Effects of the Zizyphus jujuba Seed Extract on the Lipid Components in Hyperlipidemic Rats

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Zizyphus jujuba seeds were more effective as a therapeutic regimen for the control of metabolic derangements in adult disease.

  • Zizyphus jujuba, Z. jujuba var. inermis, and Z. jujuba for. hoonensis were cultured in korea. Zizyphus seeds are used for insomnia, sedation and sleeping-dropsy in many oriental medical prescriptions. Three Zizyphus species in Korea were investigated for their chemical composition. The components in methanol extracts of the fruit and seed were separated by TLC, PPC and were identified with known compounds. Sucrose, galactose, glucose, fructose and ${\beta}-sitosterol$ were isolated from the fruit of Zizyphus jujuba for. hoonensis.

Studies on the Compositions of Mineral Components, Total Amino Acid, Free Amino Acid and Fatty Acid of Zizyphus jujuba Seed (대추씨(Zizyphus jujuba Seed)의 무기질, 구성 총 아미노산, 유리 아미노산 및 지질 구성 지방산조성에 관한 연구)

  • Chemical components of the physiological activity substance in Zizyphus jujuba seed were analyzed for the use as an functional processed products. The proximate compositions of Zizyphus jujuba seed were carbohydrate 89.40%, moisture 8.23%, crude protein 1.22%, crude fat 0.65% and crude ash 0.50%, respectively. the richest mineral contained in Zizyphus jujuba seed was Mg(198.3mg/100g), followed by Mn(113.0mg/100g), K(100.2mg/100g), Ca(25.3mg/100g), P(25.6mg/100g_, Fe(23.5mg/100g), Zn(18.7mg/100g), Cu(7.4mg/100g) and Na(5.3mg/100g). Total amino acid contents of Zizyphys jujuba seed were 1103.12mg/100g, and the major amino acids were proline(235.17mg%), aspartic acid(175.941mg%), gltamic acid(101.46mg%), leucine(50.27mg%), serine(47.71mg%), aspartic acid(47.27mg%), glycine(41.60mg%) and lysine (33.78mg%), respectively. The amount of free amino acids in the protein of Zizyphus jujuba seed were 178.62 mg/100g, and the major free amino acids were proline(121.30mg%), alanine(6.01mg%), valine(2.63mg%) and arginine(2.04mg%), respectively. The compositions of major total fatty acid in the lipid Zizyphus jujuba seed were C18:2(n5c) 28.72%, C18:1(n9t) 23.76%, C18:1(n9c) 13.45%, C16:0 13.45% and C20:5(n3) 1.77% in order, respectively.

Effects of the Zizyphus jujuba Seed Extract on the Blood Glucose and Serum Lipid Components in Streptozotocin-Induced Diabetic Rats (대추씨(Zizyphus jujuba Seed) 추출액이 Streptozotocin 유발 당뇨성 흰쥐의 혈당 및 지질성분에 미치는 영향)

  • The purpose of this study was designed to observe the effects of the feeding Zizyphus jujuba seed extract on the improvement of the blood glucose, lipids in the serum of streptozotocin (STZ)-induced diabetic rats fed the experimental diets for 4 weeks. Concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG) and phospholipid (PL) in serum were significantly higher in the STZ (55mg/kg B.W.)-induced diabetic group (group 2) and STZ (I.P.)+ Zizyphus jujuba seed extract group (group 3) than those in the control group (group 1, basal diet + water). But the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG and PL in serum were remakably lower in the group 3 than those in the group 2. In the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group (group 3) were higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the STZ- induced diabetic group (group 2). From the above results, it was suggested that the Zizyphus jujuba seed were effective on the improvement of the blood glucose, lipid compositions in serum of STZ-induced diabetic rats. Moreover, in Zizyphus jujuba seed was effective therapeutic regimen for the control of metabolic derangements in adult disease.

Physiological Activity of Zizyphus jujuba Leaf Extracts (대추잎 추출물의 생리활성 작용)

  • This study was designed to investigate the possible utilization of Zizyphus jujuba leaves as a source of functional ingredients. The physiological activity of different solvent fractions prepared from ethanol extract of Zizyphus jujuba leaves were analyzed. Xanthine oxidase inhibitory effect was very high in all fractions except chloroform fraction. The very high electron donating ability was observed in the ethylacetate fraction and the effect was similar to 0.1% tocopherol. Nitrite scavenging effect of all fractions was more than 40% even at low concentration of 1mg/ml and was increased with increasing concentration. Angiotensin I converting enzyme inhibitory activity was appeared in ethyl acetate and chloroform fractions only at high concentraton.

