Antidepressant-like effects of Nelumbo nucifera leaves extract in chronic mild stress model (만성스트레스 모델에서 하엽추출물의 항우울 효과)

  • Objectives : Chronic mild stress (CMS) model is currently recognized as a better animal model of depression. The purpose of this study was to investigate the antidepressant-like effects of the Nelumbo nucifera leave extract using CMS model. Methods : The antidepressant-like effects of Nelumbo nucifera leaves extract was determined by using animal models of depression. Male ICR mice were divided into four groups: saline-treated normal, without CMS; saline-treated stress control; CMS+ Imipramine(20mg/kg); CMS+Nelumbo nucifera leaves extract(200mg/kg). All mice except the normal group exposed an unpredicted sequence of chronic mild stressors for 5 weeks. The behavior of mice were detected by sucrose preference test, forced swim test and tail suspension test. Then concentration of corticosterone in serum was detected by enzyme immunoassay. Results : Nelumbo nucifera leaves extract administration by daily gavage from the 3rd week exhibited an antidepressant-like effect on CMS-induced depression in mice. Nelumbo nucifera leaves extract administration at dose of 200 mg/kg significantly increased the sucrose consumption, and decreased the immobility durations in forced swim test and tail suspension test. Furthermore the corticosterone level decreased than control group. Conclusions : Chronic mild stress can affect mouse behavior and corticosterone level and cause depression. The present experiments not only further confirm the antidepressant-like effects of Nelumbo nucifera leaves extract in the sucrose preference test, forced swimming test and tail suspension test, but also the improving effects of Nelumbo nucifera leaves extract on the depression-like symptoms in the CMS model. Nelumbo nucifera leaves extract has the antagonism on CMS and produce antidepressive effects.

  • Antioxidative, anticoagulant, and α-glucosidase inhibitory effects of 60% (v/v) ethanolic extract from lotus (Nelumbo nucifera) leaves were investigated in vitro. The yield and polyphenol content of the lotus leaves extract were 35.3±1.1% and 186.2±5.2 ㎍ gallic acid equivalents/mg, respectively. The lotus leaves extract effectively scavenged free and cation radicals, and nitrite in a concentration-dependent manner, and possessed potent reducing power. The lotus leaves extract had remarkable anticoagulant and α-glucosidase inhibitory activities as a function of the extract concentration.

Antioxidative Activity and Nitrite Scavenging Ability of Ethanol Extract from Nelumbo nucifera Leaves (연잎 에탄올 추출물의 항산화 효과 및 아질산염 소거능)

  • To confirm possibility of Nelumbo nucifera leaves as biofunctional material, we investigated the antioxidant activity and nitrite scavenging ability of ethanol extract from Nelumbo nucifera leaves. Nelumbo nucifera leaves were extracted with ethanol and concentrated under vacuum using rotary evaporator. Then, antioxidative activity and nitrite scavenging ability of the extract were examined in vitro. Electron -donating ability of the extract at RC50 was 90.19 ${\mu}g/mL$. After addition of 0.96 mg/mL, autooxidation of pyrogallol decreased to 66.19% by superoxide dismutase-like activity. In antioxidative activity of the extract against linoleic acid during incubation times of 24, 48, and 96 hours at $40^{\circ}C$, lipid peroxidation values significantly decreased to 72.53%, 82.00%, 84.69% with addition of 0.2 mg/mL, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and the value revealed to be $282.84\;{\pm}\;9.03$ GAE ${\mu}g/mg$ of the extract. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, exhibited to 45.55% by addition of 0.2 mg/mL. These results suggest that ethanol extract from N. nucifera leaves can be used as bioactive and functional material.

