Evaluation of the Effect of Sipjeondaebo-Tang By-Product Powder on Broiler Production (십전대보탕부산물을 활용한 육계생산성 향상 평가)

  • This study was conducted to evaluate the effects of sipjeondaebo-tang by-product powder on the growth performance of broilers. A total of 120 broiler chicks (Arbor acres) at 0 days old were assigned to one of two treatments with three replications: control and 1% sipjeondaebo-tang by-product powder. Weight gain at 3 weeks and feed intake at 5 weeks were significantly (p<0.05) influenced by the sipjeondaebo-tang by-product powder treatments. However, there were no significant differences in the feed conversion ratio between treatments as a function of treatment time (p>0.05). Overall, weight gain and the feed conversion ratio were slightly greater for the sipjeondaebo-tang by-product powder treatment than the control treatment. It was concluded that sipjeondaebo-tang by-product powder may be an appropriate source of feed additive, without any negative effects on the growth performance of the broilers.

Micronucleus test of Sipjeondaebo-tang extract using the bone marrow cells in ICR mice (ICR 마우스 골수세포를 이용한 십전대보탕 소핵시험)

  • Objectives : This study was to assessment the toxicity of Sipjeondaebo-tang(Shiquan dabu-decoction) by micronucleus test. Methods : Sipjeondaebo-tang(Shiquan dabu-decoction) water-extract in vivo micronucleus test was performed using 7 weeks ICR mice. At 24 hours after with Sipjeondaebo-tang extract at the doses of 0, 500, 1000 and 2000 mg/kg/day by peritoneal route mice were sacrificed and bone marrow cells were prepared for smear slides. Results : As a result of counting the micronucleus polychromate erythrocyte of 2000 polychromate erythrocyte, all treatment groups did not show statistically significant increase than negative control group. and there was no clinical sign and body weight connected with injection of Sipjeondaebo-tang(Shiquan dabu-decoction) extract. Conclusions: It was concluded that Sipjeondaebo-tang extract did not induce micronucleus in the bone marrow cells of ICR mice

Single Oral Dose Toxicity Test of Fermented Sipjeondaebo-tang Extracts in Mice (마우스를 이용한 십전대보탕(十全大補湯) 발효물의 단회투여 독성 연구)

  • Objectives : Sipjeondaebo-tang is a medicine traditionally prescribed as a restorative. The aim of this study was to investigate the single oral dose toxicity and safety of extract of fermented Sipjeondaebo-tang in ICR mice. Methods : In single oral dose toxicity study, non-fermented or fermented Sipjeondaebo-tang were administered by oral gavage to ICR mice (5 males, 5 females) at single doses of varying concentrations: 1250, 2500 and 5000 mg/kg. Changes of body weight, general behavior, adverse effects and mortality were determined throughout the experimental period. Hematological parameters, organ weights and necropsy findings were evaluated at the end of the experiment. Results : There were no mortality or signs of toxicity in single oral dose toxicity studies. There were also no significant differences in body weight, organ weight, or hematological parameters between the treatment and control groups. Conclusions : Fermented Sipjeondaebo-tang did not cause remarkable adverse effects in ICR mice. The oral lethal dose of fermented Sipjeondaebo-tang is more than 5000 mg/kg and no-observed-adverse-effect level (NOAEL) of the extract for both male and female mice is 5000 mg/kg.

Bacterial Reverse Mutation Test of Sipjeondaebo-tang (세균을 이용한 십전대보탕 복귀돌연변이 시험)

  • Objectives: This study was to assess the toxicity of Sipjeondaebo-tang by bacterial reverse mutation test. Methods: In this study, to evaluate the bacterial reverse mutation of Sipjeondaebo-tang water-extract, the in vitro Ames test using Salmonella typhimurium (TA98, TA100, TA1,535, TA1,537) and Escherichia coli(WP2uvrA) were performed with Sipjeondaebo-tang water extract at the concentrations 0, 312, 625, 1250, 2500 and 5000 ${\mu}g/plate$. Results: Sipjeondaebo-tang water extract was negative in Ames test with both Salmonella typhimurium or Escherichia coli with and without rat liver microsomal enzyme (S9- fraction and S+ fraction). Conclusions: According to these results, we concluded that a Sipjeondaebo-tang water extract did not cause bacterial reverse mutation.

  • Objective: To evaluate the effects of Sipjeondaebo-tang (十全大補湯) extract on a stage two pressure sore. Methods: Sipjeondaebo-tang extract (5 g) was given three times a day to a treat the pressure sore. Results: The pressure sore in the sacrococcygeal region was improved. A second pressure sore ofthe right greater trochanter region, which emerged during the study, was also improved. Conclusions: Sipjeondaebo-tang extract may represent an effective treatment for pressure sores, but further studies are needed.

  • Objectives : This study was to assessment the toxicity of Sipjeondaebo-tang(Shiquan dabu-decoction) by Chromosomalanomaly test. Methods : Sipjeondaebo-tang(Shiquan dabu-decoction) water-extract in vivo Chromosomalanomaly test was performed using chiness hamster lung cell line. Results : Sipjeondaebo-tang water extract was negative in Chromosomalanomaly test at the doses of 0, 625, 1250 and $2500{\mu}g/m\ell$ at 6h and 24h.(S9- fraction). Chromosomalanomaly test(S+fraction) was also negative at the doses of 0, 1250, 2500 and $5000{\mu}g/m\ell$. Conclusions : It was concluded that Sipjeondaebo-tang extract did not induce Chromosomalanomaly in the chiness hamster lung cell.

