Candida albicans에 대한 계지(桂枝), 백두옹(白頭翁), 백선피(白鮮皮), 백작약(白芍藥), 빈낭, 인진(茵蔯)의 항진균효과 (Antifungal Effects of Cinamon Ramulus, Pulsatillae Radix, Dictamni Radicis Cortex, Paeoniae Radix, Arecae Semen, Artemisiae Capillaries Herba against Candida albicans)

  • The purpose of this thesis is to dedicate to get the objectivity of herbal medicine treatments by choosing herbs likely to work as antifungal agents against Candida albicans which is the causes of Candida vaginitis, making experiments on them and getting the significant results. Each herb's efficacy on control the number of Candida vaginitis is noticed by using Disk Susceptibility test with six herbs medicine and Broth dilution assay of the culture. Cinamon Ramulus, Pulsatillae Radix, Dictamni Radicis Cortex, Paeoniae Radix, Arecae Semen, Artemisiae Capillaries Herba extracts experimented on the efficiency of antifungal activities against Candida albicans by disk susceptibility test. Pulsatillae Radix has the strongest efficiency. In Dictamni Radicis Cortex never appeared antifungal effect on treatment. MIC (minimal inhibition concentration) in vitro antifungal agents of Cinamon Ramulus, Pulsatillae Radix, Paeoniae Radix, Artemisiae Capillaries Herba extracts against Candida albicans was determined by broth dilution assay. MIC against Candida albicans is that Pulsatillae Radix is 2.5 mg/ml and Cinamon Ramulus is 5 mg/ml and Paeoniae Radix is 10 mg/ml and Artemisiae Capillaries Herba is 40 mg/ml.

  • Pulsatillae Radix, the root of Pulsatilla koreana Nakai (Ranunculaceae), has been traditionally used as a remedy for leukorrhea and amoebic dysentery by taking internally‘ and as an antimicrobial for external use. We carried out this study to examine the anti-dandruff effect of 70% Ethanol-Extract of Pulsatillae Radix (EPX) compared with 1 % zinc pyrithione used commercially as anti-dandruff. The EPX showed significant antimicrobial activity against Pityrosporum ovale, major cause of dandruff, and also showed significant anti-dandruff and itch-improvement effect of head skin.

백두옹(白頭翁) 및 선퇴(蟬退) 추출 혼합물의 분자 개선 효과 (Acne-remedy Effects of Extract Mixture of Pulsatillae Radix and Cicadidae Periostracum)

  • Pulsatillae Radix and Cicadidae Periostracum have long been used for treatment of injuries, inflammations or itchiness in Korea. This study was carried out to examine the acne-remedy effects of the mixture made with equal parts of Pulsatillae Radix extract and Cicadidae Periostracum extract (PRCP) compared with 2%-benzoyl peroxide (Standard) used commercially as anti-acne. The results showed the mixture (PRCP) was more effective than each crude drug extract. The 10%-PRCP showed significant antimicrobial activity against Propionibacterium acnes, the major cause of acne, and also showed significant inhibition from type 1 $5{\alpha}$-reductase ($5{\alpha}R$), another cause of acne, And the cream made with PRCP demonstrated anti-sebum and acne-improvement effects on examinee's facial skin.

백두옹 추출물이 Streptococcus mutans의 비수용성 글루칸형성 및 부착억제에 미치는 영향 (Inhibitory Effects of Radix Pulsatillae Extract on Insoluble Glucan Synthesis and Adhesion of Streptococcus mutans)

  • Streptococcus mutans plays a virtal role in trigering dental caries establishment due to its ability to synthesize two significant factors. The two factors are organic acids and glucans. The former demineralized dental enamel and the latter mediates the attachment of bacteria to tooth surface. It is believed that demineralization of dental enamel and attachment of bacteria are the crucial events that indicate and develop dental caries. For this reason, we studied the effect of the ethanol extracts of Radix Pulsatillae on the growth and acid production of S. mutans. Ethanol extracts of the Radix Pulsatillae showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition compared to the control groups (p<0.05). The extracts inhibited S. mutans adherence to hydroxyapatite treated with saliva, and cell adherence was repressed by Radix Pulsatillae. the ethanol extract of Radix Pulsatillae showed remarkable inhibition of glucosyltransferase, which synthesizes water insoluble glucan form sucrose. Phytochemical analysis showed Radix Pulsatillae contained major components such as phenolic compounds, glycosides, steroids, terpenoid, and saponin. These results suggest that Radix Pulsatillae may have anti-cariogenic properties, which may be related with major components such as phenolic compounds, glycosides, steroids, terpenoid, and saponin.

