Experimental Studies on Hemostatic Effect of Yongdamsagantang Subtracted Bupleuri radix and Yongdamsagantang Subtracted Bupleuri Radix Added Biotae Cacumen, Imperatae Rhizoma, Typhae Pollen (龍膽瀉肝湯加減方의 止血效果에 關한 實驗的 硏究)

  • Based upon our laboratory experiments of Yongdamsagantang subtracted Bupleuri Radix and Yongdamsagantang subtracted Bupleuri Radix added Biotac Cacumen, Imperatae Rhizoma, Typhae Pollen on male rats to examine the medicines' hemostatic effect, we draw conclusions as follows: 1. Compared to the control group, Yongdamsagantang subtracted Bupleuri Radix and Yongdamsagantang subtracted Bupleuri Radix added Biotae Cacumen, Imperatae Rhizoma, Typhae Pollen increased the number of platelet counts meaningfully. 2. Compared to the control group, Yongdamsagantang subtracted Bupleuri Radix and Yongdamsagantang subtracted Bupleuri Radix added Biotae Cacumen, Imperatae Rhizoma, Typhae Pollen suppressed prothrombin time without any meaning. 3. Compared to the control group, Yongdamsagantang subtracted Bupleuri Radix and Yongdamsagantang subtracted Bupleuri Radix added Biotae Cacumen, Imperatae Rhizoma, Typhae Pollen, all of these medicines elevated the fibrinogen level, but only Yongdamsagantang subtracte Bupleuri Radix is recognized to raise the fibrinogen level meaningfully. 4. Compared to the control group, Yongdamsagantang subtracted Bupleuri Radix and Yongdamsagantang subtracted Bupleuri Radix added Biotae Cacumen. Imperatae Rhizoma, Typhae Pollen repressed partial thromboplastin time meaningfully. In conclusion, Yongdamsagantang subtracted Bupleuri Radix and Yongdamsagantang subtracted Bupleuri Radix added Biotae Cacumen, Imperatae Rhizoma, Typhae Pollen can be used for stopping blood effectively in terms of platelet count increase, prothrombin time control, fibrinogen level rise, and partial thromboplastin time suppression.

Application of prescription of Bupleuri Radix mixture in the Uibanghwaltu chapter of the Bangyakhappyeon (방약합편(方藥合編) 의방활투(醫方活套) 중(中) 시호(柴胡)가 배합(配合)된 방제(方劑)에 관한 고찰(考察))

  • Objectives : Bangyakhappyeon was classified into three tongs of prescription. When juxtaposed with existing publications on oriental medicine, this composition was considerably original. Due to its ease of searching for diseases and prescription, it has been read by not only doctors and researchers but also among general citizens alike. Based on the fundamentals of Bangyakhappyeon composition, this thesis focused on the thorough research on the prescription of Bupleuri Radix mixture. Methods : 42 prescriptions of Bupleuri Radix mixtures have been divided in to three tongs based on the composition of Bangyakhappyeon. Then, these prescriptions, dosage, and composition were researched and organized. Results : After analyzing the prescription of Bupleuri Radix mixture in the Uibanghwaltu chapter of the Bangyakhappyeon text, the following results were attained. 1. Of the 42 prescriptions of Bupleuri Radix mixtures, jungtong constitutes the most prescription at 30, 8 at hatong, and 4 at sangtong. 2. 42 prescriptions are documented into 18 medical divisions and 44 disease symptoms. 3. After analyzing the prescription dosage of Bupleuri Radix mixture, the dosage of Bupleuri Radix was 3 puns to 4 jeons used in 14 sections. Most use was 1 jeon 14 prescriptions, 33.3% were recorded. 4. Invigorating vital function, catharsis, malaria, mediation method, and clearing away method of Bupleuri Radix have a close relationship with the dosage. 5. The 42 prescriptions formation of using Bupleuri Radix consisted of decoction at 39 prescriptions (92.9%) and the others at 3 prescriptions (7.1%). 6. As a result of researching the component of the prescription of Bupleuri Radix mixture, Sosihotang, Soyosan and Bojungikgitang were used as the basic prescription. Conclusions : prescription of Bupleuri Radix mixture in Uibanghwaltu of Bangyakhappyeon as based on Sosihotang, Soyosan and Bojungikgitang, combined with various oriental medicine was found to treat many kinds of diseases.

