermi radix reported with the clinical uses in the oriental medicine. Crude Lithospermi radix uesd for the treatment of burn, eczema, blister, diuretic, scarlet fever and septicemia as antipyretic, antidotic and antiphlogistics. Therefore we tested the effects of Lithospermi radix water extract on the liver-protective activities in the rats. The results obtained from enzyme assay, measurement of serum and liver alanine. aspartate aminotransferase(ALT, AST) and lipid composition indicated that Lithospermi radix water extract showed significant liver-protective activities against benzo(a)pyrene-induced hepatotoxicity.

Antimetastatic effect of several crude drugs with antitumor activity on B16-Fo and A549 cells ( I ) (항암활성 수종생약의 B16-Fo와 A549 암세포에 대한 항전이 효과( I ))

  • For the development of antimetastatic agent 41 kinds of crude drugs were used for the evaluation of inhibitory effect of several crude drugs on cell adhesion of pulmonary cancer cells and platelet aggregation. Results were obtained as follows: 1. Water extracts of crude drugs inhibited cell adhesion of A549 to complex extracelluar matrix over 40 % of contol were Houttuyniae Herba, Mylabris, Rhei Radix et Rhizoma, Meliae Cortex, Ferula Resina, Oldenlandiae diffusae Herba at the higher concentration of $10^{-3}g/ml$ while those inhibiting cell adhesion of Bl6-Fo over 40 % of control were $10^{-5}g/ml$ of Houttuyniae Herba, Aurantii Fructus, Lithospermi Radix, Zedoariae Rhizoma. Prunellae Spica, Foeniculi Fructus, Rbei Radix, Scutellariae Radix, Meliae Cortex, Ferula Resina and Oldenlandiae diffusae Herba. 2. MeOH extracts of crude drugs at the concentration of $4{\times}10^{-4}g/ml$ inhibiting cell adhesion of A549 specifically to single extracelluar matrix over 40 % of control were Lithospermi Radix, Agrimoniae Herba, Rhei Radix and Ferula Resina to collagen I, Houttuyniae Herba, Lithospermi Radix, Bupleuri Radix, Salviae miltiorrhizae Radix, Orostachys Herba, Sappan Lignum, Meliae cortex ferula Resina and Coicis Semen to collagen Ⅳ, Mylabris, Agrimoniae Herba to laminin, Houttuyniae Herba and Meliae Cortex to fibronectin. 3. NeOH extracts of crude drugs at the concentration of $4{\times}10^{-4}g/ml$ inhibiting cell adhesion of B16-Fo specifically to single extracelluar matrix over 60 % of control were Lithospermi Radix, Salviae miltiorrhizae Radix, Meliae Cortex and Ferula Resina to collagen I, Lithospermi Radix, Bupleun Radix, Saiviae miltiorrhizae Radix, Ferula Resina and Acanthopanacis Cortex to collagen Ⅳ, Bupleuri Radix, Orostachys Herba to laminin, Houttuyniae Herba to fibronectin. 4. MeOH extracts of crude drugs inhibiting platelet aggregation over 40% of ADP control were at the concentration of $50{\mu}g/m{\ell}$ of Houttuyniae Herba, Angilicae gigantis Radix, Zedoariae Rhizoma. Coicis Semen and $100{\mu}g/m{\ell}$ of Ferula Resina, Orostachys Herba, Salviae miltiorrhizae Radix, Curcumac Radix, Carthami Flos, Lithospermi Radix, Gleditsiae Spina, Sappan Lignum, Acanthopanacis Cortex. These results suggest that several crude drugs including Ferula Resina, Houttuyniae Herba, Lithospermi Radix and Salviae miltiorrhizae Radix chiefly have more possibility to exert antimetastatic activity and require in vivo antimetastatic study.

