Effect of Water Extract of KH-304 on Anti-oxidant and Erectile dysfunction in Old Rat (KH-304의 항산화효과 및 Old rat model 에서의 Erectile dysfunction에 미치는 영향)

  • The fallowing are the result of the experimental studies of KH-304 on the penile nitric oxide synthase (NOS) activity, SOD and the level of blood testosterone in old rat. KH-304 was tested for the expression and activity of eNOS, nNOS, SOD/Mn, caveol in-1, 3 in penis of old-rat. The penile expression level of the five enzyme were increased significantly after oral administration of the KH-304 100mg/kg for 100ays and the concentration of testosterone in the blood were increased. In vitro, the effect of SOD and free radical scavenging were increased significantly. Conclusively, KH-304 is capable of improving of sexual ability in old-rat.

Effect of Ginseng Ethanol Extract on Lactate Dehydrogenate-5 in Rat Brain with Age (인삼 Ethanol Extract가 쥐의 뇌 Lactate Dehydrogenase-5에 미치는 영향)

  • Attempts were made to see if feeding of ginseng ethanol extract could affect proper- ties of rat brain lactate dehydrogenase such as specific activity, heat stability, Km for substrate, inactivation by 3-bromopyruvate and trypsin, and immune response. The following results were obtained. Specific activity of LDH was observed to reach maximum in 5 month after birth and then decrease steadily. However, that of LDH from rat fed with ginseng ethanol extract was found in rat fed with ginseng ethanol extract. 3-bromopyruvate was shown to inactivate LDH-5 from old rat fed. Inactivation of LDH-5 by trypsin was remarkable in old rat fed. Km value for pyruvate in old rat fed was remarkably decreased. Cumulative results suggest that ginseng ethanol extract could affect conformational change of LDH responsible for altered properties through unknown mechanism.

Comparison of the Determinants in the Differences in Force-Frequency Relationships between Rat and Rabbit Left Atria

  • The underlying mechanism commonly applicable for both the positive and negative force-frequency relationships (FFR) was pursued in left atria (LA) of rat and rabbit. The species differences in the roles of $Na^+/Ca^{2+}$ exchanger and sarcoplasmic reticulum (SR), which are major intracellular $Ca^{2+}$ regulatory mechanisms in the heart, were examined in the amplitude accommodation to the frequency that changed from 3 Hz to the variable test frequencies for 5 minutes in the electrically field stimulated left atria (LA) of rat and rabbit. Norepinephrine strongly increased the frequency-related amplitude accommodation in both of rat and rabbit LA, while monensin, oubain or the reduced $Na^+$ and 0 mM $Ca^{2+}$ containing Tyrode solution increased the frequency-related amplitude accommodation only in the rabbit LA. Monenisn was also able to increase the frequency-related amplitude accommodation only in 1-day old rat LA but not in 4-week old rat LA that had 75% less $Na^+/Ca^{2+}$ exchanger with 97% higher SR than 1-day old rat LA. Taken together, it is concluded that the differences in the prevalence between myocardial $Na^+/Ca^{2+}$ exchanger and SR in the amplitude accommodation to the frequency-change determine the difference in the FFR between rat and rabbit heart.

Correlation between microsomal lipid peroxidation levels and drug metabolizing enzymes in rats on various ages (연령증가에 따른 마이크로솜 막지질 과산화수준의 변화와 해독효소계의 관계)

  • The studies were carried out on the correlation between microsomal lipid peroxidation level and drug metabolizing enzyme activities in rat liver microsomal suspensions on various ages (2-week-old, 2, 4, 8, and 12-month-old). The lipid peroxidation levels of liver homogenates tended to be elevated in a 4-month-old rat livers, but it was a little decreased in 8 and 12-month-old rat livers. The lipid peroxidation levels of microsomal suspension was not shown any significant differences by ages. Lipid peroxidation levels and microsomal cytochrome P450 and NADPH-cytochrome c reductase activity showed a direct correlation (r=0.72 and r=0.64), respectively. The activities of cytochrome P450-dependent aminopyrine-N-demethylase and benzpyrene hydroxylase in rat liver microsomes were increased by ages up to 8-month-old rats and maintained in 12-month-old rats. The correlation between lipid peroxidation levels and these cytochrome-dependent enzyme activities showed a high direct correlation (r=0.97 and r=0.81), respectively.

