Study on the Effects of Germinated Rhynchosia Volubilis on Osteosarcoma HOS-TE85 Related to Bone Morphogenesis (발아 서목태가 골육종세포 중 HOS-TE85의 골형성에 미치는 영향)

  • The aim of this study was to find out the effectiveness on germinated Rhynchosia Volubilis for Female Bone Morphogenesis. For this purpose, experiments using germinated Rhynchosia Volubilis(GRV) according to germinating days were conducted to measure the polyphosphate contents and to examine the effects of the transcription activity of gene related to bone morphogenesis on the formation of bone in female. The quantitative analysis of the polyphosphate contents showed that 1 day geminated Rhynchosia Volubilis(GRV) group is treble better contents of polyphosphate than non-germinated Rhynchosia Volubilis, 2 day and 3 day GRV groups. The active of the COL1, OTN, MGP, BMP genes was less than the increase of the polyphosphate contents.

  • The aim of this study is to find out what inhibition effects the Germinated Seed of Rhynchosia Volubilis(GSRV) has by its feeding period on the bone absorption in ovariectomized white rats. Close examination of the amounts of the Interleukin-6 in the rats by the groups of one, two and three-day germination periods led to the following conclusions. Feeding ovariectomized white rats with GSRV by the groups of one, two and three-day germination periods showed less amounts of Interleukin-6 in their bone marrow in all the feeding periods of two, three and four weeks than in the case of control group, revealing a statistical significance. Therefore, the Germinated Seed of Rhynchosia Volubilis(GSRV) is believed to have inhibition effects on bone resorption.

  • The aim of this study was to find out the effectiveness on germinated seed of Rhynchosia Volubilis for Female Bone Morphogenesis. For this purpose, We compared two methods. water extract and alcohol extract using germinated Rhynchosia Volubilis(GRV) according to germinating days were conducted to measure the polyphosphate contents and investigate HOS-TE85 propagation rate. Both water and alcohol extract two methods were not toxicant. And if not excessively treated, alcohol extract rate were more about $5{\sim}15$ times than water extract rate. So usually water extract were better than alcohol extract. but in case of osteoporosis, alcohol extract were effective.

  • Objective : The purpose of this studies was to determine the effect of Carthami semen, Bogol-Tang and Rhynchosia volubilis Lour. on changes of trabecular area and physiological metabolites in the ovariectomized osteoporotic Rats. Methods : In order to induce estrogen deficient osteoporosis, ovariectomy was done on rats. Then the Carthami semen, Bogol-Tang and Rhynchosia volubilis Lour. were orally administerd: Such indexes were measured as the changes of body weight, bone mineral density, trabecular area in tibia, and levels of osteocalcin, bone alkaline phosphatase, calcium, phosphorus, cholesterol, triglyceride, aspartate aminotransferase and alanine aminotransferase in serum. Results : 1. The change of bone mineral density in Bogol-Tang group and Rhynchosia volubilis lour. group. was significantly increased compare to control group. 2. The change of trabecular area % in epiphysis of tibia in Carthami semen group was significantly increased compare to control group. 3. The change of trabecular area % in diaphysis of tibia in Bogol-Tang group was significantly increased compare to control group. 4. The change of serum osteocalcin in Bogol-Tang group was significantly decreased compare to control group. 5. The change of serum bone alkaline phosphatase in Bogol-Tang group was significantly decreased compare to control group. 5. The change of serum bone alkaline phosphatase in Bogol-Tang group was significantly decreased compare to control group. 6. The change of1 serum calcium in Rhynchosia volubilis lour. group was significantly decreased compare to control group. 7. Texture and Storage Stability of Tofu Incorporated with Rhynchosia volubilis

  • The effects of Rhynchosia volubilis (yakong) incorporation (0%, 10%, 20%, and 30%, w/w of soybeans) on quality and shelf life of tofu were investigated. Moisture content of tofu increased slightly with the increase in the level of yakong incorporation from 10% to 30% and no apparent relationships between yakong incorporation and the yield were found. Turbidity of soybean whey tended to increase with increased level of yakong incorporation where the values of 20% and 30% samples were significantly different from those of control and 10% sample (p<0.05). The different levels of yakong incorporation were found to have significant influence (p<0.05) on all the color characteristics of tofu. Tofu texture varied with the level of yakong incorporation in consistent pattern; however, there was no significant difference (p>0.05) in most cases. Tofu incorporated with yakong ($10{\sim}30%$) had a shelf life of above at least 1 day longer than that of the control tofu.

Comparative Study of 3 kinds of Black Soybean on Murine Immune Cells (생쥐의 면역세포에 대한 검은콩 3종의 비교 연구)

  • The purpose of this research was the comparative study of 3 kinds of black soybean on murine immune cells. The 3 kinds of black soybean are Glycine max Merr. with inner color-yellow (GY), Glycine max Merr. with inner color-greenish (GG) and Rhynchosia volubilis Lour. (RV). All of the black soybean increased the viability of murine thymocytes in vitro. The combined treatment of GY or GG and mitogen did not affect the viability of splenic T- and B-lymphocytes compared with mitogen-treated group, but the combined treatment of RV and mitogen increased their action compared with mitogen-treated group. Also, the 3 kinds of black soybean were given p.o. once a day for 7 days, respectively. RV increased the population of thymic-$CD8^+$, splenic-$CD8^+$ and $B220^+$ cells in vivo. Furthermore, GY and GG did not affect the phagocytic activity and the production of nitric oxide in peritoneal macrophages in vitro, but RV enhanced their action. These results suggest that immunopotentiative action of Rhynchosia volubilis Lour. is more potent than their of Glycine max Merr.

