산사(山揸)가 흰쥐의 식이성 고지혈증(高脂血症)에 미치는 영향 (Effects of Crataegii Fructus on the Diet-induced Hyperlipidemia in Rats)

  • The purpose of this study was to investigate the effects of Crataegii Fructus on the diet-induced hyperlipidemia in rats. Rats were divided into 4 groups, normal group(supplied enough water and feeds only), high fat diet administered group(supplied high fat diet for 4 weeks, Control group) and Crataegii Fructus administered group(supplied high fat diet and Crataegii Fructus lyophilization extract for 4 weeks, 397.3 mg/kg(rat) in sample A, 662.5 mg/kg(rat) in sample B). Body weight, liver weight and serum lipid levels were evaluated. The water extract of Crataegii Fructus decreased liver weight and triglyceride in high fat diet induced hyperlipidemia in rats, and increased HDL-cholesterol.

  • Crataegii Fructus is commonly used as a improving digestion, removing retention of food, promoting blood circulation and resolving blood stasis agent in East Asia. Cadmium (Cd) is widely distributed in the environment due to its use in industry. An exposure to Cd causes dysuria, polyuria, chest pain, hepatic and renal tubular diseases. The liver is the most important target organ when considering Cd-induced toxicity because Cd primarily accumulates in the liver. This study investigated the protective effect of Crataegii Fructus water extract against cadmium ($CdCl_2$, Cd)-induced liver toxicity in H4IIE cells, a rat hepatocyte-derived cell line and in rats. Cell viability was significantly reduced in Cd-treated H4IIE cells in a time and concentration-dependent manner. However, Crataegii Fructus water extract (CFE) protected the cells from Cd-induced cytotoxicity via inhibition of PARP cleavage. To induce acute toxicity in rats, Cd (4 mg/kg body weight) was dissolved in normal saline and intravenously injected into rats. The rats then received either a vehicle or silymarin (as a positive control) or CFE (50, 100 mg/kg/day) for 3 days, and were subsequently exposed to a single injection of Cd. Alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) were significantly increased by Cd treatment. In contrast, pretreatment with CFE reduced ALT, AST and LDH. In histopathological analysis, CFE reduced the hepatic degenerative regions and the number of degenerative hepatocytes. These are considered as direct evidences that Crataegii Fructus has favorable inhibitory effects on the Cd-intoxicated liver damages. The efficacy of Crataegii Fructus shows slight lower than that of silymarin in the present study.

산사(山査)가 노화유발(老化誘發) 흰쥐의 항산화능(抗酸化能)에 미치는 영향(影響) (Effect of CRATAEGII FRUCTUS(山査) on Antioxidant Capacity in D-galactose induced Aging Rats)

  • Objectives : CRATAEGII FRUCTUS(山査) is known as the substance which delays aging by the antioxidant effects. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on antioxidant enzyme activities such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group (D-galactose 50mg/kg and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 6 weeks, SS Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, neutral fat and cholesterol were measured in rat erythrocytes and blood plasma. Results : There was no significant difference in the level of TBARS in the blood plasma between each experimental group. Red blood cell SOD activities were significantly different in each group. They were significantly decreased in the Control group compared to those of Normal and there was an increasing phenomenon in the SS group compared to the Control group. There was a significant difference in the activities of the red blood cell - glutathione peroxides in each group. There was a significant increase in the SS group compared to the Control group. Red blood cell catalase activities was no significant difference in each group. Plasma total lipid concentration was significantly different in each group. It was significantly increased in the Control group compared with the Normal group and it was decreasing in the SS group compared to the control group. Plasma triglyceride was not significantly different in each group. Plasma total cholesterol and Plasma HDL -cholesterol concentrations in the blood plasma were not significantly different in each group. Conclusions : According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in inhibiting lipid peroxides and increasing the activities of antioxidative(anti aging) enzyme in D-galactose induced aging rat.

  • Objectives: CRATAEGII FRUCTUS(山査) is known as the substance which decreases concentration of sarum lipid. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on decreasing concentration of sarum lipid which are triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol in rat high cholesterol diet inducede hyperlipide rats. Methods: Sprague-Dawley rats were divided into 3 groups, Normal group(supplied enough water and feeds only. Normal Group), high fat diet administered group(AIN-76 high fat diet #100496 used beef tallow for 4 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group(AIN-76 high fat diet #100496 used beef tallow and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 4 weeks. SS Group). Rats were sacrificed and concentration of triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol were measured in rat's sarum lipid Results: There was no significant difference in the level of waight between each experimental group(F=0.243, p=0.787, ANOVA test), Concentration of triglyceride has no significant difference in each group(F=0.210, p=0.813, ANOVA test). There was significant increase of total cholesterol in the Control group compared to those of Normal but there was no significant difference in the SS group compared to the Control group(F=5.237, p=0.0l9, ANOVA test). There was a significant increase of HDL-cholesterol in the SS group compared to the Control group and decrease in Control group compared Normal group(F= 11.437, p=0.001, ANDYA test). Concentration of LDL -cholesterol was significantly different in each group(F= 16.377, p=0.0001, ANDYA test). It was significantly increased in the Control group compared with the Normal group and it has no difference in the SS group compared to the control group. TBARS values(Thiobarbituric Acid Reactive Substances) were significantly different in each group(F=5.242, p=0.0019, ANDYA test). It was significantly increased in the Control group compared with the Normal group and significantly decreased in SS group compared with the Control group. Conclusions: According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in decreaseing concentration of sarum lipid in high cholesterol diet induced hyperlipidemic rats.

