  • The purpose of this study was to investigate the effect of Gleditsiae Spina on the pro life-ration of human cancer cell-lines. The effects of Gleditsiae Spina on the proliferation of A431, HeLa. MOLT-4, K562 cells, Balb/c 3T3 cells, mouse thymocytes, splenocytes and human lymphocytes were estimated by MTT colorimetric assay. The results were as follows; 1. Gleditsiae Spina increased the proliferation of HeLa, MOLT-4 and K562 cells. 2. The cytotoxicity of mitomycin C on K562 cells was increased by the combination of Gleditsiae Spina. 3. Gleditsiae Spina did not effect the proliferation of Balb/c 3T3 cells. 4. Gleditsiae Spina stimulated the proliferation of thymocytes. 5. • In this study, antineoplastic activity against human hepatocellular carcinoma cell line(Hep G2) was tested in Gleditsiae Spina. Gleditsiae Spina was extracted with water, and the cytotoxic activity was tested using a calorimetric tetrazolium assay(MTT assay), the apoptosis was tested using a DNA electrophoresis and flow cytometry. The nitric oxide production from mouse peritoneal macrophage was tested using a Griess method. Gleditsiae Spina extracts against the proliferation of Hep G2 cells not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$, and Gleditsiae Spina extracts not showed the cytotoxicity of mitomycin C and the cytotoxicity of cisplatin on Hep G2 cells. Gleditsiae Spina extracts aginist the proliperation of BALB/c 3T3 cells not showed cytotoxicity, the proliperation of mouse thymocytes and splenocytes not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$. Gleditsiae Spina extracts not showed nitric oxide production from mouse peritoneal macrophage in vitro. Gleditsiae Spina was administered orally for 7 days at 300mg/kg increased nitric oxide production from mouse peritoneal macrophage.

  • Objectives : This research was investigated the anticoagulant effect of the Gleditsiae spina extract. Methods : We researched prothrombin time (PT) assay, activated partial thromboplastin time (APTT) assay in vitro and in vivo using arteriovenous (A-V) shunt rat model and shortening Rat tail bleeding time (BT). A-V shunt and BT were treated with extract of Gleditsiae spina (GS) 400 mg/kg for a week. Results : Bleeding time of Gleditsiae spina extract in vivo had a significant increase as about 1.2 times and thrombus weight of Gleditsiae spina extract had a significant reduction of thrombus weight as 26%. Gleditsiae spina extract represented an effect of anticoagulation by operating on extrinsic pathway factor II, V, VII, X and intrinsic pathway factor VIII, IX, X, XI, XII in the coagulation system. Conclusions : Considering the above mentioned results, it is judged that a Gleditsiae Spina extract has a control effect of thrombus creation.

  • The anti-inflammatory activity of the aqueous extract from Gleditsiae spina was investigated utilizing carrageenin-induced edema, granuloma pouch and adjuvant arthritis in rats. The effects of this agent on vascular permeability and acetic acid-induced writhing in mice were also examined. Its anti-inflammatory activity on carrageenin edema was observed with oral administration. The aqueous extract from Gleditsiae spina(400 mg/kg, 7 days) showed significant inhibitory effects on granuloma and exudate formation in rats. In the method of adjuvant arthritis, the aqueous extract(400 mg/kg), orally administered for 21 days, inhibited the development of hind paw edema in rats. The aqueous extract was also inhibited the increase in vascular permeability and the number of writhings induced by acetic acid in mice. In the present study, the aqueous extract of Gleditsiae spina was indicated to have the anti-inflammatory action.

  • Objectives : Gleditsiae Spina has the effects of expelling toxins, draining pus, invigorating blood and resolving abscesses. Some clinicians apply the herb for patients suffering from cancer. However, its anti-cancer activities are not well understood. In the present study, anti-tumor agents from Gleditsiae Spina were purified. Methods : The viability of the PC-3 cell line was determined using MTT assay, and the induction of apoptosis by Gleditsiae Spina extract in PC-3 cells was measured by Annexin-V/propidium iodide double staining assay detected by flow cytometry. TLC and HPLC analysis were used to separate and identify the anti-cancer agents. Results : Treatment of the extract resulted in significant decreased cell viability of PC-3 cells in a dose- and time-dependent manner. Dose-dependent apoptotic cell death was also measured by flow cytometry analysis. The anti-cancer agents were successfully separated and identified by using TLC and HPLC analysis and the most potential agent among them was separated from EtOAC fraction. Conclusions : These results might be applied in developing new drugs from natural resources like Korean traditional medicine, and also support the clinical usefulness of herbal medicine.

  • The purpose of this research was to investigate cytotoxity of Sunbanhwalmyungeum extract. Cytotoxity was determined by MTT assay method. After tumor cell lines(G361, BI6F10, MDA, A549) transplantation, the extracts of SunBangHwalMyungEum and its composition oriental medicines were administered, cytotoxity was measured by absorbance. The results were obtained as follows. 1. Sunbanhwalmyungeum extract and its composition oriental medicines extracts showed the concentration was higher, the more cytotoxity increased. 2. Both water and ethanol extracts of Sunbanhwalmyungeum showed excellent cytotoxity against G361, B16F10, MDA, A549 and high cytotoxity over 80$\%$ against G361, B16F10, MDA except A549 at the concentration of 1000ppm. 3. In water extract, Rhei Radix et Rhizoma, Gleditsiae Spina, Trichosanthis Radix, Glycyrrhizae Radix, Ledebouriellae Radix showed excellent cytotoxity. • GLEDITSIAE SPINA (GS) has been used as folk remedies traditionally for treatment of antiphlogistic and antifebrile agents. An ethanol extract and its fraction of GS were assessed to determine the mechanism of its antioxidant activity. Also, inhibitory effect of extract from GS and its fraction measuring the inhibitory effect on $Cu^{2+}$-induced human low-density lipoprotein (LDL) oxidation. GS ethanol extract and its fraction exhibited a concentration-dependent reactive oxygen species (ROS) and reactive nitrogen species (RNS) scavenging activities, including trolox equivalent antioxidant capacity (TEAC), OPPH radical, superoxide anion, hydroxyl radicals, peroxynitrite and nitric oxide, using different assay systems. Furthermore, the GS ethanol extract and its fraction showed dose-dependent protection of LDL oxidation induced by $CuSO_4$. In addition, the GS ethanol extract and its fraction were characterized as containing a high amount of total phenolics. These results suggest that GS ethanol extract and its fraction might be helpful for preventing oxidative stress and protecting LDL oxidation.
