IN VITRO EFFECT OF PANAX GINSENG ON PHYTOHAEMAGGLUTININ-INDUCED LYMPHOCYTE TRANSFORMATION AND ITS IN VIVO EFFECT (식물성 혈구 응집소(Phytohaemagglutinin)로 유도한 림파구 변형에 관한 인삼의 in vitro 및 in vivo효과)

  • The effect of hydrocortisone or Panax ginseng, and/or a combination of hydrocortisone and Panax ginseng on phytohaemagglutinin (PHA-P)-induced transformation of peripheral blood lymphocytes were studied in 4 normal healthy adult volunteers. Increasing concentrations of Panax ginseng 0.16-1.60${\mu}g$/ml caused a doserelated inhibition of PHA-P transformation of lymphocytes. A combination of 500${\mu}g$/ml hydrocortisone and 0.80${\mu}g$/ml Panax ginseng produced a greater suppression of PHA-P stimulation than either drug used alone. This suggests that Panax ginseng has a steroid-like effect in vitro, and may have a potentiating effect with hydrocortisone on T-cell mediated immunity. The in vivo effects of Panax ginseng were further studied in 6 healthy adult volunteers. PHA-P transformation studies were performed before and after Panax ginseng capsules were taken for 2 weeks and at a higher dosage at 4 weeks. Our results showed that in 3 subjects, there was significant inhibition of PHA-P transformation at 4 weeks.

Inhibitory Effects of Onchungeum on Cytokine Production from Phytohaemagglutinin-stimulated Peripheral blood Mononuclear cells of Behcets Patients (Behcet 환자의 단구세포에서 온청음의 Cytokine 분필억제 효과)

  • Behcets disease is a systemic inflammatory disorder. The etiology and pathogenesis of Behcets disease has yet been fully elucidated but might involve immune dysfunction. Cytokines involved in the regulation of inflammatory reactions and immune responses may play a role in the pathogenesis of Behcets disease (BD). Onchungeum is an Oriental herbal medication, which has been successfully used in Korea for the treatment of BD. This report describes modulation effects of Onchungeum on cytokine production from phytohaemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMC) of Behcets patients by ELISA. Onchungeum significantly inhibited the production of pro-inflammatory cytokines. TNF-α and IL-1β, compared to absence of Onchungeum (by 52.3 1.4 % inhibition for TNF-α and 113.5 3.3 % for IL-1β, p < 0.001). Onchungeum also inhibited the production of IFN-γ, immunoregulatory Th1 cytokine, by 89.4 0.8 % (p < 0.001). The inhibitory effects of Onchungeum on cytokine production showed dose-dependent manner, and the pre-treatment of 1 mg/ml Onchungeum had better effects than immunosuppressive drug for treatment of BD, cyclosporin A. Our results suggest that Onchungeum treatment for Behcets disease patients may have pharmacologic activities and abilities of regulation of immune and inflammatory responses by cytokine modulation.

Effect of 65 kDa Buffalo Placental Protein on B-Cell Proliferation and Antibody Response

  • Immunosuppressive potential of 65 kDa buffalo placental protein (bPP65) on B-cell proliferation in vitro and antibody response in vivo was evaluated. B-cell proliferation was estimated by measuring incorporation of tritiated thymidine in buffalo lymphocytes while primary antibody responses against phytohaemagglutinin (PHA) or keyhole limpet haemocyanin (KLH) were evaluated in mice. bPP65 suppressed proliferation of lipopolysaccharide (a B-cell specific mitogen)-stimulated buffalo lymphocytes in vitro indicating suppression of B-cells. This suppression was dose dependent over the protein concentration range $25-100 {\mu}g/ml$. Primary antibody responses in mice against PHA and KLH in presence of bPP65 were lower as compared to in its absence but these were not statistically significant. Amino acid composition data of bPP65 and BSA suggested that bPP65 is different from BSA.

