The Antibacterial Component from Cinnamomi Cortex against a Cariogenic Bacterium Streptococcus matans OMZ 176

  • The methanol extract of Cinnamoni Cortex showed antibacterial action against cariogenic bacterium, Streptococcus mutans OMZ 176. The active principle of the extract was identified to be trans-cinnamaldehyde, which was bactericidal in the minimal inhibitory concentration (MIC) of $100\;\mu$g/ml against the strain. From the results of antibacterial activity of cinnamaldehyde and its derivatives, the acrolein group in the cinnamaldehyde was elucidated to be an essential element for the activity.

  • Naturally occurring diallyldihydroxybiphenyl compounds, magnolol and honokiol were reported to have potent antibacterial activities against most of Gram positive bacteria (1-4). To developmore potent antibacterial agents, some allyhydroxybiphenyl derivatives were synthesized from the starting compounds, p-phenyl-phenol (I) p-phenylphenol (IV), o, o-biphenol (VII) and p. p'-biphenol (XII). Among the newly synthesized compounds (III, VI, IX, XI, XIV and XVI), the antibacterial activities of 2-allyl-p-phenylphenol (VI), 6-ally-o, 0'-biphenol (IX), 2, 2'-diallyl-o, o'-biphenol (XIV) and 2, 2', 6-triallyl-p, p'-biphenol (XIV) were more potent than those of magnolol and bonokiol against a cariogenic bacterium, Stroptococcus matans ATCC OMZ 176.

Purification and Characterization of Streptococcus mutans Cell Wall Hydrolase from Bacillus subtilis YL-1004

  • Bacillus subtilis YL-1004 was isolated from soil for the development of agents to control dental caries. This strain produced an extracellular lytic enzyme that hydrolyzed the Streptococcus mutans cell wall. The lytic enzyme was purified to homogeneity by affinity chromatography and gel permeation chromatography to give a single band on SDS-PAGE and non-denaturing polyacrylamide gel electrophoresis. The molecular weight of the enzyme was deduced from SDS-PAGE and gel chromatography to be 38 kDa and the PI to be 4.3 from isoelectric focusing. Sirty $\%$ of its lytic activity remained after incubation at $50^{\circ}C$ for 30 min, and its optimal temperature was $37^{\circ}C$ . The enzyme showed its highest activity at pH 8.0 and was stable at pHs ranging from 4.0 to 9.0. Treatment with several modifiers showed that a cysteine residue was involved in the active site of the enzyme. This lytic enzyme from Bacillus subtilis YL-1004 exhibited specificity towards Streptococci and also showed autolytic activity on Bacillus subtilis YL-1004.

Streptococcus salivarius 119의 인공치태 억제효과에 대한 연구 (THE INHIBITORY EFFECT OF STREPTOCOCCUS SALIVARIUS 119 ON THE FORMATION OF ARTIFICIAL PLAQUE)

  • 경조직 우식으로 발생하는 치아우식증은 다인성 질환으로서 치태내 세균중에서도 Streptococcus mutans가 주 원인균이며, 치면에 부착, 증식 및 산생성 과정을 거쳐 치아우식을 유발한다. Streptococcus salivarius는 사람의 구강에 정상적으로 존재하는 세균이다. 본 연구에서는 소아의 구강으로부터 분리된 Streptococcus salivarius 119의 특성과 Streptococcus mutans 및 Streptococcus oralis에 대한 영향을 연구하여 다음과 같은 결과를 얻었다. 1. Streptococcus matans를 일회용 큐벧에서 배양시 550nm에서의 흡광도가 0.327이었으나, Streptococcus mutans와 Streptococcus salivarius 119의 혼합 배양시에는 0.119로 감소되었다. 2. 비커 와이어 검사에서 Streptococcus mutans 배양시 형성된 인공치태 무게는 84.7mg이었으나, Streptococcus mutans와 Streptococcus salivarius 119 혼합 배양시에는 12.3mg으로 감소되었다. 3. BHI broth에서 배양된 Streptococcus salivarius 119 배양 상청액을 가한 비커 와이어 검사에서 형성된 인공치태 무게는 100.5mg인데 반해, 5% 자당이 함유된 BHI broth에서 배양된 Streptococcus salivarius 119 배양 상청액을 가한 비커 와이어 검사에서 20.4 mg이었다. 4. Streptococcus oralis와 Streptococcus salivarius 119 단독 배양시에는 각각 ml당 $4.8\times10^7,\;7.5\times10^8$이었으나, 혼합 배양시에는 Streptococcus oralis는 $4.2\times10^7$, Streptococcus salivarius 119는 $5.8\times10^7$로 감소하였다. 5. Streptococcus salivarius 119 배양 상청액을 thin layer chromatography를 실시한 결과, Streptococcus salivarius 119가 형성한 polymer는 글루캔이었다. 6. Streptococcus salivarius 119가 만드는 글루캔을 처리하여 thin layer chromatography를 실시한 결과, $1\rightarrow6$ 결합이 주된 결합인 수용성 글루캔이었다. 이상의 결과를 종합하면 구강에서 분리된 Streptococcus salivarius 119에 의한 Streptococcus mutans의 인공치태 형성 억제작용은 수용성 글루캔 형성에 의한 것으로 사료되었다.

천연약물을 이용한 액제형 의치세정제 개발 (Development of Liquid Denture Cleaner Formulation using Natural Antimicrobial Agents)

  • 노인용 의치세정제는 실제 사용자의 특성을 고려하여 제품의 최종 형태가 중요하다. 각종문헌조사를 통해 의치세정제에 요구되는 사용자의 니즈를 반영한 제품의 기본요건을 탐색하였고, 의치성 구내염 원인균인 Candida albicans에 적합한 천연항균제를 스크리닝 하기위해 항균력 시험을 아울러 실시 하였다. 시투루스추출물을 0.04%로 희석하여 항균테스트 결과 우수한 결과가 도출되었다. 최종적으로 의치세정제 제조를 위한 최적의 레시피를 개발하였으며 아래와 같은 결과를 도출하였다. 1. 단순 의치세정제로서의 제형은 액제형이 가장 경제적이며 효과적이다. 2. 식물성 약용성분인 Citrus 추출물은 의치성 구내염 원인균인 Candida albicans에 대해 탁월한 항균성을 나타내었다. 3. 효소가 함유된 의치세정제는 효소 비함유 의치 세정제보다 세정력 및 항균력이 우수하였다.

삼백초 추출물의 항치아우식 효과 (Anticariogenic Properties of the Extract of Saururus chinensis)

  • It has been well established that S. mutans is the major etiological agent in dental caries, one of the most common oral diseases worldwide. The present study was designed to investigate the effect of Saururus chinensis (S. chinensis) ethanol extracts on the growth, acid production, biofilm formation, adhesion, and insoluble glucan synthesis of S. mutans. The ethanol extracts of S. chinensis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2 and 0.4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 80% at the concentration of 0.05 mg/ml and complete inhibition was observed at the concentration of 0.4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of S. chinensis showed more than 10% inhibition over the concentration of 0.025 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.025 ${\sim}$ 0.4 mg/ml of the ethanol extract of S. chinensis. Our research strongly suggested S. chinensis was a promising natural product for the prevention of dental caries.
