Studies on the effect of Sophora flavescens extract on the hair growth stimulation and acne inhibition (苦蔘抽出物이 毛髮成長 促進 및 面疱 抑制에 미치는 영향)

  • In the course of screening natural extracts for hair growth, we found that the extract of dried root of Sophora flavescens has the prominent hair growth promoting effect. After topical application of Sophora flavescens extract to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. In addition, the Sophora flavescens extract revealed to possess potent inhibitory effect on $5{\alpha}$-reductase Ⅰ and Ⅱ activity. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by Sophora flavescens extract treatment. RT-PCR analysis showed that Sophora flavescens extract induced mRNA levels of growth factors such as insulin-like growth factor-Ⅰ and keratinocyte growth factor in dermal papilla cells, suggesting hair growth promoting effect of Sophora flavescens extract is mediated through inhibition of $5{\alpha}$-reductase type Ⅱ activity and the regulation of growth factors in dermal papilla cells. Furthermore, Sophora flavescens extract also showed anti-bacterial effect on Propionibacterium acnes. These results suggest that Sophora flavescens can be used as a potent treatment agent for helping hair growth stimulation and acne inhibition.

  • Objective : The objective of this study is to investigate the inhibitor effects of an traditional oriental herb, Sophora flavescens on the hepatic and renal side effects of chemotherapy by using B16-BL6 melanoma-injected C57BL6 mouse tumor model. Methods : In this study, the effects of an traditional oriental herb, Sophora flavescens, on the side effects of chemotherapy were studied using B16 melanoma-injected C57BL6 mouse tumor model. Results : Sophora flavescen has significant effect on the reduction of the side effects of chemotherapy. Sophora flavescen recovered the reduction of WBC and RBC during cisplatin chemotherapy. Water extract of Sophora flavescens significantly inhibited cisplatin-induced increase of serum blood urea nitrogen (BUN) which is a good indicator of renal toxicity. Sophora flavescens extract does not decrease the anti-tumor activity of cisplatin showing that it can selectively inhibit side effects of anticancer drugs preserving beneficial effort. Conclusion : Theses results suggest a possibility that Sophora flavescens extract can be used for cancer patients for the reduction of the side effects and improving the quality of life during chemotherapy of cancer patients.

Antimicrobial Activity of Medicinal Plant Extracts against Streptococcus mutans II

  • This study was carried out to research antimicrobial agents from medicinal plants, Pueraria thunbergiana, Sophora flavescens, Lonicera japonica, Pinellia ternata, Dictamnus dasycarpus, Paeonia lactiflora, Angelica dahurica, Spirodela polyrhiza, Cryptotympana pustulata, Pinus densiflora, Cimicifuga heracleifolia, Bupleurum falcatum, Magnolia kobus, Artemisia princeps, Houttuynia cordata, Forsythia koreana, Arctium lappa, Castanea crenata, Aster tataricus, Citrus unshiu, Gardenia jasminoides, Ledebouriella seseloides, Hovenia dulcis, Prunus sargentii. The ethanol extracts of 24 medicinal plants were tested for the antimicrobial activity against Streptococcus mutans. The extracts of Sophora flavescens, Pinus densiflora, Cimicifuga heracleifolia, Gardenia jasminoides and Ledebouriella seseloides showed antimicrobial activity against Streptococcus mutans. The extracts of Pinus densiflora and Sophora flavescens among these medical plants showed significant antimicrobial activity against Streptcoccus mutans. These results suggested that the extracts from Pinus densiflora and Sophora flavescens could be the potential source of antimicrobial agents against S. mutans.

  • This study was carried out to evaluate cytotoxicity of the methanol extract from Sophora flavescens Ait. against L1210 (lymphocytic leukemia) and $P388D_1$ (lymphoid neoplasma) Cells in vitro. We have determined cytotoxicity by the MTT (3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H- tetrazolium bromide) assay. The order of cytotoxicity of Sophora flavescens Ait. extracts against L1210 and $P388D_1$ cells in vitro is as follows: Fr. 4 > Fr. 3 > Fr. 5 > Fr. 2 > Fr. 1. These results suggest that the fraction 4 of the methanol extracts from Sophora flavescens Ait. may be a valuable choice for the development of antitumor agents. In order to develop an antimicrobial agent, dried Sophora flavescens Ait. was extracted with hot methanol, and then antimicrobial activity (MIC test) was investigated. In this study, the fraction 3 of the methanol extracts from the roots of S. flavescens showed strong growth inhibition activity against gram-positive and gram-negative bacteria (MIC, $3.125\;{\mu}g/ml$) such as S. mutans, S. epidermidis and P. putida. These results indicate that fractions 3 and 4 inhibit tumor cells and bacteria.

  • As part of our screening of anticariogenic agents from medicinal plants, the ethanol extracts of 25 herbs widely used in the fork medicine were tested for the antimicrobial activity against a cariogenic bacterium Streptococcus mutans ATCC 25175. The ethanol extracts of Coptis chinensis, Sophora flavescens, Glycyrrhiza uralensis, Mentha arvensis, and Saururus chinensis showed antimicrobial activities against S. mutans. The extracts of Coptis chinensis and Sophora flavescens among these medicinal plants showed significant antomicrobial activity against S. mutans. These results suggested that the extracts of Coptis chinensis and Sophora flavescens could be the potential source of antimicrobial agent against S. mutans ATCC 25175.

  • The purpose of this study is to investigate antibacterial activities of Sophora flavescens Ait. extracts for the prevention of dental caries caused by Streptococcus mutans. The fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. Showed strong growth inhibition activity against Streptococcus mutans (MIC, 3.13 ${\mu}$g/ml). The glucosyltransferase activity inhibited the formation of glucan by the fraction 5-4-3 and showed 77% of the antiproliferative effect at 100 ${\mu}$g/ml. This showed a significant activity (p<0.05). These results suggest that the fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. may be a valuable material as the prevention of dental caries.

  • Sophorae Radix, the dried roots of Sophora flavescens Aiton (Leguminosae), has been used in oriental traditional medicine for treatment of skin and mucosal ulcers, sores, gastrointestinal hemorrhage, diarrhea, inflammation and arrhythmia. Present study was carried out for the gastroprotective effect of trifolirhizin from Sophora flavescens. This reports were evaluated antibacterial activity against Helicobacter pylori and HCl ethanol-induced gastric lesion in rats and showed the significant effectiveness. In pylorus ligated rats, the treatments of trifolirhizin showed decrease in the volume of gastric secretion and acid output. Also we evaluated the antibacterial activity against H. pylori treated with methanol extract and trifolirhizin from Sophora flavescens had a equivalent antibacterial activity with ampicillin against H. pylori at the dose of $100{\mu}g/mL$. It may be regarded that the antigastritic effects and antibacterial activity of trifolirhizin from Sophora flavescens are originated from reduction of total acid output identified by gastric secretion reduce, free radical scavenging effects and the antibacterial activity against H. pylori.

  • A simple reversed phase HPLC method was developed for extracting pharmacologically active compounds kurarinone and trifolirhizin from the roots of Sophora flavescens using a binary gradient of acetonitrile : $H_{2}O$ with UV detection at 254 nm. The kurarinone and trifolirhizin contents of the roots of S. flavescens collected from ten district markets in Korea and China were $77.24{\sim}686.89$ ${\mu}g/g$ and $46.89{\sim}288.58$ ${\mu}/g$, respectively.
