The Study of the Anti-oxidative Effects of Polygonati Multiflori Caulis on Oxidized Brain and Liver Cells in Rats (야교등(夜交藤)이 흰쥐 뇌와 간에서의 산화 억제에 미치는 영향)

  • Objectives : This study was purposed to the anti-oxidative effects of Polygonati Multiflori Caulis(henceforth PMC) on oxidized brain and liver cells in rats. Methods : After extraction of PMC with water, the water extract was divided into five fractions : hexane, ethyl ether, ethyl acetate, butanol and an aqueous fraction. The phenol contents of each fraction were measured. The lipid peroxidation inhibition effect were then investigated. Results : After processing PCM water and PCM fractionations on oxidized brain cells in rats, the SOD (super oxide dismutase) activity and glutathione content were increased, and the NO (nitric oxide) content was decreased. It had much higher SOD activity than liver cells in rats excluded in the n-BuOH and aqueous fractions. In case of oxidized liver cells in rats, the SOD activity and glutathione content increased, while both the NO content and the MDA (malondialdehyde) content decreased. It had much higher glutathione content than brain cells in rats in the every fractions. It had much lower MDA content than brain cells in rats in the Aqueous fractions and brain cells in rats had much lower MDA content than liver cells in rats in the total extract, n-hexane, EtOEt, EtOAc and n-BuOH fractions. Conclusions : PMC has anti-oxidative effect on oxidized liver cells and brain cells in rats, through there are differences in fraction. Additionally, Anti-oxidative effect of brain cells can be relaxed the mental nerve and it is related PMC effect.

Experimental Study of Fraction of Citri Reticulatae Viride Pericarpium extract on the Brain Damage in Cerebral Ischemia (청파활성분획이 허혈성 뇌손상에 미치는 실험적 연구)

  • Citri Reticulatae Viride Pericarpium extract(CRVP) have been used in oriental medicine for many centuries as a therapeutic agent for smoothing the liver and regulating the circulation of qi, and promoting digestion and removing stagnated food. The effects of CRVP on the inhibition of brain damage in cerebral ischemia is not known. Therefore, this Study was designed to investigate the cerebral protective effects of CRVP on the transient cerebral ischemia using modern techniques, and further to provide the possibility of scientification of oriental medicine. The size of cerebral infarct size was measured by morphometry, and brain edema was measured by morphometry and brain water content determination. The results were a$ follows ; 1. Water fraction of CRVP was reduced infect area of rats brain slices which were subjected to a transient cerebral ischemia in a dose-dependent manner. 2. Methylene chloride fraction and hexane fraction of CRVP was significantly reduced infarct area of rats brain slices which were subjected to a transient cerebral ischemia in a dose-dependent manner. 3. Methylene chloride fraction and hexane fraction of CRVP was significantly reduced infarct volume of rats brain which was subjected to a transient cerebral ischemia in a dose-dependent manner. 4. Methylene Chloride fraction and hexane fraction of CRVP was significantly decreased brain edema induced by a transient cerebral ischemia in a dose-dependent manner. 5. Methylene chloride fraction and hexane fraction of CRVP was significantly decreased brain water content of rats which were subjected to a transient cerebral ischemia. It is suggested that CRVP has an anti-ischemic effect through the inhibition of brain damage in a transient cerebral ischemia, and that in future further development of main effective constituent in CRVP can provide a novel therapeutic strategy for cerebral ischemia.

Effects of Hwangnyeonhaedok-tang on Cerebral Hemorrhage and Edema in Collagenase Induced-ICH Rats (황련해독탕(黃連解毒湯)이 Collagenase에 의한 흰쥐의 뇌출혈 및 뇌부종에 미치는 영향)