Acute Toxicity Study on Sipjeondaebo-tang in Rats (SD 랫드를 이용한 십전대보탕의 급성 독성 연구)

  • Sipjeondaebo-tang has been traditionally prescribed a medicine as a restorative. In this study, we investigated the acute toxicity about water-extracted Sipjeondaebo-tang. Thirty rats completed 14 days of oral Sipjeondaebo-tang at the respective doses of 0(control group), 2000 and 5000 mg/kg. We observed survival rates, general toxicity, change of body weight and autopsy. To be confirmed the data for the toxicity and safety problems of oriental medicine prescription. Compared with the control group, we could not find any toxic alteration in all treated groups (2000 and 5000 mg/kg). LD50 of Sipjeondaebo-tang was over 5000 mg/kg and it is very safe to SD rats.

Mitigative Effect of Sipjeondaebo-tang on RhoA Activation in Cold-Exposed Vascular Cells (저온 노출된 혈관 세포 내 RhoA 단백질 활성화에 대한 십전대보탕의 억제 효과)

  • Objective: To investigate the vasodilatory effect of Sipjeondaebo-tang by inhibiting RhoA activity in vascular cells during cold exposure. Methods: Human vascular endothelial cells and pericytes were pretreated with Sipjeondaebo-tang for 30 min, followed by incubation at 37 ℃ (control) or 25 ℃ (cold exposure) for 30 min. Activation of endothelin-1-mediated RhoA in pericytes was assessed by pretreating the cells with Sipjeondaebo-tang for 30 min, followed by incubation with endothelin-1 at 37 ℃ for 30 min. Western blotting was performed to measure the expression of active RhoA. Endothelin-1 and nitric oxide release from endothelial cells was examined with enzyme-linked immunosorbent assay kits. The formation of stress fibers and focal adhesion complexes was analyzed by immunocytochemistry. Results: Cold treatment activated RhoA in both pericytes and vascular endothelial cells, whereas Sipjeondaebo-tang treatment inhibited this activation. Sipjeondaebo-tang treatment also reversed the cold-mediated production of endothelin-1 and nitric oxide. Cold exposure promoted the formation of stress fibers and focal adhesion complexes by increasing the expression of phospho-focal adhesion complex kinase, whereas Sipjeondaebo-tang treatment suppressed this response. Conclusions: These findings suggested that Sipjeondaebo-tang inhibits cold-induced RhoA activation and its related pathway components, including endothelin-1 and nitric oxide, in vascular cells. Therefore, Sipjeondaebo-tang could be beneficial for the treatment of Raynaud's phenomenon.

Pancytopenia Improves with Compromised Liver Function in A Hepatocellular Carcinoma Patient Treated by Modified Sipjeondaebo-tang (십전대보탕가감방을 투여한 간세포암종 환자에서 범혈구감소증 호전 및 간수치 상승 증례)

  • Objectives : To observe the therapeutic effects of an oriental herbal prescription in a hepatocellular carcinoma patient with splenomegaly and pancytopenia. Methods : Modified Sipjeondaebo-tang was prescribed three times a day to a hepatocellular carcinoma patient with splenomegaly and pancytopenia. Laboratory tests were carried out regularly to observe the therapeutic effects of the oriental herbal prescription for pancytopenia. Results : When treated with modified Sipjeondaebo-tang including Cinnamomi Cortex, the levels of white blood cells, red blood cells, hemoglobin, and hematocrit increased. However, aspartate transaminase, alanine transaminase, alkaline phosphatase, gamma-glutamic transpeptidase, total bilirubin, direct bilirubin levels also increased. Conclusions : Administering modified Sipjeondaebo-tang with Cinnamomi Cortex showed effect in improving pancytopenia but an increase in liver enzyme levels was also observed.

Physicochemical Properties of Pekin Duck Breast Meat from Ducks Fed Diets Containing Different Types of Sipjeondaebo-Tang by- Products and Red Ginseng Marc with Fermented Red Koji (십전대보탕 부산물과 발효홍국 홍삼박의 혼합제재를 여러 가지 제형에 따라 오리사료에 첨가 시 오리 가슴육의 변화)

  • This study investigated the physicochemical properties of Pekin duck breast meat obtained from ducks fed diets consisting different types of sipjeondaebo-tang by-products and red ginseng marc with fermented red koji during storage. A total of 180 Pekin ducks (0-day old) were divided into four groups, each consisting of three replicates (15 ducks per pen). This study investigated diets with four types of treatments: control (basal diet),1% blend powder, pelleted 1% blend, and coated pellets of 1% blend; the blend was a mixture of sipjeondaebo-tang by-products powder and red ginseng marc with fermented red koji. There was no significant difference (p>0.05) in duck breast meat pH on storage days 3 and 7, TBARS on storage days 0 through 7, and DPPH radical scavenging on storage days 0 and 7. However, the pH values on storage day 0 and DPPH radical scavenging on storage day 3 were significantly different (p<0.05) in the meats from control and treated diet fed ducks. Especially, on storage day 7, the breast meat from ducks treated with different types of sipjeondaebo-tang by-products and red ginseng marc with fermented red koji showed lower TBARS values and increased DPPH radical scavenging activity compared to the control. In conclusion, addition of different types of sipjeondaebo-tang by-product and red ginseng marc with fermented red koji to 1% blend might be helpful in increasing antioxidant effects and reducing product wastage.
