Detection of Serum Anti-Extracellular Protein Kinase a Autoantibodies as a Potential Tumor Marker

  • In previous studies, it has been discovered that cancer cells not only overexpress regulatory subunit I (Rl)/protein kinase type I (PKA-I) but also secrete outside the cell an extracellular form of PKA (ECPKA) and that the ECPKA secretion detected in patients' serum is obviously greater than that found in non-cancer patients or healthy subjects. We now found that ECPKA elicits the formation of serum autoantibodies that can serve as a cancer diagnostic and prognostic marker. To measure the presence of anti-ECPKA autoantibody in the human sera, basic methodology for ECPKA assay was established an enzyme-linked immunosorbent assay (ELISA). We obtained serum samples from 199 patients with different types of cancer, and also obtained 31 serum samples to compare with ECPKA concentrations from non-cancer patients and 119 normal volunteers. Compared with normal or non-cancer patient sera, we found that the frequency of anti-ECPKA autoantibody was significantly higher in cancer patients (88%) than in those without cancer (17%). Furthermore the presence of anti-ECPKA autoantibodies in the serum of cancer patients was highly correlated with the site of metastasis. The immunoassay developed for anti-ECPKA antibodies is highly sensitive and specific. Therefore, this discovery of an autoantibody-based cancer diagnostic may have serious clinical application and may become an important advance over current technology.

Effects of Intravenous Administration of Taurocholate on Serum Aryl Sulfotransferase Activity in Cholestatic Rats

  • Possible mechanisms of increased serum aryl sulfotransferase (AST) isozyme activities in cholestatic rats were studied. Serum AST-I, II and -III, IV isozymes activities were determined from the experimental rats with common bile duct ligation (CBDL) or choledocho-caval shunt (CCS). The activities of serum AST-I, II and -III, IV isozymes were found to be increased significantly in both the CCS plus taurocholic acid (TCA) injected group, and the CBDL plus TCA group than those in each control group, such as CCS or CBDL alone groups. The above results suggest that the elevated serum AST most likely due to increased hepatocyte membrane permeability caused by TCA mediated liver cell necrosis.

  • Zinc is an essential trace element. Previos methods for the determination of zinc in serum have been difficult and reported findings must be treated with caution. The normal range of zinc in serum varies significantly according to different methods and author. The scope of this work was to establish the normal range of zinc in serum values in healthy Korean persons. Methods are described for the determination of zinc in serum diluted ten-fold with deionised water using Hitachi Model 207, atomic absorption spectrophotometer. The values of zinc in serum of 23 healthy Korean persons by atomic absorption spectroscopy ranged from 87.6 to $131.2\;{\mu}g/100\;ml$ with the mean value $109.1\;{\mu}g/100\;ml$ and standard deviation 10.86. In this method, satifactory result was obtained with regard to the coefficient variation and recoveries of added zinc in the measurenents.

Effect of Calcium on Estrogen and Follicle Stimulating Hormone Secretion in Rabbits

  • The effect of calcium gluconate on estrogen (estradiol) serum level as well as follicle stimulating hormone level was studied. Our results revealed that oral administration of calcium gluconate (100 mg/kg body wight) to adult non-pregnant female rabbits caused a significant increase of serum levels of estradiol and follicle stimulating hormone. On the other hand, oral contraceptive (Norminest tablets) decreased significantly follicle stimulating hormone serum level, while combined administration of calcium gluconate and oral contraceptive caused significant increase of serum level of follicle stimulating hormone compared with control values. Also, concurrent administration of calcium gluconate and Norminest tablet increased significantly the rate of conception compared with group recieved Norminest tablets only. These results indicated that combined administration of calcium and oral contraceptives must be cautiously.

  • Ghrelin and obestatin are a single gene products and are a multiple functional peptides that regulates energy homeostasis, and food intake. In the present work, we studied the secretion of ghrelin and its co-secreted peptide obestatin in 44 patients with ischemic heart disease with that of 27 healthy matched controls. Here we first conducted using an immunohistochemistry assay to screen whether human salivary glands have any obestatin immunoreactivity. Then, serum and saliva obestatin and acylated ghrelin levels were determined by using Radioimmunoassay. Our immunohistochemical analysis demonstrated that obestatin was localized in the striated and excretory duct of human salivary gland. We also report for the first time that obestatin, like ghrelin, is present in human salivary gland and saliva. No evidence of the role of obestatin or ghrelin saliva levels in the context of ischemic heart disease was found. Salivary ghrelin and obestatin levels are correlated in controls with the blood levels. Determination of salivary values could represent a non-invasive alternative to serum ones that can be useful in clinical practice.

  • This study find out the effect of red Ginseng extract(1.0g/kg) against TBTO(10, 20 and 40mg/kg) poisoning on each organs, hematological, serum biochemistry in rat for 3 weeks. 1. Each organ weight per body weight ration in treated group, all rats liver were significantly increased. (P<0.05,P<0.01)2. Iin TBTO treated group, the hematological parameters such as WBC, RBC, Hgb, Hct, PLt, MCV, MCH and MCHC in serum were remarkably decreased in comparison to that of control group. Only added red Ginseng extract group were slightly decreased but not significanted. 3. In TBTO treated group, the biochemical parameters such as AST, ALT in serum were remarkable elevated in comparison to that control group (P>0.05).4. In TBTO treated group, the activity of Triglyceride in serum of male rat were significantly increased in comparison to that of control group (P<0.05, P<0.01).5. In case of B.U.N activity in treated group(10, 20 and 40 mg/kg) were almost increased in comparison to that of control group, but not added red Ginseng extract.