The Effect of Oral Administration of Yongyukjowi-tang on the Immune Activity in Aged Rat (용육조위탕(龍肉調胃湯) 경구투여가 노화 흰쥐의 면역활성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of oral administration of Yongyukjowi-tang(YJT) on the immune activity in aged Sprague-Dawley rat(SD rat). Methods : SD rats were divided into three groups: Yongyukjowi-tang groups(YJT groups), distilled water groups(control groups) and Vitamin C groups(positive control groups) which were administered an oral dose(1ml/1day) to 6, 48 and 68 weeks old SD rats for four weeks. After four weeks, the number of total leukocyte, CD3+, CD4+, CD8+, and the level of cytokine (IL-2, IL-4, IL-10, IFN-${\gamma}$) were measured in spleen tissue of each SD rats. Results and Conclusions : 1) The number of total leukocyte significantly increased in 52 weeks old YJT group and 72 weeks old YJT group in comparison with those of the control group. 2) The number of CD3+ cell significantly increased in 72 weeks old YJT group in comparison with those of the control group and the positive control group. 3) The number of CD8+ cell significantly increased in 52 weeks old YJT group in comparison with those of the control group. 4) The level of IL-2 significantly increased in 72 weeks old YJT group in comparison with those of the control group and the positive control group. 5) The level of IL-4 significantly increased in 52 weeks old YJT group in comparison with those of the positive control group. These results suggest that oral administration of YJT has an effect on increase of immune activity in aged rat.

Histologic Analysis of Vocal Folds in Aging Rats (노화 쥐 성대의 조직학적 분석)

  • Background and Objectives Presbyphonia is characterized by hoarse, breathy, weak vocal intensity. Extracellular matrix (ECM) in lamina propria (LP) of the vocal folds play an important role in voice production, and change of ECM according to the aging leads to the presbyphonia. The aim of this study was to investigate the histologic analysis of aging vocal fold of rat. Materials and Method Six and twenty two months old Sprague-Dawley rats (n=8, each group) were used and classified into young (six months old rats) and old (twenty two months old rats) group. Histologic analysis and immunohistochemical staining for ECM of LP were performed. Results Overall cellular density was significantly decreased in old rat group. Elastin fibers of LP were significantly decreased in old rat group. Type I collagen was significantly increased in old rat group. Type III collagen did not show significant difference. Hyaluronic acids did not show significant difference in Alcian blue staining and immunohistochemical staining. Conclusion Decreased general cellular density and elastin fiber and increased type I collagen were observed in the LP of vocal folds of aging rats. These ECM changes might to contribute the aging voice.

Studies on the Roles of $Na^+\;-Ca^{2+}$ Exchange according to Postnatal Age in the Negative Staircase Effect of the Rat Heart (흰쥐 심근의 역 사다리 효과에 있어서 생후 연령에 따른 $Na^+\;-Ca^{2+}$ 교환의 역할에 관한 연구)

  • Recent reports revealed that the $Na^+-Ca^{2+}$ exchangers and feet structures of sarcoplasmic reticulum(SR) are located in close vicinity in the specific compartment. Therefore, we investigated the possibility that the $Na^+-Ca^{2+}$ exchanger may decrease the tension development by transporting the $Ca^{2+}$ out of the cell right after it released from SR, on the basis of this anatomical proximity. We exammined the negative force-frequency relationship of the developed tension in the electrically field stimulated left atria of postnatal developing rat(1, 3 day, 1 week and 4 week old after birth). Cyclopiazonic $acid(3{\times}10^{-5}\;M)$ treatment decreased the developed tension further according to postnatal age. $Monensin(3{\times}10^{-6}\;M)$ treatment did not increase the maximal tension in 4 week-old rat, preserving negative staircase, while the negative staircase in the younger rat were flattened. $Ca^{2+}$ depletion in the buffer elicited more suppression of the maximal tension according to the frequency in all groups except the 4 week-old group. The % decrease of the maximal developed tension of 4 week-old group at 1 Hz to that of 0.1 Hz after $Na^+$ and $Ca^{2+}$ depletion was only a half of those of the yonger groups. Taken together, it is concluded that the $Na^+-Ca^{2+}$ exchange transports more $Ca^{2+}$ released from SR out of the cell in proportion to the frequency, and this is responsible for the negative staircase effect of the rat heart.