  • Objectives: Brain edema is brain swelling that occurs due to the accumulation of excess water in the brain parenchyma. AQP4 and AQP9 are water-channel proteins expressed strongly in the brain and control water fluxes into and out of the brain parenchyma. This study was conducted to evaluate effect of Hwangnyeonhaedok-tang on brain edema and intracerebral hemorrhage. Methods: Intracerebral hemorrhage was induced by intrastriatal injection of type IV collagenase(0.23 U/${\mu}l$, 0.1 ${\mu}l$/min) into Sprague-Dawley rat brains. Hwangnyeonhaedok-tang water extract(1,000 mg/kg) was administered orally three times every 20 hours from 4 hours after ICH operation. Then hematoma volume, brain edema percentage, and water content of brain tissue were measured. Immuno-histochemistry was performed for AQP4 and AQP9 expressions in the brain sections and area % of immuno-labeling was analyzed with image analyzing system. Results: 1. Water extract of Hwangnyeonhaedok-tang reduced hematoma volume of ICH induced rat. 2. Water extract of Hwangnyeonhaedok-tang reduced MPO positive neutrophils in the perihematoma of the ICH induced rat. 3. Water extract of Hwangnyeonhaedok-tang reduced brain edema percentage and water content of brain tissue of ICH induced rat. 4. Water extract of Hwangnyeonhaedok-tang reduced AQP4 immuno-positive cells in the perihematoma of the ICH induced rat. 5. Water extract of Hwangnyeonhaedok-tang reduced AQP9 immuno-positive cells in the perihematoma of the ICH induced rat. Conclusions: These results suggest that Hwangnyeonhaedok-tang decreases intracerebral hemorrhage and brain edema by means of downregulating AQP4 and AQP9 expressions in the brain.

Effects of Cheongyoung-tang on CHT-Induced Brain Edema and Neurological Disturbance Symptom in Rats (청영탕이 CHT로 유발된 백서의 뇌부종 및 신경장애 증상에 미치는 영향)

  • This study was carried out to evaluate Effects of Cheongyoung-tang on CHT-Induced Brain Edema and Neurological Disturbance Symptom in Rats. we make a comparative study of the such parameters as neurological severity score(NSS) and changes in water content, lactate, glucose and hematological substance. 1. ΔNSS increased in CYT1 and CYT2 as compared with the control group, which showed no efficacy. 2. In water content of both hemispheres, CYT1 and CYT2 as compared with the control group have no difference. 3. Water content of the left hemisphere decreased in CYT1 and CYT2 as compared with the control group. 4. The lactate level in serum decreased in CYT1 and CYT2 as compared with the control group. 5. In hematological changes, RBC, PLT and MCHC increased in CYT1 and CYT2 as compared with the control group, but MCV decreased. According to these results, CYT has an effect on CHT-induced brain edema and neurological symptom.

Effects of Salviae Miltiorrhizae Radix on Blood-Brain Barrier Impairment of ICH-Induced Rats (단삼(丹蔘)이 뇌조직출혈 흰쥐의 혈액뇌관문 손상에 미치는 영향)

  • Objectives : This study was performed in order to evaluate the effects of Salviae Miltiorrhizae Radix (SMR) water extract against the cerebral hemorrhage and the blood-brain barrier (BBB) impairment in the intracerebral hemorrhage (ICH). Method : ICH was induced by the stereotaxic intrastriatal injection of bacterial collagenase type IV in Sprague-Dawley rats. SMR was orally given three times every 20 hours during 3 days after the ICH induction. Hematoma volume, water content of brain tissue and volume of evans blue leakage were examined. Myeloperoxidase (MPO) positive neutrophils and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) were observed with immunofluorescence labeling and confocal microscope. Results : SMR significantly reduced the hematoma volume of the ICH-induced rat brain. SMR significantly reduced the water content of brain tissue of the ICH-induced rat brain. SMR reduced the percentage of the evans blue leakage around the hematoma on the caudate putamen compared to the ICH group, especially on the cerebral cortex. SMR significantly reduced the volume of the evans blue leakage level in the peri-hematoma regions of the ICH-induced rat brain. SMR significantly reduced MPO positive neutrophils in the peri-hematoma regions of the ICH-induced rat brain. SMR reduced the TNF-${\alpha}$ expression in peri-hematoma regions of the ICH-induced rat brain. TNF-${\alpha}$ immuno-labeled cells were coincided with MPO immuno-labeled neutrophils in peri-hematoma regions of the ICH-induced rat brain. Conclusion : These results suggest that SMR plays a protective role against the blood-brain barrier impairment in the ICH through suppression of inflammation in the rat brain tissues.

