Inhibitory Effect of Retinoic Acid on lipid Synthesis in Human Sebocyte (피지선세포에서 Retinoic Acid의 피지생성억제효과)

  • The differentiation of the sebaceous gland is remarkably species-specific and sebocytes may play crucial parts in the pathophysiologic processes and disorders of pilosebaceous unit SZ95 cell is an immortalized human sebaceous gland cell line that shows characteristics of normal human sebocytes, In this study, we investigated the effect of testosterone and the anti-androgenic effect of 13-cis-retinoic acid (13-cis-RA) on lipid synthesis in SZ95 cells. Cytoplasmic lipid droplets were shown by Oil-red staining. The majority of the SZ95 cells positively labeled with Oil-red dye, while HaCaT cells negatively labeled with Oil red dye. Total lipid level of SZ96 cells is higher 4 times than that of HaCat cells. Testosterone markedly increased 2 times lipid synthesis of SZ95 cells in compared with control. 13-cis-RA significantly inhibited lipid synthesis and cell proliferation in SZ95 cells. Combined treatment with testosterone and 13-cis-RA resulted in a lower total lipid levels than that with androgen alone. In conclusion, SZ95 cells well resembled the morphologic and functional characteritics of normal human sebocytes. This in vitro model could provide a valuable tool for the study of sebocytes with a key role in pathophysiology and differentiation of sebaceous glands.

  • Sebum is an oily substance produced by sebaceous glands in human skin. The differentiation of the sebaceous gland is remarkably species-specific and sebocytes may play crucial parts in the pathophysiologic processes and disorders of pilosebaceous unit. In this study, we investigated the inhibitory effect of Ficis folium on the lipid production using the human sebocyte cell line 5195. Our results showed that Ficis folium significantly inhibits synthesis of sebaceous lipids such as cholesterol esters, triglycerides, and total lipid. And Ficis folium supressed cytoplasmic lipid droplets. On the other hand, Ficis folium didn't effect on cell proliferation. These results suggest treatment with Ficis folium resulted in a lower lipid synthesis in SZ95 cells without cytotoxicity.

  • The aqueous Green tea comes to be used with the Oriental medicine plant, it has the numerous health benefits, including antioxidant, anti-inflammatory, and anti- carcinogenic properties. Epidermal progenitor cells give rise to multiple skin lineages: hair follicle, sebaceous gland and the overlying interfollicular epidermis. Sebocytes are the cells of the sebaceous gland, which synthesize and accumulate lipid dropolets. In order to determine the effect of Green tea on lipid production, several experiments were performed in SZ95 cells (sebocytes). We found that Green tea increased lipid droplets compared with control in a dose-dependent manner. Human sebaceous glands produce sebum, a lipid mixture of squalene, wax esters, triglycerides, cholesterol esters, and free fatty acids that is secreted onto the skin. Therefore, to investigate the effects of Green tea on intracellular lipid levels, we treated SZ95 cells with Green tea, and then examined cholesterol and triglyceride levels. After treatment of the cells with Green tea, the cholesterol and triglyceride levels of SZ95 cells were increased significantly in a dose-dependent manner.

Ponciri Fructus Extract Induces Lipogenesis through Transcription Factor SREBP-1 Activation (지실 추출물의 전사인자 SREBP-1 활성에 의한 지질 생성 촉진)

  • This study was to explore the lipogenic effect by ethanol extract of ponciri fructus (EPF) and possible molecular mechanisms in sebocyte. When SZ95 sebocyte cell line were treated with the EPF, lipid droplets were accumulated in the majority of cells. EPF increased expression of sterol regulatory element-binding protein-1 (SREBP-1) and fatty acid synthase (FAS) in the SZ95 cells. EPF augmented expression of PPAR-${\beta}$ and PPAR-${\gamma}$ but not that of PPAR-${\alpha}$. These results suggest that EPF induces lipogenesis in SZ95 cells through SREBP-1, PPAR-${\beta}$ and PPAR-${\gamma}$ activations.

  • SZ95 cell is an immortalized human sebaceous gland cell line that shows the morphologic, phenotypic and functional characteristics of normal human sebocytes. Sebocytes may play crucial parts in the pathophysiologic processes and disorders of the pilosebaceous unit. The secretory activity of the sebaceous gland is remarkably species-specific and acne is an exclusively human disease. Thus, this SZ95 cells offer possibilities for investigations on the physiology of the sebaceous gland and its role in sebum-associated skin disease such as acne. In this study, we investigated the effects of 13-cis-retinoic acid (13-cis-RA) and spironolactone, frequently used as therapeutic agents of acne, on the lipid synthesis and proliferation of human sebocytes. Cell proliferation was determined by MTT assay and cytoplasmic lipid droplets was shown by Oil-red a staining. Total lipid levels were biochemically estimated by the sulfo-phospho-vanilline reagent. 13-cis-RA and spironolactone significantly inhibited proliferation and lipid levels in a dose-dependent manner. Combined treatment with testosterone and 13-cis-RA or spironolactone resulted in a lower total lipid levels than that with androgen alone. These observations indicate that 13-cis-RA and spironolactone are potent inhibitors of both cell proliferation and lipid synthesis in human sebocytes. We will provide experimental evidence that this human sebocyte cell line serves as an adequate tool for evaluating the anti-lipogenic activity of various compounds potentially useful for the bioactive cosmeceutical ingredients on acne skin, and studying the intracellular biochemical markers depending on the types of compounds from various sources.

  • Lavandula vera is indispensable member of the herb family, used for perfumes and potpourri. Androgens have profound effects on the physiology of the sebaceous gland. Sebum is secreted due to the effect of androgen, which starts to be secreted at puberty. Using the human sebocyte cell line SZ95. the author investigated the inhibitory effect of Lavandula vera on the lipid production. Light microscopic finding were examined numerous cytoplasmic lipid droplets SZ95 cells by Oil red staining and lipid droplets were increased markedly by testosterone. On the other hand, combined treatment with Lavandula vera and testosterone resulted in a lower lipid droplets than with testosterone alone in a dose-dependent manner. These findings indicate that Lavandula vera acts antagonistically to testosterone and inhibits the lipid synthesis of SZ95 cells at the cellular level.

  • Sebum is secreted due to the effect of androgen, which start to be secreted at puberty. Androgens have profound effects on the physiology of the sebaceous gland, Using the human sebocyte cell line SZ95, we investigated the inhibitory effect of Chelidonii Harba (CH) on the subum production. Our results showed that numerous cytoplasmic lipid droplets were examined by Oil red staining and lipid droplets were increased markedly by testosterone. Cell viability was dose-dependently decreased by CH as compared with untreated cells, while total lipid content and cholesterol slightly were increased by CH. Testosterone significantly stimulated the synthesis of total lipid and the synthesis of specific sebaceous lipids such as cholesterol and triglyceride. Combined treatment with CH and testosterone resulted in a lower lipid synthesis than with testosterone alone. Especially cholesteol content was reduced by combined treatment with CH and testosterone. These results indicate that CH inhibits the testosterone-induced lipid synthesis in SZ95 cells and acts antagonistically to androgen at the cellular level.