  • The natural plants were screened for new cosmetic materials. Many parts of Nelumbo nucifera has been investigated for anti-wrinkling and anti-furrowing effects. The leaves. flowers and stamens of Nelumbo nucifera were extracted with ethanol or distilled water. The effects of cell viability and $H_2O_2-induced$ cytotoxicity were studied on CCD-986sk human fibroblast cell line by MTT assay. We measured various effects related to skin such as the synthesis of MMP-1 and type I procollagen. The ethanol extract of Nelumbo nucifera leaves (ENL) at concentrations of 30 ${\mu}g/ml$ showed maximum cell viability on CCD-986sk cells by 129.70%. and $H_2O_2-induced$ cytotoxicity was blocked by ENL (5. 10. 20. 30. and 40 ${\mu}g/ml$) concentration-dependently. The synthesis of MMP-1 and type I procollagen were positive in ENL. These results suggest that ENL has anti-oxidant and anti-furrowing effects.

  • Objectives : The purpose of the study is to investigate the activities of Nelumbo nucifera leaf extracts on Streptococcus mutans, Streptococcus mitis, Streptococcus sobrinus, Porphyromonas gingivalis, Prevotella intermedia, and Treponema denticola. Methods : The inhibitory effect of lotus leaf extracts on the growth of oral bacteria was assessed in experiments with extracts from freshly harvested and pulverized lotus leaves and bacterial cultures of dental caries. Results : The results showed that N. nucifera extracts possess antimicrobial activity on all bacterial strains. The minimal inhibitory concentration (MIC) values varied from 4 mg/ml to 10 mg/ml against antimicrobial activity. The relative growth ratio (RGR) against of N. nucifera extracts were determined as 50% in concentration of 4.0 mg/ml. The extract of N. nucifera was effective in reducing on the glucosyltransferase (GTase) activity of six strains in vitro. Conclusions : Methanol extracts of lotus leaves showed antimicrobial effects on three bacterial species causing dental caries and three bacterial species causing periodontitis, as well as inhibitory effects on GTase activity.

Antioxidant and Anti-wrinkling Effects of Extracts from Nelumbo nucifera leaves (하엽(荷葉) 추출물이 항산화 효능 및 피부노화에 미치는 영향)

  • Objective : The purpose of this study was to investigate anti-aging and antioxidant effects of extracts of Nelumbo nucifera leaves (NN-L) using ethanol on skin .Methods : Each part of leaves(NN-L), flowers(NN-F) and stem(NN-S) was extracted with 70% ethanol. We performed radical scavenging assay(DPPH, ABTS+, Superoxide anion radical), elastase inhibition assay, collagenase inhibition assay. NN-L extracts were tested for cell viability(MTT assay), MMP-1 inhibition and MMP-1 protein expression on CCD-986sk cells (human fibroblast line).Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. We measured its free radical scavenging activity, elastase inhibitory activity and expression of MMP-1 (matrix metalloprotease-1) in human fibroblast cells. Among the parts of Nelumbo nucifera, NN-L showed the highest antioxidant activities and in radical scavenging. DPPH, ABTS+ and Superoxide anion radical scavenging activity of NN-L at concentration of 1,000 μg/mL were 91.43%, 99.31% and 73.7% respectively. In vitro elastase and collagenase inhibition effects of NN-L at concentration of 1,000 μg/mL was 42.8% and 55.3% respectively. The ethanol extract of NN-L showed cell viability of 95.4% in 50 μg/mL concentration. In addition, The results from Western blot assay showed that NN-L decreased the expression of MMP-1 protein in a dose-dependent manner (by up to 35.0% at 50 μM).Conclusion : The findings suggest that the NN-L great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

  • The cell surface sculpture of the plant epidermis has received great interest recently. It has also been an active area of research, as the biological microstructures of the surface, such as papillae and waxes, exhibit several unique properties, including self-cleaning character; namely the "Lotus effect" first described in the leaves of the lotus, Nelumbo nucifera. The Lotus effect is the phenomenon in which the super-hydrophobic and water-repellent nature of lotus leaves allow water drops to run off easily on the surface in a rolling and sliding motion thereby facilitating the removal of dirt particles. It is well-known that surface roughness on the micro- and nanoscale is a primary characteristic allowing for the Lotus effect. This effect is common among plants and is of great technological importance, since it can be applied industrially in numerous fields. In the present study, Nelumbo nucifera leaf and stem epidermal surfaces have been examined with a focus on the features of papillae and wax crystalloids. Both young and mature Nelumbo nucifera leaf epidermis demonstrated the Lotus effect on their entire epidermal surface. The central area of the upper epidermis, in particular, formed extremely papillose surfaces, with an additional wax layer, enabling greater water repellency. Despite the presence of wax crystalloids, epidermal surfaces of the lower leaf and stem lacking papillae, were much more easily wetted.

