Changes of Antioxidant Activity and Ingredient in Medicinal Plants by Processing (수치에 의한 생약의 항산화 활성 및 성분 변화)

  • This study was conducted to investigate the change of antioxidant activity and ingredient in 4 species medicinal plant by processing. As a result, EtOAc extract and n-BuOH extract obtained from Cistanche deserticola Y. C. Ma and processed Cistanche deserticola Y. C. Ma showed difference on the experiment of antioxidant activity by DPPH method. Change of ingredient also was confirmed by TLC and HPLC analyses. Two main compounds of antioxidant activity change were isolated by column chromatography from the EtOAc extract and n-BuOH extract of Cistanche deserticola Y. C. Ma. The chemical structure of the compound 1 isolated from EtOAc extract of Cistanche deserticola Y. C. Ma was elucidated as 2'-acetylacteoside by means of IR and NMR analyses. Also, the chemical structure of the compound 2 isolated from n-BuOH extract of Cistanche deserticola Y. C. Ma was confirmed as acteoside. The antioxidant activity of processed Cistanche deserticola Y. C. Ma. was declined by 3 time decrements of 2'-acetylacteoside such as 1.89 mg/g to 0.62 mg/g and 2 time decrements of acteoside such as 5.81 mg/g to 2.67 mg/g by processing.

Effects of Cistanche Deserticola on Thyroid Function in Hypothyroidism Rat Model Induced by PTU(6-Propyl, 2-thiouracil) (육종용이 PTU로 유발된 갑상선 기능저하증 동물모델에 미치는 영향)

  • Hypothyroidism is the most common disease in the endocrine system. Cistanche Deserticola has been used traditionally in the treatment of kidney deficiency and Yang deficiency syndrome. In this study, we investigated the therapeutic effects of Cistanche Deserticola on Hypothyroidism rat model induced by PTU. 24 Two-month-old Sprague-Dawley male rats were divided into 4 groups : 1) normal(n=6), 2) PTU-induced hypothyroidism control(n=6), 3) hypothyroidism rat treated with Cistanche Deserticola(n=6), 4) hypothyroidism rat treated with levothyroxine(n=6). PTU was administered for 4 weeks. Cistanche Deserticola and levothyroxine was administered 2 weeks after PTU was initiated for a total duration of 2 week. Body weights were checked every week and after 4 weeks, biochemical analysis was performed and T3, T4 and TSH were measured by ELISA kits. In comparison with the normal group, the control group showed hypothyroidism with lower T3, T4 level and higher TSH level. In the Cistanche group the level of T4 was significantly increased and TSH level was significantly decreased. There was no significant difference in biochemical labs and weight between the Cistanche group and the control group. These findings suggest that Cistanche Deserticola could help the production of thyroid hormones under PTU suppression. And there is no harmful effect on liver and kidney function, and other metabolism. According to these results Cistanche Deserticola could be an useful agent for treating hypothyroidism.

Whitening and anti-aging effects of Cistanche deserticola extract (육종용 추출물의 미백과 항노화 효과)

  • This study assessed the whitening and anti-aging effects of the Cistanche deserticola extract, to develop a cosmetic substance. The cell viability of the Cistanche deserticola extract was evaluated in B16F10 melanoma cells by the MTT (3-(4,5-dimethylthaiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay. The cell viability of the extract was determined to be 90% at 4mg/ml concentration. Furthermore, the tyrosinase, collagenase, and elastase mRNA expression level were measured by RT-PCR, using the Cistanche deserticola extract treated B16F10 melanoma cells. At 4 mg/ml concentration, mRNA expression level of tyrosinase, collagenase, and elastase was dramatically decreased to 80.9%, 37.6%, and 70.9%, respectively. The antioxidant activity of the Cistanche deserticola extract was determined by DPPH free radical scavenging. The DPPH free radical scavenging capacities ranged from 70.6% to 82.6%, when evaluated from 2 mg/ml to 10mg/ml concentrations. The effects of whitening and anti-aging of the Cistanche deserticola extracts were examined at 2, 4, 6, 8, and 10 mg/ml concentration. Tyrosinase activities were inhibited from 66.8% to 78.5%, elastase activities were inhibited from 67.6% to 79.3%, collagenase activities were inhibited from 72.3% to 83.6%, and hyaluronidase activities were inhibited from 65.8% to 69.2%, respectively. These data suggest that the Cistanche deserticola extract is effective in whitening and anti-aging; therefore, it is considered to be a functional cosmetic material in cosmetic products.

Anti-inflammatory Effects of Cistanche deserticola Water Extracts in LPS-stimulated RAW 264.7 Macrophages (육총용(肉蓯蓉) 열수추출물이 RAW 264.7 대식세포에 미치는 항염증 효과)

  • Objectives : The purpose of this study is to confirm the anti-inflammatory effects of Cistanche deserticola Water Extracts(CDWE). Methods : In this study, cell viability was determined by MTT assay. NO production was determined with Griess reagent, and IL-6 production was determined by ELISA. And western blot analysis was performed to confirm the protein expression of NOS2, COX-2, I𝜅B-𝛼, p-I𝜅B-𝛼, and NF-𝜅B. Results : CDWE was not cytotoxic at 15.625-1,000㎍/㎖ in RAW 264.7 cells. CDWE inhibited NO production in a concentration-dependent manner, with inhibitory effect on expression levels of IL-6. Expression level of NOS2, NF-𝜅B p65, and p-I𝜅B-𝛼 decreased at 250-1,000㎍/㎖. mRNA expression of COX-2 suppressed at 250, 1,000㎍/㎖. Conclusions : These results suggest that CDWE has anti-inflammatory effects and can be used as an anti-inflammatory therapeutic material.

A Study on the Latest Research Trends in Natural products with Anti-Aging Effects (항노화 효능을 가진 천연물에 대한 최신 연구 동향에 관한 연구)

  • In this study, four types of retinol, retinyl palmitate, adenocin, and polytoxylate-dretinamide, which are the ingredients of the Ministry of Food and Drug Safety Notice, included in the study. also we looked at trends in research on Sciadopitys verticillata, Prunella vulgaris, Celosia cristata L., Brazilin, Persicaria hydropiper, Astragalus membranaceus Bunge, Forsythiae Fructus, Lithospermum root, Rheum undulatum L. and Cistanche deserticola Y. C. Ma a natural material that has the efficacy of antioxidant aging. The anti-aging study so far has been found to be centered mainly on collagen production and elastase synthesis inhibition mechanisms. However, given that the aging process of the skin is caused by various ageing processes, it is believed that anti-aging studies using safe and effective natural materials that can help the skin age with various mechanisms should be conducted.
