Endotoxin에 의한 혈전증에 미치는 홍화자의 효과 (Effect of Carthamus tinctorius L. Semen on Endotoxin-induced Thrombosis in Rats)

  • In the advanced age, cardiovascular disease is more serious than any other disease. Especially, the thrombus causes the serious disease like apoplexia, carebri and myocardial infarction. Thrombosis is caused by the injury of endothelium and the alterations in normal blood flow. To investigate activities of Carthamus tinctorius L. Semen butanol fraction for blood coagulation system, endotoxin (4000EU/kg) was injected (i..v) to rats at 1hr after administration of Carthamus tinctorius L. Semen butanole fraction (500 mg/kg, p.o.). Carthamus tinctorius L. Semen butanol fraction was found to have antiplatelet activity in vitro. In vivo it showed a delay of blood clotting time, and prothrombin time, and reduction of fibrinogen and FDP It also increased SOD activity, and decreased MDA content. These results suggest that the antithrombosis effect of Carthamus tinctorius L. Semen butanol frac tion results from suppressive activity for a blood coagulation system and antioxidative activity.

Physiological Effects of Hot Water Extract and Solvent Fractions of Carthamus tinctorius L.

  • In this study, we investigated the biological activities of Carthamus tinctorius including antioxidative, fibrinolytic, thrombin inhibitory, ${\alpha}$-glucosidase inhibitory and collagenase inhibitory activities. Carthamus tinctorius, hot water extract was fractionated into hexane, chloroform, ethyl acetate, butanol and water fraction. Each of these was assayed individually. The hot water extract showed high antioxidative activity and thrombin inhibitory activity at 90.17% and 97.10% respectively. In the fraction activity tests, chloroform fraction showed the highest antioxidative activity at 81.85%. The fibrinolytic activity was strong only in the butanol fraction at 0.70 plasmin units/ml. The thrombin inhibitory activities of hexane, ethyl acetate and butanol fractions were 97.35%, 86.74% and 93.18% respectively. In collagenase inhibitory activity test, hexane fraction showed the highest activity at 87.78%. In conclusion, the hot water extract and solvent fractions of Carthamus tinctorius L can be used as a material for the development of biofunctional tea and foods respectively.

사염화탄소에 의한 간독성에 미치는 홍화자의 보호작용 (Protective Effect of Carthamus tinctorius L. Semen on Hepatotoxicity by Carbon Tetrachloride in Rats)

  • The protective effect of Carthamus tinctorius L. Semen on the carbon tetrachloride induced liver damaged rats were studied. First, methanol extract was prepared and the extract was fractionated with hexane, $CHCl_3$, BuOH and $H_2O$ respectively. Animals were divided into 6 groups and each group was treated with each fraction respectively. To investigate the hepato-protective effect of Carthamus tinctorius L. Semen AST, ALT, albumin, TP, cholesterol, TG, creatinine and total bilirubin values were measured in each treated group and compared with those of control group. GST activity was increased in BuOH group compared with the control group. In malondialdehyde levels, all fractions was decreased compared with the control group. In histopathologic examination, hexane and $H_2O$ fractions of Carthamus tinctorius L. Semen observed mild degree of ballooning degeneration. The results show the protective effect of Hexane,$CHCl_3$, BuOH and $H_2O$ fractions on hepatotoxicity of $CHl_4$ by decreasing ALT, AST, bilirubin, cholesterol, TG and BUN. It seems that the decrease of MDA are related to the recovery effect. The protective effects of Carthamus tinctorius L. Semen fractions in hepatotoxic pathogenesis by $CHl_4$ was suggested in blood chemistry analysis and histopathologic examination.

홍화씨의 식품학적 성분 분석 및 항균활성 (Analysis of Food Components of Carthamus Tinctorius L. Seed and its Antimicrobial Activity)

  • 본 연구는 홍화씨의 식품학적 성분 및 항균활성을 분석하였다. 홍화씨의 수분함량은 $5.58{\pm}0.09%$, 조단백질 $37.16{\pm}0.85%$, 조회분 $3.52{\pm}0.04%$, 조지방 $13.69{\pm}8.98%$, 탄수화물 40.05%이었다. 홍화씨의 총 아미노산은 391.99 mg%이고, 이중 필수아미노산의 함량은 122.97 mg%이었다. 홍화씨의 아미노산 중 alanine (75.43 mg%), arginine (37.93 mg%) 및 aspartic acid (33.24mg%)을 많이 함유하고 있었다. 홍화씨의 주요 유리당은 fructose가 3.29%이었고, linoleic acid (C18:2)는 약 79.46%의 비율을 차지하여 홍화씨의 조지방 중 가장 많은 것으로 나타났다. 또한, Oleic acid (C18:1)도 약 10.42%를 함유하여 불포화지방산의 함유량도 매우 높게 나타났다. 홍화씨의 무기질 함량 측정 결과, K 및 Ca의 함량이 1,528 및 1,005 mg%으로 가장 많이 함유되어 있었다. 총 페놀 및 총 플라보노이드 함량은 $55.52{\pm}0.99$ mg GAE/g, $78.69{\pm}0.91$ mg QE/g으로 나타났다. 홍화씨의 항균활성은 농도가 증가함에 따라 Staphylococcus aureus, Salmonella typhimurium, Escheria coli, Candida albicans, Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus 및 Clustridium perfringens 균에 대한 항균 효과가 높아지는 것으로 나타났다.

