대회향(大茴香) 물추출물이 마우스 대식세포주(RAW 264.7 cell line)의 hydrogen peroxide 생성에 미치는 영향 (Effect of Anisi Stellati Fructus Water Extract on Hydrogen Peroxide Production in RAW 264.7 Mouse Macrophages)

  • The purpose of this study is to investigate effects of Anisi stellati Fructus Water Extract on hydrogen peroxide production in RAW 264.7 mouse macrophages. Anisi stellati Fructus were extracted by hot water. Effects of Anisi stellati Fructus water extract (AS) on hydrogen peroxide production in RAW 264.7 were measured by dihydrorhodamine 123 assay after 20, 24, 28, 44, 48, and 52 h incubation at the concentrations of 10, 25, 50, and $100{\mu}g/mL$. For 20 h incubation, AS significantly increased hydrogen peroxide production in RAW 264.7 cells by $108.6{\pm}1.56%$, $109.5{\pm}1.94%$, $108.4{\pm}1.14%$, and $107.3{\pm}3.06%$ at the concentrations of 10, 25, 50, and $100{\mu}g/mL$ (P < 0.05) respectively. For 24, 28, 44, 48, and 52 h incubation, AS also significantly increased hydrogen peroxide production in RAW 264.7 cells at the concentrations of 10, 25, 50, and $100{\mu}g/mL$ (P < 0.05). These results suggest that Anisi stellati Fructus has the immune - enhancing property related with its increase of hydrogen peroxide production in macrophages.

대회향의 시스템 약리학적 분석과 항균작용 (Systemic Analysis of Antibacterial and Pharmacological Functions of Anisi Stellati Fructus)

  • 시스템 약리학적 분석을 통해 대회향(Anisi Stellati Fructus)의 활성성분 스크리닝, 표적유전자 확보 및 관련 질병과의 네트워크를 구축한 후 대회향의 항균작용을 중점적으로 분석하였다. Traditional Chinese Medicine Systems Pharmacology (TCMSP) Database 와 Analysis Platform을 통해 대회향의 잠재적 활성성분 49개를 확보하였으며, 그 중 설정한 조건에 부합하는 9개 활성성분을 스크리닝 하였다. TCMSP Database는 활성성분의 약물 동태학적 특성 및 약물-표적-질병 간의 관련성을 네트워크 수준에서 파악할 수 있는 획기적인 in silico적 접근을 가능하게 해준다. 활성성분과 반응하는 201개의 유전자 정보를 UniProt database를 통해 확인하고, 취합한 유전자들이 관여하는 348개의 생물학적 과정을 David 6.8 Gene Functional Classification Tool에서 확보하였다. Chemokine ligand 2, Interleukin-10, Interleukin-6, Tumor Necrosis Factor를 포함한 총 47개의 유전자가 항균작용에 관여하였고 이들을 표적으로 하는 luteolin, kaempferol, quercetin 등이 대표적 항균 관련 활성성분이었다. 이와 같이 확보된 데이터는 연구 재료 선정에 정확성과 시간, 노력, 비용 절감의 효과를 제공함과 더불어 추후 실험적 증명으로 이어져 감염병의 예방과 치료 전략에 과학적인 근거를 제시할 수 있을 것이다.

생쥐에서 5-Hydroxytryptophan에 의한 설사에 대한 각종 전통 생약 추출물의 억제 효과 (Inhibitory Effects of Extracts from Traditional Herbal Drugs on 5-Hydroxytryptophan-Induced Diarrhea in Mice)

  • To find serotonin(5-hydroxytryptamine, 5-HT)-antagonizing activities in traditional herbal drugs, crude extracts from 66 kinds of traditional herbal drugs were randomly screened for inhibitory effects on 5-hydroxytryptophan(HTP)-induced diarrhea in mice. Intraperitoneal injection of 5-HTP(2.5 mg/kg) induced diarrhea in 92% of mice, when observed from 10 to 15 min after injection. Crude extracts(2 g/kg) from 66 kinds of traditional herbal drugs were orally pretreated for 1 h before 5-HTP injection. Of the 66 herbal drugs screened, Ephedrae Herba(麻黃), Cimicifugae Rhizoma(升麻), Anisi stellati Fructus(八角茴香), Aurantii Fructus(枳實), Polygalae Radix(遠志) showed the most potent inhibiting activities against 5-HTP(2.5 mg/kg)-induced diarrhea in mice. There are at least 3 possible mechanisms that would be responsible for the inhibitory effect of crude extracts on 5-HTP-induced diarrhea; 1) crude extract-induced inhibition of the activity of aromatic aminoacid decarboxylase catalyzing the conversion of 5-HTP to 5-HT, 2) crude extract-induced blockade of 5-HT receptor(s) in the gastrointestinal tract responsible for 5-HTP-induced diarrhea, 3) crude extract-induced inhibition of gastrointestinal activity, irrespective of 5-HT system. The exact mechanisms and molecules, responsible for the inhibitory effect of crude extracts on 5-HTP-induced diarrhea remain to be clarified.
