Gene Expression Profile in the Liver Tissue of High Fat Diet-Induced Obese Mice

  • The purpose of this study was to investigate the gene profiles that were up- or down-regulated in the livers of high-fat diet-induced obese mice and $db_-/db_-$ mice with deficient leptin receptor. C57/BL6 normal mice and $db_-/db_-$ mice, respectively, were divided into two groups and fed a standard or high-fat diet for four weeks. Liver weight was unchanged in the normal mice but the high-fat diet led to a 10% weight increase in the $db_-/db_-$mice. Adipose tissue mass increased by about 88% in the normal mice that were fed a high-fat diet and by about 17% in the $db_-/db_-$mice on the high-fat diet. In terms of serum lipids, total cholesterol significantly increased in mice on the high-fat diet. Microarray analysis was carried out using total RNA isolated from the livers of standard or high-fat diet-fed mice of the normal and $db_-/db_-$ strains. The change of gene expression was confirmed by RT-PCR. About 1.6% and 6.8% of total genes, respectively, showed different expression patterns in the normal mice fed the high-fat diet and $db_-/db_-$ mice. As a result of microarray, many genes involved in metabolism and signal pathways were shown to have different expression patterns. Expression of Mgst3 gene increased in the livers of normal and $db_-/db_-$ mice that were fed a high-fat diet. Wnt7b and Ptk9l were down-regulated in the livers of the normal mice and $db_-/db_-$ mice that were fed a high-fat diet. In conclusion, a high-fat diet induced obesity and affected gene expression involved in metabolism and signal pathway.

Corn silk extracts did not alter the adiposity and underlying substrate utilization in high-fat diet-induced obese C57BL/6 mice

  • Corn silk (Okmi-su) was anciently adopted as a material for tea or beverage. Corn silk extracts (CSE) contain bioactive phytochemicals such as phenolic acid, flavonoids, ascorbic acid, tannins, and glycosides. Under the impact of these functional components, CSE has benefits for antioxidation, diuresis, anti-diabetes, and dyslipidemia recovery. Nonetheless, its role in whole-body adiposity was not investigated; therefore, the effects of CSE on obesity were evaluated in high-fat diet-induced obese mice. Mice were assigned to either group (n=12); 1) normal diet (18% kcal from fat), 2) high-fat diet (45% kcal from fat, the control), 3) high-fat diet with CSE (800 mg/kg diet), and 4) high-fat diet with orlistat (500 mg/kg diet, a comparable control for weight loss). Our results showed that body weight, adiposity, and energy expenditure in obese mice were not altered by CSE. Lean body mass tended to decrease by CSE, which can be explained by stimulation of diuresis (p=0.06). In conclusion, our results suggest that dietary consumption of CSE does not influence the adiposity and underlying substrate utilization in high-fat diet-induced obese mice.

  • Objectives : In this study, the aim was to investigate the inhibitory effects of Bangpungtong-seoung-san on the obese gene and obese inhibitory about obese-mouse induced by high fat diet. Methods : In order to investigate the effects of Bangpuangtongseoung-san(BPTS) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with BPTS extract) and fed for 13weeks. Results : 1. The change of body weight and the final increase of body weight were decreased significantly. 2. The amount of the adipocyte in body weight was decreased significantly. 3. In primary adipocytes, ${\beta}3AR$ gene expression was increased significantly and leptin gene expression was decreased in $100{\mu}g/ml$ density significantly. 4. In primary adipocytes, the amount of TNF-${\alpha}$ was decreased in $100{\mu}g/ml$ density significantly and the amount of leptin was decreased but did not show significance. 5. In adipocytes tissue, the expression of leptin was decreased and the expression of ${\beta}3AR$ was increased. Conclusions : These results suggest that BPTS may inhibit the expression of the obese gene mad obese inhibitory about obese-mouse induced by high fat diet.

Cissus quadrangularis Extracts Decreases Body Fat Through Regulation of Fatty acid Synthesis in High-fat Diet-induced Obese Mice

  • The current study investigated the anti-obesity effect of Cissus quadrangularsis extracts (CQR-300) and its molecular action mechanism on obese mice induced high-fat diet (HFD). To induce the obesity, mice were fed a HFD for 6 weeks and then fed HFD only or HFD with CQR-300 at 50 and 200 mg/kg. Then, body weight gain and white adipose tissue weights were measured. We investigated the reduction in body fat and the regulation of fatty acid synthesis was measured by dual energy X-ray absorptiometry and real-time PCR with Western blot, respectively. In vitro study, CQR-300 inhibited pancreatic lipase activity. The CQR-300 treatment was significantly decreased the body weight gain and adipocytes size as well as white adipose tissues weights in HFD-induced obese mice. Furthermore, CQR-300 reduced the body fat and fat mass with regulating of adipose tissue hormones as leptin. Treatment with 50 mg/kg CQR-300 showed effectively lower expression levels of adipogenesis/lipogenesis related genes and proteins such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), Sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) in white adipose tissue (WAT) as compared with the HFD fed only mice. These results suggest that the CQR-300 has an anti-obesity effect via inhibition of lipase activity, decrease the body fat mass by regulating the adipogenesis and lipogenesis related genes and proteins in epididymal adipose tissue with evaluate body fat reduce in the HFD-induced obese mice.

