자궁경부암세포에 대한 천화분(天花粉)의 성장억제 및 세포사멸효과 (Growth Inhibition and Apoptosis Induction of Trichosanthis Radix Extract on Human Uterine Cervical Carcinoma Cells)

  • Purpose : Trichosanthis Radix is traditional medical herb which has been shown to inhibit tumor cell proliferation. In this study, the effects of Trichosanthis Radix extract were investigated on inducing growth inhibition and apoptosis of human uterine cervical carcinoma cells. Methods : Human uterine cervical carcinoma cells line, ME-180, was used for the study. The cells were treated with varying concentrations of Trichosanthis Radix extract. Cell growth and inhibitory rate were measured by MTT assay. Apoptosis induction was detected by fluorescence microscopy, DNA ladder formation and flow cytometry. Results : Trichosanthis Radix extract inhibited the growth of human uterine cervical carcinoma cells in a dose-dependent manner. It induced ME-180 cells to undergo apoptosis including fragmented nuclei and nucleosome-sized DNA fragmentation. Flow cytometric analysis showed the increasing rate of apoptotic cells by Trichosanthis Radix extract. Reduction of mitochondrial membrane potential and increase in caspase-3 activity and were found in ME-180 cells treated with Trichosanthis Radix extract. Conclusion : Our data suggest that Trichosanthis Radix extract inhibit the growth and proliferation of ME-180 cells by apoptotic induction and facilitates its activity via caspase-3 activation initiated by depolarization of mitochondria.

과루인이 자궁경부암세포의 성장억제 및 세포고사에 미치는 영향 (Growth Arrest and Apoptosis of Human Uterine Cervical Carcinoma Cells Induced by Trichosanthes Semen Extract)

  • To investigate the effects of Trichosanthes semen extract on the growth and apoptosis of human uterine cervical carcinoma cells. Effects of Trichosanthes semen extract on the growth of ME-180 cells were assayed by MTT assay. Apoptosis induced by Trichosanthes semen extract was detected by fluorescent microscopy, DNA fragmentation analysis and flow cytometry. Caspase-3 and caspase-8 activities were assayed. Trichosanthes semen extract induced ME-180 cells to die in a dose- and time-dependent manner. ME-180 cells treated with Trichosanthes semen extract exhibited typical characteristics of apoptosis. The population of Sub-G1 cells increased significantly, and the cells represented the reduced size, condensed chromatin and apoptotic bodies. They showed the decreased mitochondrial membrane potential and increased activities of caspase-3 and caspase-8. The results suggest that Trichosanthes semen extract induced ME-180 cell apoptosis and the activation of caspase and mitochondrial pathway were involved in the process of Trichosanthes semen extract-induced apoptosis.

  • Purpose : Polygonum cuspidatum extract is an oriental herb which has been used for uterine diseases. In this study, the effects of Polygonum cuspidatum extract were investigated on inducing growth inhibition and apoptosis of human uterine cervical carcinoma cells. Methods : Viability of Polygonum cuspidatum extract-induced ME-180 cells was measured by MTT assay. Apoptotic cells were visualized by EtBr/AcOr staining under fluorescent microscope. Nucleosomal DNA fragmentation was assayed by agarose gel electrophoresis. Cell cycle distribution and changes in mitochondrial membrane potential were observed by flow cytometry. Results : Polygonum cuspidatum extract induced ME-180 cell death in a dose- and time-dependent manner. In the cells treated with Pc, the population of cells at sub-G1 phase significantly increased, and the condensed nuclei, apoptotic bodies and nucleosome-sized DNA were detected. Moreover, reduction in mitochondrial membrane potential was detected. Conclusion : Polygonum cuspidatum extract inhibits the growth and proliferation of ME-180 cells by apoptotic induction and facilitates its activity initiated by depolarization of mitochondria.

