복합한약제제가 Pentobarbital에 의해 유도된 수면시간에 미치는 영향 (Effects of the Combined-Preparation of Crude Drugs on Pentobarbital-induced Sleeping Time)

  • This experiment was performed to investigate whether 50% ethanol extracts of the combined-preparation of Longanae Arilus, Chrysanthemi Flos, Zizyphi Fructus and Ginseng Radix alba (CPE) has hypnotic effects and/or enhances pentobarbital-induced sleeping time. Locomotor activity was evaluated using a ambulometer of tilting-type. The sedative-hypnotic effects were evaluated by measuring the sleeping onset time and sleeping time in pentobarbital-treated mice 30 min. after oral administration of CPE and muscimol. The intracellular $Cl^-$ concentration of cerebellar granule cells was estimated using $Cl^-$ sensitive fluorescence probe N-(ethoxycarbonylmethyl)-6-methoxyquinolinium (MQAE). CPE (150 mg/kg) decreased the locomotor activity, but CPE itself did not induce sleep. However, CPE reduced sleeping onset and prolonged sleeping time induced by pentobarbital (42 mg/kg). In addition, CPE (2 ${\mu}g/ml$) and pentobarbital (2.5 ${\mu}M$) itself did not affect on the chloride influx in primary cultured cerebellar granule cells, but the combination of CPE and pentobarbital (2.5 ${\mu}M$) increased the chloride influx onto the cells. In conclusion, it is suggested that CPE might augment pentobarbital-induced sleep through the increase of chloride influx.

  • Prednisolone acetate was administered in deers sedated with xylazine hydrochloride. Sleeping time in deers given prednisolone after xylazine sedation was shortened a little in Red deers, approximately one half in Elk and Sika deers comparing with deers sedated with xylazine alone. It was proved that prednisolone acetate shortened the recovery time of deers sedated with xylazine hydrochloride.

수종 중추성 약물이 Barbiturates 및 Non-barbiturate의 수면효과에 미치는 영향 (The Influence of Various Drugs Acting on the Central Nervous System on Sleeping Time of Barbiturates and Non-barbiturate)

  • The effects of 5 different drugs (amphetamine, caffeine, serotonin, sod. salicylate and pentazocine) on the duration of action of two barbiturates (thiopental and pentobarbital) and an intravenous anesthetic (propanidid) were determined in rats. Duration of action was determined by the time elapsing between loss and return of the righting reflexes. All drugs were injected intraperitoneally except propanidid which was administered by the intravenous route. Preliminary experiments indicated that at a dose of 40 mg/kg either of the two barbiturates or propanidid produced loss of the righting reflexes without death. At this dose, however, the duration of action of propanidid was extremely short. However, this dose was selected for subsequent studies. 1. At the dose employed amphetamine shortened the sleeping time of three compounds. 2. Caffeine and theophylline shortened the sleeping time of thiopental and prolonged the duration of action of pentobarbital. 3. Serotonin had no effect on duration of action of the barbiturates but prolonged the sleeping time produced by propanidid. 4. Sod. salicylate significantly prolonged the sleeping time of the barbiturates whereas pentazocine exhibited this effect only in relation to thiopental.

Tranquilizer-like Effects of Sanjoinine A: Possible GABA/Benzodiazepine Receptors Complex Involvement

  • Zizyphi Spinosi Semen (ZSS) has been widely used for the treatment of anxiety and insomnia in Korea and China. This experiment was performed to know whether sanjoinine A, one of major alkaloid compounds of ZSS has anxiolytic and hypnotic effects through the GABAergic systems. Our results showed that administration of sanjoinine A increased open arm entries and spent time in open arm in the elevated plus-maze and increased head dips in hole board test. Different from traditional anxiolytic, diazepam, sanjoinine A itself did not decrease locomotor activity and strength level in mice. Furthermore, Sanjoinine A (0.5-2.0 mg/kg) prolonged sleeping time and reduced sleeping latency induced by pentobarbital in a dose-dependent manner similar to muscimol, a $GABA_A$ receptor agonist. Sanjoinine A (0.25-1.0 mg/kg) also increased sleeping rate and sleeping time in the combined administration at the sub-hypnotic dose of pentobarbital and showed synergic effects with muscimol in potentiating sleeping onset and enhancing sleeping time induced by pentobarbital. However, sanjoinine A itself did not induce sleeping at the higher dose. In addition, both of sanjoinine A and pentobarbital increased chloride influx in primary cultured cerebellar granule cells. Sanjoinine A decreased the $GABA_A$ receptor ${\alpha}$-subunit expression and increased ${\gamma}$-subunit expression, and had no effects on abundance of ${\beta}$-subunit in primary cultured cerebellar granule cells, showing different expression of subunits from pentobarbital. In conclusion, sanjoinine A shows anxiolytic-like effects and augments pentabarbital-induced sleeping behaviors through the modification of GABAergic systems. 청소년의 앉아 있는 시간과 저체중 및 비만의 관계 (Association between sitting time and BMI-defined low weight and obesity in Korean adolescences)