Melanogenesis-Promoting Effects of Rhynchosia nulubilis and Rhynchosia volubilis Ethanol Extracts in Melan-a Cells

  • We evaluated the antioxidant activity and melanogenic effects of black soybean ethanol extracts, including Rhynchosia nulubilis bean ethanol extract (RNBEE), R. nulubilis leaf ethanol extract (RNLEE), R. volubilis bean ethanol extract (RVBEE), and R. volubilis leaf ethanol extract (RVLEE). The total polyphenol contents of RNBEE, RNLEE, RVBEE, and RVLEE were 16.0, 57.7, 365.9, and 260.1 mg/g, respectively. The total flavonoid contents of RNBEE, RNLEE, RVBEE, and RVLEE were 40.4, 91.7, 84.7, and 216.5 mg/g, respectively. The electron-donating abilities of RNBEE, RNLEE, RVBEE, and RVLEE at $1,000{\mu}g/mL$ were 32.4%, 12.7%, 83.5%, and 84.5%, respectively. RNBEE, RNLEE, RVBEE, and RVLEE at $50{\mu}g/mL$ significantly increased (p < 0.01) melanin contents by 30.4%, 32.1%, 35.5%, and 37.4%, respectively, compared to that of the control. RNBEE, RNLEE, RVBEE, and RVLEE at $50{\mu}g/mL$ significantly increased (p < 0.01) intracellular tyrosinase activity by 18.4%, 21.8%, 21.5%, and 21.1%, respectively, compared to that of the control. These results demonstrated that black soybean ethanol extracts promote melanogenesis in melan-a cells. Among the black soybean ethanol extracts, R. volubilis was found to be more effective than R. nulubilis, and leaf extract was found to be more effective than bean extract. The potential mechanism underlying the hyperpigmentation effects of black soybeans is the promotion of tyrosinase activity.

Physiological Activity of the Fermented Small Black Soybean (Rhynchosia volubilis) with a Solid State Culture of the Bearded Tooth Mushroom (Hericium erinaceum) Mycelia (쥐눈이콩-노루궁뎅이버섯 균사체 발효물의 생리활성)

  • To enhance the physiological activity of the Rhynchosia volubilis (RV), R. volubilis (RVHE-A) and R. volubilis-added herbal powder (RVHE-B) were fermented with a solid state culture of Hericium erinaceum mycelia (HE). The total isoflavone contents of the non-fermented RV-A ($489.9{\mu}g/g$) and RV-B ($571.1{\mu}g/g$) were remarkably increased in fermented RVHE-A ($1,836.4{\mu}g/g$) and RVHE-B ($1,276.7{\mu}g/g$). In particular, aglycone isoflavones such as daidzein and genistein were significantly higher in the RVHE-A than any other sample. When hot-water (HW) and EtOH extracts (E) were fractionated from the RV and RVHE, both extracts from the RVHE-A were higher than those from the RV-A in total polyphenol and flavonoid contents. However, the RVHE-B-HW showed a lower polyphenol and flavonoid content level than did RV-B-HW. RVHE-A-HW and -E also had more potent ABTS radical scavenging activity than any extract from the non-fermented RV and other ferments (RVHE-B). In the meanwhile, RVHE-A-HW potently stimulated the production of macrophage activation-related cytokines such as $TNF-{\alpha}$, IL-6 and IL-12 ($841.7{\pm}71.3pg/mL$, $3.9{\pm}0.1ng/mL$, $179.3{\pm}30.2pg/mL$) from peritoneal macrophage more than RV-A-HW ($92.5{\pm}1.5pg/mL$, $0.1{\pm}0.0ng/mL$, $37.4{\pm}5.4pg/mL$) as well as RVHE-B-HW ($557.0{\pm}21.3pg/mL$, $1.8{\pm}0.0ng/mL$, $90.0{\pm}10.0pg/mL$). However, all the EtOH extracts did not show significant activity. In addition, the RVHE-A-HW showed a significantly higher intestinal immune system modulating activity through Peyer's patch and GM-CSF production than did any other extract from RV and RVHE-B. In conclusion, these results suggest that the fermented R. volubilis with H. erinaceum mycelia possesses a possible use as an industrial application as functional food or material.

Rhynchosia volubilis Lour. and Beta vulgaris Modulate Extracts Regulate UV-Induced Retinal Pigment Epithelial Cell and Eye Damage in Mice (약콩, 비트 추출물의 자외선에 의한 망막 상피세포와 마우스의 눈 손상 조절 효능)

  • Ultraviolet (UV)-induced damage plays a major role in ocular diseases, such as cataracts and retinal degeneration. UV irradiation can generate free radicals including reactive oxygen species (ROS), which are known to cause lipid peroxidation of cellular membranes. It has also been shown that UV can damage DNA directly and induce apoptosis. Rhynchosia volubilis Loureiro (the small black bean or yak-kong, RV) and Beta bulgaris (beet, BB) are used as health supplements. In this study, we explored the protective effects of RV and BB against UVA-induced damage in human pigment epithelial (ARPE-19) cells and in mice. RV and BB mixture and their effective constituents (cyanidin, delphidin, petunidin glycosides) improved cell viability and suppressed intracelluar ROS generation. Phosphorylation of p38 mitogen-activated protein kinase (MAPK), c-Jun N-terminal kinase (JNK), Erk1/2 was analyzed by immunoblotting. RV and BB mixture inhibited UVA-induced phosphorylation of p38 MAPK, JNK, Erk1/2 in APRE-19 cells. RV and BB treatment also showed protective effects on ocular damage in UVA-irradiated mice by increasing the levels of endogenous antioxidants such as superoxide dismutase and glutathione. RV and BB have the potential to be used in a range of ocular diseases and conditions, based on in vitro and in vivo study.
