산약의 Mouse embryonic fibroblast cell에 대한 자외선 손상 방어효과 (The Protective Effects of Dioscoreae Rhizoma on the Exposure to UVA of MEF cells)

  • Purpose: This study was to determine the protective effects of Dioscoreae Rhizoma on the Mouse Embrio Fibroblast (MEF) cells exposed to the ultraviolet rays(UVA). Methods: The samples were assigned randomly to five groups; control group without any treatments, UVA group exposed only to UVA, DR group exposed only to the Dioscoreae Rhizoma, UVA-DR group exposed to UVA before being treated with the Dioscoreae Rhizoma, and DR-UVA group treated with the Dioscoreae Rhizoma before being exposed to UVA. The survival rate of cells, metabolic rate of cells, transformation of nucleus within cells, alteration of cell cycle, effects on the apoptosis, the change of the amount of protein related to cell cycle were measured in order to determine the cell protective effects of the Dioscoreae Rhizoma on each group. Results: 1. DR-UVA group has more cell protective effects compared to the UVA group in all experiments, indicating that the Dioscoreae Rhizoma protects skin from UVA physically and chemically. 2. UVA-DR group shows more efficiency compared to UVA group in rapid recovery of damaged cell and leading highly damaged cells to apoptosis, preventing the expression of abnormal cells. Conclusions: Dioscoreae Rhizoma has effects of protecting MEF cells from UVA, of recovering cells damaged by UVA, and of prohibiting the expression of abnormal cells.

Propolis Inhibits UVA-Induced Apoptosis of Human Keratinocyte HaCaT Cells by Scavenging ROS

  • Propolis is a resinous material collected by honeybees from several plant sources. This research aimed at showing its protective effect against UVA-induced apoptosis of human keratinocyte HaCaT cells. Using Hoechst staining, it was demonstrated that propolis (5 and $10{\mu}g/mL$) significantly inhibited the apoptosis of HaCaT cells induced by UVA-irradiation. Propolis also showed the protective effect against loss of mitochondrial membrane potential induced by UVA-irradiaiton in HaCaT cells. Propolis also inhibited the expression of activated caspase-3 induced by UVA-irradiation. To investigate the role of ROS in UVA-induced apoptosis and protection by propolis, the generation of ROS was determined in cells. The results showed that the generation of ROS was markedly reduced in cells pretreated with propolis. Consequently, propolis protected human keratinocyte HaCaT cells against UVA-induced apoptosis, which might be related to the reduction of ROS generation by UVA-irradiation.

여름용 모자의 UVA 투과량 (UVA radiation transmittance in Summer Hats)

  • The purpose of this study was to investigate the UVA radiation protection effects of summer hats currently on the market with the purpose of making it possible to choose a hat with suitable UVA protection. Twelve different summer hats from the market were selected for the experiment. The results are summarized as follows: It is more effective to wear a hat than not wear a hat to block UVA radiation. Summer hats with the greatest degree of protection, from highest to lowest, are cotton, straw2, and straw1. In the area of the forehead, which is rarely influenced by the irradiation angle, the cotton hat was the most effective in protecting from UVA radiation because the material density was greater than that of the straw hats. A hat with a 8.5 cm brim was more effective at blocking UVA radiation on the jaw than 6 cm, 4 cm, and 0 cm wide brims, but it still couldn't block the radiation completely. Irradiation amounts at 11:00 AM on the forehead, jaw! s and left and right cheeks were lower than amounts measured on the back of the neck. This revealed that irradiation amounts depend on the shape of the hat and time of day. A hat with a brim encircling the head was found to be more effective in blocking UVA radiation than a hat with only a front or side brim.

Topical Irradiation of UVA to The Eye Induces Immunosuppression in The Mice via Nitric-Oxise Dependent Neuronal Pathways

  • It has been well documented that dermal irradiation by ultraviolet A (UVA) locally decreases the number of Langerhans cells and suppresses contact hypersensitivity of the skin. We found that topical irradiation of UVA to the eye systemically decreased the number of Langerhans cells (LC) in the dorsalskin and lymph nodes and elicited lymphocyte apoptosis in the latter tissues but not in the thymus. Optic nerve resection, but not ciliary ganglionectomy, eliminated the UVA-induced decrease in dermal Langerhans cells by a mechanism that was partially inhibited by hypophysectomy. The immunosuppressive effect of UVA was not observed in knockout mice lacking inducible-type of nitric oxide synthase (iNOS). These results suggested that topical irradiation of UVA to the eye induced immunosuppression via NO-dependet neuronal pathways.