Protective Effect of Bupleuri Radix on Hypoxia Reperfusion Induced by PC12 Cell Damage and Global Ischemia in Gerbil (PC12 손상 세포 및 전뇌허혈 유발 Gerbil에 대한 시호 세포보호효과)

  • Objects: This research was conducted to investigate the protective effect of Bupleuri Radix against ischemic damage using PC12 cells and global ischemia in gerbils, Methods: To observe the protective effect of Bupleuri Radixon ischemic damage, viability and changes in activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase and production of malondialdehyde (MDA) were observed after treating PC12 cells with Bupleuri Radix during ischemic damage. Gerbils were divided into three groups: a normal group, a 5-minute two-vessel occlusion (2VO) group and a Bupleun Radix administered group after 2VO. The CCAs were occluded by microclip for 5 minutes, Bupleuri Radix was administered orally for 7 days after 2VO. Histological analysis was performed on the 7th day. For histological analysis, the brain tissue was stained with 1 % of cresyl violet solution. Results: 1. Bupleuri Radix has a protective effect against ischemia in the CA1 area of the gerbil's hippocampus 7 days after 5-minute occlusion. 2. In the hypoxia/reperfusion model using PC12 cells, the Bupleuri Radix has a protective effect against ischemia in the dose of 0.2{\;}\mu\textrm{g}/ml,2{\;}\mu\textrm{g}/ml{\;}and{\;} 20{\;}\mu\textrm{g}/ml$. 3. Bupleuri Radix increased the activities of glutathione peroxidase and catalase. 4. The increased activity of superoxidedismutase (SOD) by ischemic damage might have been induced as an act of self-protection. This study suggests that Bupleuri Radix has some neuroprotective effect against neuronal damage following cerebral ischemia in vivo with a widely used experimental model of cerebral ischemia in Mongolian gerbils. Bupleuri Radix also has protective effect on a hypoxia/reperfusion cell culture model using PC12 cells. Conclusions: Bupleuri Radix has protective effect against ischemic brain damage during the early stages of ischemia.

  • Effects of siho(Bupleuri Radix) extracts on lipid composition and antioxidant system were investigated in rat fed high fat diet. body weight gain, concentration of plasma triglyceride, total cholesterol, LDL-cholesterol showed a tendency to decrease in siho(Bupleuri Radix) extracts groups. However, the concentration of HDL-cholesterol showed no significantly different among treatments. Plasma and liver TBARS concentration showed a low values in siho(Bupleuri Radix) extracts groups. Activities of SOD, GSH-Px and CAT showed a tendency to increase in siho(Bupleuri Radix) extracts groups.

A Study of Neuroproctective Effect of Bupleuri Radix on Hippocampal Neurons (시호(柴胡)의 뇌해마 신경세포 보호효능에 대한 연구)

  • Objective : This study was performed to investigate neuroprotective effects of Bupleuri Radix against oxidative and ischemic damages. Method : To observe the neuroprotective effects against ischemic damage, ischemic insult was induced by oxygen/glucose deprivation (OGD) on organotypic hippocampal slice cultures (OHSC) from 1 week-old Sprague-Dawley rats. Propidium iodide (PI) fluorescence-stained neuronal dead-cell areas, area percentages and TUNEL-positive apoptotic cells in CA1 and dentate gyrus, and LDH levels in culture media of the OHSC were measured following Bupleuri Radix extract treatment. Result : The following results were obtained: (1) The $5\;{\mu}g/ml$ of Bupleuri Radix treatment demonstrated a significant decrease in PI fluorescence-stained neuronal dead-cell areas and area percentage in CA1 region of the OHSC from 18 hrs to 48 hrs following the OGD. The $50\;{\mu}g/ml$ of Bupleuri Radix treatment was also significant from 6 hrs to 48 hrs following the OGD and was more effective. (2) The 5 and $50\;{\mu}g/ml$ of Bupleuri Radix treatment demonstrated a significant decrease in PI fluorescence-stained neuronal dead-cell areas and area percentage in DG region of the OHSC from 6 hrs to 48 hrs following the OGD. The $50\;{\mu}g/ml$ treatment was more effective than the $5\;{\mu}g/ml$ treatment. (3) Bupleuri Radix treatment demonstrated a significant decrease in TUNEL-positive apoptotic cells in CA1 region (with 5 and $50\;{\mu}g/ml$) and in DG region (with $50\;{\mu}g/ml$) of the OHSC damaged by the OGD. (4) Bupleuri Radix treatment demonstrated a significant decrease in LDH concentrations in culture media of the OHSC damaged by the OGD. Conclusion : These results suggest that Bupleuri Radix has neuroprotective and control effects on inflammatory and immune responses where there has been ischemic damage to the central nervous system.

Effects of the Methanol Extract of Bupleuri Radix on Primary Cultured Brain Cells, DRG and Hepatocytes (시호의 메탄올 추출물이 일차배양한 뇌, DRG 및 간세포에 미치는 영향)

  • Effects of the methanol extract of Bupleuri Radix on primary cultured chicken embryonic brain cells, dorsal root ganglia (DRG) and rat hepatocytes were studied. The methanol extract of Bupleuri Radix at the concentration ranging from $10{\;}{\mu}g/ml\;to\;100{\;}{\mu}g/ml$ significantly recovered the cytotoxicity of rat hepatocytes induced by the treatment of galactosamine; at the concentration of $100\;{\mu}g/ml$, values of GOT and GPT in the culture medium were reduced by the 60% and 75%, respectively of those in the absence of the methanol extract of Bupleuri Radix. The addition of the methanol extract of Bupleuri Radix. into chicken embryonic brain cells which were cultured with a deficient medium significantly increased the number of cells promoting the neurite outgrowth. However, the methanol extract of Bupleuri Radix showed no effect on the activities of PDHC and acetylcholinesterase in primary cultured brain cells.

