Anti-inflammatory Effect of Euphorbiae kansui Radix Extract in Lipopolysaccharide-stimulated Mouse Peritoneal Macrophages (LPS로 유도된 마우스 복강 대식세포에서 감수(甘遂)추출물의 염증억제 효과)

  • This study is aimed to investigate the anti-inflammatory effect of Euphorbiae kansui radix methanol extract (ERE) in lipopolysaccharide(LPS)-stimulated mouse peritoneal macrophages. Peritoneal macrophages were obtained from thioglycollate-injected Balb/c mice. Cells were stimulated with LPS or LPS plus interferon-gamma (IFN-${\gamma}$) in the presence of ERE and various inflammatory markers were assayed. Finally, LPS-induced signaling molecules were measured. ERE up to $400{\mu}g/m{\ell}$, was not cytotoxic to ERE inhibited LPS/IFN-${\gamma}$-induced nitric oxide (NO), inducible NO synthase. ERE also reduced the levels of cyclooxygenase-2 and the proinflammatory cytokines such as tumor necrosis factor-${\alpha}$, interleukin(IL)-6 and IL-12. The inhibitory effect of ERE on LPS-induced $I{\kappa}B{\alpha}$ degradation was weak but phosphorylation of JNK, p38 and ERK1/2 was strongly suppressed. Our data indicated that the anti-inflammatory effect of ERE in LPS-stimulated macrophages was partly mediated by its inhibition of JNK, p38 and ERK1/2.

Experimental Study on Anti-allergic and Anti-inflammatory Effects of Soeumin-Googwihyangso-san Methanol Extract in Vitro (소음인(少陰人) 궁귀향소산(芎歸香蘇散)의 항(抗) 알레르기 및 항(抗) 염증에 미치는 실험적 연구)

  • Soeumin-Googwihyangso-san(SGGHSS) has been used for the prevention or treatment of Soeumin-allergic rhinitis. This study was performed to demonstrate anti-allergic and anti-inflamatory effects of SGGHSS methanol extract in HMC cell lines and activated mouse B cells and CD4+ T cells. SGGHSS inhibited the production of TNF-$\alpha$ in PMA plus A23187 activated HMC-1 cells but not that of IL-6, as measured by ELISA. SGGHSS inhibited the expression of CD23 and surface IgE in B cells as determined by flowcytometry. It also inhibited secretion of IFN-$\gamma$ and IgG1, the Th1 related IgG type, but increased that of IL-4 in anti-CD40 and IL-4 treated B cells as measured by ELISA. As for Th cell differentiation, SGGHSS did not much affect IL-4 or IFN-$\gamma$. Taken together, our data showed that SGGHSS exerted an anti-inflammtory effect by inhibiting TNF-$\alpha$ in mast cells and has anti-allergic activity not via inhibition of CD4+ T cell, but via inhibition of B cells. These results suggest some evidence that SGGHSS can be applied to allergic disease.

The Effect of Bangpungtongsungsan on Allergic Rhinitis Of Obese Mouse Fed High Fat Diet. (고지방식이로 비만을 유발한 마우스의 알레르기 비염에 대한 방풍통성산(防風通聖散)의 효과)

  • Objective : For various reasons, Obesity & Rhinitis are constantly rising. So Interest of treatment has been expanding. We want to verify The Chungyeol (fire extinguishing) Lisup (Draining) effect of Bangpungtongsungsan on obese model of allergy rhinitis.Material and Methods : BALB/c mouse were divided four groups: control(CON), allergic induction(ARE), Bangpungtongsungsan extract administration(BTT), Bangpungtongsungsan double concentration extract administration (BT2T). Every group except control group were caused allergic rhinitis by Ovalbumin. BTT & BT2T were orally administered the Bangpungtongsungsan for 21days. Since then we observed the liver tissue cell and the nasal mucous membrane.Results : In comparison with ARE, experimental groups show relief of the nasal mucous membrane damage(secretion of mucus decrease, Itching decrease), Th2 eruption control(IL-4 decline), effect of anti-inflamatory(reducing TNF-α creation, decreasing of iNOS through NF-κB activation-inhibition). In addition, experimental groups show a loss in weight, diminished accumulation of fat. (decreasing within liver tissue, reducing TNF-α creation) BT2T is more effective to BTT.Conclusion : Bangpungtongsungsan treat obese model on allergy rhinitis thereby control fat augmentation, relieving inflammation.

  • Acupuncture therapy has demonstrated efficacy in several clinical areas, and of these areas the understanding of pain has progressed immensely in the last two decades. The underlying mechanisms of acupuncture in general and the analgesic effect in particular are still not clearly delineated. The leading hypothesis include the effects of local stimulation, neuronal gating, release of endogenous opiates, and the placebo effect. Accumulating evidence suggests that the central nervous system(CNS) is essential for the processing of these effects, via its modulation of the autonomic nervous system, neuro-immune system, and hormonal regulation. These processes tap into basic survival mechanisms. As such, understanding the effects of acupuncture within a neuroscience-based framework becomes vital. We propose a model which incorporates the stress-induced hypothalamus-pituitary-adrenal axis(HPA-axis) model of Akil et al., the cholinergic anti-inflamatory observations of Tracey et al., and Petrovic et al.

The Effect of Bee-venom Acupuncture on Heel Pain (족근통(足根通)에 봉약침료법(蜂藥針療法)이 미치는 영향(影響))

  • Objetive : To evaluate the effect of treatment for Heel Pain by using Bee-venom Acupuncture that is well known for anti-inflamatory effect and function of activating immunine system. Methods : We investigated 32 cases of patient with Heel Pain. From 1th November 2001 to 30th June 2002, 32 cases of patient with Heel Pain treated at the Department of Acupuncture & Moxibustion of Kyung-won University Oriental Hospital were selected for two group. One group was treated by Bee-venom Acupuncture therapy (Bee-venom Acupuncture Group : BAG), the other group was treated by Common Acupuncture therapy (None Bee-venom Acupuncture Group : NBAG). Both group were composed of 16 patients. After treatment, we evaluated the effects per each group. Results : 1. In the distribution by sex & age : 5 cases were male, 11 cases were female in BAG. 10 cases were male, 6 cases were female in NBGA. Forty aged cases were the most in BAG. Thirty aged cases were the most in NBAG. 2. Assessment of treatment by therapeutic frequency : 14 cases(87.5%) were above "Fair" within 6 times in BAG, 7 cases(43.8%) were above "Fair" beyond 10 times. 3. Assessment of treatment : Excellent were 9 cases, Good 5 cases, Fair 2 cases in BAG, Excellent were 2 cases, Good 7 cases, Fair 2 cases, Poor 5 cases in NBAG. Therapeutic effect above "Fair" were 16 cases(100%) in BAG, 11 cases(68.8%) in NBAG. 4. Comparing BAG with NBAG by therapeutic frequency and assessment in Grade II state : 11 cases(91.7%) were above "Good" within 6 times in BGA, 3 cases (57.1%) above "Fair" beyond 10 times. 5. About average frequency of treatment in the distribution of Grade : Grade III was 5.3 times, Grade II 4.0 times in BGA, whereas Grade III 16.0 times, Grade II 8.4 times in NBGA. Conclusions : In the treatment of Heel Pain, the Bee-venom Acupuncture can be regarded as more useful method in the clinical practice, because it has comparatively shorter duration of treatment and is more effective.