Effects of Amino Acid Composition of Diet and Environment on RNA, Protein Content in Brain and Learning Ability in Rats (식이내 아미노산의 조성과 환경이 흰쥐의 두뇌중 RNA 단백질함량 및 학습능력에 미치는 영향)

  • This study was performed to investigate the influence of the amino acid composition of diet and environment on RNA, protein content in brain and learning ability in rats. Forty-two Sprague-Dawley male rats were divided into six groups according to type of diet, casein, soybean meal, or corn gluten and rearing condition, isolated or enriched. They were fed foods ad libitum for 6 weeks. A water maze was used to test behavioral performance for 3 weeks from 4th week. The rats were sacrificed at 6th week and their whole brains were taken and frozen for assay of RNA and protein. The results were summarized as follows : 1) The body weight gain for the experimental periods of corn gluten group was significantly lower than the casein and the soybean meal group. 2) The brain weight of the corn gluten group was significantly lower than the casein and the soybean meal group and the environmental enrichment slightly increased it among rats fed the corn gluten diet. 3) The total RNA contents were the greatest in the environmentally enriched casein group. The brain protein contents of the isolated corngluten group was the smallest. However, the contents of the enriched corn gluten group were similar to those of the others. 4) In the water maze test, the isolated corngluten group spent significantly more time than the others. Environmental enrichment could decrease time to perform the task of the maze.

Effects of ${\omega}-3$ and ${\omega}-6$ Fatty Acids from Prenatal to Growing Period on the Brain Growth and Behavioral Development of the Rats ($\omega3$$\omega6$계 지방산이 흰 쥐의 뇌 성장과 지능발달에 미치는 영향)

  • This study was designed to find out the effects of $\omega$-3 and $\omega$-6 polyunsaturated and saturated fatty acid from prenatal to growing period on the brain growth and behavioral development of rats. Rats(Sprague-Dawley strain) were fed experimental diets-fish oil, corn oil or beef tallow-with different contents of $\omega$-3 and $\omega$-6 fatty acids throughout the prenatal and lactational period and up to 10 weeks of age. DNA and RNA concentration of rat brain were determined at 0, 3, 6 weeks of age and choline and acetylcholine concentrations were analyzed at 10 weeks of age. When the rats were 7 weeks of age, position reversional test in a Y-shaped water maze for 4 weeks was measured. The experimental results obtained are summarized as follows. Food intakes were significantly lower in fish oil group and body weight gain was low in the group fed beef tallow and the groups fed fish oil and corn oil were somewhat good. Food efficiency ratio was not significantly different among the groups. Brain weight was not affected by the fatty acid composition of experimental diets and DNA and RNA concentration of the rat brain were consistently maintained at the same level. It was not different significantly among the dietary groups in the DNA and RNA concentrations of the rat brain during the experimental period. The acetylcholine concentration in the fish oil group was somewhat higher than the other groups. The position reversional test in a Y-shaped water maze showed a significant difference the score of test among the experimental groups. The score of the rats fed the fish oil diet was significantly higher than the other groups and the concentration of acetylcholine in brain were too. Therefore the correlatin between the Y-shaped water maze test score and the acetylcholine concentratin in the brain was found. Above finding support the content that dietary fatty acid composition does not affect to the brain cell number and cell size but the behavior development is influenced. Therefore, the improvement of behavior development is required the effective usage of finny tribe.

Effect of Riboflavin on the Metabolism of Lipids and Neurotransmitter in Rat Brain (리보플라빈이 뇌조직이 지방과 신경전달 물질대사에 미치는 영향)

  • Rats were fed for an 8-week period a low riboflavin diet(5ug riboflavin/day) or a control diet(30ug/day) supplied either ad libitum or by pair feeding in order to study the effect of riboflavin on the metabolism of lipids and neurotransmitters. Erythrocyte glutathione reductase (EGR) and monomine oxidase(MAO) activity in the liver and brain were assayed. EGR activity coefficient in riboflavin deficient rats was significantly higher than in ad libitum controls whereas MAO activity was decreased in the deficient rats. Fatty acid composition showed a different trend in the serum, liver and brain. In the serum, the concentrations of essential fatty acids and $\omega$-3 fatty acids(eicosapentaenoic acid, docosahexaenoic acid)were decreased about 20-40% in the deficient and pair-fed than in the ad libitum controls. Brain serotonin and 5-HIAA(5-hydroxyindole acetic acid) concentrations were decreased in the riboflavin deficient rats. Learning ability measured by a water maze and exploratory activity using the open field test were not impaired in the deficient rats. These results indicate that brain lipid metabolism was protected in subclinical riboflavin deficiency, however, riboflavin deficiency affected brain serotonin content.

