Effects of Picrorrhiza Rhizoma on Acute Inflammation in Mice

  • The effects of Picrorrhiza Rhizoma (PR) aqueous extracts were observed on xylene-induced acute inflammation. The xylene was topically applied 60 min after administration of 500, 250 and 125 mg/kg of PR extracts, and all animals were sacrificed 2 hrs after xylene application. The changes on ear weights, histolopathological analyses of ear were evaluated as compared with indomethacin and dexamethasone 15 mg/kg treated groups - well known anti-inflammatory agents. Xylene application resulted in marked increases in induced ear weights as compared with intact control ear. Severe vasodilation, edematous changes of ear skin and increase in the thickness of the ear tissues as acute inflammation were detected in xylene-treated control ears at histopathological observation. However, these xylene-induced acute inflammatory changes were dosedependently decreased by oral treatment of PR extracts. Therefore, it is concluded that PR extracts has favorable anti-inflammatory effects on xylene-applicated acute ear inflamed mice.

Anti-Inflammatory Effect of Jungcheonhwadamgangki-tang ga Antler in Xylene-Application Mouse Ear Acute Inflammation Model (정천화담강기탕가녹용(定喘化痰降氣湯加鹿茸)이 Xylene으로 유발된 마우스의 급성 염증에 미치는 영향)

  • In the present study, the anti-inflammatory effect of "Jungcheonhwadamgangki-tang ga Antler" water extract was tested in Xylene-Application mouse ear acute inflammation model. The test articles were once dosed before Xylene-Application, and the changes on body weight and ear weights and histopathological observation of induced ear were conducted with ear histomorphometry. The obtained results were as follows. The increases of absolute and relative ear weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, thickness and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler. In addition, the increases of ear thickness half and thickness full detected in the vehicle control were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang ga Antler-dosing groups. Base on these results, it is concluded that Jungcheonhwadamgangki-tang ga Antler extracts has clear anti-inflammatory effect on the acute inflammation such as bronchial asthma.

In addition, the increases of ear thickness half and thickness full detected in the vehicle control, were also dose-dependently decreased in the all Haepyoeiin-tang plus Antler-dosing groups. Base on these results, it is concluded that Haepyoeiin-tang plus Antler water extracts have a clear anti-inflammatory effect on the acute inflammation. However, somewhat lower anti-inflammatory effects were detected in Haepyoeiin-tang plus Antler water extracts compare to that of Dethamethason and Dicrofenac. About 500 mg/kg of Haepyoeiin-tang plus Antler water extracts have similar effect compared to that of Dicrofenac 15 mg/kg.

Expression of cytochrome P-450(CYP)2B1/2 in lymphocytes of workers exposed to toluene and xylene (Toluene과 Xylene 노출 근로자의 림포사이트에서 Cytochrome P-450(CYP)2B1/2의 발현)

  • In order to develop the methods for exposure assessment and find susceptibility markers for the workers who are exposed to low doses of toluene, xylene and other chemical in petroleum industries, we investigated the application of P-450 expression in human lymphocytes utilizing mouse monoclonal anti-rat CYP2B1/2, the levels of toluene and xylene in air and their metabolite levels in urine with the levels of expressed CYP2B1/2 proteins. The general characteristics such as age, smoking and drinking habit were no significant difference between the control and exposed workers, but the working durations and working hours were significantly different. Workers in exposed group were exposed to the mean of 2.1 ppm (range, 0.00-4.54) of toluene and 0.3 ppm (rang, 0.00-1.23) of xylene. The mean concentration of urinary hippuric acid was low and less than 1/5 of the biological exposure index recommended by the Ministry of Employment and Labor Korea. Methyl hippuric acid in urine was not detected in control and exposed workers. Also, there were no significant differences in the levels of the urinary metabolites between the control and exposed group. When chemiluminescence dot blottings were carried out utilizing mouse monoclonal antibody against CYP2B1/2, the strong density dots corresponding to a mouse monoclonal antibody was observed in the human lymphocytes from the exposed workers. These results suggested that the chemiluminescence dot blot assay for CYP of lymphocytes should be valuable for identifying CYP expression as biomarkers in the workers exposed to toluene and xylene.

Effects of DHU001, a Mixed Herbal Formula on Acute Inflammation in Mice

  • The effects of DHU001, a mixed herbal formula consisted of 7 types aqueous extracts for treating respiratory disorders were observed on xylene-induced acute inflammation. The xylene was topically applied 60 min after administration of 500, 250 and 125 mg/kg of DHU001, and all animals were sacrificed 2 hrs after xylene application. The changes on ear weights, histolopathological analyses of ear were evaluated and compared to those of indomethacin and dexamethasone(15 mg/kg treated)-Both of drugs are well-known by anti-inflammatory agents. Xylene application resulted in marked increases in induced ear weights as compared with intact control ear. Severe vasodilation, edematous changes of ear skin and increase in the thickness of the ear tissues, neutrophil infiltration as acute inflammation were detected in xylene-treated control ears at histopathological observation. However, these xylene-induced acute inflammatory changes were dose-dependently decreased by oral treatment of DHU001. Therefore, it is concluded that DHU001 has favorable anti-inflammatory effects on xylene-applicated acute ear inflamed mice.