행인(杏仁)에서 추출한 Amygdalin의 자궁경부암세포 ME-180에 대한 항암 효과 (Anti-tumor Effect of Amygdalin extracted from Armeniacae Amarum Semen on Human Cervical Cancer Cell ME-180)

  • Objectives: Amygdalin is abundant in the seeds of bitter almond and apricots of the Prunus genus and other rosaceous plants. Amygdalin is known to have antitussive and anticancer activities. Apoptosis, also known as programmed cell death, is an important mechanism in cancer treatment. Methods: In the present study, we investigated whether the aqueous extract of Amygdalin induces apoptotic cell death in ME-180 cervical cancer cells. For this study, 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay, terminal deoxynuclotidyl transferase (TdT)-mediated dUTP nick end-labeling (TUNEL) assay, 4,6-diamidino-2-phenylindole (DAPI) staining, flow cytometric analysis, DNA fragmentation assay, Western blot, and caspase-3 enzyme assay were performed on ME-180 cervical cancer cells. Results: Through morphological and biochemical analyses, it was demonstrated that ME-180 cells treated with Amygdalin exhibit several apoptotic features. The treatment of Amygdalin increased the Bax expression and caspase-3 enzyme activity and decreased Bcl-2 expression. Here, we have shown that Amygdalin induces apoptotic cell death in ME-180 cervical cancer cells through Bax-dependent caspase-3 activation. These results suggest the possibility that Amygdalin exerts anti-tumor effect on human cervical cancer.

  • Tumor necrosis factor-${alpha}$ (TNF) induced a cytotoxic response in ME-180 cervical carcinoma cells in vitro. This cytotoxic response was accompanied by a temporal series of mitogenic stimuli : increased c-fos, c-jun and jun-B expression. Depletion of protein kinase C (PKC) by exposure of ME-180 cells to 100ng/ml phorbol myristate acetate (PMA) for 24hours almost completely abolished TNF-mediated increase in these signals, indicating that a PKC-dependent pathway is involved in TNF-mediated increases in the expression of c-fos, c-jun and jun-B. Characteristics of TNF receptors after exposure to 100ng/ml PMA or 24hours were not altered, suggesting that diminished induction of these oncogenes by TNF after PMA treatment is not due to any changes at the receptor level. To examine whether a PKC-dependent pathway is involved in TNF-mediated cytotoxicity in ME-180 cells, cytotoxicity was measured after depletion of PKC. No apparent changes in cytototoxicity after PKC depletion suggest that a PKC-dependent pathway is not involved in TNF-mediated cytotoxicity. Furthermore, results from cytotoxicity tests after exposure to staurosporine (PKC inhibitor) did not show any changes in the TNF-mediated cytotoxicity, confirming that a PKC-dependent pathway is not involved in this process. These data indicate that 1) TNF induces expression of c-fos, c-jun and jun-B oncogenes via a PKC-dependent pathway and 2) PKC-dependent expression of these three oncogenes by TNF may not be involved in TNF-mediated cytotoxicity in ME-180 cells.

TNF에 대한 내성획득에서 MnSOD의 역할에 관한 연구 (The Role of MnSOD in the Mechanisms of Acquired Resistance to TNF)