Korean Red Ginseng Up-regulates C21-Steroid Hormone Metabolism via Cyp11a1 Gene in Senescent Rat Testes

  • Ginseng (Panax ginseng Meyer) has been shown to have anti-aging effects in animal and clinical studies. However, the molecular mechanisms by which ginseng exerts these effects remain unknown. Here, the anti-aging effect of Korean red ginseng (KRG) in rat testes was examined by system biology analysis. KRG water extract prepared in feed pellets was administered orally into 12 month old rats for 4 months, and gene expression in testes was determined by microarray analysis. Microarray analysis identified 33 genes that significantly changed. Compared to the 2 month old young rats, 13 genes (Rps9, Cyp11a1, RT1-A2, LOC365778, Sv2b, RGD1565959, RGD1304748, etc.) were up-regulated and 20 genes (RT1-Db1, Cldn5, Svs5, Degs1, Vdac3, Hbb, LOC684355, Svs5, Tmem97, Orai1, Insl3, LOC497959, etc.) were down-regulated by KRG in the older rats. Ingenuity Pathway Analysis of untreated aged rats versus aged rats treated with KRG showed that the affected most was Cyp11a1, responsible for C21-steroid hormone metabolism, and the top molecular and cellular functions are organ morphology and reproductive system development and function. When genes in young rat were compared with those in the aged rat, sperm capacitation related genes were down-regulated in the old rat. However, when genes in the old rat were compared with those in the old rat treated with KRG, KRG treatment up-regulated C21-steroid hormone metabolism. Taken together, Cyp11a1 expression is decreased in the aged rat, however, it is up-regulated by KRG suggesting that KRG seems enhance testes function via Cyp11a1.

Effect of Exercise Training on Aging Atrophy in Rat Skeletal Muscle II. Effect of Long Term Weight-Training (흰쥐 골격근의 노화성 위축에 대한 운동훈련의 영향 - II. 장기간에 걸친 체중부하 훈련의 영향 -)

  • The present study was designed to examine effect of long term weight-training on aging atrophy in the rat skeletal muscle. Male rats of 8, 15, and 24 month old were used. Each age groups included control and weight-training for 5 months by using body press apparatus. The histo- and cytochemical, ultrastructural and stereological changes in aging skeletal muscles of the rat were observed in the present study. During the training period the body weight and muscular weight in all groups except the rectus femoris and the gastrocnemius in young age groups remained constant, but muscular weights were increased in the rectus femoris and the gastrocnemius muscles in young age groups. In trained rat, the volume density of muscle fiber type IIA and IIB were increased, but those of type IIC was decreased. Type I remained constant in 8 and 15 month old age groups, but reduced in the tibialis anterior and the gastrocnemius muscles in the 24 month old groups. Some histotological and ultrastructural changes associated with age were found: numerical increase of cytiplasmic vacuoles, lysosomes, lipofuscins, and irregularity of myofibrils. At 24 month old groups some unusual formation of contraction band and muscle splitting were observed. After weight-training, ultrastructural degenerative changes occured in the type I muscle fiber, such as splitting of muscle fiber, disorganization of myofilaments, swelling of mitochondria, accumulation of many lipid droplets, appearance of many lysosomes and residual bodies and necrotic fibers, in the old age groups. But, in the type II muscle fibers hypertrophy of muscle fiber appeared without any noticible damage as the type I. The activities of $Mg^{++}$ -ATPase decreased with age and this enzyme activities in the trained rat were significantly decreased with age. Activities of the acid phosphatase were increased with age and significantly in the trained rat. In stereological analysis, volume density of the myofibrils and the tubular system were increased, on the other hand there mitochondrial capacity was decreased. These experimental results suggested that old rats are not susceptible to be affected by weight-training as young rats, and that physical capacity of the rats must be considered when old rats are exercised for training.

  • The present study was conducted to investigate the structural changes in rat cornea. Sixty eyes from one-day-old uneyed rats, fourty eyes from 4-weeks-old rats, and foully eyes from 10-weeks-old adult rats were used. With the increase of age, the epithelial layer was thickened by the addition of new successive cellular layers. Then, the new-born rat's epithelial cells formed a pentagonal shape, and the quality of decidual cells showed a high electron-density, although the boundary between cells was distinctive. The newly produced cells showed a low electron-density so that there was the distinctive difference between light and darkness. In Bowman's layer, collagen fibrils demonstrate a regularly arranged structure along with the age. In stroma's layer, the density of keratocytes was decreased and thereby Progressively flattened during the development. The collagenous layer of the adult rats was more distinctive than that of the new-born rats in a form of parallel alignment running vertically and horizontally.
