Effect of Water Extract of Saussureae Radix in RANKL-induced Osteoclast Differentiation (파골세포 분화에 목향 물 추출물의 효과)

  • Osteoporosis is social problem around the world, because fracture of old age may lead to critical medical condition. Osteoclast is a main target for prevention and treatment of osteoporosis due to their responsibility for bone resorption. Saussureae Radix has been known that has gastro-protective, bronchodilatory effect and has a anti-biotic effect. Saussureae Radix has been widely used in Oriental medicine. However, the effect of extract of Saussureae Radix in osteoclast differentiation remains unknown. Thus, we examined the effect of Saussureae Radix in receptor activator of nuclear factor-${\kappa}$B ligand (RANKL)-induced osteoclast differentiation. From the results of our study, Here we found that Saussureae Radix significantly inhibited osteoclast differentiation induced by RANKL. Saussureae Radix suppressed the activation of NF${\kappa}$B in bone marrow macrophages (BMMs) treated with RANKL. Also, Saussureae Radix significantly inhibited the mRNA expression of c-Fos, tartrate-resistant acid phosphatase (TRAP), osteoclast-associated receptor (OSCAR), nuclear factor of activated T cells (NFAT)c1 and cathepsin K in BMMs treated with RANKL. Particularly, Saussureae Radix greatly inhibited the protein expression of c-fos and NFATc1. especially in the case of NFATc1 expression, a master transcription factor of the differentiation of osteoclasts is very important step for osteoclastogenesis. These results demonstrate that Saussureae Radix may be useful treatment option of bone-related disease such as osteoporosis and rheumatoid arthritis.

Effects of YongdamSaganTangGamibang(YSTG) on the Inflammatory Reactions. (용담사간탕가미방이 염증치료 및 예방에 미치는 영향에 대한 연구)

  • This study was carried out to investigate the effects YSTG on the skin inflammatory reactions. Among the herbal ingredients of YSTG, ethanol extracts of Cinnamoni Ramulus, Polygoni Avicularis Herba and Scutellariae Radix showed potent radical scavenging activity, more than 90$\%$ at a concnentration of 0.01$\%$, tested by DPPH(I,1-diphenyl-2-picryl-hyrazyl) method. And ethanol extract of Saussureae Radix inhibited the lipopolysaccharide-induced release of nitric oxide(NO), 86$\%$ at 50㎍/㎖, by the macrophage RAW 246.7 cells. In addition, the ethanol extracts of Cinnamoni Ramulus and Polygoni Avicularis Herba showed antibacterial activities on Staphylococcus aureus that was reported to cause impetigo and atopic disease, Cinnamoni Ramulus, Saussureae Radix and Plantaginis Semen on Propionibacterium acnes that causes acne, and Cinnamoni Ramulus on Candida albicans that causes cadidiasis, Saussureae Radix on Corynebacterium xerosis that cause osmidrosis axillae. Taken together, I expect that YSTG may be used as an effective drug for treatment on skin inflammation.

Effects of Radix Saussurea on hepatoprotection (목향(木香)함유 DHL과 ML이 간세포 보호에 미치는 영향)

  • Dehydrocostus lactone (DHL) and Mokko lactone (ML) were isolated from Saussureae Radix, and their effects on heme oxygenase-1 (HO-1) expression and hepatoprotection in the liver cell line HepG2 were investigated. DHL induced HO-1 expression and HO activity in a dose-dependent manner, whereas ML lacking one double bond property at 11 and 13 carbons on its own chemical structure had no apparent effects. DHL also induced Nrf2 nuclear translocation and enhanced antioxidant response element (ARE) activation which mediated HO-1 gene transcription. Pretreatment with DHL protected HepG2 cells against oxidative damages caused by H2O2. Interestingly, the hepatoprotective effects of DHL appeared to be associated with HO enzymatic activation, HO-1 expression and Nrf2 activation, because blockage of HO activity by a HO inhibitor and inhibition of HO-1 and Nrf2 cellular synthesis by small interfering RNA abolished heptoprotection afforded by DHL. Taken together, this investigation provides evidence supporting that Saussureae Radix is hepatoprotective against oxidative stress that causes abnormal liver damages.