Inhibitory Effects of Nelumbo nucifera on Tyrosinase Activity and Melanogenesis in Clone M-3 Melanocyte Cells (연잎, 연꽃 및 연꽃 수술 추출물의 Tyrosinase 활성억제 및 Melanin 생성억제에 의한 미백 효과)

  • Objectives : We examined three parts of Nelumbo nucifera that might be useful for skin-whitening effect. Each parts of leaves, flowers and stamen were extracted with water or 70% ethanol. Methods : This study investigated inhibitory effects of each extracts on tyrosinase activity. Extracts were tested for cytotoxicity on clone M-3 melanocyte cells, melanin inhibitory activities were further assessed. Results : The water extract of Nelumbo nucifera stamen. ethanol and water extracts of flowers exhibited inhibitory effects on tyrosinase (IC50 values 726.16, 1063, and 1732.36 ug/mL, respectively). The water extract of Nelumbo nucifera stamen, ethanol extract of flowers, both ethanol and water extracts of leaves showed relatively lower cytotoxicity, with cell viability above 80% with a concentration of 20 ${\mu}g/mL$. In addition, The water extract of Nelumbo nucifera stamen inhibited the melanin production in clone M-3 melanocyte cells. Conclusions : Among each parts of Nelumbo nucifera. the water extract of stamen was the strongest candidates for skin-whitening cosmetic application.

  • Phytochemical investigation of the MeOH extract of the leaves of Nelumbo nucifera resulted in the isolation of five norsesquiterpenes, four flavonoids, two triterpenes and one alkaloid. Their chemical structures were characterized by spectroscopic methods to be (E)-3-hydroxymegastigm-7-en-9-one (1), (3S,5R,6S,7E)- megastigma-7-ene-3,5,6,9-tetrol (2), dendranthemoside B (3), icariside $B_2$ (4), sedumoside $F_1$ (5), luteolin (6), quercetin 3-0-${\beta}$-D-glucuronide (7), quercetin 3-0-${\beta}$-D-glucoside (8), isorhamnetin 3-0-rutinoside (9), alphitolic acid (10), maslinic acid (11), and N-methylasimilobine (12). Norsesquiterpenoids (1-5) and triterpenes (10-11) were isolated for the first time from this plant. Compounds 6 and 10-12 exhibited considerable cytotoxicity against four human cancer cell lines in vitro using a SRB bioassay.

Nelumbo nucifera Leaves Extract Reduced the Production of Hepatitis B Surface Antigen on HepG2.2.15 (연잎추출물의 B형 간염 표면 항원 발현 억제 효과)

  • Nelumbo nucifera (lotus) is known to be a useful medicinal plant and leaf extract contains several flavonoids and alkaloids. To analyze the effect of Nelumbo nucifera leaves extract (NNL) on the HBsAg production, we treated NNL on HepG2.2.15 cells which contain the hepatitis B viral genome and secrete surface antigen into media. NNL suppressed the production of hepatitis B surface antigen as a dose-dependent manner. To analyze the effect of NNL on HBV DNA replication, PCR analysis was performed. NNL was not affected the HBV DNA replication and HBsAg mRNA expression. To understand the effect of NNL on the production of HBsAg, we carried out the analysis of lipid-metabolizing gene expression using one-step RT-PCR. NNL reduced the gene expression of FASN and SREBP2 and increased the expression of LDLR. Triglyceride content of HepG2.2.15 cells was not decreased by treatment of NNL. This result suggests a possibility that NNL may have an effect for the inhibition of hepatitis B surface antigen by modulation of lipid and cholesterol metabolism.