홍화의 항혈전작용에 대한 실험적 연구 (An Experimental Study on the Antithrombotic Effects of CARTHAMUS TINCTORIUS)

  • Objectives: The purpose of this study was to investigate the antithrombotic effect of Carthamus tinctorius. Methods: SD rats were used to investigate the inhibitive effect of platelet aggregation (in vitro & in vivo), prothrombin time, platelet count. Mice were used to investigate the survival rate after injection of collagen & epinepbrine. Results: 1. The inhibitive effect of platelet aggregation (in vitro & in vivo) were increased significantly in all groups. 2. Prothrombin time was decreased in both groups. 3. Platelet count was decreased significantly in sample A. 4. Survival rate after injection of collagen & epinepbrine was increased in sample B. Conclusion: According to the above results, it is suggested that Carthamus tinctorius has antithrombotic effects.

홍화씨 추출물이 치주인대세포와 조골유사세포의 골 광물화 작용에 미치는 효과 (Effect of Extract of Seeds of Carthamus tinctorius L. on Mineralization in Periodontal Ligament Cells and Osteoblastic Cells)

  • The main goal for the treatment of periodontal diseases is the regeneration of lost cementum, bone and connective tissue. Clinical and histological research suggests that it is possible to restore periodontal structures. Seeds of Carthamus tinctorius L. has been used for the treatment of bone fracture and osteoporosis in traditional Korean medicine. The purpose of this study is to examine the effect of extract of seeds of Carthamus tinctorius L. on mineralization in periodontal ligament cells and osteoblastic cells. Periodontal ligament cells were primarily obtained from a extracted premolars with non-periodontal diseases, Osteoblastic cells were obtained from calvariae of a fetal rat, Cells were cultured with DMEM at $37^{\circ}C$ with 5% $CO_2$ in 100% humidity incubator. Alkaline phosphatase(ALP) level and the number of calcification nodules were examined and western blot analysis using osteonectin was performed, Measurements of ALP levels and calcification nodules showed that extract of seeds of Carthamus tinctorius L. had significantly higher activity than control in all of both cells. In western blot analysis, protein expression of osteonectin indicated that extract of seeds of Carthamus tinctorius L. showed an increased pattern than control in all of both cells. From the above results, it seems that extract of seeds of Carthamus tinctorius L. has excellent effect on mineralization in periodontal ligament cells and osteoblastic cells.

홍화 추출물이 생쥐 골수 유래 중간엽 줄기세포의 지방분화에 미치는 영향 (Effects of Carthamus Tinctorius Extract on Adipogenic Differentiation of Mouse Bone Marrow-Derived Mesenchymal Stromal Stem Cells)

  • Objective: This study investigated the effect of purified Carthamus tinctorius (C. tinctorius) extracted with a hot water and ethanol method on adipogenic differentiation of mouse bone marrow-derived mesenchymal stromal stem cells (mBMSCs). Methods: The C. tinctorius was extracted using hot water and ethanol. The samples were concentrated by a rotary evaporator and were then dried using a freeze-dryer. The mBMSCs were cultured and maintained in a minimum essential medium eagle alpha (${\alpha}-MEM$) supplemented with 10% FBS and 1% antibiotic antimycotic solution. To induce adipogenic differentiation, the cells were treated with Dulbecco's modified eagle's medium-low glucose (DMEM-LG) containing 1 mg/mL insulin, 1 mM dexamethasone, and 0.5 mM 3-isobutyl-1-methylxanthine. To evaluate the adipogenic differentiation ability, oil-red O staining was performed after adipogenic differentiation for 21 days. The mRNA expression and protein level of adipogenic-related genes were quantified by quantitative real-time PCR and western blotting, respectively. Results: In the results of the MTT assay, no concentrations of C. tinctorius extracts showed toxicity on mBMSCs, so we fixed the treatment concentration of the extract at 100 ng/mL. In oil-red O staining, the water-C. tinctorius extract treatment significantly decreased adipogenic differentiation compared with the control and ethanol extract groups. The water-C. tinctorius extract group in particular showed reduced mRNA and protein expression of Peroxisome proliferator-activated receptor gamma ($Ppar{\gamma}$) and CCAAT/enhancer-binding protein alpha ($C/ebp{\alpha}$), which are adipogenic-related transcription factors. Conclusion: These data suggest that extract of C. tinctorius decreased the adipogenic differentiation of mBMSCs, while only water-C. tinctorius extract had an effect on different adipogenesis in mBMSCs. The C. tinctorius will be a useful therapeutic reagent for the prevention of obesity-related diseases such as diabetes, hyperlipidemia, coronary artery disease, and osteoporosis.
