Analysis of the Chemical Constituents of Agaricus brasiliensis

  • This study examined the chemical composition of A. blasiliensis and the chemical structural properties of an immuno-stimulating polysaccharide. The amino acids, free sugars, and organic acids by HPLC and fatty acids by GC were analyzed. The immuno-stimulating substance from A. blasiliensis was extracted with hot water and purified by ethanol precipitation. It underwent ion exchange chromatography on DEAE-cellulose and gel filtration on Toyopearl HW 65F. Through GP-HPLC, the substance was found to be homogeneous. Its chemical structure was determined by $^{13}C-NMR$. Fatty acids, organic acids, and sugar alcohol composition consisted exclusively of linoleic acid, fumaric acid and mannitol, respectively. The amino acids were mainly glutamic acid, glycine, and arginine. By $^{13}C-NMR$ analysis, the immuno-stimulating substance was identified as ${\beta}-(1{\rightarrow}3)\;(1{\rightarrow}6)$-glucan, composed of a backbone with $(1{\rightarrow}3)$-linked D-glucopyranosyl residues branching a $(1{\rightarrow}6)$-linked D-glucopyranosyl residue. The ${\beta}$-glucan from A. blasiliensis showed pronounced immuno-stimulating activity on the antibody-production ability of B-lymphocytes by the hemolytic suspension assay. In these results, A. blasiliensis was estimated to have potent pharmacological properties and potential nutritional values.

  • Objectives : This experimental study was designed to investigate the effects of Acanthopanax, sessiliflorus SEEM extracts following gamma-ray irradiation on immuno-stimulating and anti-tumor activity in terms of proliferation of tumor cells, thymocytes, splenocytes, and NO production from peritoneal macrophages in mice. Methods: 10AS and 100 AS were the bark powders of Acanthopanax sessiliflorus $S_{EEM}$ stems which were exposed in 10 kGy or 100 kGy of electron beam respectively. Results : Treatment with either 10AS or 100AS increased proliferation rates of thymocytes and splenocytes significantly, and treatment with l0AS also decreased proliferation rates of tumor cells significantly. Treatment with either l0AS or l00AS promoted NO production from peritoneal macrophages significantly. Conclusion : These results suggested that AS has direct inhibition effect of tumor growth and immuno-stimulating activity. In conclusion, we demonstrate that AS could be used to treat cancer patient as complementary or alternative medicine to typical anti-cancer medication.

Immuno-stimulating, Anti-stress and Anti-thrombotic Effects of Unossified Velvet Antlers

  • Both ethanol and water extracts of unossified velvet antler were found to exhibit a significant immuno-stimulating activity as measured by carbon clearance test in mice, a remarkable anti-fatigue effect in weight-loaded forced swimming performance in mice, a significant anti-stress effect on immobilization in rats. The antler extracts also showed a weak but significant anti-thrombotic activity. These findings are indicative of adaptogenic properties of antlers and their normalizing effects during stressful condition.

  • A potent immuno-stimulating activity was detected from the watersoluble and ethanol-precipitated crude extract (AG-1) of the root of Angelica gigas Nakai. The crude extract was fractionated into two fractions, an acidic AG-2 and a neutral AG-3 fraction by DEAE-cellulose adsorption. The two fractions contained polysaccharides of which M.W. were 10 Kdal and >2,000 Kdal respectively, proteins, and various inorganic components. The immunostimulating activities of two fractions were not reduced by proteinase K, acid or alkali treatment. The polysaccharides obtained from the root of A. gigas were mainly composed of arabinose, galactose, and galacturonic acid. These results indicated that immuno- stimulating components of the root of A. gigas was a kind of pectic polysaccharides or arabinogalactans.

Anti-tumor and immuno-stimulating activity of fungal polysaccharides

  • Low molecular weight molecules and high molecular weight substances were found to have anti-tumor and immuno-modulating activity. Previously polysaccharides have been received much attention because of adhesives, food additives or animal foods (Whistler et al., 1976). In effort of developing new anti-tumor substances with low toxicity, numerous polysaccharides from yeast, algae, bacteria, higher plants and especially fungi have been investigated for anti-tumor and immuno-modulating activities. Thus the high molecular weight molecule was reported to have anti-tumor activity through host mediated immunity. In this brief article, attention will be paid to polysaccharides which is especially fungal origin.