Immunomodulatory activities of ethanolic extract of Drynariae Rhizoma (골쇄보(骨碎補) ethanol 추출물의 면역 조절 작용에 관한 연구)

  • In the traditional Chinese medicine, Drynariae Rhizoma (DR) has been reported as a good enhancer for bone healing. DR, a plant widely used in the traditional medicinal systems of Korea, has been reported to possess antiviral, antibacterial and anti-inflammatory activities. Modulation of immune response to alleviate disease has been of interest for a long time. Plant extracts have been widely investigated for possible immunomodulatory properties. Thus, I have evaluated the anticellular and immunomodulatory properties of ethanolic extract of DR. DR extract inhibited proliferation of mitogen (phytohaemagglutinin; PHA) and antigen (purified protein derivative; PPD)-stimulated human peripheral blood mononuclear cells (PBMCs). In addition, DR inhibited growth of several cell lines of mouse and human origin. It also inhibited production of nitric oxide (NO), interleukin-2 (IL-2) and tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$. Intracytoplasmic $interferon-{\gamma}\;(IFN-{\gamma})$ and expression of cell surface markers, CD16 and HLA-DR, on human PBMC, were not affected on treatment with DR but CD25 expression was down regulated. This study demonstrates the antiproliferative and immunosuppressive potential of ethanolic extract of DR in vitro.

Regulatory effects of Seogakjihwang-tang on Cytokines and Growth Factor Production in PBMC from the Patient with Cerebral infarction under Consciousness Disorders

  • Seogakjihwang-tang (SJT) was widely used to treat patients suffering from cerebral infarction. But scientific investigation has been carried out very little. The aim of the present study is to investigate the effect of SJT on the production of various cytokines in the patients with cerebral infarction (CI). We investigated interleukin (IL)-4, IL-10 and transforming growth factor (TGF)-1 in the sera of 27 patients with cerebral infarction under consciousness disorders and 10 normal controls using an originally devised sensitive sandwich enzyme-linked immunosorbent assay (ELISA). We found that plasma levels of IL-4 were slightly elevated in patients with cerebral infarction, whereas plasma levels of IL-10 (P<0.001) and TGF-1 were reduced. Peripheral blood mononuclear cells (PBMC) obtained from the patient with CI were cultured for 24 h in the presence or absence of lipopolysaccharide (LPS) or phytohaemagglutinin (PHA). The amount of IL-4, IL-10 and TGF-1, in culture supernatant, was significantly increased in the LPS or PHA treated cells compared to unstimulated cells (P<0.05), We also show that increased cytokines IL-4, and IL-10 level was significantly inhibited by SJT in a dose-dependent manner. Maximal inhibition rate of IL-4 and IL-10 production by SJT was 45.63.3% and 614.7% for LPS-stimulated cell and 27.31.2% and 83.62% for PHA-stimulated cells, respectively (P<0.05). On the other hand, SJT significantly increased the LPS or PHA-induced TGF-1 production (P<0.05). These data suggest that SJT has a regulatory effect on the cytokines production, which might explain its beneficial effect in the treatment of CI.

The inhibitory effects of Gami-Phedoc-San on TNF-α, IL-1β and IFN-γ secretion from human monocytes

  • In our study, the several cytokines were determined in phytohaemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMC) of Adamantiades-Behcets patients. Adamantiades-Behcets disease (ABD) is a systemic inflammatory disorder and might involve immune dysfunction. High levels of $TNF-\alpha,\;IL-1\beta$ and $IFN-{\gamma}$ indicate the activation of inflammatory reactions and immune system in ABD. Gami-Phedoc-San (GPS) is an Oriental herbal medication, which has been used in Korea for the treatment of ABD. GPS (1 mg/ ml) significantly inhibited the secretion of proinflammatory cytokines, $TNF-\alpha\;and\;IL-1\beta$, compared to absence of GPS (by $50.5{\pm}1.9%$ inhibition for $TNF-\alpha$ and $106.9{\pm}16.8%$ for $IL-1\beta$). GPS also inhibited the production of $IFN-\gamma$, immunoregulatory Th1 cytokine, by $78.4{\pm}2.8%$. The inhibitory effects of GPS on cytokine secretion showed dose-dependent manner, and the pre-treatment of 1 mg/ml GPS had better effects than immunosuppressive drug for treatment of ABD, cyclosporin A. Our results suggest that GPS treatment for ABD patients might have pharmacological activity of immune and inflammatory responses through the cytokine modulation.