  • 연구배경 : 종양괴사인자(tumor necrosis factor ; TNF)는 다양한 생물학적 기능을 가지고 있는 바, 그 중 생체 외에서 증명된 뚜렷한 항암 효과로 말미암아 최근 항암 유전자요법의 중요한 대상으로 관심을 모으고 있다. 현재 유전자 이입의 기술적 문제로 생체 외에서 암세포에 유전자 이입을 시행한 후 이를 다시 환자의 생체내로 이식하는 방법이 연구의 주종을 이루고 있다. 그러나 저자들의 과거의 연구를 포함한 여러 연구에서 TNF가 이입된 암세포는 TNF에 대해 내성을 보이는 이에는 새로이 방어 단백질을 합성하는 것이 관여할 것이라는 시사가 있었다. 이 획득내성의 기전을 밝히는 것이 종양생물학의 이해를 넓히고 보다 효과적인 항암 유전자요법을 개발하기위한 매우 중요한 과제로 생각된다. 저자들은 TNF 유전자 이입에 따른 암세포의 TNF에 대한 획득내성에, 일부 세포에서 TNF에 의해 발현이 유도된다는 것이 밝혀진, 항산화효소의 하나인 MnSOD의 발현의 변화가 관여하는 지를 규명하고자 본 실험을 수행하였다. 방 법 : TNF에 다양한 감수성을 보이는 인체 및 생쥐 기원의 4가지 암세포주(WEHI164, NCI-H2058, A549, ME180)에 TNF-$\alpha$ 유전자를 retroviral 이용하여 이입하고 TNF의 발현을 시도하여 PCR, ELISA, MTT assay로 확인하였고, TNF 유전자가 이입된 세포(WEHI164-TNF, NCI-H2058-TNF, A549-TNF, ME180-TNF)는 TNF에 내성을 보이는지 역시 MTT assay로 검증하였다. TNF 유전자 이입 전후의 MnSOD mRNA 발현의 차이는 Northern blot analysis를 통하여 비교하였다. 결 과 : 1) TNF-$\alpha$ 유천자 이입 및 발현 확인 PCR을 시행한 결과 TNF 유전자가 이입된 각 세포 790 base pair 표기의 진한 DNA band를 보인 반면 모세포주는 보이지 않아서 retroviral vector를 이용한 유전자 이입이 DNA 수준에서 이루어 있었다. 그리고 TNF 유전자가 이입된 세포의 배양상층액에서 TNF양을 ELISA로 측정한 결과 TNF를 세포에 따라 1.91ng/24hr/$10^6\;cells$에서 3.91ng/24hr/$10^6\;cells$ 생산함을 알 수 있었다. 2) TNF 유전자 이입 전후, 암세포의 TNF에 대한 감수성 비교 TNF 농도 100ng/ml에서 WEHI164-TNF와 ME180-TNF 세포는 통계적으로 유의하게 (p<0.01) TNF에 대한 내성을 획득함을 알 수 있었다. 3) TNF 유전자 이입 전후외 MnSOD mRNA 발현양상 TNF에 감수성을 보이는 WEHl164와 ME180세포는 TNF 유전자 이입 후에 MnSOD mRNA 발현이 증가되지 않았으며, TNF에 내성을 보이는 NCIH2058과 A549 세포는 TNF 유전자 이입 후에 MnSOD mRNA 발현의 증가가 관찰되었으나 이는 모세포에 외부에서 TNF를 주었을 때도 관찰되었다. 결 론 : 세포주에 TNF 유전자를 이입하여 TNF를 발현하게 하였을때 세포 자신은 TNF에 대해 내성을 하게 되는데, 이 획득내성은 MnSOD 발현 능력의 향상에 의한 것은 아닐 것으로 판단되나 각 세포주의 자연 상태의 TNF에 대한 감수성 여부는 MnSOD의 발현 차이가 관련될 것으로 생각된다.

Amygdalin Extract from Armeniacae Semen Induces Apoptosis through Bax-dependent Caspase-3 Activation in Human Cervical Cancer Cell Line ME-180