  • SPEX SkinSkan is a newly developed spectrofluorometer that can calculate transmission of UVA on the surface of human skin as a non-invasive method. We have investigated UVA protection factors of Japanese over-the-counter sunscreens using SPEX SkinSkan. This non-invasive method is useful to know UVA protection of sunscreens in vivo.

  • Prolonged exposure to solar ultraviolet A (UVA) radiation has been known to cause premature skin aging (photo-aging). UVA radiation generates ROS thereby induce degenerative changes of skin such as degradation of dermal collagen, elastic fibers. Matrix metalloproteinases (MMPs), the proteolytic enzymes have been implicated as a major player in the development of UVA-induced photo-aging. Many studies have been conducted to block the harmful effects of UV radiation on the skin. Recently, we are interested in the availability of fermented red ginseng (FRG) as natural matrix metalloproteinases inhibitors (MMPIs). The efficacy difference between red ginseng and FRG has been compared. Both RG and FRG have no cytotoxic effects below the concentration of $300{\mu}g/ml$. Human dermal fibroblasts (HDFs) were pretreated with FRG or RG for 24h, followed by irradiation of UVA. Then, we measured the intracellular ROS production and the expression of MMP, $IL-1{\beta}$ at the mRNA level. We also examined the intracellular localization of $NF-{\kappa}B$ and MMP-9 on the FRG or RG treated and UVA-irradiated HDFs. FRG decreased the intracellular ROS production elicited by UVA. In addition, FRG decreased the mRNA expression of MMP-3, MMP-9, and $IL-1{\beta}$ more efficiently than RG. Furthermore, FRG suppressed the nuclear localization of $NF-{\kappa}B$, and the expression of MMP-9. Taken together, our results suggest that FRG is promising agents to prevent UVA-induced photo-aging by suppressing MMP expression and inflammation.

적색 및 청색 LED광과 UVa광 처리가 착색단고추 과실의 착색에 미치는 영향 (Effect of Red LED, Blue LED and UVa Light Sources on Coloration of Paprika Fruits)

  • 본 실험은 미성숙 즉 미착색과의 착색 증진을 위해 LED광원의 적색광, 청색광 그리고 UVa를 각각 두 가지 광도로 처리하여 처리에 따른 착색정도를 비교하였다. 실험에 사용한 착색단고추 품종은 적색 계통의 'Special'과 황색 계통의 'Helsinki'으로 춘천 근교 농가에서 수경재배 하였다. LED의 경우 Red_High는 $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 내외, Red_Low는 $20{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 내외, Blue_High는 $70{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 내외, Blue_Low는 40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 내외, UVa의 경우 고광도는 $3{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 저광도는 $0.25{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$로 처리하였다. 품종에 상관없이 처리 2일 후에 과실의 착색이 급격히 진행되었고 3일째에 처리별로 큰 차이를 보였다. 'Special'의 경우 2일째 고광도 UVa광이 제일 급격한 착색 변화를 나타내었고, 저광도 UVa를 제외한 나머지 처리구에서는 비슷한 착색의 변화을 나타내었다. 처리 3일째에는 고광도 LED 적색광이 급격한 착색변화를 나타내며 고광도 UVa광보다 높은 착색 값을 나타내었다. 'Helsinki' 경우에는 2일째에 청색광, 적색광, UVa의 순으로 착색 변화가 빠르게 진행되었다. 두 품종 모두 3일째에는 'Special'의 $a^*/b^*$ 값과 황색품종 'Helsinki'의 착색 정도를 나타내는 $b^*$ 값이 모두 저광도보다 고광도에서 높은 값을 나타내며 착색 증진 효과를 나타내었다. 착색종료일인 처리후 6일째 두품종 모두 무처리구와 저광도 UVa광을 제외한 나머지 모든 처리구에서는 비슷한 값을 나타내었다. UVa 처리구 과실의 표면이 주글거리는 피해를 발생하였다. 이상의 결과로 저광도보다는 고광도의 LED 광원을 사용하는 것이 착색이 빠르게 진행될 수 있도록 도우며, UVa 파장은 과실의 외관에 피해를 입히기 때문에 적합하지 않게 생각된다.