Esperimental Infection of Infectious Bovine Rhinotracheitis Virus to Rabbits Immunosuppressed with Dexamethasone (Dexamethasone으로 면역기능(免疫機能) 저하(低下)된 토끼에 Infectious Bovine Rhinotracheitis Virus 감염시험(感染試驗))

  • To establish a laboratory animal model for study on development of diagnostic methods for infectious bovine rhinotracheitis virus(IBRV), experimental infection of the virus to rabbits immunosuppressed with dexamethasone(DX) were carried out. Results obtained throughout the experiments were as follows. When lymphocyte activity was measured by lymphocyte transformation to phytohaemagglutinin in parallel with total and differential leucocyte counting, both groups treated with 2.0mg DX once and 1.0mg DX daily showed marked immunosuppression between 5 to 72 hrs. after administration. The degree of suppression of lymphocyte activities was more remarkable in the latter group. IBRV PQ7 strain at $10^{7.5}\;TCID_{50}/0.2ml$ was inoculated into conjunctival sacs of rabbits immunosuppressed with DX and non-treated. During 3 weeks observation, the immunosuppressed groups revealed mild conjunctivitis, viremia and virus recovery by 33.3 to 100%, whereas the DX nontreated group showed viremia and virus recovery with no clinical conjunctivitis by one of three rabbits(33.3%). In conclusion, it was indicated that experimental infection of IBRV PQ7 strain to rabbit was limited in prerequisite to immunologic modification by administration of immunosuppressive drugs.

Cytokines Expression and Nitric Oxide Production under Induced Infection to Salmonella Typhimurium in Chicken Lines Divergently Selected for Cutaneous Hypersensitivity

  • In the present study, the impact of Salmonella Typhimurium on cell-mediated immunity (CMI) was investigated in 5 week-old immuno divergent broiler lines selected for the high and low response to phytohemagglutinin-P. The immune response was assessed in peripheral-blood mononuclear cells (PBMCs) induced with Salmonella Typhimurium at different time intervals (0 h, 0.5 h, 2 h, 4 h, 6 h, 12 h and 24 h). The differential mRNA expression patterns of IFN-${\gamma}$, IL-2 and iNOS were evaluated by quantitative real time PCR. In-vitro production of nitric oxide (NO) was also estimated in the culture supernatant and correlated with iNOS mRNA expression. Present study showed higher production of NO in the high cell-mediated line (HCMI) as compared to the low cell-mediated line (LCMI) upon stimulation with Salmonella Typhimurium. Correspondingly, higher mRNA expression of iNOS and IFN-${\gamma}$ were observed in high response birds (HCMI); but IL-2 was down regulated in this line compared to the low response birds (LCMI). Significantly (p<0.05) higher expression of iNOS, IFN-${\gamma}$ and higher production of NO in high line indicated that the selection for PHA-P response might be employed for increasing the immune competence against Salmonella Typhimurium in chicken flocks.

Inadequacy of application of cytokinesis-blocked cells in fish (Rock fish, Sebastes schlegeli) and fowl(chicken) as biological dosimeter for radiation exposure (방사선 피폭의 생물학적 선량측정에 어류(조피볼락, Sebastes schlegeli) 및 조류(닭)의 세포질분열 차단 세포 적용의 부적절성)

  • The purpose of the present experiment was to investigate the micronuclei (MN) frequency in cytokinesis-blocked (CB) cells after various doses of gamma-rays in two species (fish and fowl) and so to contribute to the clarification of the question whether these species are suitable as a target organism in the test system. The frequencies of binucleated cells, and gamma-ray-induced MN in CB cells at several doses were measured in three donors of two species. No binucleated cell was noted in erythrocyte. The peaks of binucleated lymphocyte formation were found at a concentration of 2% phytohaemagglutinin (PHA) and $3{\mu}g/m{\ell}$ cytochalasin B (Cyt-B) in fish at 144 hours after incubation and 2% PHA and $6{\mu}g/m{\ell}$ Cyt-B in fowl at 72 hours after incubation. But the micronucleus counts failed to show any evidence of radiation damage. Measurements performed after irradiation showed a dose-related decrease in the formation of binucleated cells in each of the donors studied. Results indicated that the assays were not suitable for this due to blastization inhibition (binucleation failure) after irradiation. We concluded that the use of CB cell from fish and fowl for detecting the results of mdiation exposure was highly questionable.
