  • A new lupane triterpenoid saponin, 3, 11-dihydroxy-lupan-20-en-28-oic acid 28-o-$\alpha$-L-rhamnopyranosyl-(1$\rightarrow$4)-$\beta$-D-glucopyranosyl-(1$\rightarrow$6)-B-D-glucopyranosyl ester, have been isolated from the leaves of Acanthopanax koreanum Nakai (Araliaceae) together with one known flavonol glycoside, rutin. The structure were elucidated on the basis of spectral and chemical evidence.

Nutritional Components and Physiological Activity of Centella asiatica Cultured in Chungju by Drying Method (건조방법에 따른 충주산 병풀의 영양성분과 생리활성)

  • Centella asiatica (CA) has been used as a nutritional plant as well as a traditional herbal medicine around the world. This study, quality component (proximate composition, total polyphenol, and triterpenoid compound), and antioxidant and anti-inflammatory activities of CA dried using various methods were investigated. Proximate compositions of CA with different drying methods included a large amount of carbohydrates, crude protein, crude ash, and crude fiber. Among the drying methods, cold drying provided the highest total polyphenol content and antioxidant activity, while hot-air drying at 75℃ provided the lowest total polyphenol content and antioxidant activity. In addition, when the major triterpenoid compounds of CA were analyzed, the highest content of asiaticoside of triterpenoid glycoside was obtained with all drying methods. With respect to the total triterpenoid, the highest content was obtained with cold drying (68.8 mg/g) whereas natural drying (31.4 mg/g) provided the lowest content. In anti-inflammatory activity of LPS-stimulated RAW 264.7 cells, EtOH extract of cold drying showed a significantly higher inhibitory activity in comparison to the other drying methods. In conclusion, it is considered that the cold drying method is suitable for industrial preparation of functional materials with high physiological ingredients, and antioxidant and anti-inflammatory activities from CA.

  • A triterpenoid glucoside and five phenolic compounds have been isolated from the roots and leaves of Rosa davurica. They were elucidated as rosamultin and gallic acid from the roots, and methyl gallate, gallic acid, protocatechuic acid, quercetin and hyperoside from the leaves of this plant, respectively.

Alcohol-induced Hyperlipidemia Is Ameliorated by Orally Administered DWP208, a Sodium Succinate Form of ZYM201

  • DWP208 is a sodium succinate form of ZYM-201 which is a triterpenoid glycoside isolated from Sanguisorba officinalis, a medicinal plant prescribed for various diseases, such as duodenal ulcers and bleeding in East Asian counties. We demonstrated that this compound is able to normalize the altered lipid metabolism induced by hyperglycemia and a high fat diet. In this study, we determined whether hyperlipidemic conditions induced with chronically treated alcohol can also be restored by DWP208. Similar to our previous results, orally administered DWP208 (1 to 10 mg/kg) also ameliorated the hyperlipidemia that was induced by alcohol. This compound reversed the alcohol-induced hyperlipidemia including (i) up-regulated hyperlipidemic parameters such as low-density lipoprotein (LDL), very low-density lipoprotein (VLDL), atherosclerotic index (AI), triglyceride, and total cholesterol, and (ii) down-regulated hyperlipidemic parameters such as absolute body weight, superoxide dismutase (SOD) activity, and high-density lipoprotein (HDL) in serum and liver. According to our data, the ameliorative activity of DWP208 is due to its indirect anti-oxidative activity as a result of which lipid peroxide and hydroxyl radical levels were reduced and the activity of SOD was enhanced. Therefore, our data strongly suggest that DWP208 can be used as a remedy against alcohol-induced hyperlipidemia.

Analysis of Triterpene Glycoside Levels and Antioxidant Activity in the Different Shoot Tissues of Centella asiatica (L.) Urban (병풀 지상부 조직에서 시기별 triterpene glycoside 함량 및 항산화활성 분석)

