Effect of Mushroom Extracts on Tyrosinase Promoter (수종의 버섯 추출물이 티로시나아제 프로모터에 미치는 효과)

  • To estimate the inhibitory effect of some mushroom extract on melanin biosynthesis, we tested its inhibitory effects of five mushroom extracts on tyrosinase promoter in Bl6 mouse melanoma cells. In five mushroom extracts, Cordyceps militaris and Poria cocos exhibited low repression effect on tyrosinase promoter. However, Ganoderma lucidum, Paecilomyces japonicus, Phellinus linteus showed high repression effect, Especially, Paecilomyces japonicus and Phellinus linteus extracts had very higher repression effect approximately $81{\sim}83%\;at\;100{\mu}g/mL$. In the MTT assay, Paecilomyces japonicus and Phellinus linteus extracts exhibited high cytotoxicity. Therefore, repression effect of the extracts were closely connected with cytotoxicity.

Immuno-modulatory and Anti-carcinogenic Property of Cordyceps militaris and Paecilomyces japonicus (동충하초의 면역조절 및 항암효과)

  • The purpose of this research was to investigate the immuno-modulatory effect and anti-carcinogenic property of Cordyceps militaris(CM) and/or Paecilomyces japonicus (PJ). The proliferation of cultured splenocytes and thymocytes were enhanced by the addition of 10 ㎍/ml of CM and/or PJ. B lymphocytes subpopulation in splenocytes were increased both CM and/or PJ administered(p.o. for 7 days)-mice. Thymic T lymphocytes, especially TH cells were significantly increased in CM-administered mice. CM and/or PJ treatment inhibited the cell viability of L 1210 mouse leukemia and HL60 human leukemia cells and induced the apoptosis of L1210 and HL60 cells. In addition, CM and/or PJ increased the hemaggutination(HA) titer against SRBC. These results suggest that CM and/or PJ have an immuno-modulatory action and anti-carcinogenic property.

Inhibition of Free Radical-Induced Lipid Oxidation by the Extract from Submerged-Liquid Culture of Mushrooms in the Medium Containing Mulberry Tree Powders (뽕나무가루 첨가 배지에서 배양한 버섯균사체 배양물의 자유라디칼 유도 산화 억제)

  • Antioxidant activity of extracts from the submerged-liquid culture of mushrooms was measured using two systems : linoleic acid and mouse liver microsomes induced by various free radical sources. Mushrooms of Pleurotus ostreatus (Neutari), Phellinus linteus (Sanghwang), Paecilomyces japonicus (Dongchunghacho), Hericicum erinacium (Norugungdengyee) and Agaricus blazei (Shinryeong) in 1% mulberry tree powder-supplemented medium were incubated in a shaking incubator (200 rpm, $25^{\circ}C$) for 3 days. Hot water extracts of mycelial cultures were freeze-dried, followed by fractioning with hexane, chloroform, ethylacetate and butanol in the order. Antioxidant activity of each sample was examined in free radical-induced linoleic acid oxidation in phosphate-buffered saline (PBS ) solution by measuring the amount of malonaldehyde (MA), and mouse liver microsomal systems by measuring the amount of thiobarbituric acid reactive substances (TBARS). In linoleic acid oxidation system, hot water extracts from the cultures of Pleurotus ostreatus, Phellinus linteus, and Paecilomyces japonicus exhibited stronger antioxidant activity than aqueous or butanol fraction and the combined fraction of hexane, chloroform and ethylacetate, but the hot water extract from Pleurotus ostreatus culture was the strongest activity. The antioxidant activity of the hot water extract from Pleurotus ostreatus culture was stronger than any other fractions in mouse microsomal system. These results suggest that hot water extract of Pleurotus ostreatus culture, and the cultures of Phellinus linteus and Paecilomyces japonicus could be useful for functional materials to reduce the oxidation of lipids in food systems induced by free radicals.