Expression of eotaxin in 3T3-L1 adipocytes and the effects of weight loss in high-fat diet induced obese mice

  • Eotaxin is an important inflammatory chemokine in eosinophil chemotaxis and activation and, thus, is implicated in asthma. Recently, obesity was associated with an increased prevalence of asthma, but the relationship between obesity and eotaxin expression has only been partially understood in obese mice and human studies. Therefore, we studied the expression patterns of eotaxin in 3T3-L1 preadipocytes/adipocytes to determine whether eotaxin levels are influenced by body weight gain and/or reduction in diet-induced obese mice. First, we investigated eotaxin expression during differentiation in 3T3-L1 adipocytes. Then, we treated 3T3-L1 preadipoeytes/adipoeytes with tumor necrosis factor-alpha (TNF-${\alpha}$), eotaxin, interleukin (IL)-4, IL-5, or leptin. To examine the effects of weight loss in high-fat diet induced obese mice, we fed C57BL/6 mice a high-fat diet or a normal diet for 26 weeks. Then, half of the high-fat diet group were fed a normal diet until 30 weeks to reduce weight. Epididymal adipose tissue, visceral adipose tissue, serum, and bronchoalveolar fluid of mice were examined for eotaxin expression. The results showed that eotaxin expression levels increased with adipocyte differentiation and that more eotaxin was expressed when the cells were stimulated with TNF-${\alpha}$, eotaxin, IL-4, IL-5, or leptin. An in vivo study showed that eotaxin levels were reduced in visceral adipose tissues when high-fat diet fed mice underwent weight loss. Taken together, these results indicate a close relationship between eotaxin expression and obesity as well as weight loss, thus, they indirectly show a relation to asthma.

대황(大黃)이 고지방식이로 비만이 유발된 C57BL/6 mouse의 지방조직 염증 및 인슐린 저항성에 미치는 영향 (Effects of Rhei Radix et Rhizoma on Insulin Resistance and Adipose Tissue Inflammatory Response in High Fat Diet Induced Obese C57BL/6 Mice)

  • Objectives : The purpose of this study was to investigate how Rhei Radix et Rhizoma affects on insulin resistance and adipose tissue inflammatory response in high fat diet induced obese C57BL/6 mice. Methods : Obesity was induced in C57BL/6 mice by high fat diet for 12 weeks. Models were divided into 3 groups (n=6) of normal diet, high fat diet (HFD), and high fat diet with Rhei Radix et Rhizoma and investigated for 12 weeks. We measured body weight, FBS and oral glucose tolerance test (OGTT), serum insulin, homeostatic model assessment-insulin resistance (HOMA-IR), weight of liver and epididymal fat pad. Inflammatory markers such as adipose tissue macrophage (ATM), tumor necrosis factor-${\alpha}$ and interlukin-10 and CD68 of epididymal adipocyte were determined to evaluate the effect of Rhei Radix et Rhizoma on adipose tissue inflammation. Results : Compared with the HFD group, we observed loss of body weight and epididymal fat pad weight, improvement of glucose level and HOMA-IR, reduction of ATM and gene expression of TNF-${\alpha}$, CD68 in the high fat diet with Rhei Radix et Rhizoma group. Conclusions : This study suggests that Rhei Radix et Rhizoma has effects on insulin resistance and adipose tissue inflammatory response in high fat diet induced obese mice.

고지방식이 동물모델에서 통곡물 시리얼의 근감소성 비만 예방 효과 (Preventive Effects of Whole Grain Cereals on Sarcopenic Obesity in High-fat Diet-induced Obese Mice)

  • 통곡물 시리얼(Whole grain cereal, WGC)이 함유된 식이는 에너지 대사 조절에 중요한 다량영양소(macronutrients)를 제공한다. 본 연구는 고지방식이(high-fat diet, HFD)로 유발된 비만 마우스를 이용하여 WGC의 근감소성 비만 예방 효과에 대해 평가하였다. C57BL/6N 마우스에 정상식이(normal diet, ND), ND+WGC, HFD, HFD+WGC를 12주 동안 제공하였다. WGC는 체중, 식이효율, 체지방 및 지방세포의 크기를 감소시켰다. 또한, WGC는 간 무게 및 간에 축적된 지방을 감소시킴으로써 HFD에 의한 비알코올성 지방간을 개선시켰다. 더욱이, WGC는 비만 마우스 및 정상 마우스의 근육 무게 및 근력을 증가시켰다. 따라서, WGC는 지방 축적을 억제하고 근육량을 증가시키므로 근감소성 비만 예방을 위한 기능성 식품으로 사용될 수 있을 것으로 기대된다.