  • Bupleuri radix (Umbelliferae), the dried root of Bupleurum Chinense DC, has been clinically used to mitigate pain sensation. The descending pain control system consists of three major components, and modulation of pain in the periaqueductal gray is the most extensively studied descending pain control system. However, the relation of Bupleuri radix on the descending pain control system has not been clarified. In the present study, modulation of the aqueous extract of Bupleuri radix on glycine-induced ion current in the acutely dissociated periaqueductal gray neurons was investigated by using nystatin-perforated patch-clamp technique under voltage-clamp condition. In the present results, the glycine-induced ion current was significantly suppressed by 0.1 mg/ml Bupleuri radix, while treatment with $10^{-5}\;M$ naltrexone, opioid receptor antagonist, alleviated Bupleuri radix-induced inhibition on glycine-induced ion current. The present study showed that the aqueous extract of Bupleuri radix may activate descending pain control system through inhibition on glycine-induced ion current in the periaqueductal gray neurons and this effect is mediated by opioid receptors.

The Effect of Bupleuri Radix Herbal-acupuncture at GB34(Yangneungcheon) on Hyperlipidemia in Rats (양릉천 시호 약침이 고지혈증에 미치는 영향)

  • Objectives & Methods : The purpose of this study is to observe the effects of Bupleuri Radix Herbal-acupuncture at GB34(Yangneungcheon) of hyperlipidemic rats. The author performed several items to analyze body-weight of the rats, weight of its liver, serum as well as urine changes in each group. Results : 1. Bupleuri Radix herbal-acupuncture(BR-HA) infused solution (MCR-HAS) increased both DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. BR-HA at GB34 significantly decreased the level of serum total cholesterol and atherogenic index(AI), while significantly increased the ratios of high-density lipoprotein (HDL)/total cholesterol and phospholipid/total cholesterol in hyperlipidemic rats. 3. BR-HA at GB34 significantly decreased urine glucose in hyperlipidemic rats. Conclusions : From the results above, it is suggested that Bupleuri Radix herbal -acupuncture at GB34 has a therapeutic effect on hyperlipidemia.

Effects of Bupleuri Radix on Rat Hepatic COMT by Common Bile Duct Ligation and Taurocholate Load after Common Bile Duct Ligation (시호가 총담관결찰 및 Taurocholate 부하 흰쥐 간의 COMT 활성에 미치는 영향)

  • Object : This study was earned out to examine the effect of Bupleuri Radix on experimental cholestasis, and make clear a part of this mechanism. Methods : Two models of common bile duct ligation group and taurocholate load group after common bile duct ligation were induced, and Bupleuri Radix extract was taken orally for 14 days. In the 1, 2, 4, 7 and l4days after treatment, cytosolic, mitochondrial and microsomal catechol-O-methyltransferase(COMT) activities in liver were measured. Results : The activities of cytosolic, mitochondrial and microsomal COMT increased in the Blupleuri Radix treated group after common bile duct ligation and after taurocholate load and common bile duct ligation. The activities of cytosolic and mitochondrial COMT increased particularly in Blupleuri Radix treated group after taurocholate load and common bile duct ligation. Conclusions : According to the result, it is considered that Blupleuri Radix not only improves cholestatis in liver, but also decreases a genetic synthesis of taurocholic acid.

Antihyperlipidemic Effects of Bupleuri Radix, padix and Uncariae Ramulus et Uncus on Experimental Hyperlipidemia in rats (고지혈증 랫트를 이용한 시호, 작약, 조구등의 항고지혈 효과)

  • Effects of Bupleuri Radix, Paeonie Radix or Uncariae Ramulus et rncus on experimental yperlipidemia were studied. Hyperlipidemia was induced on male Wistar rats by feeding them on high holesterol diet for one week, as previously described by the authors. Blood lipid profile was verified on these ats by measuring total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL) and low ensity lipoprotein cholesterol(LDL). Then, the rats were followed by feeding normal diet. At the same time, ethanol extracts of the three individual herb medicines were orally administered to the rats for 4 weeks and the parameters above mentioned were monitored. Methanol extract of 5Bupleuri radix reduced the TC value significantly at 2 week point and Paeoneae Radix reduced the TC value at 4 week point in compare to control group, suggesting the antihyperlipidemic effect of the two herbal medicines in vivo. The extract of Uncariae Ramulus et Uncus however did not show antihyperlipidemic effect in our experiment.