  • 고품질, 기능성 청국장 제조를 위한 균주 개발 연구의 일환으로, 재래식 청국장으로부터 생육이 빠르며, 면역증강활성이 우수한 JB-1 균주는 Bacillus pumilus 로 확인되었으며 Bacillus pumilus KCTC 10461BP로 균주기탁 되었다. JB-1으로 발효시킨 청국장은 관능성과 영양성이 우수하며 물추출물 2mg/ml의 농도에서 410% 정도의 면역세포 증식율을 나타내었으며, 산업적 규모의 6 kg 의 대두증자에 20 ml 배양액을 접종함으로서 16시간 이내에 저이취와 높은 생균수 및 아미노산 함량의 청국장 제조가 가능함을 확인하였다.

  • In recent studies on cefodizime, it may potentially have the capability of stimulating chemotactic activity of neutrophils and monocytes as well as the strong immuno-modulator. In turn, infection can result in a drastic change of mediators. which lead to initiate an immune response in an indirect way. With this backgrounds, we have studied to see if cefodizime can be a potential substance to induce an immunological function in dendritic cells and peritoneal macrophages. (omitted)

감식초에서 분리한 면역활성다당의 특성 (Characterization of Immuno-stimulating Polysaccharides Isolated from Korean Persimmon Vinegar)

  • 한국 전통발효음료 중에 존재하는 특이다당류의 화학적 특성 및 생물활성을 규명할 목적으로, 농가에서 직접 발효한 감식초로부터 다당류를 분리하고 이들의 각종 면역증강활성에 대해 검토하였다. 감식초에 80% ethanol 침전을 행하여 얻어진 조다당 획분 PV-0을 이용, 연속적인 3회의 column chromatography를 행하여, 감식초의 주요 다당인 PV-1b-I으로 정제할 수 있었다. PV-1b-I획분은 HPLC상에서 대칭을 유지하는 단일 peak로 검출되었으며, 분자량은 약 110 kDa으로 평가되었다. 정제다당인 PV-1b-I의구성당 조성을 확인한 결과, mannose(46.8%), galactose(28.5%) 및 arabinose(19.1%)를 높은 비율로 함유하고 있었다. 또한 본 정제다당은 ${\beta}$-glucosyl Yariv reagent와 강하게 반응하는 특성을 보여주었으며 이러한 사실로부터 PV-1b-I이 arabino-3,6-galactan 부위를 소유함을 확인할 수 있었다. 한편 감식초에서 정제한 다당 PV-1b-I은 in vitro상에서 macrophage의 활성화를 높은 비율로 유도하였으며, 비장세포에 대해 높은 증식능을 보였다. 또한 PV-1b-I을 정맥 투여한 경우, NK cell을 활성화하여 YAC-1 종양세포에 대한 세포독성을 증가시킴이 관찰되었다. 한편 고분자 정제 다당인 PV-1b-I는 비특이적 면역계에 있어 중요 역할을 담당하고 있는 보체계에 대하여 농도 의존적인 활성화 경향을 보였다. 이들은 $Ca^{2+}$ 이온이 제거된 상태에서의 항보체 활성과 anti-human C3를 이용한 2차원 면역전기영동에 의하여 C3 산물을 동정한 결과로부터 보체계의 classical pathway와 alternative pathway 양 경로를 모두 경유하여 활성을 나타냄을 확인할 수 있었다.