  • For experimental studies on the anticonvulsion effect and it was measured in mice that toxigenic effect, influence on the central nervous system, anticonvulsion effect, mechanism of anticonvulsion effect by change of GABA level and glutamic acid in brain, effect of the creation and degradation system of brain oxygen free radicals in convulsion. The results were obtained as follows: 1. Samulanshintang was perfect medicine without toxigenic effect. 2. Pretreatment of Samulanshintang did not influence on the central nervous system. 3. Pretreatment of Samulanshintang did not influence on maximal electric seizure(MES), strychnine, bicuculine and picrotoxin, but pentylenetrazol(PTZ)-induced convulsion significantly decreased. 4. Effect of Samulanshintang except for Jinsa on the PTZ-Induced convulsion decreased. 5. Effect of Samulanshintang fragrance(SMATF) and Samulanshintang distiled water(SMATW) on the PTZ-induced convulsion did not influence. 6. Decrease of brain GABA level in PTZ-induced convulsion was increased by pretreatment of Samulanshintang. 7. Decrease of brain glutathione content in PTZ-induced convulsion was increased by pretreatment of Samu- lanshintang. 8. GABA-T activity increased by PTZ-induced was controlled by the pretreatment of Samulanshintang. 9. Increase of brain lipid peroxide content in PTZ-induced convulsion was decreased by pretreatment of Samulanshintang. 10. Significant increase of brain xanthine oxidase and aldehyde oxidase activities in PTZ-induced was controlled by pretreatment of Samulanshintang. 11. Decrease of brain superoxide dismutase(SOD), catalase and glutathione peroxidase activities in PTZ-induced was decreased by pretreatment of Samulanshintang. From the above results, Samulanshintang was perfect medicine without toxigenic effect and was recognized anticonvulsion effect by decreasing brain glutamic acid level and increasing brain GABA level. Samulanshintang have an effect on creation and degradation system of brain oxygen free radicals in convulsion, thus it was considered that Samulanshintang could be applied in convulsive disorder as epilepsy, febrile seizure and spasm etc.

Effects of Dandelion on Oxygen Free Radical Generating and Scavenging System of Brain in Streptozotocin-Induced Diabetic Rats (서양민들레가 Streptozotocin으로 유발한 당뇨 횐쥐의 뇌조직 중 유해 활성산소 생성 및 제거 효소계에 미치는 영향)

  • Many studies have shown that hyperglycemia leads to an increase of lipid peroxidation in diabetic patients and animals, reflecting a rise reactive oxygen species production. It is increasingly recognized that brain is another site of diabetic organ damage. Accordingly, this study was to investigate the effect of dandelion on oxygen free radical generating and scavenging system of brain in streptozotocin (STZ)-induced diabetic rats. Male Wistar rats were divided into diabetic (control) and diabetic-dandelion supplemented groups. Dandelion was supplemented for 4 weeks with dandelion leaf and root powder (DLP, DRP) or dandelion leaf and root water extract (DLW, DRW) based on 11.4 g of raw dandelion/kg diet. Diabetes was induced by single injection STZ (55 mg/kg B.W., i.p.)in a citrate buffer. Oxygen free radical generating enzymes, cytochrome P-450, amino-pyrine N-demethylase, aniline hydroxylase and xanthine oxidase, were lowered in dandelion supplemented-groups compared to the control group. Superoxide dismutase, catalase and gluthathione peroxidase activities of brain were also lower in dandelion leaf and root supplemented-group than in the control group, whereas glutathione S-transferase activity and gluthathione content were increased in dandelion supplemented-groups compared to the control group. With regard to the lipid peroxidation products, the malondialdehyde content of brain was lower in dandelion supplemented groups. Therefore, it could be suggested that powder and water extract of dandelion leaf or root are beneficial in preventing diabetic complication from lipid peroxidation and free radical in brain of diabetic rat brain.