Quality Characteristics of Teriyaki Sauces containing Medicinal Herb Extracts

  • This study determined the effects of addition of medicinal herb extracts (Zanthoxylum piperitum extract(TZ), Acanthopanax sessiliflorus(TA), Panax ginseng(TP), Cinnamomum lureitri Nees(TC), Angelica gigantis radix(TAG) and Crataegii fructus extract (TCF) to teriyaki sauce with regard to taste and health benefit. After adding the medicinal herb extracts to teriyaki sauce, moisture content was found to be inversely proportional to the viscosity. The sauce with Panax ginseng extract(TP) had lowest sweetness, but highest salinity. The results showed that the sauce with Korean herb extracts had lower salinity content. In terms of colour value, the sauce with Crataegii fructus extract(TCF) had much higher lightness, redness, and yellowness than sauce prepared with other herb extracts. It had the strongest acidic taste and lowest pH. Sensory evaluation test revealed that the sauce with Anthoxylum piperitum extract(TZ) was the most preferred sauce among all the sauces tested. The degree of brown color of teriyaki sauces was correlated positively with taste preference(r=.570, p<0.01) and overall preference(r=.505, p<0.01).

Antihyperlipidemic Effects of Red Ginseng, Crataegii Fructus and Their Main Constituents Ginsenoside Rg3 and Ursolic Acid in Mice

  • Anti-hyperlipidemic effect of red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE), which are used frequently in China and Korea as herbal medicines to treat arteriosclerosis, were investigated. Treatments of RG and CF significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. Ginsenoside Rg3 and ursolic acid, the main constituents of RG and CF, respectively, also reduced TG and TC levels in hyperlipidemic mice. RG and CF significantly lowered the high blood TG and TC levels and body and epididymal mass weights induced by long-term feeding of a high-fat diet and increased the high-fat diet-induced decrease in blood HDL cholesterol levels. RG and Rg3 reduced the blood TC levels more than CF and ursolic acid. However, blood TG level were reduced by CF and ursolic acid more than RG and Rg3. RG, CF, and their constituents also inhibited pancreatic lipase and HMG-CoA reductase activities. The most potent inhibitor was Rg3. These findings suggest that RG and CF may be suitable for the therapies of hypercholesterolemia and triglyceridemia, respectively.

산사(山査)가 토끼 음경해면체의 수축에 미치는 영향 (Effects of Crataegii fructus on the Contractile Response of Rabbit Corpus Cavernosum)

  • This study was aimed to evaluate the cavernosal relaxation effect of Crataegii fructus(CF) in the contracted rabbit penile corpus cavernosum by agonists.In order to study the effect of CF on the vasoconstriction of rabbit penile corpus cavernosum, isolated rabbit penile corpus cavernosum tissues were used for the experiment using organ baths containing Krebs solution.To investigate the cavernosal relaxation of CF, CF extract at $0.01{\sim}3.0mg/m{\ell}$ was added after penile corpus cavernosum were contracted by norepinephrine(NE) $1{\mu}M$. To analyze the mechanism of CF's vasorelaxation, CF extract infused into contracted penile tissues by NE after each treatment of indomethacin(IM), $N{\omega}$-nitro-L-arginine(L-NNA), methylene blue(MB), tetraethylammonium chloride(TEA).To study the effect of CF on influx of extracellular calcium chloride($Ca^{2+}$) in penile tissues, in $Ca^{2+}$-free krebs solution, $Ca^{2+}$ 1 mM infused into contracted penile tissues by NE after pretreatment of CF. Cytotoxic activity of CF on human umbilical vein endothelial cell(HUVEC) was measured by MTT assay, and nitric oxide(NO) prodution was measured by Griess reagent. CF relaxed cavernosal strip with endothelium contracted by NE, but in the strips without endothelium, CF-induced relaxation was significantly inhibited. The pretreatment of L-NNA, MB, TEA decreased significantly on the cavernosal relaxation than not-treatment of them. But the pretreatment of IM had no significant effect on the cavernosal relaxation. In $Ca^{2+}$-free krebs solution, when $Ca^{2+}$ infused into contracted penile tissues by NE, pretreatment of CF inhibit contraction induced by adding $Ca^{2+}$.NO production wasn't increased by treatment of CF on HUVEC. This findings showed that CF is effective for the relaxation of rabbit penile corpus cavernosum, and we suggest that CF relax rabbit corpus cavernosal smooth muscle through multiple action mechanisms that include increasing the release of nitric oxide from corporal sinusoidal endothelium, inhibition of $Ca^{2+}$ mobilization into cytosol from the extracellular fluid, and maybe a hyperpolarizing action.