Antidiabetic Effect of Cordyceps militaris and Paecilomyces japonicus in Streptozotocin-induced Diabetic Rats (Streptozotocin 유발 당뇨 흰쥐에서 Cordyceps militaris와 Paecilomyces japonicus의 항당뇨 효과)

  • To elucidate the effect of water and methanol extracts of Cordyceps militaris, Paecilomyces japonicus and their mycelin on diabetes and organs in STZ-induced diabetic rats, weight of organs (liver, kidney, spleen, thymus), plasma level of blood glucose, total protein, triglyceride, free fatty acid, hepatic total protein, triglyceride and glycogen were determined as compared with those of negative control group. The blood glucose level of CM-1 and CM-M group showed significantly reduced, and all groups except CM-2 increased in body weight. CM-1 decreased the liver weight, and PJ-2 decreased the kidney weight. In all groups except PJ-2, plasma total protein level was increased, and the triglyceride, and CM-3 and CM-H decreased the free fatty acid was decreased in CM-3, PJ-1 and PJ-2 treated groups. In hepatic tissue, total protein was significantly increased in CM-H and CM-M treated group, and in all groups except CM-2, the triglyceride were significantly decreased and glycogen was increased. In conclusion, CM-1 and CM-M that possess potential antidiabetic activity increased glycogen and lowered serum glucose level, thus they might improve metabolic disorder originated from diabetes by increasing serum protein and reducing excess triglyceride in serum and liver tissue.

Phylogenetic Analysis of the Entomopathogenic Fungal Species and Taxonomical Positions of Their Commercial Products (동충하초의 계통분류 및 시판동충하초의 분류학적 위치)

  • This study was carried out to identify the phylogenetic relationship and to know the distribution of the entomopathogenic fungi by comparing the DNA sequences of internal transcribed spacer regions (ITS1 and ITS2) and 5.8S ribosomal DNA (rDNA) repeat unit. The entomopathogenic fungi had their specific sequences in ITS1 and 2 regions depending on species. The comparison of the ITS sequences of standard strains indicated that the sequences ITS1 were more variable than those of ITS2. It seems that Paecilomyces tenuipes, Isaria japonicus and P. japonicus are the same species but called as different names because of very similar sequences, and unidentified Paecilomyces sp. KACC 40220 and KACC 40656 showed identical sequences to P. tenuipes. Thirty six strains of the commercial products of entomopathogenic fungi used in this study were divided into four groups by the phylogenetic analysis based on 5.85 rDNA and ITS regions. We found twenty-three strains were P. tenuipes / japonica, eleven strains were C. militaris, and other two strains were Beauveria bassiana and C. multiaxialis, respectively.

Study on the Inoculation Augmentation of paecizomyces japonicus to the Silkworm, Bombyx mori, Using Dexamethasone (Dexamethasone을 이용한 누에(Bombyx mori)에 대한 동충하초균 (Paecilomyces japonicus)의 접종율 제고에 관한 연구)

  • Entomopathogenic fungus, Paecilomyces japonicus, has been commercially used as medicinal purpose . The silkworm, Bombyx mori, as an optimal host for the fungi, has been selected and used for the production of the fungal fruit bodies. In current method, newly molted fifth instal larvae should be exposed to the adverse stress environment of high temperature (3$0^{\circ}C$), high relative humidity ( 90%), and starvation for 24h for better fungal inoculation to the host insects. In this study, an alternative method using chemical agent, dexamethasone (DEX: an eicosanoid biosynthesis inhibitor), was tried to get the immunodepressive effect on the larvae to elevate the inoculation rate of the fungi to the silkworm without any harsh rearing environment. DEX (100$\mu\textrm{g}$) showed significantly synergistic effect on the hemocyte lethality of the fungus, and was effective to decrease cellular immune responses measured by the number of hemocyte microaggregation and phenoloxidase activity of the fifth instar larvae in response to the fungal injection. A detergent of 0.05% Triton-X was effective to increase the in- oculation rate of the fungi to the larvae and used in all fungal spraying solutions. Without any environ- mental stress treatment, only DEX (100$\mu\textrm{g}$) injection to the fifth instar larvae followed by the fungal spray was effective to get the inoculation rate equivalent to the current fungal spray method requiring harsh rearing environment. These results suggest that the inoculation of P. japonicus can be elevated by the help of DEX and that the silkworms use eicosanoids to elicit cellular immune response against fungal pathogen.