감잎에서 분리한 다당의 면역자극 활성 (Immuno-stimulating Activities of Polysaccharide Fractions Isolated from Persimmon Leaves)

  • In order to develop new physiologically active polysaccharides from persimmon leaves, two different crude polysaccharides were prepared using hot water (PLW-0) and pectinase digestion (PLE-0) and their immuno-stimulating activities were estimated. PLW-0 and PLE-0 showed similar sugar compositions with 15 different sugars, including rarely observed sugars in general polysaccharides such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose, aceric acid, 3-deoxy-D-manno-2-octulosonic acid, and 3-deoxy-D-lyxo-2-heptulosaric acid, but the uronic acid content of PLE-0 was lower than that of PLW-0 caused by pectinase treatment. Both PLW-0 and PLE-0 showed potent anti-complementary activity in a dose-dependent manner which was similar to a known immuno-stimulating polysaccharide, PSK, from Coriolus versicolor. The activity of PLE-0 at a low concentration ($100{\mu}g/m{\ell}$) was higher than that of PLW-0. In an in vitro cytotoxicity analysis, PLW-0 and PLE-0 (up to $1,000{\mu}g/m{\ell}$) did not affect the growth of peritoneal macrophages and Colon 26-M3.1 carcinoma cells. In contrast, they enhanced lymphocyte proliferation activity. Peritoneal macrophages stimulated with PLW-0 and PLE-0 produced various cytokines, such as IL-6 and IL-12. However, PLE-0 was more effective on the cytokine production. Intravenous administration of PLW-0 and PLE-0 significantly augmented natural killer (NK) cell cytotoxicity against Yac-1 tumor cells 3 days after the treatment of polysaccharide fractions. But NK cells obtained from the PLE-treated group showed higher tumoricidal activity even at a low dose of $40{\mu}g$/mouse. In experimental lung metastasis of Colon 26-M3.1 carcinoma cells, prophylactic administration of PLW-0 and PLE-0 significantly inhibited lung metastasis in a dose-dependent manner and PLE-0 was more effective on the inhibition of cancer metasasis. The results lead us to conclude that the pectinase-treated process is indispensable to preparing polysaccharides with higher immune-stimulating activity from persimmon leaves.

복분자주에서 분리한 Mannan 다당의 면역증진 활성 (Immuno-stimulating Activities of Mannose-rich Polysaccharides Isolated from Korean Black Raspberry Wine)

  • 한국 전통발효음료 중에 존재하는 특이 다당류의 화학적 특성 및 생물활성을 규명할 목적으로, 농가에서 직접 발효한 복분자주로부터 다당류를 분리하고 이들의 각종 면역증강활성에 대해 검토하였다. 복분자주에 80% ethanol 침전을 행하여 얻어진 조다당획분 RB-0를 이용, 연속적인 3회의 column chromatography를 행하여, 복분자주의 주요 다당인 RB-1b-I으로 정제할 수 있었다. RB-1b-I획분은 HPLC상에서 대칭을 유지하는 단일 peak로 검출되었으며, 분자량은 약 180 kDa으로 평가되었다. 정제다당인 RB-1b-I의 구성당 조성을 확인한 결과, mannose가 76.8%로 주를 이루고 있었으며, 그 외에 galactose(15.8%), arabinose(3.8%), glucose(2.6%) 및 rhamnose(1.2%)의 순으로 함유되어 있었다. 이러한 결과는 복분자 중에 존재하는 주 다당이 발효 효모의 세포벽에서 기원한 mannan임을 추정하게 하였다. 한편 복분자주에서 정제한 다당 RB-1b-I은 in vitro상에서 macrophage의 활성화를 높은 비율로 유도하였으며, 비장세포에 대해 높은 증식능을 보였다. 또한 RB-1b-I을 정맥 투여한 경우, NK cell을 활성화하여 YAC-1 종양세포에 대한 세포독성을 증가시킴이 관찰되었다. 한편 고분자 정제 다당인 RV-1b-I은 비특이적 면역계에 있어 중요 역할을 담당하고 있는 보체계에 대하여 농도 의존적인 활성화 경향을 보였다. 이들은 Ca++ 이온이 제거된 상태에서의 항보체 활성과 antihuman C3를 이용한 2차원 면역전기영동에 의하여 C3 산물을 동정한 결과로부터 보체계의 classical pathway와 alternative pathway양 경로를 모두 경유하여 활성을 나타냄을 확인할 수 있었다. 이상의 결과로부터 복분자주에는 인체 건강에 유익한 면역증진 활성을 제공하는 다당류가 함유되어 있음을 알 수 있었다.