A Study on Cytotoxicity of Dangkwi-Yonghoe-Hwan (당귀용회환의 세포독성(細胞毒性)에 관(關)한 연구(硏究))

  • The purpose of this research was to investigate cytotoxicity of DangkwiYonghoe-Hwan(DYH) and the constitutive crude drugs on several cancer cell-lines, thymocytes, splenocytes and 3T3 cells. The DYH consists of Coptidis Rhizoma, Scutellariae Radix, Phellodendri Cortex, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus, Saussureae Radix and Angelicae Gigantis Radix. The cytotoxicity was determined by MTT method. The DYH inhibited the proliferation of MOLT-4, K562, HL-60, Jurkat, L1210, P815, S180 and Yac-1, thymocyte, splenocyte and 3T3 cells. The cytotoxicity of Coptidis Rhizoma on the cancer cell-lines was the most potent in the constitutive crude drugs. The proliferation of cancer cell-lines was partly inhibition and partly increase by the treatment of Scutellariae Radix, Gardeniae Fructus, Gentianae scabrae Radix, Indigo pulverata Levis, Aloe, Rhei Rhizoma, Moschus and Angelicae Gigantis Radix. Phellodendri Cortex and Saussureae Radix had a poor cytotoxicity on cancer cell-lines. Coptidis Rhizoma and Phellodendri Cortex inhibited the proliferation of thymocyte, splenocyte and 3T3 cells.

Isolation and Quantitative Determination of Costunolide from Saussurea Root (목향(Saussureae Radix)으로부터 Costunolide의 분리 및 함량분석)

  • Isolation and quantitative determination of costunolide from Saussurea lappa Clarke (Compositae) has been conducted by using HPLC method. Costunolide in an acetone extract from the crude drug was separated on a RP-18 column using a $MeOH-H_20$ mixture (65:35) as an eluent and the average content is about $1.32{\sim1.42%.$ The content of costunolide in dried extract was decreased by about 24% in seven days. However it showed a slight decrease in solution. It is highly recommended that quantitative determination of costunolide from Saussureae Radix should be conducted as early as possible after solvent extraction.

  • Nitric oxide (NO) can mediate numerous physiological processes, including vasodilation, neurotransmission, cytotoxicity, secretion and inflammatory response. The regulation of NO production by inducible NO synthase (iNOS) is considered to be the possible target of the development of anti-inflammatory agent, based on the observation that NO can activate cyclooxygenase, which results in the synthesis of prostaglandins. In an effort to screen new inhibitor of NO production from about 352 species of herbal extracts, we found 9 species with 50% or more inhibitory effect on NO production. Especially, the dose-dependent inhibition of NO production in lipopolysaccharide-treated macrophages by two of the herbal extracts (Artemisiae asiaticae Herba and Saussureae Radix) was due to the decrease in the expression of iNOS.

  • Saussureae Radix, the dried root of Saussurea lappa Clarke, has traditionally used for alleviating pain in abdominal distention and tenesmus, indigestion with anorexia, dysentery, nausea, and vomiting. Here we observed that methanol extracts of Saussurea Radix inhibited CDK2 activities in vitro. This inhibitory compound was isolated and identified as dehydrocostuslactone, one of the major constituents of Saussurea Radix. It is well known that dehydrocostuslactone induces apoptotic cell death. In this study, we also showed that dehydrocosruslactone inhibited cellular Rb phosphorylation and blocked cell growth at the concentration below $12\;{\mu}g/ml$ at which apoptotic cell death was not observed. Taken together, these results indicated that dehydrocostuslactone showed its anti-proliferative effects through the inhibition of CDK2 activity as well as the induction of apoptotic cell death.