  • Objectives: This study aimed to examine the association between sitting time and BMI-defined low weight and overweight/obesity in Korean adolescents. Methods: Data were from Adolescent Health Behavior Online Survey (KYRBS) in 2015 including 63,345 adolescents aged 12-18 years. The analysis investigated low-weight and overweight/obesity odds ratio(OR) according to study sitting time and leisure sitting time adjusted for sex, school grade, social-economic status(SES), physical activity and sleeping time. Results: After adjusting for sex, school grade, SES, physical activity and sleeping time, study sitting time and leisure sitting time were not significantly associated with low-weight. However, leisure sitting time was positively associated with overweight/obesity. Conclusions: Health program to less sitting time should be established with consideration for differences in risk according to sex, age, SES and active leisure time through the school physical class should be provided to adolescents. Additionally, strategies to promote healthy weight are required to integrate sitting time, physical activity and sleeping time in Korean adolescents.

동물모델에서 시호가용골모려탕(柴胡加龍骨牡蠣湯)의 수면유도 및 항불안 효과 (Sleep-promoting and Anti-anxiety Effects of Shihogayonggolmoryo-tang in Mice)

  • Shihogayonggolmoryo-tang (ST) is a Korean medical herb cocktail which has been used to treat anxiety induced insomnia. In this study, we will examine sleep-promoting and anti-anxiety effects of ST, and investigate its mechanism. ICR mice were divided into three groups for the first examination : control group (n=11), ST50 group (50 mg/kg, po, n=11), ST200 group (200 mg/kg, po, n=11). Sleep-promoting effect was confirmed by measuring the sleeping duration time and sleeping onset time after thiopental sodium treatment (50 mg/kg, ip). ICR mice were divided into five groups for the second examination : control group (n=11), ST200 group (200 mg/kg, po, n=11), ST200+Flumazenil group (ST 200 mg/kg, po, flumazenil 0.3 mg/kg, ip, n=11), diazepam group (1 mg/kg, ip, n=11), diazepam+flumazenil group (diazepam 1 mg/kg, ip, Flumazenil 0.3 mg/kg, ip, n=11). Anxiety behavior and sleep-promoting effect was confirmed by open field test and measuring the sleeping duration time and sleeping onset time. Expression levels of c-fos in tuberomammillary nucleus (TMN) and ventrolateral preoptic nucleus (VLPO) were analyzed by immunohistochemistry. ST treated group showed significantly decreased anxiety behavior and enhanced sleeping duration time and sleeping onset time concentration dependently. The expression of c-fos was significantly upregulated in VLPO as sleep-inducing center and TMN as downregulated in arousal center by ST treatment. In addition, all effects of ST were reversed by flumazenil. Our results suggest that ST has sleep-promoting and anti-anxiety effects through regulating arousal center (TMN) and sleep-inducing center (VLPO).

지역사회 일부 대학생의 수면습관이 건강에 미치는 영향 요인 (Effects on health by sleeping habits from few local university students)

  • Background & Objective, Methods: This study aims to analyze the reason why if university students feel lack of sleep and study how much we know about correct sleeping habits and thus increase attention on correct sleeping habits and prepare plans for applying this into our lives thus 210 male and female students from 2 universities in Daegu were studied during May 10 and 15 of 2006 and the summary and results are as follows. Results: Sleeping is very important since it as much as takes about 1/3 of our lives. However, most of students do not sleep well. Therefore, this study researched the reason for not sleeping well and the status on incorrect sleeping habits subjecting 210 university students. The results of this research showed that average sleeping hours are for $6{\sim}8$ hours and majority of students went to bed before lam and got up before 7am. Majority of students went to bed late to watch TV or doing computers from both male and female students followed by attending various gatherings. It took $10{\sim}20$ minutes to fall asleep and reasons for waking up while sleeping was with no special reason. The most sleepy time was after meals at 39% followed by during class, during daytime, while using transportations. More female students answered to take nap than male students and majority of them answered as taking nap for less than 2 hours. And, 51% answered that they slept well and 49% answered that they did not, and majority of them answered the reason for not sleeping well as being sensitive, or because of dreams and nightmares. Chronic fatigue was the biggest cause for lack of sleeping followed by fall in concentration, skin diseases. Conclusions: The results of this study showed that we need to take plenty of sleep by exercising regularly or sleeping for daily appropriate hours, maintaining peaceful mind before going to bed, avoid taking naps and limiting the time for watching TV and doing computers to develop correct sleeping habits. Also, we hope that it will Provide an opportunity to live healthy and quality lives mentally as well as physically by realizing and practicing the importance and necessity of sleeping.