A Case of Liver Cirrhosis Patient`s Senile Pruritus Treated by External Preparation Containing Ponciri fructus and Radix lithospermi (지실과 자초 전탕액 외치법을 병행한 간경변 환자의 노인성 가려움증 치험 1례)

  • Objectives : Pruritus is a skin disorder that usually occurs in the elderly. The purpose of this study is to report the effect of external preparation containing Ponciri fructus and Radix lithospermi on senile pruritus and xeroderma with liver cirrhosis.Methods : We treated 85-years-old liver cirrhosis patient suffered from itching sensation on the whole body with external preparation containing Ponciri fructus and Radix lithospermi and oral administration of Sodalgeonbitang for 37 days.Results : Despite discontinuing western medical treatment of topical steroids and oral antihistamine, pruritus and erythema of the patient were significantly improved.Conclusions : As for external application, decoction of Ponciri fructus and Radix lithospermi might be effective used for treatment senile pruritus and xeroderma.

Enantiomeric Ratio of Shikonin Derivatives as a Possible Key for the Determination of the Origin of Lithospermi Radix

  • An HPLC method was developed to resolve the enantiomers of shikonin derivatives of the Lithospermi Radix. The optimum mobile phase on a Chiracel AD column was 5% isopropanol in n-hexane with folw rate of 1 ml/min. Establishment of this method made possible to determine the ratios of shikonin/acetylshikonin or alkanin/acetylalkanin in the same root. The correlation of the ratios of these substance pairs appeared characteristic for the country where they were originated from. All of the Korean species showed significantly higher ratios of shikonin/acetylshikonin and alkanin/acetylalkanin than the Chinese ones. this method would be useful to determine the orgin of Lithospermi Radix.

An Active Fraction on Phagocytosis of Murine Peritoneal Macrophage in Lithospermi Radix (자근에 함유된 복강 마크로파지의 탐식능 활성분획)

  • The phagocytic activity of murine peritoneal macrophages was determined by lucigenin chemiluminescence with luminometer and engulfment of fluorescein-conjugated E. coli particles. 70% MeOH extract of Lithospermi Radix was fractionated successively with hexane, methylene chloride, n-BuOH and water. The water fraction (m.w. 500 to 1,000) enhanced the lucigenin chemiluminescence and the engulfment of fluorescein-conjugated E. coli particles in murine peritoneal macrophages. The water fraction suppressed the production of nitric oxide in the macrophages. These results suggest that an active fraction of phagocytosis in Lithos-permi Radix is the water fraction and the molecular weight is 500 to 1,000.

  • Ethyl ether fractions of Ferula Resina(EFR) and Lithospermi Radix(ELR) and hexane fraction of Salviae miltiorrhizae Radix(HSR) exerted an inhibitory effect on cell adhesion of A549 to extracelluar matrix most effectively in vitro cell adhesion assay. Thus, with above fractions for the evaluation of antitumor activity, T/C% was monitored in ICR bearing S-180 and for antimetastatic effect, pulmonary colonization assay, weight of organs, changes of WBC and platelet were studied. In pulmonary colonization assay incidence rate to control was 73 % 42 %, 14 % in ELR, HSR and EFR-treated groups repsectively. Gain of lung weight was significantly inhibited in all groups while spleen weight was significantly reduced only in SMR group, but no changes in kidney and liver as compared with control. Number of platelet was significantly increased in all groups to normal range as compared with thrombocytopenic contol. WBC was significantly reduced only in LR group. These results suggest that ethyl ether fraction of Ferula Resina has more effective antimetastatic activity.

Isolation of Constituents of Lihtospermum erythrorhizon and Development of an Analytical Method of its Constituents by HPLC-UV (자근의 성분 분리 및 이들 성분의 HPLC-UV 동시분석법 개발)

  • Lithospermi radix, the dried roots of Litospermum erythrorhizon Sieboid et Zuccarini (Boraginaceae), has long been used to treat detoxification and inflammation. In this study, we isolated two main quinoid compounds, ${\beta}$-hydroxyisovalerylshikonin (1) and acetylshikonin (2) from L. erythrorhizon. As acetylshikonin is considered as a marker compound of L. erythrorhizon, a rapid analysis method for the simultaneous determination of quinoid compounds including 2 was also developed by HPLC (High Performance Liquid Chromatography) and validation of this analytical method. By the developed method, two quinoid marker compounds (${\beta}$-hydroxyisovalerylshikonin and acetylshikonin) were successfully quantified in 31 commercial samples which were collected from different regions. The contents were 0.20% (${\beta}$-hydroxyisovalerylshikonin) and 0.22% (acetylshikonin), respectively.