Cytotoxicity in HL-60 cells and human lymphocytes and effect of leukemia cell differentiation induced by Saussureae Radix extract (정상사람림프구와 HL-60 cell에서 목향의 세포독성과 백혈병세포 분화효과에 관한 연구)

  • Objectives : This study was focused to investigate the toxicity of Saussurea lappa (SL) extracts in HL-60 cells and human lymphocytes. We also examined the differentiation effect of SL against leukemia cells. Methods : For examining the toxicity of SL, cytokinesis-block micronucleus (CBMN) assay and single cell gel eletrophoresis (SCGE) assay were used in present study. The cell differentiation effect of SL was evaluated by nitroblue tetrazolium (NBT) reduction assay. Results : The inhibition of cell growth in HL-60 cells was observed in a dose-dependant manner after SL treatment for 24 h. According to SCGE assay, HL-60 cells treated with SL increased DNA damage at $10{\mu}g/m{\ell}$, while DNA damage was induced by 0.1, 1, $10{\mu}g/m{\ell}$ concentration of SL in human lymphocytes. Our results indicated that SL have no genotoxic effect in HL-60 cells and human lymphocytes. Additionally, the differentiation effect was induced in $1{\mu}g/m{\ell}$ SL-treated HL-60 cells. Conclusions : From above results it is suggested that SL could be beneficial for the preparation of the useful agent for treating leukemia.

Spasmolytic and Anti-peptic Ulcer Activities of Crude Drugs Acting on Gastrointestinal Tract in Rats (흰쥐에서 위장관에 작용하는 생약의 진경 및 항위궤양 효능)

  • The water extracts of ten crude drugs were tested for the spasmolytic and anti-peptic ulcer activities on rat ileum smooth muscle contraction and aspirin-induced acute hemorrhag ic erosive gastritis respectively. The water extract of Aurantii immaturi pericarpium(AIP)($IC_{50}=1.5{\times}1O^{-2}$g/l), Aurantii nobilis pericarpium(ANP)($IC_{50}=2.5{\times}1O^{-2}$g/l), Cyperi rhizoma(CR)($IC_{50}=3.3{\times}1O^{-2}$g/l). Linderae radix(LR) ($IC_{50}=6.8{\times}1O^{-2}$), Aurantii fructus immaturus(AFI)($IC_{50}=11.8{\times}1O^{-2}$), Saussureae radix(SR)($IC_{50}=13.2{\times}1O^{-2}$g/l) and Ponciri fructus(PF)($IC_{50}=23.3{\times}1O^{-2}$g/l) showed inhibitory activity on the isometric contraction of rat ileum smooth muscle induced by electrical stimulation in a concentration-dependent manner, whereas the water extracts of Arecae pericarpium(AP), Agastachis herba(AH) and Magnoliae cortex(MC) potentiated the isometric contraction. In the aspirin-induced acute gastritis, the water extracts of MC, AP and CR reduced significantly the gastric juice secretion, gastric juice acidity and pepsin activity. They also showed protective activity of gastric mucosal layer from erosion and petichial hemorrhage in gross and histological examination.

Detection of Aristolochic Acid from Medicinal Herbs

  • Aristolochic acid has been known to be a carcinogenic compound and to cause Chinese herbs nephrophathy. To determine the content of aristolochic acid in various medicinal herbs marketed domestically, samples of Aristolochia fangchi, Aristolochia fructus, Aristolochia radix, Asiasari radix, Saussureae radix, and Akebia caulis were analyzed using TLC and HPLC. The optimal conditions for TLC and HPLC were established for the quantitative analysis of aristolochic acid. This study showed that Aristolochia fangchi and Aristolochia fructus examined in this study contain aristolochic acid of 3.9 and 2.3 mg/g sample, respectively. In contrast, aristolochic acid was not detected in other samples.