방기복령탕(防己茯嶺湯)이 비만유도생쥐의 혈당 및 체지방개선에 미치는 영향 (Effects of Bangkibokryeong-tang (Fangjifuling-tang) on the Reduction of Blood Glucose and Body Fat in High Fat Diet Induced Obese Mice)

  • Objectives This study aimed to investigate the effect of Bangkibokryeong-tang (BBT, Fangjifuling-tang) on blood glucose and body fat in high-fat diet-induced obese mice. Methods The experimental animals were divided into five groups- normal diet-fed control (ND), high-fat diet-fed control (HFD), HFD+BBT 75, HFD+BBT 150, and HFD+olistat as a positive drug control group. Markers of obesity, such as body weight, organ weight, diet efficiency, and serum levels of total cholesterol, triglycerides, lipid content, leptin, adiponectin, glutamic oxaloacetic transaminase (GOT)/glutamic pyruvic transferase (GPT)/lactate dehydrogenase (LDH), blood glucose, and insulin, were measured. Furthermore, results of the oral glucose tolerance test and ${\alpha}-glucosidase$ inhibition activity were examined in obese mice. Results Mice treated with BBT demonstrate lower body and organ weight, and reduced weight gain and food efficiency than that in the HFD-only control group. In addition, BBT decreased lipid accumulation in the liver and the levels of enzymes such as GOT, GPT, and LDH in the serum. Furthermore, the levels of triglycerides, total cholesterol, low density lipoprotein (LDL), and leptin were decreased in the serum but the levels of high density lipoprotein (HDL) and adiponectin were increased in the BBT-treated group compared with the control group. The BBT-treated group also demonstrated decreased blood glucose and insulin concentrations induced by feeding on a high-fat diet and improved glucose tolerance. Conclusions Based on the results above, BBT may reduce body fat and hyperglycemia in HFD-induced obesity. This suggests that BBT may be clinically useful in the treatment of obesity.

방풍통성산이 고지방식이 유도 어린 비만 생쥐의 지방 축적에 미치는 영향 (Effect of Bangpungtongsungsan on Suppressing Fat Accumulation in High-fat Diet-Induced Young Obese Mice)

  • Objectives The aim of this study was to determine the inhibitory effects of Bangpungtongsungsan extract (BTS) on fat accumulation in high-fat diet-induced young obese mice. Methods The extract was administered to 3-week-old C57BL/6 male mice fed with a high-fat diet. The experimental groups were divided into a control group (Ctrl), high-fat diet group (HFDF), and BTS treated group after high fat diet feeding (BTST), with 10 mice assigned to each group. Lipid synthesis was observed to confirm the inhibition of fat synthesis. Changes in body weight, body fat percentage, and total cholesterol in the blood were observed to confirm weight control. Peroxisome proliferator-activated receptor gamma (PPAR-γ) and sterol regulatory element-binding protein (SREBP)-1 positivity was observed to confirm the inhibition of fat accumulation in liver tissue. Results Bangpungtongsungsan significantly inhibited lipid synthesis. Changes in body weight, body fat percentage, and total cholesterol in the blood were significantly lower in BTST rats than in HFDF rats. PPAR-γ and SREBP-1 positivity were significantly lower in BTST rats compared to HFDF rats. Conclusions This study confirms the potential of BTST to inhibit fat accumulation in obesity.

Anti-inflammatory and anti-diabetic effects of brown seaweeds in high-fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: Seaweeds have been reported to have various health beneficial effects. In this study, we investigated the potential anti-obesity and anti-inflammatory effects of four types of domestic brown seaweeds in a high-fat diet-induced obese mouse model and bone marrow-derived macrophages (BMDM). MATERIALS/METHODS: Male C57BL/6N mice were fed low-fat diet (LFD), high-fat diet (HFD) or HFD containing Undaria Pinnatifida, HFD containing Laminaria Japonica (LJ), HFD containing Sargassum Fulvellum, or HFD containing Hizikia Fusiforme (HF) for 16 weeks. RESULTS: Brown seaweed supplementation did not affect long-term HFD-associated changes in body weight or adiposity, although mice fed HFD + LJ or HFD + HF gained slightly less body weight compared with those fed HFD at the beginning of feeding. Despite being obese, mice fed HFD + LJ appeared to show improved insulin sensitivity compared to mice fed HFD. Consistently, we observed significantly reduced blood glucose concentrations in mice fed HFD + LJ compared with those of mice fed HFD. Although no significant differences in adipocyte size were detected among the HFD-fed groups, consumption of seaweeds decreased formation of HFD-induced crown-like structures in gonadal adipose tissue as well as plasma inflammatory cytokines. BMDM from mice fed HFDs with seaweeds showed differential regulation of pro-inflammatory cytokines such as IL-$1{\beta}$ and IL-6 compared with BMDM from mice fed HFD by LPS stimulation. CONCLUSION: Although seaweed consumption did not prevent long-term HFD-induced obesity in C57BL/6N mice, it reduced insulin resistance (IR) and circulation of pro-inflammatory cytokines. Therefore, seaweeds may ameliorate systemic inflammation and IR in obesity partially due to inhibition of inflammatory signaling in adipose tissue cells as well as bone marrow-derived immune cells.