Screening of Natural Products for Endothelial and Renal Nitric Oxide Production

  • Natural products, which have been used for the treatment of hypertension, diuresis and nephritis in traditional oriental medicine, were selected for the screening of nitric oxide (NO) production in endothelial cells and kidney tissues in vitro as well as in vivo by measuring the conversion of [$\^$14/C]-L-arginine to [$\^$14/C]-L-citrulline, a coproduct of the enzyme reaction with NO. Confluent monolayer of endothelial cells were used for the screening of 16 natural products. Among the natural products, Zizyphus jujuba and Codonopsis pilosula stimulated endothelial NO synthase activity. Thus, both confluent monolayer of endothelial cells and kidney homogenates (glomeruli, cortical tubules, meudllae) were treated with Zizyphus jujuba and Codonopsis pilosula (final concentration 10 $\mu\textrm{g}$/$m\ell$) and NO releases were compared with those by receptor - dependent agonists, bradykinin and ADP and receptor - independent calcium ionophore A23187 in vitro. In rat experiment, NO releases in glomeruli, cortical tubules and medullae and plasma renin activity were assessed after intraperitoneal injection of Zizyphus jujuba and Codonopsis pilosula (10 mg/kg/day for 4 days). As a result, both Zizyphus jujuba and Codonopsis pilosula significantly increased NO releases in cultured endothelial cells, kidney tissues in vitro as well as in vivo. Stimulation of NO releases by Zizyphus jujuba and Codonopsis pilosula was similar to those by receptor - dependent agonists, bradykinin and ADP and receptor - independent calcium ionophore A23187 in cultured endothelial cells. However, plasma renin activity was not influenced by these two natural products. In conclusion, stimulatory effects of Zizyphus jujuba and Codonopsis pilosula on NO release in kidney may contribute their hypotensive effects and antinephritic action possibly by increasing renal blood flow.

Effects of Dietary Zizyphus jujuba Seed Meal on Broiler Performance (대추씨(Zizyphus jujuba Seed) 급여가 육계의 생산성에 미치는 영향)

  • The study was conducted to investigate the effect of dietary Zizyphus jujuba seed meal on broiler performance, small intestine and cecal length, gizzard weight and fecal gas emission. A total of 180 day-old broiler chicks (Ross) were randomly divided into 4 groups, fed the commercial diet containing 0, 0.3 (T1), 0.6 (T2) and 0.9 % (T3) of Zizyphus jujuba seed meal from two to 5 weeks of age. Body weight gain and feed efficiency (feed/gain) were significantly increase in T2 than other groups at 3 weeks age (P<0.05). There was no statistical difference in body weight gain and feed efficiency (feed/gain) by feeding Zizyphus jujuba seed meal, although they tended to be improved by treatment during whole experimental period (5 weeks age). The gizzard weights of three treatment groups were higher than control. But no difference among three treatment groups was observed. The small intestine and cecal length was no change by feeding Zizyphus jujuba seed meal. Emission of $NH_3$ and VFA gas from excreta were significantly decreased from 5 to 10 days of storage period (P<0.05). In conclusion, the results of this study showed that the addition of Zizyphus jujuba seed meal at 0.3 to 0.6 % to the diet has increased digestibility of feed and the reduction of $NH_3$ emissions from feces, there was a possibility of improvement in body weight gain of broiler chickens.

Sedative Activity of Aporphine and Cyclopeptide Alkaoids Isolated from the Seeds of Zizyphus Vulgaris var. Spinosus, and the Fruits and Stem Bark of Zizyphus Jujuba var. Inermis in mice (산조인 및 대추, 대추나무로부터 단리한 아포르핀과 환상 펩티드 알칼로이드의 생쥐에 대한 진정작용)

  • The objective of this study was to evaluate the sedative activity of four aporphine alkaloids (APA) and nine cyclopeptide alkaloids(CPA), which had been isolated from the seeds (sanjoin) of Zizyphus vulgaris var. spinosus, and the fruits and stem bark of Zizyphus jujuba var. inermis. The assessment of sedative activity was carried out, employing a hexobarbital-induced sleeping time method in mice. When the relative sedative potency of sanjoinine-A(CPA) was given as one unit, those of nuciferine (APA), lysicamine (APA), chlorpromazine (positive control), and sanjoinine -Ahl (an epimer of sanjoinine-A) were 13, 6.5, 5, and 3, respectively. The sedatvie activities of other CPAs were much lower than those of sanjoinine-A and -Ahl, and other APAs were not active. On heat treatment, nuciferine and lysicamine were degraded into some artifacts which exhibited no sedative activity, while sanjoinine-A was converted into sanjoinine-Ahl which showed more potent sedative activity. These results suggested that nuciferine and sanjoinine-A were major sedative components of native sanjoin, and that sanjoinine-A and its epimeric artifact, sanjoinineAhl were the active principles of roasted sanjoin. It provides a scientific basis for heat-processing (roasting) of this Oriental medicine.