초등학생의 컴퓨터 사용 습관이 수면 및 식생활 습관에 미치는 영향 (An Analysis Relationship Between computer using habit and sleeping and eating habits in case of children)

  • 본 연구의 목적은 초등학교 학생들의 컴퓨터 사용습관과 이에 따른 수면패턴 및 식생활 습관의 실태를 분석하여 문제점을 분석하고 개선 방향을 제시하는데 있다. 먼저 컴퓨터 사용습관과 이에 따른 수면패턴 및 식생활 습관의 실태를 분석하여 문제점을 분석하기 위하여 초등학생들을 대상으로 설문조사를 실시하였다. 설문 대상은 초등학생 3~6학년의 학생들로 농촌 지역과 도시 지역으로 구분하였다. 설문의 주요 내용은 컴퓨터 사용의 목적, 컴퓨터 사용 시간, 평소 수면 시간, 수면의 질, 수면 부족의 원인 등과 컴퓨터 사용으로 인한 식사 습관의 변화 등으로 구성하였다. 설문 조사 결과 일부 초등학생의 경우 컴퓨터의 사용 기간과 시간이 식습관과 수면 패턴에 영향을 주고 있는 것으로 나타났으며, 학생들의 올바른 식습관과 수면을 위해서는 관련 교과에서의 컴퓨터 사용에 따른 기초 생활지도가 필요한 것으로 나타났다.

임상시험 중 수면행위가 심박변이도에 미치는 영향 분석 (Assessment of covariate-effect of sleeping behavior on heart rate variability during a clinical trial)

  • The object of this study is to evaluate confounding effect of sleeping behavior on heart rate variability (HRV) in healthy volunteers during a randomized clinical trial. From 6 healthy volunteer, we acquired circadian electrocardiography(ECG) with a Holter monitoring device(DigiTrak Plus, Philips). The compatible analytical program, Zymed, was used for calculation of RR intervals of 24h ECG signal. Then, we generated HRV signals and calculated 9 HRV parameters in time domain and frequency domain for every 30 minute with Matlab 6.x software. We also encoded participants' activities as follows; stable condition(1), sleeping behavior(2), eating behavior(3), and undefined condition(4). Using SPSS 12.0, we tested if there was any difference between HRV parameters of day-time and night-time and between those of a subject in stable condition and in sleeping behavior. As a result, one of the most important parameter-LF/HF ratio, which indicates the autonomic balance between sympathetic and parasympathetic nervous systems, as same the other parameters, showed large inter-individual variations in circadian profiles. And, the mean LF/HF ratio of each participant was different between in stable condition and in sleeping behavior (p<.05). Consequently, the effect of sleeping behavior must be considered when one or more of HRV parameters, especially LF/HF ratio, are used as assessment index in clinical studies.

온돌에서의 욕의 쾌적성에 관한 연구 (A Study on the Comfort, Physiological Responses and Microclimate in Beding Pad)

  • The purpose of this study was to investigate the effects of thickness of the sleeping pad on the physiological responses and the microclimate in the heating ondol room during sleeping time. The measuring points were skin temp., weight loss and body movement as physiological responses and the compression ratio, the temp. on/under the sleeping pad, the inside limp. of the sleeping quilts and subjective sence while 7 hours sleeping. Thickness of sleeping pads was 27.2 mm(A), 34.8 mm (B), 47.9 mm(C) used for 90 days and no used pad 60.7 mm(D). The floor surface temp. was keeping $29{\pm}1^{\circ}C$ while the environmental conditions was at $23{\pm}1^{\circ}C$ ($50{\pm}5\%$ R.H.) The results were as followings; 1) The mean compression ratio after using the sleeping pads for 45 days was about $70\%$ , and it wasn't increased any more thereafter 2) After 2 hours sleeping the temp. under the sleeping pads was shown that C and D were higher than A and B, and the temp. on the sleeping pads was shown that A and C pads were higher than B and D. But after 3-4 hours sleeping the skin temp. of legs was shown that B and D pads were higher than A and C. 3) Weight loss for C pad was significantly higher than others. Body movement and side position in tying was higher in A and C pads than B and D. 4) The inside temp. of quilt for A and C pads were higher in the early part of sleeping period than those of B and C pads and relative humidity was lower in C than others. 5) D pad was softer than others and fatigue degree was turned out to be lowest in D, the highest in A. From this point of view, we concluded that thickness of the sleeping Pad in the heating ondol room had different characteristic from that in no heating ondol room. And 35 mm used pad was turned out to be proper.