Effect of troglitazone on radiation sensitivity in cervix cancer cells

  • Purpose: Troglitazone (TRO) is a peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) agonist. TRO has antiproliferative activity on many kinds of cancer cells via G1 arrest. TRO also increases $Cu^{2+}/Zn^{2+}$-superoxide dismutase (CuZnSOD) and catalase. Cell cycle, and SOD and catalase may affect on radiation sensitivity. We investigated the effect of TRO on radiation sensitivity in cancer cells in vitro. Materials and Methods: Three human cervix cancer cell lines (HeLa, Me180, and SiHa) were used. The protein expressions of SOD and catalase, and catalase activities were measured at 2-10 ${\mu}M$ of TRO for 24 hours. Cell cycle was evaluated with flow cytometry. Reactive oxygen species (ROS) was measured using 2',7'-dichlorofluorescin diacetate. Cell survival by radiation was measured with clonogenic assay. Results: By 5 ${\mu}M$ TRO for 24 hours, the mRNA, protein expression and activity of catalase were increased in all three cell lines. G0-G1 phase cells were increased in HeLa and Me180 by 5 ${\mu}M$ TRO for 24 hours, but those were not increased in SiHa. By pretreatment with 5 ${\mu}M$ TRO radiation sensitivity was increased in HeLa and Me180, but it was decreased in SiHa. In Me180, with 2 ${\mu}M$ TRO which increased catalase but not increased G0-G1 cells, radiosensitization was not observed. ROS produced by radiation was decreased with TRO. Conclusion: TRO increases radiation sensitivity through G0-G1 arrest or decreases radiation sensitivity through catalase-mediated ROS scavenging according to TRO dose or cell types. The change of radiation sensitivity by combined with TRO is not dependent on the PPAR ${\gamma}$ expression level.

Antitumor and Immunopotentiating Effects of Manda Enzyme

  • This study was to evaluate the antitumor and immunopotentiation effects of Manda Enzyme (ME). Oral administration of ME (0.2ml/mouse) to tumor bearing mice significantly prolonged survival rate compared to the control group with the prolongation ratio of 40%. The inhibition ratios for the first and the second experiments were 51.8% and 26.4%, respectively. Only the spleen index was significantly increased in the MEF-treated group, but not in the control group. Gamma globulin level of the MEF-treated group was elevated when mice were injected with sarcoma-180 cells on the left groin. Activities of natural killer (NK) and lymphokineactivated killer (LAK) cells were observed by $^{51}Cr-release$ method. Activities of NK cell against YAC-1 cells were significantly increased in the MEF treated group. And LAK cell activities against P815 cells were also significantly increased in the experimental group. These observations, therefore, suggest that ME may have an anticancer effect and immunopotentiating effect in vivo.

저령(Grifola umbellata)의 균핵에서 추출한 조다당류의 면역활성 및 항암 효과 (Immuno-modulatory and Antitumor Effect of Crude Polysaccharides Extracted from Sclerotium of Grifola umbellata)

  • 저령의 균핵으로부터 중성염용액, 열수 및 메탄올 추출물을 분리하였다. 세포독성 실험 결과, 중성염용액 추출물은 $0{\sim}2,000\;{\mu}g/ml$의 농도에서 NIH3T3, Sarcoma 180 및 MCF-7에 대한 세포독성이 없었으나, 메탄올 추출물에서는 $1,000\;{\mu}g/ml$ 이상의 농도에서는 독성을 나타내었다. Sarcoma 180 복수암에 대한 항암 효과는 중성염용액 추출물을 투여한 실험군에서 66.74%의 높은 생명 연장 효과를 나타내었으며, 암세포의 생 세포 수 또한 54.2% 감소시키는 효과를 나타내었다. 중성염용액 추출물은 대조군에 비해 보체 대체 경로에서의 항보체 활성을 $85.05{\sim}88.73%$, B 임파구의 alkaline phosphatase 활성을 6배 이상 증가시킴으로써 면역 활성 효과를 향상시켰다. 또한 중성염용액 추출물을 50 mg/kg body weight의 농도로 마우스 복강에 투여하였을 때 대조군에 비하여 복강세포수가 1.7배 증가하였으며, 혈액 내 백혈구 수 또한 3.6배의 증가를 나타내었다. 간, 비장 및 흉선 등의 면역 관련 장기의 체중에 대한 중량을 측정한 결과, 중성염용액 추출물 투여군은 대조군에 비해 증가된 수치를 보였으며, 혈액생화학적 검사를 시행한 결과, 대조군과 유사한 경향을 나타내었다. 중성염용액 추출물의 총 다당류와 단백질의 함양은 각각 98.25%와 1.44%로 측정되었다. 따라서 저령균핵의 중성염용액 추출물의 항암 효과가 암세포에 대한 직접적인 세포독성에 의한 것이 아니고 면역활성에 의한 것으로 판단된다.