  • Centella asiatica is one of the local herbs that is claimed to possess various physiological effects. C. asiatica also accumulates large amounts of pentacyclic triterpenoid saponins known as centelloids. These terpenoids usually include asiatic acid, asiaticoside, madecassoside, and madecassic acids. In the present study, to understand the changes of triterpene glycoside levels in the different shoot tissues of C. asiatica during seasonal cultivation, we investigated the High-Performance Liquid Chromatography (HPLC) analysis via different extraction methods, such as water, 20% ethanol and methanol extracts. Significant increases were observed in the levels of madecassoside and asiaticoside in the leaf extracts by methanol compare with extracts using water or 20% ethanol. Additionally, we also analyzed the various antioxidant activity in the different shoot tissues of C. asiatica using different extracts, such as leaves, petioles and both materials. Among these petiole extracts showed high 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTs) scavenging activity in all extracts, whereas 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity exhibited high activity levels in the leaf part using methanol and ethanol extracts. Levels of total phenolics and flavonoid also showed the highest levels in the leaf tissues using all extracts such as methanol, ethanol and water. Our results indicated that the increased levels of triterpene glycoside and antioxidant activity in the leaf parts of the C. asiatica were indicating that useful metabolites were mainly maintained through seasonal cultivation such as madecassoside, asiaticoside.

Comparison of Triterpenoid Contents of the Four Rubus Plants in Korea Using TLC-DM (TLC-DM을 이용한 Rubus속 4종 식물의 Triterpenoid 함량 비교)

  • The extraction yield of the methanolic extracts and 19${\alpha}$-hydroxyursane-type triterpenoid (19${\alpha}$-HUT) fraction were investigated in the unripe and ripe fruits and the leaves of the four Rubus plants (Rubus coreanus, R. crataegifolius, R. phoenicolasius, R. pungens var. oldhami) to develop the biomaterial 19${\alpha}$-HUT mixture as functional foods. Thin layer chromatogaphy-Densitometer (TLC-DM) was used to analyze the individual quantity of 19${\alpha}$-HUTs using standard compounds (euscaphic acid, tormentic acid, 23-hydroxytormentic acid, kaji-ichigoside F$_1$, rosamultin, niga-ichigoside F$_1$). The content of methanolic extract of the fruits were higher in the ripe stage than in the unripe stage whereas the content of 19${\alpha}$-HUT mixture varied with each Rubus species. The Rubus plants containing the highest amount of 19${\alpha}$-HUTs in the leaves were R. coreanus, R. phoenicolasius and R. pungens var. oldhami while only R. cratagefolius showed the highest content in the ripe fruits. The mean of total genin content of 19${\alpha}$-HUTs was 0.94 mg/g; that of the glycosides was 0.60 mg/g. The genin quantity was found in the order of 23-hydroxytormentic acid> euscaphic acid> tormentic acid; the glycoside was observed in the order of niga-ichig-oside F$_1$> kaji-ichigoside F$_1$> rosmaultin, by which the biosynthetic pathway of 23-hydroxytormentic acid and its glucoside niga-ichigoside F$_1$ via the intermediates tormentic acid and/or rosamultin was presumed. It is also suggested that the ripe fruits of R. crataegifolius will be desirable to use as functional foods rather than unripe fruits.

  • From the roots of Caragana sinica (Buc'hoz) Rehder (Leguminosae), a new saponin named caraganoside A was isolated and elucidated as 3-0-$\beta$-D-xylopyranosyl (1$\rightarrow$2)-[$\beta$-D-glucopyranosyl (1$\rightarrow$3)]-$\alpha$-L-arabinopyranosyl oleanolic acid 28-O-[$\beta$-D-glucopyranosyl ester by means of chemical and spectral studies. Kalopanax-saponin F, hemsloside Ma3 and araloside A were also isolated and characterized. The former two compounds were separated as their methylesters.

A New Triterpenoid Saponin from the Tropical Marine Sponge Lipastrotethya sp. (열대 해면동물 Lipastrotethya sp.에서 분리된 사포닌 화합물)

  • Marine sponges have been a remarkably rich source of pharmacologically active and structurally diverse natural products. As a part of our continuing search for novel secondary metabolites of biomedical importance from marine invertebrate, we encountered the sponge Lipastrotethya sp. from Chuuk, Micronesia. The crude organic extract of this animal exhibited considerable cytotoxicity against the K562 cell line. Guided by the $^1H$ NMR analysis, flash chromatography of the crude extract followed by HPLC yielded a new triterpene glycoside, along with ten known saponins of the sarasinoside class. The structure of this new compound was determined by combined spectroscopic methods such as COSY, HSQC and HMBC experiment. Among these metabolites, six compounds exhibited moderate cytotoxicity against ACHN, MDA-MB-231, NCI-H23 and NUGC-3 cell lines.