In vitro 실험법에 의한 천연물 중의 UVA 광독성 억제제 검색 (In vitro Screening of UVA Phototoxicity Inhibitors using the Natural Products)

  • The phototoxicity inhibitory activity of 15 natural products having antiinflammatory effect was screened by three in vitro methods: yeast growth inhibition test with Candida albicans, RBC photohemolysis and MTT assay. We induced phototoxic reaction by irradiating UVA (365 nm) on chlorpromazine (CPZ) that has been widely documented as phototoxic agent in clinical and experimental studies and then observed the effects of the natural products after treating them with CPZ. In yeast growth inhibition test, X. stramonium showed the inhibitory effect on the UVA phototoxicity and E. officinalis, Yeast, P. suffruticosa showed phototoxicity inhibitory effect in that their % hemolysis compared with control were 36.14${\pm}$ 2.69, 42.82${\pm}$1.35, 36.41${\pm}$0.48 on UVA. In MTT assay, all tested natural products increased cell viability compared with the control.

장파장의 자외선 LED 광원을 이용한 안질환 세균의 살균효과 (A Study of Sterilization Effect of Long-wavelength UVA-LED Irradiation on Bacteria Causing Eye Diseases)

  • 목적: 본 연구는 UVA영역의 LED를 이용하여 안질환을 일으키는 대표적인 유발균인 녹농균과 포도상구균에 대한 살균효과를 알아보고, UVA영역의 LED를 콘택트렌즈 살균에 활용하고자 하였다. 방법: 안질환의 원인 세균인 녹농균과 포도상구균에 대한 살균효과 실험은 400 nm의 UVA-LED를 이용하여 소형 살균기기를 제작하여 녹농균은 $10^{-7}$ CFU/ml, 포도상구균은 $10^{-5}$ CFU/ml 로 각각 액체배지에 희석하여 넣은 후 시간대별로 15분, 30분, 60분, 120분, 240분, 360분, 480분 동안 각각 400 nm의 UVA를 조사하였다. 결과: 살균실험 결과를 직선회귀방정식(regresson line equation)으로 변환하여 목표 살균치(target log inactivation)를 도출한 결과 녹농균에 대한 3log inactivation의 UV조사량($mJ/cm^2$)이 54,847 UV dose($mJ/cm^2$)이었으며, 조사시간(irridiation time)은 135.42(min) 이었다. 또한 포도상구균의 3log inactivation은 39,066 UV dose($mJ/cm^2$)이었으며 조사시간은 98.72분이었다. 결론: 이러한 실험 결과 400 nm의 UVA-LED를 이용한 녹농균과 포도상구균에 대한 뛰어난 살균효과를 확인하였으며 콘택트렌즈의 살균에 유용하게 이용될 것으로 사료된다.

진피 콜라겐의 노화에 대한 활성산소와 자외선의 영향 (THE ROLE OF REACTIVE OXYGEN SPECIES ON UVA-INDUCED AGING OF DERMAL COLLAGEN)

  • Collagen은 세포 외에 존재하며, 분자의 수명이 매우 긴 단백질이므로, 노화와 관련된 비효소적 당화의 대상으로 많은 관심이 모아지고 있다. 본 연구에서는 진피 collagen의 비효소적 당화와 분해 및 가교결합에 대한 자외선(UVA)과 활성산소종들의 영향을 검토하였다. 암소 피부로부터 얻은 collagen 과 glucose의 혼합물에 몇가지 활성산소 소거제를 첨가하고 UVA를 조사하여 비효소적 당화, 단백질 분해, 가교결합의 정도를 관찰하였다. 비효소적 당화는 자외선에 의해 증가되지 않았으며, 수퍼옥사이드 라디칼과 일중항 산소의 소거제에 의해서는 감소하였으나, 히드록시 라디칼 소거제에 의해서는 변화가 없었다. 단백질 분해와 가교결합은 자외선에 의해 증가되었으며, 분해는 세가지 활성산소의 소거제 모두에 의해 감소되었으나, 가교결합은 특히 히드록시 라디칼 소거제의 영향이 컸다. 즉 수퍼옥사이드 라디칼과 일중항 산소는 자외선과는 관계 없이 당의 자동산화에 의해 발생되어 당화로 인한 변화의 초기 단게에 관여하고, 반응의 뒷부분에서는 당단백질과 자외선에 의해 만들어진 히드록시 라디칼이 주된 역할을 하고 있다. 상기의 결과로부터 활성산소종들과 자외선은 다른 조직 및 세포물질들을 손상시키듯이 노화와 관련된 collagen 변화를 증가시키는 것을 알 수 있었다. 이는 자외선 차단제와 함께 활성산소 소거 물질들을 외용도포하는 것이 태양광선의 해로운 영향으로부터 진피를 보호하는데 도움이 될 수 있음을 의미한다.