Antitumor compounds from fruiting bodies of cultivated fungus of Paecilomyces japonica

  • As part of our continuing attempts to evaluate biologically active compounds from fruiting bodies of cultivated fungus of Paecilomyces japonicus Yasuda, we conducted series of experiments on various fractions and compounds isolated by systematic fractionations. Our main efforts were concentrated on searching for compounds showing antitumor activities, which were tested on mice carrying Sarcoma-180 ascitic tumor. The antitumor activity was assessed by the life spans after these mice were administered Lp. with test compounds for consecutive 20 days. One of two pure compounds, which we have isolated to date, demonstrated significant prolongation of life span. ( Mean Survival Time: 30.3 days compared to that of control: 23.6 days). Structural analysis showed that this compound corresponds to D-mannitol. On the other hand, Ergosterol, another isolated pure compound didn't show efficient antitumor activity. We also obtained water-soluble fractions containing protein-bound polysaccharides and n-butantol fractions, which showed strong antitumor activities, 35.4(150%) and 32.1(136.0%) days of MST, respectively. In SRB assay, however, the test materials didn't show any toxic effects, but the level of acid phosphatase increased significantly when they were applied in cultured macrophage in vitro. Therefore, we concluded that antitumour activities might be attributed to immunostimulating rather than cytotoxic effects. Further experiments are underway to purify and structurally characterize new antitumour compounds from the active fractions.

Preparation of Mushroom Mycelia-cultured Traditional Meju with Enhanced Anticaricinogenicity and Sensory Quality (항암성과 향미가 개선된 재래식 버섯균사체메주의 제조)

  • Mushroom mycelia-cultured traditional meju (MTM) was prepared by inoculating 10% submerged-liquid culture of mushroom strains to five holes (1$\times$3 cm) per side of the traditionally-fermented meiu (10$\times$10$\times$10cm), followed by incubating additional 4 weeks at $25^{\circ}C$. Mushroom strains used were Neutari (Pleurotus ostreatus, PO), Yeongji (Ganoderma lucidum, GL), Synryeong (Agaricus blazei, AB), Ypsae (Grifola frondosa, GF), Pyogo(Lentinus edodes, PE), Dongchunghacho (Paecilomyces japonicus, PJ) and Sanghwang (Phellinus linteus PL). All MTMs showed an enhanced anticarcinogenicity against S-180 cell-induced mouse ascites cancer antimutagenicity against aflatoxin B$_1$ (AFB$_1$) and 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), and sensory qualities, relative to control meju. Such positive effects of MTM prepared with Sanghwang, Yeongji, or Synryeong were superior to those of MTM with Ypsae, Pyogo, Dongchunghacho, or Neutari.

  • Effect of mulberry tree powders on the antioxidant activity of submerged -liquid culture of mushrooms was investigated. Agaricus blazei (AB), Hericicum erinacium (HE), Pleurotus ostreatus (PO), Phellinus linteu (PL) and Paecilomyces japonicus (PJ) were cultured in a shaking incubator (200 rpm, $25^{\circ}C$) for 7 days in culture media: 1) basal medium (BM) and 2) BM+1% mulberry tree powders (BMM). Hot water extracts from the submerged-liquid cultures of mushrooms and BMM were freeze-dried for the measurement of antioxidant activity, of which reaction mixture (25 mL: 10 mL of 0.2 M sodium phosphate buffer, pH 8.0; 4.5 mL distilled water; and 10.5 mL ethanol) contained 275 $\mu$mol linoleic acid and one mg test sample. The reaction mixture was incubated in a shaking incubator (200 rpm, 4$0^{\circ}C$) for 16 days. Peroxide value (POV) was measured for a period of over 16 days, and malonaldehyde (MA) was determined only for samples from the day 16 of incubation. Mycelial weight of mushroom strains cultured in BMM was greater than BM. The antioxidant activities of AB-cultured in BW (AB-BMM) and HE-cultured in BMM (HE-BMM) were superior to those of other mushroom strains-cultured in BMM or BM and of BMM. These results suggest that mulberry tree powders enhance the antioxidant activity of submerged-liquid culture of mushroom strains. The AB-BMM and HE-BMM were the most active cultures.