5종 단미제의 Staphylococcus aureus에 대한 in vitro 항균력 평가 (A Study on Antibacterial Effects of Five Single Herbs Aqueous Extracts against Staphylococcus aureus)

  • Objectives: The object of this study was to observe the in vitro antibacterial effects of five single(Pulsatillae Radix, Patrinae Radix, Sanguisorbae Radix, Sophorae Flos, and Sophorae Radix) aqueous herbal extracts, traditionally used for treating various gynecological diseases including mastitis in Korea, against Staphylococcus aureus. Methods: Antibacterial activities against Staphylococcus aureus of aqueous extracts of Pulsatillae Radix, PatrinaeRadix, Sanguisorbae Radix, Sophorae Flos, and Sophorae Radix were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were also monitored at Minimal Incubation Concentration(MIC) and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using murine macrophage(Raw 264.7) and human mammary gland carcinoma cell(MCF-7). Results: MIC of aqueous extracts of Pulsatillae Radix, Patrinae Radix, Sanguisorbae Radix, Sophorae Flos, and Sophorae Radix against Staphylococcus aureus were detected as $0.215{\pm}0.107$ mg/ml, $0.273{\pm}0.107$ mg/ml, $0.469{\pm}0.297$ mg/ml, $11.850{\pm}8.406$ mg/ml, and $0.664{\pm}0.546$ mg/ml, respectively. MIC of Ciprofloxacin was detected as $0.469{\pm}0.297{\mu}g/ml$ at same conditions. In addition, all five single aqueous herbal extracts were also showed marked dosage-dependent inhibition of bacterial growth. The effects of intracellular killing with Raw 264.7 and inhibition of bacterial invasion with MCF-7 cells were detected, in the order of Sophorae Flos, Pulsatillae Radix, Patrinae Radix, Sanguisorbae Radix and Sophorae Radix aqueous extracts in the present study. Conclusions: The results obtained in this study suggest that all five single aqueous herbal extracts showed antibacterial effects against Staphylococcus aureus and they also showed dosage-dependent inhibitory effects on the bacterial growth. They showed the significant intracellular killing and inhibition of bacterial invasion effects. It means, all five single aqueous herbal extracts may show potent anti-infectious effects against Staphylococcus aureus for mastitis.

청열약류(淸熱藥類)의 면역약리학적(免疫藥理學的) 고찰(考察) (The immunopharmacologic study on drugs for clearing away heat and detoxicating(II))

  • In the result of investigating traditional chinese medical literatures to understand definite immun opharmacologic effects of drugs for clearing away heat and detoxicating such as Oldenlandiae Diffusae Herba, Fel Ursi, Fraxini Cortex, Pulsatillae Radix, Bruceae Fructus, Portulacea Herba, Patriniae Radix, we could reach conclusions as follows: 1. Oldenlandiae Diffusae Herba can increase voracity of leukocytes and immune function of splen ocytes. 2. Fel Ursi, Patriniae Radix can inhibit acute, chronic inflammation by decreasing voracity of macrophages, monocytes and recover lymphocytes. 3. Fraxini Cortex have anti-inflammatory effect then applied to treat with arthritis. Pulsatillae Radix, Bruceae Fructus, Portulacea Herba have anti-cancer, anti-biotic effects. Above results indicates that drugs for clearing away heat immunosuppressive effect that they can apply to all sorts of inflammatory diseases such as arthritis, DTH, SLE, and cancer.

백두옹의 혈당강하 성분연구 (I) (Studies on the Hypoglycemic Constituent of Pulsatillae Radix ( I ))

  • For the investigation of bioactive natural products with hypoglycemic effect, we have evaluated various extracts of Pulsatillae Radix(Ranunculaceae), which has been used in traditional medicine for hematochezia due to intense evil heat, malaria, chills and fever, epistaxis and internal hemorrhoids. The ethylacetate extract of the radix of this plant was found to show a significant hypoglycemic effect on alloxan diabetogenic mice. Using bioactivity-guided chromatographic purification of the ethylacetate extract, hypoglycemic constituent: 2${\beta}$, 3${\beta}$, 14${\alpha}$, 20, 22R, 25-hexahydroxy-cholest-7-en-6-one was isolated and structurally identified by physico-chemical properties and spectroscopic evidences.

Trichomonicidal Activity of Herbal Extracts Used in Traditional Medicine in Korea

  • Trichomonicidal activity of thirty methanolic herbal extracts used in traditional medicine in Korea was evaluated. Trichomonas vaginalis was used as experimental model, and anti-Trichomonas activity was determined over cultures of the parasite in TYM Diamond medium. Six methanolic extracts such as Acanthopanacis Cortex, Agrimoniae Herba, Pulsatillae Radix, Sanguisorbae Radix, Sophorae Radix, and Torilidis Fructus showed more than 50% trichomonicidal activity at the concentration of 200 g/ml. These extracts were further fractionated into n-butanol soluble and aqueous phases. Except for Acanthopanacis Cortex, all of n-butanol soluble phases showed potent trichomonicidal activity, while none of aqueous phases exhibited trichomonicidal activity.