  • Iron deficiency anemia (IDA) is the world's most common nutritional problem. It is characterized by a low hemoglobin (Hb) level and low iron status. A study was conducted to investigate the incidence of iron deficiency anemia in day scholar girls belonging to different socioeconomic strata at Punjab University, Lahore. Iron status of the subjects was estimated by measuring Hb, hematocrit (Hct), red blood cell (RBC) count, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCR), serum iron, serum ferritin, total protein and albumin. Results indicated that females belonging to low socioeconomic strata had lower values for Hb, Hct, RBC count, total protein and albumin. Serum iron, serum ferritin, MCV and MCH values fell within the normal range for all of the socioeconomic groups. However, serum iron and ferritin varied with socioeconomic status and higher-income groups had significantly higher serum iron and ferritin. It was concluded that anemia may develop due to poor intake and absorption of iron and that those in the low-income bracket are the most affected group.

  • The ovarian follicular fluid was found to contain steroidogenesis stimulatory protein similar to albumin from human and buffalo. Therefore, the albumins from various species, commercial and purified, were studied for their steroidogenic effect on progesterone secretion by granulosa cells from buffalo ovaries, during culture. A dose of $20{\mu}g$ of bovine serum albumin was optimum to exhibit maximum progesterone secretion on day 6 of culture, in medium ($350{\mu}l$) containing $10^5$ cells. Among commercial albumins, chicken albumin showed highest effect on progesterone secretion, which was followed by albumins from goat, bovine, human, sheep and rat, respectively at day 6 of culture. The albumins were also purified from blood serum of buffalo, goat and rat using salt fractionation, ion-exchange chromatography, gel filtration and SDS-PAGE. The highest stimulatory effect on progesterone secretion was shown by albumin purified from buffalo blood serum and lowest by that from rat blood. Comparatively the buffalo and goat albumins were more biologically active than commercial albumins. The presence of some active molecules conjugated with freshly purified albumins may be responsible for better stimulatory effect.

Antidiabetic effect of Enicostemma littorale Blume aqueous extract in newly diagnosed non-insulin-dependent diabetes mellitus patients (NIDDM): A preliminary investigation

  • The antidiabetic efficacy of Enicostemma littorale Blume (chhota chirayata) aqueous extract was examined in newly diagnosed non-insulin-dependent diabetes mellitus (NIDDM) patients taking only the extract and was administered as two divided doses, half an hour before meal as 5g of aqueous extract per single dose. Out of the 20 patients volunteered, 11 successfully completed the 2 month trial and a significant decrease in fasting & postprandial blood glucose and glycosylated haemoglobin levels were observed along with a significant improvement in the antioxidant parameters of the patients. There was also a significant increase in serum insulin levels in 7 patients after extract treatment as compared to levels before treatment. Serum total cholesterol and serum triglyceride levels were decreased significantly with a significant increase in serum HDLCholesterol levels. Other vital parameters remained stable and no side effects were observed. This is the first report showing the hypoglycemic, antioxidant and hypolipidemic properties of the aqueous extract of E. littorale Blume in non-insulin dependent diabetes mellitus patients.

EFFECTS OF POLYPHENOLS OF Cocos Nucifera HUSK FIBRE ON SELECTED KIDNEY FUNCTION INDICES IN MICE

  • Decoction of Cocos nucifera husk fibre is used indigenously in Nigeria for malaria treatment. Polyphenols have been identified as the phytochemicals responsible for the antimalarial activity of Cocos nucifera husk fibre, though their toxicity has not been evaluated. The polyphenols of Cocos nucifera husk fibre were therefore evaluated for their effects on selected kidney function indices in mice. Fifty mice were randomly divided into five groups (A-E) of ten mice each. Mice in group A were orally administered 5% DMSO solution while those in groups B, C, D and E were orally administered 31.25, 62.5, 125 and 250 mg/Kg body weight of the polyphenols respectively for seven days. Serum urea, creatinine and uric acid concentrations were determined. Serum levels of sodium, potassium, chloride and calcium ions and kidney alkaline phosphatase (ALP), glutamate dehydrogenase (GDH) and gamma-glutamyltransferase (GGT) activities were also determined. The results showed that the polyphenols significantly reduced (p<0.05) urea concentration at 250 mg/Kg body weight and creatinine concentration at all doses compared to controls. The polyphenols caused no significant alteration (p>0.05) in serum uric acid concentration and kidney ALP, GGT and GDH activities compared to controls. There was significant increase (p<0.05) in serum sodium ion concentration at 31.25, 125 and 250 mg/Kg body weight of polyphenols whereas significant increase (p<0.05) in serum potassium and chloride ions was observed at 62.5 and 250 mg/Kg body weight compared to controls. Thus, polyphenols of Cocos nucifera husk fibre may adversely affect some osmoregulatory functions of the kidney, especially at higher concentrations.