역미셀계를 이용한 지용성 식물체 추출물의 tyrosinase 저해효과 분석 (Ability of Lipophilic Extract Obtained from Plants to Inhibit Tyrosinase Activity in Reverse Micelles)

  • 체내에서 멜라닌 생합성을 촉매하는 tyrosinase에 대한 천연의 지용성 저해제 발굴을 위하여, isooctane/AOT(100 mM)/인산완충용액(20 mM, pH 8.0)으로 구성되고 tyrosinase(105.3 units/mL)와 3,4-dihydroxyphenyl-alanine(0.18 mM)을 함유한 역미셀계에서 식물체 75종의 petroleum ether 추출물이 tyrosinase 반응을 억제하는 효과를 분석하였다. 조사대상 식물체중에서 마늘이 in vitro 멜라닌 합성 반응을 완전히 저해하였고(100%), 모과, 고구마, 양파, 무순, 사과의 저해활성이 60% 이상이었다. 한약재와 향신료 중에서는 로즈마리, 고수, 계피, 산사자, 측백엽, 오매, 두충, 박하, 정향이 60% 이상의 저해능을 보였다. 그리고 각 식물체의 tyrosinase 저해효과와 더불어 추출수율을 함께 고려했을 때, 단위 원재료로부터 tyrosianse 저해제 생산을 많이 할 수 있는 소재를 선정할 수 있었다. 본 연구에서는 역미셀계를 이용함으로서 기존에는 분석이 어려웠던 지용성 물질의 tyrosinase 저해효과를 측정할 수 있었다.

Effects of Insamsansa-eum (Renshenshanzha-yin) on Hypercholesterolemia and Analysis of Its Effects according to the Pattern Identification

  • Backgrounds : Hyperlipidemia is a major cause of cardiovascular disease (CVD). Lowering serum cholesterol levels could reduce the risk of CVD. Insamsansa-eum (Renshenshanzha-yin, ISE), composed of Ginseng Radix and Crataegii Fructus, is a new medicine developed to treat hyperlipidemia and CVD. Objectives : In this study, we intended to explore the clinical effects of ISE on patients with hypercholesterolemia, and moreover we also compared its effects according to the pattern identification. Methods : Subjects were administered ISE with the dose of 600 mg three times a day for 4 weeks. Patterns of subjects were identified with diagnostic scoring system for Yin-Yang and the condition of Excess-Deficiency before treatment. Serum lipids were measured at baseline and after 4 weeks of medication. Results : ISE lowered total cholesterol(TC), triglyceride(TG), total lipid(TL), phospholipid(PL) and low density lipoprotein cholesterol(LDL) significantly. Compared with the data of our previous study, it was less effective than Atorvastatin but showed equal lipids-lowering effect to Chunghyul-dan (Qingxue-dan, CHD). In Yang pattern group, ISE was less effective in lowering TG and LDL than it was in not-Yang-not-Yinpattern group. On safety assessment, there was no adverse effect, hepatic or renal toxicity. Conclusions : We suggest that ISE is a safe and useful herbal medicine for hypercholesterolemia, and moreover it could be more useful when it is used for patients with not Yang pattern.

수종의 한약재가 HepG 2.2.15 Cell의 HBeAg발현 억제에 미치는 효과(效果) (The Effect of Herbs on Inhibition of HBeAg Production in HepG2.2.15 Cell line)

  • Purpose : Hepatitis B virus DNA transfected cell line(HepG2.2.15) was cultured to evaluate the effect of herbs on the expression of HBeAg and the replication of HBV. HepG2.2.15 produces HBV particles as well as viral proteins into cell culture media. Methods : Extracts of herbs were adminitered to the cells on the proper concentration. Culture media was collected 48 hours after the herbal administration and HBeAg level in the media was examined by ELISA method. To confirm that the anti-viral effect was not due to direct cytotocixity of the extracts, normal cell proliferation was shown by cell counting. And as of the interference in protein synthesis of HepG2.2.15 by herb-extracts, we used the result of study that we performed before by ${\alpha}FP$ assay using EIA method. Results& Conclusion : Herb medicines like 地楡(Sanguisorbae Radix) and 覆盆子(Rubi Frusctus) showed significant inhibitory effect on HBeAg expression at p<0.01 and 五味子(Acanthopanacis Cortex) at p<0.05. Whereas, though some herbs such as ?草根(Rubiae Radix), 山査(Crataegii Fructus), 白芍藥(Paeoniae Radix Alba), and 大黃(Rhei Radix et Rhizoma) showed the tendecy to suppress HBeAg. most of them were not significant statistically. From the above, we could conclude that those herb medicines can be applied to patients effectively and further studies on effective fraction of some herbs are thought to be needed.
